Polyphenol-rich fraction from Ecklonia cava (a brown alga) processing by-product reduces LPS-induced inflammation in vitro and in vivo in a zebrafish model

  • Ecklonia cava is a common edible brown algae that is plentiful in Jeju Island of Republic of Korea. Polyphenols from E. cava have strong anti-inflammatory activity. However, a large number of the by-products from E. cava processing are discarded. In the present study, to utilize these by-products, we assessed the anti-inflammatory activity of the polyphenol-rich fraction (PRF) from E. cava processing by-product (EPB) in lipopolysaccharide (LPS)-induced RAW264.7 macrophage cells. Four compounds, namely eckol, eckstolonol, dieckol, and phlorofucofuroeckol-A, were isolated and identified from PRF. We found that PRF suppressed the production of nitric oxide (NO), inducible nitric oxide synthase, and cyclooxygenase-2 in the LPS-induced cells. Furthermore, the protective effect of PRF was investigated in vivo in LPS-stimulated inflammation zebrafish model. PRF had a protective effect against LPS-stimulated toxicity in zebrafish embryos. In addition, PRF inhibited LPS-stimulated reactive oxygen species and NO generation. According to the results, PRF isolated from EPB could be used as a beneficial anti-inflammatory agent, instead of discard.

Probiotics를 이용하여 발효시킨 감귤 가공부산물 발효물의 특성 (Characteristics of Citrus By-Product Ferment Using Probiotics as Starter)

  • 감귤 과괴 뿐만 아니라 가공부산물 내에는 carotenoid, bioflavonoids, pectin 등의 풍부하게 함유되어 있으며, 특히 flavonoids는 항산화 또는 금속이온과 착염을 형성하여 free radical이나 electrophiles 및 과산화 지질의 생성을 막아 세포, 조직, 기관이 손상을 억제 해주며, 노화방지, 순환계 질환을 예방 해주며 항암 효과가 있는 것으로 밝혀지고 있다. 이에 본 연구에서는 미생물을 첨가하여 발효시킨 감귤 가공부산물의 열수 추출액과 감귤 가공부산물의 열수 추출액을 가지고 어류 질병 세균에 대한 항균활성과 항산화활성을 측정하였는데, 어류 질병 세균에 대한 항균 실험에 있어서는 미생물을 첨가하여 발효시킨 감귤 가공부산물 추출액에서 저해 효과가 뚜렷하게 나타났다(Table 3). 반면 항산화활성 측정에서는 감귤 가공부산물 자체도 항산화 활성이 높다는 것을 알 수가 있다. 그러나 E. faceium, P. pentosaceus, L. plantarum, L. rhamnosus, B. subtilis, S. cerevisiae를 각각 첨가하여 발효시킨 감귤 가공부산물을 기존 합성 항산화제인 BHT와 BHA와 비교하여 보았을 때 전자공여능을 이용한 항산화 활성 측정에서는 전체적으로 높은 항산화 활성 효과를 가지고 있는 것으로 나타났고, 특히 B. subtilis, S. cerevisiae를 이용하여 발효시킨 실험구에서는 합성 항산화제인 BHA보다 높은 89%와 같은 값인 87%에 활성을 나타내고 있다. SOD유사활성 측정에 대한 결과는 Fig. 2에서처럼 L. rhamnosus를 첨가하여 발효시킨 실험구를 제외하고 나머지 5개의 실험구에서는 기존 합성항산화제인 BHT와 BHA보다 높거나 유사한 활성을 나타내었다. 그리고 hydroxyl radical 소거활성 측정에서는 미생물을 첨가하여 발효시킨 산물에 추출액의 활성이 높게 나타났으며, 1%에 농도에서는 합성 항산화제보다도 좋은 활성을 나타내는 것으로 조사 되었는데, 특히 B. subtilis를 첨가하여 발효시킨 실험구에서는 가장 높은 활성을 나타내고 있다. 이상의 결과를 종합하여 볼 때 미생물을 첨가하여 발효과정을 거친 감귤 가공부산물의 어류 질병 세균에 대한 항균활성이나 항산화활성에 있어서 그 효과가 탁월하다는 것을 알 수가 있다. 그리고 발효과정에 있어 첨가되어진 미생물 중에서는 B. subtilis와 S. cerevisiae가 발효 미생물로서 적합하다는 것을 알 수가 있었고, 본 연구에서 Bacillus subtilis 같은 경우는 가장 높은 항균활성과 항산화활성을 나타내고 있어 최적 발효 미생물로 말할 수가 있을 것이다.

