To produce anti-malarial drugs, natural products were extracted from 18 species of marine algae by various mechanical methods. Twelve species of marine algae were found to have antiplasmodial activity by inhibiting the growth of the chloroquine-resistant Plasmodium falciparum strain FCR-3 with EC/sub 50/ values less than 100 ㎍/㎖. The methanol extract of Neoholmeria japonica had the strongest antiplasmodial activity with EC/sub 50/ value of 62 ㎍/㎖.
As part of an effort to discover bioactive natural products from marine sources, we investigated the bioactive secondary metabolites from two marine isolates of the fungi, Trichoderma viride and Rhizopus stolonifer. Three cyclopentenones, myrothenones A (1) and B (2) and trichodenone A (3), were isolated from T. viride and two cyclopentenones, 2-bromomyrothenone B (4) and botrytinone (5), were isolated from R. stolonifer. The molecular structures and absolute stereochemistries of the cyclopentenones were determined from chemical and physicochemical evidence, including quantum chemistry calculations, X-ray analysis, and the circular dichroism (CD) exciton chirality method. Myrothenone A (1) displays tyrosinase inhibitory activity, with an I$C_{50}$ value of 6.6 ${\mu}M$, and is therefore more active than the positive control, kojic acid.

Two new cyclic depsipeptides wewakamide A (1) and guineamide G (2) have been isolated from the marine cyanobacterium Lyngbya semiplena and Lyngbya majuscula, respectively, collected from Papua New Guinea. The amino and hydroxy acid partial structures of wewakamide A and guineamide G were elucidated through extensive spectroscopic techniques, including HR-FABMS, 1D $^1H$ and $^{13}C$ NMR, as well as 2D COSY, HSQC, HSQC-TOCSY, and HMBC spectra. The sequence of the residues of wewakamide A was determined through a combination of ESI-MS/MS, HMBC, and ROESY. Wewakamide A possesses a ${\beta}$-amino acid, 3-amino-2-methylbutanoic acid (Maba) residue, which has only been previously identified in two natural products, guineamide B (3) and dolastatin D (4). Although both new compounds (1,2) showed potent brine shrimp toxicity, only guineamide G displayed significant cytotoxicity to a mouse neuroblastoma cell line with $LC_{50}$ values of 2.7 ${\mu}M$.
The adsorption characteristics of mixed heavy metals (Cr(III), As(VI)) in aqueous solution were investigated using a sediment amendment composite. Sediment amendment composite was composed of clean sediment (40%), zeolite (20%), recycled aggregate (10%), steel slag (10%), oyster shell (10%), and cement (10%). The experimental results showed that the adsorption equilibrium was attained after 180 mins. Heavy metal adsorption was characterized using Freundlich and Langmuir equations. The equilibrium adsorption data for the sediment amendment composite better fitted with the Langmuir model than the Freundlich model. The maximum adsorption capacity of Cr(VI) (36.07 mg/g) was higher than As(III) (25.54 mg/g); and the adsorption efficiency of the Cr(VI) and As(III) ions solution decreased with decreasing pH from 2 to 10. The collective results suggested that the sediment amendment composite is a promising material for a reactive cap that controls the release of Cr(VI) and As(III) from contaminated sediments.

The Federated States of Micronesia (FSM) is an island country in the western Pacific and is a known biodiversity hotspot. However, a relatively small number of fungi (236 species) have been reported till July 2021. Since fungi play major ecological roles in ecosystems, we investigated the fungal diversity of FSM from various sources over 2016 and 2017 and constructed a local fungal inventory, which also included the previously reported species. Fruiting bodies were collected from various host trees and fungal strains were isolated from marine and terrestrial environments. A total of 99 species, of which 78 were newly reported in the FSM, were identified at the species level using a combination of molecular and morphological approaches. Many fungal species were specific to the environment, host, or source. Upon construction of the fungal inventory, 314 species were confirmed to reside in the FSM. This inventory will serve as an important basis for monitoring fungal diversity and identifying novel biological resources in FSM.

Here, we evaluated the anti-glycation effects and renal protective properties of 70% (v/v) ethanolic extract of Colpomenia sinuosa (CSE) against AGEs -induced oxidative stress and apoptosis at different concentrations (1, 5, and 20 ㎍/mL). At 20 ㎍/mL, CSE showed that anti-glycation activities via the inhibition of AGE formation (51.1%), inhibition of AGEs-protein cross-linking (61.7%), and breaking of AGEs-protein cross-links (33.3%), were significantly (###p < 0.001 vs. non-treated group) lower than the nontreated group. Methylglyoxal (MGO) significantly (***p < 0.001) reduced cell viability (24.4%) and increased reactive oxygen species (ROS) level (642.3%), MGO accumulation (119.4 ㎍/mL), and apoptosis (55.0%) in mesangial cells compared to the nontreated group. Pretreatment with CSE significantly (###p < 0.001) increased cell viability (57.8%) and decreased intracellular ROS (96.5%), MGO accumulation (80.0 ㎍/mL), and apoptosis (22.6%) at 20 ㎍/mL. Additionally, we confirmed intracellular AGEs reduction by CSE pretreatment. Consequently, our results suggest that CSE is a good source of natural therapeutics for managing diabetic complications by the antiglycation effect and renal protective activity against MGO-induced oxidative stress.

