Long-Duration Three-Dimensional Spheroid Culture Promotes Angiogenic Activities of Adipose-Derived Mesenchymal Stem Cells

  • Mesenchymal stem cells (MSCs) offer significant therapeutic promise for various regenerative therapies. However, MSC-based therapy for injury exhibits low efficacy due to the pathological environment in target tissues and the differences between in vitro and in vivo conditions. To address this issue, we developed adipose-derived MSC spheroids as a novel delivery method to preserve the stem cell microenvironment. MSC spheroids were generated by suspension culture for 3 days, and their sizes increased in a time-dependent manner. After re-attachment of MSC spheroids to the plastic dish, their adhesion capacity and morphology were not altered. MSC spheroids showed enhanced production of hypoxia-induced angiogenic cytokines such as vascular endothelial growth factor (VEGF), stromal cell derived factor (SDF), and hepatocyte growth factor (HGF). In addition, spheroid culture promoted the preservation of extracellular matrix (ECM) components, such as laminin and fibronectin, in a culture time- and spheroid size-dependent manner. Furthermore, phosphorylation of AKT, a cell survival signal, was significantly higher and the expression of pro-apoptotic molecules, poly (ADP ribose) polymerase-1 (PARP-1) and cleaved caspase-3, was markedly lower in the spheroids than in MSCs in monolayers. In the murine hindlimb ischemia model, transplanted MSC spheroids showed better proliferation than MSCs in monolayer. These findings suggest that MSC spheroids promote MSC bioactivities via secretion of angiogenic cytokines, preservation of ECM components, and regulation of apoptotic signals. Therefore, MSC spheroid-based cell therapy may serve as a simple and effective strategy for regenerative medicine.

  • Bone marrow (BM) derived mesenchymal stem cells (MSC) are pluripotent cells which can differentiate into osteogenic, adipogenic and other lineages. In spite of the broad interest, the information about the changes in BM cell composition, in particularly about the variation of MSC number and their properties in relation to the age of the donor is still controversial. The aim of this study was to investigate the age associated changes in variations of BM cell composition, phenotype and differentiation capacities of MSC using a rat model. Cell populations were characterized by flow cytometry using light scattering parameters, DNA content and a set of monoclonal antibodies. Single cell analysis was performed by conventional fluorescent microscopy. In vitro culture of MSC was established and their phenotype and capability for in vitro differentiation into osteogenic and adipogenic cells was shown. Age related changes in tibiae and femurs, amount of BM tissue, BM cell composition, proportions of separated MSC and yield of MSC in 2 weeks of in vitro culture were found. At the same time, neither change in phenotype no in differentiation capacities of MSC was registered. Age-related changes of the number of MSC should be taken into account whenever MSC are intended to be used for investigations.

  • MSC는 ITU에 의해 표준화된, 병행 시스템의 명세를 기술하기 위한 그래픽 형태와 텍스트 형태를 제공하는 언어로서 실시간 시스템 특히 통신 교환 시스템의 특성을 기술하기 위해 자주 사용된다. 통신 시스템이 제대로 동작함을 보이기 위해서는 정형적인 방법을 사용하여 시스템 행위를 검증할 필요가 있다. 통신 소프트웨어를 검증하는 방법 중 하나의 방법으로 유한 상태를 기반으로 하는 방법이 있다. 유한 상태를 기반으로 하는 방법에서는 먼저 시스템 명세에 해당하는 전체 상태 전이 그래프를 생성한 후 이를 바탕으로 model-checking 등의 방법을 사용하여 시스템의 특성을 검증한다. 본 논문에서는 MSC로 기술된 통신 소프트웨어 명세로부터 전체 상태 전이 그래프를 생성하는 방법에 초점을 맞추었다. 시스템 명세에 해당하는 상태 전이 그래프를 생성하기 위해 보다 직관적으로 MSC의 의미론을 표현할 수 있고, 또한 쉽게 전체 상태 전이 그래프를 생성할 수 있는 행위 종속 그래프를 제안하였다. MSC 명세는 일단 행위 종속 그래프로 변환이 되고 이 행위 종속 그래프를 이용하여 전체 상태 전이 그래프가 생성된다.Abstract Message Sequence Chart (MSC) standardized by International Telecommunication Union is a graphical and textual language for describing the specification of concurrent systems. It is frequently used both formally and informally for specifying the behavior of real-time systems, in particular telecommunication switching systems. To ensure that a communication system operates properly, the verification process showing the correctness of system's behavior formally is necessary. One of the verification methods is a finite-state method. In the finite-state method, the global state transition graph (GSTG) is constructed and then safety and liveness properties of systems are verified through a well-known method such as model checking. In this paper, we forcus on the construction of GSTG from the specifications of telecommunication software written in MSC. We suggest Action Dependency Graph (ADG) which can present the semantics of MSC intuitively and also provide a GSTG construction method from ADG. MSC specifications are translated to ADGs and, in turns, the GSTGs are constructed by using these ADGs.