훈건 굴을 이용한 분말조미소재의 가공 및 품질안전성 (Processing and Shelf-life Stabilities of Flavoring Substances of the Smoke-Dried Oysters)

  • This study was conducted to evaluate the optimal processing conditions of smoke-dried powdered oysters and to determine their shelf-life during storage for development of a natural oyster flavoring substance. The optimal conditions for processing of smoke-dried oyster powder with freshy oyster were as follows. Raw shelled oysters were rinsed with 3% saline solution, drained, boiled for 10 minutes at $98^{\circ}C$, and then smoked for 1 hour at $50^{\circ}C$, followed by drying for 4 hours at $80^{\circ}C$ Smoke-dried oyster powder with oyster scraps were prepared as flavoring material. The smoked oyster scraps were submerged in oyster sauce far 10 minutes at room temperature and then dried with hot air for 5 hours at $50^{\circ}C$. The smoke-dried oysters and smoke-dried oyster scraps were then pulverized to 50 mesh and packed in tea bags or vacuum-packed in laminated plastic film bags (PE/PVDC/CPP, $12{\mu}m/15{\mu}m/50{\mu}m$). Compared to non smoke-dried powdered oysters, the smoking and dipping in oyster sauce enhanced the flavor and prevented lipid oxidation of the smoke-dried powdered oyster product. Shelf-life tests indicated that the vacuum-packaging method preserved the quality of smoke-dried powdered oysters stored for 150 days at room temperature.

Effects of enzymatically hydrolyzed fish by-products in diet of juvenile rainbow trout (Oncorhynchus mykiss)

  • Five experimental diets were formulated to evaluate the effects of dietary enzymatically hydrolyzed tuna by-product on growth, non-specific immune responses, and hematology of juvenile rainbow trout (Oncorhynchus mykiss). A basal diet with 50% of fishmeal was used as control (CON) and four other diets replaced 12.5% ($TBB_{12.5}$), 25% ($TBB_{25}$), 37.5% ($TBB_{37.5}$), and 50% ($TBB_{50}$) of fish meal in the CON diet. Juvenile rainbow trout ($4.87{\pm}0.05g$) were randomly distributed into 15 tanks (50 L) and fed 3-4% of wet body weight two times a day. At the end of 7 weeks of feeding trial, weight gain, specific growth rate, feed efficiency, and protein efficiency ratio of fish fed CON diet were significantly higher than those of fish fed $TB_{50}$ diet (P < 0.05). But there were no significant differences among fish fed CON, $TBB_{12.5}$, $TBB_{25}$, and $TBB_{37.5}$ diets (P > 0.05). There were no significant differences in GPT levels among fish fed CON, $TBB_{12.5}$, $TBB_{25}$, and $TBB_{37.5}$ diets. Also, there were no significant differences in lysozyme, superoxide dismutase, glucose, and total protein levels in all experimental diet (P > 0.05). The broken-line analysis indicated that the minimum dietary level of enzymatically hydrolyzed tuna by-product to replace fishmeal could be 29.7% in rainbow trout. These results indicated that the optimum level of dietary enzymatically hydrolyzed tuna by-product could replace greater than 29.7% but less than 37.5% of fishmeal in juvenile rainbow trout diet.