Scuticociliates are regarded as serious pathogens in marine aquaculture worldwide. In Korea, they cause mass-mortalities in fish such as the commercially important olive flounder Paralichthys olivaceus. In particular, mixed infections of scuticociliates with pathogenic bacteria have been commonly reported. During efforts to identify natural marine-algae derived products that possess anti-bacterial and anti-scuticociliate properties, we found that an 80% methanolic extract of the red alga Polysiphonia morrowii Harvey exhibits both anti-scuticociliate activity against Miamiensis avidus, which is a major causative agent of scuticociliatosis, and anti-bacterial activities against fish pathogenic bacteria. Activity-guided fractionation and isolation of the 80% methanolic extract of P. morrowii yielded three bromophenols, which were identified as 3-bromo-4,5-dihydroxybenzyl methyl ether (1), 3-bromo-4,5-dihydroxybenzaldehyde (2) and urceolatol (3) based on spectroscopic analyses. 3-bromo-4,5-dihydroxybenzyl methyl ether (1) showed the highest anti-bacterial and anti-scuticociliate activities, with a minimal inhibitory concentration (MIC) of $62.5{\mu}g/mL$ (against Vibrio anguillarum) and minimal lethal concentration (MLC) of 62.5 ppm (in seawater). Investigations of the anti-bacterial and anti-scuticociliate activities of seventeen bromophenol derivatives, including the three isolated natural bromophenols, showed that the existence of an electron donating group or atom with a non-covalent electron pair at $C_4$ of the 2-bromophenol structure may be important in anti-scuticociliate activity. These findings suggest that the extract and bromophenol derivatives of P. morrowii may provide useful alternatives in aquaculture anti-scuticociliate therapies.

Natural product compounds are the source of numerous therapeutic agents. The marine environment produces natural products from a variety of structural classes exhibiting activity against numerous disease targets including anticancer agents. Among these, pectenotoxin-2 (PTX-2), which was first identified as a cytotoxic entity in marine sponges, which depolymerizes actin filaments, was found to be highly effective and more potent to activate an intrinsic pathway of apoptosis in p53-deficient tumor cells compared to those with functional p53 both in vitro and in vivo. However, the anti-proliferative mechanism of the compound at non-cytotoxic concentrations has not yet been explored. In the current study, we sought to investigate anti-proliferation and apoptosis of PTX-2 against U937 human leukemic cells and its underlying molecular mechanism. Exposure of U937 cells to PTX-2 resulted in growth inhibition and induction of apoptosis in dose- and time-dependent manner as measured by MTT assay, fluorescent microscopy and flow cytometric analysis. The anti-proliferative effect of PTX-2 was associated with a marked increase in the expression of cyclin-dependent kinase p21 (WAF1/CIP1) mRNA which was tumor suppressor p53-independent. The increase in apoptosis was connected with a time-dependent down-regulation of anti-apoptotic Bcl-XL and inhibitor of apoptosis proteins (IAPs) family such as XIAP and cIAP-2. Though additional studies are needed, these findings suggested that PTX-2-induced inhibition of U937 cells was associated with the induction of apoptotic cell death and the results provided important new insights into the possible molecular mechanisms of the anti-cancer activity of PTX-2.

The microalga, Tetrathelmis tetrathele, is used in the development of products for the aquaculture, food, and nutraceutical industries. In the present study, we investigated whether the T. tetrathele ethanolic extract (TTE), which has anti-inflammatory properties, can confer protection against alopecia and improve scalp health, influence the proliferation of human keratinocytes, HaCaT cells, and human hair follicle dermal papilla cells (HFDPC), or inhibit 5α-reductase activity. We found that TTE inhibited the production of the inflammatory mediator, nitric oxide (NO), and prostaglandin E2 (PGE2) without cytotoxicity in LPS-stimulated RAW 264.7 cells. In addition, TTE encouraged the proliferation of HaCaT cells and HFDPC. Our results showed that TTE had anti-inflammatory activities, proliferated HaCaT cells and HFDPC, and inhibited 5α-reductase activity. Therefore, we suggest that T. tetrathele could be a potent therapeutic agent for alopecia prevention and scalp improvement.