A Study on the Correlation between MSC and major curriculum for Efficient Engineering Education (효율적인 공학 교육을 위한 MSC 교과목과 전공 교과목의 연관성에 관한 연구)

  • The purpose of this paper is to improve the curriculum efficiently and to raise the level of students's satisfaction by studying the correlation between MSC and major courses. For this, we compared the student's grades for those courses. Furthermore the level of satisfaction of the MSC subjects was surveyed. We found that one of dissatisfactions was due to the student's low recognition about the Accredited Program. So efforts to raise the student's satisfaction abouts the program should be done continuously and an in-depth study about MSC and major courses is necessary instead of just offering the required subjects.

  • This report summarizes the results of MODTRAN model that are used for the calculation of input radiance of the KOMPSAT-2 Multispectral Camera (MSC). We have calculated the input radiances for four months: January 15, April 15, July 15 and October 15. Annual averages are the arithmetic mean of results from four months. We used the mid-latitude winter and summer for the month of January and July, respectively, while US standard atmospheres are used for April and October. The orbital characteristics of KOMPSAT-2 and the seasonal variations of solar zenith angle over the Korean peninsula were incorporated as inputs to the model. The tropospheric aerosol extinction (visibility = 50 km) was assumed. The surface albedo used in the model calculation represents the global annual mean clear-sky albedo. MSC contract values are found to be considerably greater in the MSC spectral range than the total radiances calculated with the above general conditions. From these results, it can be inferred that the forthcoming MSC images would be somewhat dark. We certainly need a countermeasure for this issue.

Analysis of the MSC(Multi-Spectral Camera) Operational Parameters

  • The MSC is a payload on the KOMPSAT-2 satellite to perform the earth remote sensing. The instrument images the earth using a push-broom motion with a swath width of 15 km and a GSD(Ground Sample Distance) of 1 m over the entire FOV(Field Of View) at altitude 685 km. The instrument is designed to haute an on-orbit operation duty cycle of 20% over the mission lifetime of 3 years with the functions of programmable gain/offset and on-board image data compression/storage. The MSC instrument has one channel for panchromatic imaging and four channel for multi-spectral imaging covering the spectral range from 450nm to 900nm using TDI(Time Belayed Integration) CCD(Charge Coupled Device) FPA(Focal Plane Assembly). The MSC hardware consists of three subsystem, EOS(Electro Optic camera Subsystem), PMU(Payload Management Unit) and PDTS(Payload Data Transmission Subsystem) and each subsystems are currently under development and will be integrated and verified through functional and space environment tests. Final verified MSC will be delivered to spacecraft bus for AIT(Assembly, Integration and Test) and then COMSAT-2 satellite will be launched after verification process through IST(Integrated Satellite Test). In this paper, the introduction of MSC, the configuration of MSC electronics including electrical interlace and design of CEU(Camera Electronic Unit) in EOS are described. MSC Operation parameters induced from the operation concept are discussed and analyzed to find the influence of system for on-orbit operation in future.