Application of Fast Atom Bombardment Collision-induced Dissociation Tandem Mass Spectrometry for Structural identification of Glycerolipids Isolated From Marine Sponge

  • Two types of glycerolipids [monoacylglycerols (MAG) and cyclitols] were isolated by reversed phase high-performance liquid chromatography from the methanol extracts of a marine sponge, and analyzed by fast atom bombardment mass spectrometry (FAB-MS) in positive-ion mode. FAB mass spectra of these compounds yielded protonated molecules $[M + H]^+$ and abundant sodiated molecules $[M + Na]^+$ from a mixture of 3-nitrobenzyl alcohol and NaI. The structures of these compounds were elucidated by FAB-collisional-induced dissociation (CID)-tandem mass spectrometry. We carried out collision-indused dissociation (CID) of these lipids in B/E-linked scan mode. The CID B/E-linked scan of $[M + H]^+$ and $[M + Na]^+$ precursor ions resulted in the formation of numerous characteristic product ions through a series of dissociative processes. The product ions formed by charge-remote fragmentation (CRF) provided important information for the identification of the acyl chain structure substituted at the glycerol backbone. Some of the product the ions were diagnostic for the presence of a glycerol backbone or acyl chain structure.

기후변화에 따른 해양생태계 변화와 어업 (Changes in Marine Ecosystem according to Climate Change and Fishery)

  • Global climate change is significant issue in marine ecosystem and fishery market. According to rising of both seawater temperature and sea-level, global fishery environment and marine ecosystem are changing drastically. Moreover, over-exploitation in fishery areas is the most important issue in the marine ecosystem conservation. In analysis and statistics of global network, major fish populations are decreasing very fastly, but fish catch are increasing annually. It means balance between product and consume is not appropriate in the global market. China as emerging new fish consumer in the world is strongly developing fishery industry and technology. In this paper, I try to review a relationship between marine ecosystem and climate change in global scale according to references. I also suggested possibility of sustainable global fishery in changing marine ecosystem by analysis of some related reports of international global fishery.

  • The assessment of the potential for the design of marine renewable energy systems is reviewed and the current situation for marine renewable energy is promising. The most studied forms of marine renewable energy are ocean wind energy, ocean wave energy and tidal energy. Wind turbine generators include mostly horizontal axis type and vertical axis type. But also more exotic ideas such as a kite design. Wave energy devices consist of designs converting wave oscillations in electric power via a power take off equipment. Such equipment can take multiple forms to be more efficient. Nevertheless, the technology alone cannot be the only step towards marine renewable energy. Many other steps must be overcome: policy, environment, manpower as well as consumption habits. After reviewing the current conditions of marine renewable energy development, the authors analyzed the key factors for developing a strong marine renewable energy industry and pointed out the huge potential of marine renewable energy.

Effect of Sarcotride A on Membrane Potential in C6 Glioma Cells

  • We tested effect of sarcotride A, a bioactive cyclitol derivative from a marine sponge, on membrane potential in C6 glioma cells. Membrane potential was estimated by measuring fluorescence change of DiBAC-loaded glioma cells. Sarcotride A increased membrane potential in a concentration-dependent manner. We tested effects of pertussis toxin, U73122, EIPA, and $Na^+-free$ media on sarcotride A-induced increase of membrane potential to investigate involvement of G proteins, phospholipase C, $Na^+/H^+$ exchanger, and $Na^+$ channels. However, we were not able to observe any significant effect of those pharmacological inhibitors, excluding the involvement of the molecules as candidate targets or signaling molecules of sarcotride A-induced increase of membrane potential. Further investigation is necessary to elucidate action mechanism of sarcotride A.

Two New Scalaranes from a Korean Marine Sponge Spongia sp.

  • Intensive chemical investigation of Korean marine sponge Spongia sp. has led to the isolation of two new scalaranes. The planar structures of the new compounds 1 and 2 were determined through 1D and 2D NMR spectral data analysis, while the relative stereochemistry of the compounds was determined based on the analysis of $^1H-^1H$ coupling constants and NOESY spectroscopic data. Compounds 1 and 2 did not display any significant biological activities on farnesoid X-activated receptor (FXR) in co-transfection assay.
