In the previous studies, the present author found that high proportion of the follicular oocytes from mouse and rabbit ovaries are able to resume their maturation division in the anterior chamber of the eye in which they have been incubated by auto- or homoplastic transplantation. Especially in the case of the homoplastic transplantation, it was known that no trouble has been detected in the process of resumption of the oval maturation in particular connection with the antigen-antibody reaction between donor and recipient. These findings provide a possibility that the follicular oocytes from various animals would be matured in the eye even after the xenoplastic transplantation. Under such an assumption, the present studies were performed to examine the behavior of the follicular oocytes in the eye chamber of the animals of different species. For the donor of the follicular oocytes, domestic rabbits, albino rats of Sprague-Dowley strain, and albino mice of A-strain bred in our laboratory were used. The oocytes obtained from the ovarian follicules were introduced to the anterior chamber of the eye of different species of animals, with an exception of rabbit in which only the female animals were used as a recipient. The procedures of collection of ova, introduction to the eye, harvest from the eye ball, fixation, and staining were the same as mentioned in the previous reports (Cho, 1967b; Cho and Kim, 1968). The conclusions obtained are summarized as below. 1. The rabbit follicular oocytes are able to mature in the eye chambers of both male mouse and rat, although the proportion of the maturation is lower than when they are incubated autoplastically in the eye. When the ova were incubated in the male mouse eye for 24 hours, 21 per cent of them showed chromosomes at metaphase I and II, whereas the rate was 32 per cent when they were incubated in the eye of the male rat. These are apparently low comparing to the rate of 52 per cent of autoplastic transplantation. 2. When rat follicular oocytes were transferred into the mouse eye chamber and recovered after 24 hours, 43 per cent of them produced the mataphase I and II chromosomes. This proportion was higher than the result of the homoplastic transplantation which yielded 23 per cent of the ova on maturation. 3. The most striking result was found in the experiment with mouse follicular oocytes. Seventy-six per cent of the oocytes resumed their maturation division within 24 hours after they were transferred into the male rat eye chamber, and this figure was significantly high compared to the result o 55 per cent obtained by the homoplastic transplantation. In the rat eye, the induction of the degenerative ova also was low (19%). On the other hand, the proportion of the oval maturation decreased to 45 per cent, while that of degeneration increased 33 per cent when they were incubated in the eye of the female rabbit. 4. It was apparent from the present experiments that the follicular oocytes can reveal their activation to maturation in the eye chamber which contains aqueous humor which is known to be composed of low protein content and of very little gamma-globulin which acts as an antibody(Oser, 1965), and that it shows higher osmolarity than blood serum(Levene, 1958). Taking these properities into consideration the humor may provide unfavourable environment to the cells and tissues incubated in. However, it could be noteworthy finding that only the follicular oocytes in the eye of the different species can grow in healthy condition although the maturation rates are varied with the animal species. The fact that the rabbit follicular oocytes show the lower proportion in maturation may be due to the greater amount of the yolk granules in the egg cytoplasm than those in the mouse and rat oocytes. That the mouse oocytes incubated in the eye of the rat mouse and rat oocytes. That the mouse oocytes incubated in the eye of the rat resumed their maturation process in greater proportion would e explained by the fact that the rat eye chamber particularly provides the better environment to the mouse oocytes than the eye chamber of mouse does.
Single intravenous and oral administration to SD rats and ICR mice of both sexes were performed to investigate the acute toxicity of DWC-751, a new parenteral cephalosporin. $LD_50$ values for ICR mice and SD rats administered intravenously with DWC-751 were as follows; 1151.1 mg/kg (male SD rat), 1183.5 mg/kg (female SD rat), 2698.1 mg/kg (male ICR mouse), 2833.0 mg/kg (female ICR mouse). It is suggested that $LD_50$ values in rats and mice of both sexes would be 5000 mg/kg in oral route. Major general symptoms induced by injection intravenously with DWC-751 are decreased motor activity, increased respiratory rate, tremor and convulsion. In oral route, piloerection and soft stool are observed to 4 day after administration. No significant body weight changes were observed at any level in the groups administered with DWC-751. The gross finding of rats administered intravenously was observed cecum distension.

This study was conducted to investigate whether the obesity of obese fa/fa male Zucker rats can be ameliorated by mulberry leaves. There was no significant difference between mulberry treatment group and control group in body weight gain and food efficiency ratio. It also showed decreases on plasma triglyceride and total-cholesterol concentration compared to the control group. But there was no change in plasma leptin concentration between the control and mulberry treatment groups. These results suggest that mulberry leaves reduce serum lipid levels, but do not effect the plasma leptin concentration.
Kami-honghwa-tang(KH-19) is a prescription for reducing the side effect of radiotherapy. In this study, safety of KH-19 was evaluated by GLP guideline of Korea Food and Drug Administration. In acute oral toxicity study on rat, transient inhibition of weight increase was shown, but change in general symptom was not detected. No dead animal was observed up to 5,000 mg/kg in both male and female animals. In acute oral toxicity study on Beagle dog, transient vomiting, diarrhea, anorexia, and weight reduction was observed. However, no dead animal was observed up to 2,000 mg/kg in both male and female animals.