Expression of HLA and Mixed Lymphocyte Reaction of Mesenchymal Stem Cells Derived from Human Umbilical Cord Blood (제대혈 유래 중간엽줄기세포에서 HLA의 발현과 Mixed Lymphocyte Reaction)

  • In recent years, the mesenchymal stem cells (MSC) derived from various tissues have been widely tested for developing cell therapies, tissue repair and transplantation. Although there has been much interest in the immunomodulatory properties of MSC and their immunologic reactions following autologous, allogeneic and xenogenic transplantation of MSC in vivo, up to date, the expression of immunogenic markers, such as class I and II human leukocyte antigens (HLA), after differentiation of human umbilical cord blood (hUCB)-derived MSC has been poorly investigated and require extensive in vitro and in vivo testing. In this experiment, the expression of the HLA-ABC and HLA-DR on hUCB-derived MSC have been tested by immunocytochemical staining. The undifferentiated MSC were moderately stained for HLA-ABC but very weakly for HLA-DR. In order to investigate the inhibitory effect of allogeneic lymphocytes on proliferation of MSC, the MSC were cultured in the presence or absence of peripheral allogeneic lymphocytes stimulated with concanavalin A. The allogeneic lymphocytes did not significantly inhibit MSC proliferation. We conclude that hUCB-MSC expressed moderately class I HLA antigen while almost negatively class II HLA antigen. The MSC have an immunomodulatory effect which can suppress the allogeneic response of lymphocytes. These in vitro data suggest that allogeneic MSC derived from cord blood can be useful candidate for allogeneic cell therapy and transplantation without a major risk of rejection.

  • 본 연구에서는 경주시 내남면 일대를 대상으로 KOMPSAT MSC(Multi Spectral Camera) 영상(2007.06.12)을 이용하여 TCT(Tasseled-Cap Transformation), NDVI(Normalized Difference Vegetation Index) 알고리즘을 적용하여 분포도를 작성 하였으며 TCT DN 값을 기초로 영상 강조 및 변환을 통한 임상분류에 적합한 밴드 추출과 NDVI 분포도에서의 DN값을 기초로 산림현장 조사 결과에서 취득된 결과와의 비교 분석을 통하여 알고리즘에 대한 임상분류에 있어서의 변별력 분석을 수행하였다. 본 연구를 통하여 KOMPSAT MSC 영상에서의 임상분류를 위한 식생 알고리즘 적용 가능성을 검토하고자 한다.

Histological Examination of Engineered Mesenchymal Stem Cells Improve Bladder Function in Rat (랫드에서 방광기능 향상의 엔지니어링 중간엽 줄기세포의 조직학적 소견)

  • This study was undertaken to examine the effects and to investigate the relevant mechanisms of overexpressing stromal cell-derived factor-1 (SDF-1) produced by engineered mesenchymal stem cells, in a neurogenic bladder (NB) rat model. Sprague-Dawley (SD) rats (N=48) were randomly divided into 4 groups comprising 12 rats each: control group, Injury group, Injury+imMSC group, and Injury+SDF-1 eMSC group. Rats in the Injury+imMSC group were treated with imMSCs, whereas the Injury+SDF-1 eMSC group were administered SDF-1 eMSCs. After 4-weeks therapy, the bladder and pelvic nerve (PN) tissues were examined by subjecting to Masson's trichrome staining and immunofluorescence. Administration of SDF-1 eMSC resulted in improved smooth muscle content in the bladder tissue, significantly increased β-III tubulin expression of the PN, and enhanced SDF-1 expression (P<0.05). The bladder wall repair can be attributed to the overexpression of SDF-1 by SDF-1 eMSCs. Significantly increased SDF-1 expression was obtained in the Injury+SDF-1 eMSC group (P<0.05). The crushed PN also showed significant recovery in the Injury+SDF-1 eMSC group (P<0.05). In conclusion, our results indicate that SDF-1 eMSCs express more SDF-1 in vivo, thereby facilitating the repair of injured nerve and recovery of NB in rats.

  • As a unique payload of Komsat-2, MSC, comprising EOS(Electro-Optical Sub-system), PMU(Payload Management Unit) and PDTS(Payload Data Transmission Sub-system), is supposed to take pictures of one panchromatic and 4 multi-spectral image between wavelength 450mm~900mm, and is being under final Satellite I&T. It will perform the earth remote sensing with applications such as acquisition of high resolution images, surveillance of large scale disasters and its countermeasure, survey of natural resources, etc.. Under the hostile influence of the extreme space environmental conditions due to deep space and direct solar flux, the thermal design is especially of major importance in designing a payload. There are tight temperature range restrictions for electro-optical elements while on the other hand there are low power consumption requirements due to the limited energy source on the spacecraft. This paper describes details of thermal control system for MSC.