Effect of Compound-A, a phenylpropanoid isolated from Arctium lappa fruit, on the reversed cutaneous anaphylaxis (RCA) and complement-dependent hypersensitivity (CDH) were studied in SD male rats and ICR male mice, respectively. RCA and hemolysin (HY) titer test are related to reaction of Type II Hypersensitivity. Experiments were carried out to determine RCA as the edema of skin two hours after injection of 0.05 ml/site of anti-rat serum rabbit serum in SD rat. Purpose: The purpose of this study was to examine effects of decreased locomotor activity on mass, Type I and II fiber cross-sectional areas of ipsilateral and contralateral hindlimb muscles 21 days after establishing the Parkinson's disease rat model. Methods: The rat model was established by direct injection of 6-hydroxydopamine (6-OHDA, 50 ${mu}g$) into the left substantia nigra after stereotaxic surgery. Adult male Sprague-Dawley rats were assigned to one of two groups; the Parkinson's disease group (PD; n=17) and a sham group (S; n=8). Locomotor activity was assessed before and 21 days after the experiment. At 22 days after establishing the rat model, all rats were anesthetized and soleus and plantaris muscles were dissected from both ipsilateral and contralateral sides. The brain was dissected to identify dopaminergic neuronal death of substantia nigra in the PD group. Results: The PD group at 21 days after establishing the Parkinson's disease rat model showed significant decrease in locomotor activity compared with the S group. Weights and Type I and II fiber cross-sectional areas of the contralateral soleus muscle of the PD group were significantly lower than those of the S group. Conclusion: Contralateral soleus muscle atrophy occurs 21 days after establishing the Parkinson's disease rat model.
Dose-dependent pharmacokinetics of acetaminophen was studied in the rat. Acetaminophen was injected intravenously at doses of 10, 30, 50, 100 and 200mg/kg to the adult male rats. The well-known dose-dependent pharmacokinetic behavior was found even at 30mg/kg dose. It implies that metabolism of acetaminophen in the liver, probably sulfation, is saturated at very low concentration of acetaminophen. Dosage regimen establishment based on this characteristics would be necessary even at usual does level (300-600mg/day/body).
Present studies were carried out in order to estabilish the bronchoalveolar lavage method and to examine the response of bleomycin and peplomycin on the total cell number and the subpoulations of bronchoalveolar lavage fluid. A total of 24 male F344 rats, weighing 300-350 mg, were divided into 3 groups. Animals recelved either belomycin (BLM` 0.75 mg/0.2 ml/rat), peplomycin (PLM` 0.25mg/0.2ml/rat) for groups 2 and 3 or an equal volume of sterile saline lacking drugs for controls (group 1).
Objective : This study was conducted to investigate the effects of Panax Ginseng (인삼) on the sperm motility and spermatogenesis in the male rat. Methods : We used 8-week-old Sprague-Dawley rats, and administered the extract powder of Panax Ginseng to 5 rats (treated group) and normal saline (control group) once a day for 28 days. We isolated their testes surgically, then observed the change of the body weights before and after administration of Panax Ginseng extracts and normal saline. We observed the weight of the testes, epididymis, vascular gland, and prostate. Also, we examined the total, normal motile sperm concentration, and the concentration of testicular catalase and peroxidase. Results : We found that the concentration of normal, motile sperm in the testes of the Panax Ginseng group showed a significant difference compared with the control group. The angiogenesis of the seminiferous tubule was increased and the increasement of the number of spermatogonia, primary and secondary spermatocyte was observed in the Panax Ginseng group through a microscope. The body weight, the weight of the testes, epididymis, prostate and the concentration of testicular catalase and peroxidase were higher in the Panax Ginseng group but showed no significant difference. Conclusion : This study shows that Panax Ginseng may have an effect on the morphology and motility of sperm, the important factor in male fertility, and can promote the concentration of antioxidants, catalase and peroxidase, which is the important factor in spermatogenesis.
Objectives The object of this study was to evaluate the effect of Yikgeebohyul-tang aqueous extracts (YKBHT) on the propylthiouracil (PTU)-induced rat hypothyroidism. Methods The rats were divided into 6 groups : intact vehicle control, PTU control, LT4, YKBHT 500, 250 and 125 mg/kg treated groups. Hypothyroidism was induced by daily subcutaneous treatment of PTU 10 mg/kg for 28 days. YKBHT aqueous extracts were administered once a day as an oral dose of 500, 250 and 125 mg/kg for 42 days. The changes were observed : weight of body, thyroid gland, liver, testis, epididymis and prostate, serum thyroid hormone levels, serum male sex hormone levels, serum lipid profiles, liver and testis antioxidant system. These results were compared with LT4 0.5 mg/kg intraperitoneally treated rats. Results These PTU induced hypothyroidism and related hepatic and male reproductive organ damages were favorably and dose-dependently inhibited by treatment of YKBHT 500, 250 and 125 mg/kg, and YKBHT also effectively regulated the PTU-induced abnormal antioxidant defense factor changes in the both liver and testis. Although, LT4 also inhibited PTU-induced hypothyroidism and relative liver damages. But it deteriorated the hypothyroidism related testis, epididymis and prostate damages through testicular oxidative damages. Conclusions : The result of this study suggests that YKBHT has favorable effects on the hypothyroidism and related liver and reproductive organ damages with augmentation of antioxidant defense factor in the testis and liver. YKBHT 500, 250 and 125 mg/kg dose-dependently inhibited PTU-induced hypothyroidism and related liver and male reproductive organ damages in rats.
