한국인 IgA 신병증 환아에서 MHC Class II유전자형과 예후와의 관계 분석 (MHC Class II Allele Association in Korean Children With IgA Nephropathy and its Role as a Prognostic Factor)

  • 목 적 : IgA 신병증은 혈뇨와 단백뇨를 보이는 사구체 질환으로 세계적으로 가장 빈도가 높은 질환중의 하나 이며 우리 나라를 포함한 아시아 지역에서도 높은 발생율을 보이고 있으나 아직 정확한 발생기전이나 원인은 불분명한 상태이다. IgA 신병증은 발병 10년 후 $20-30\%$가 말기 신부전증으로 진행됨이 보고되고 있으나 신부전으로 진행속도는 환자에 따라 매우 다양하기 때문에 특이적인 조직적합항원을 찾아냄으로서 치료 및 예후 결정에 도움을 주기 위하여 본 연구를 시행하였다. 대상 및 방법 : 세브란스 병원에 입원하여 IgA 신병증으로 진단 받은 환자를 대상으로 정상 신기능을 유지하고 있었던 소아환아 69명(Group A)과 이미 신부전으로 진행하여 신이식을 시행 받은 70명(Group B)을 포함한 신기능 저하군으로 분류하여 HLA대립유전자의 결정은 SSOP (sequence specific oligonucleotide probe)법으로 분석하여 HLA-class II(DR, DQ)대립유전자의 발현빈도와 IgA 신병증의 예후와의 관계를 분석하였다. 결 과 : 대상 환자의 진단 당시의 평균나이는 A군은 $8.8{\pm}2.9$세, B군은 $35.0{\pm}15.5$세였고, 남녀비는 각각 2.8:1과 2.5:1이었다. HLA-DQB1 유전자중 139명중 $47.5\%$$03^{**}$의 발현빈도가 가장 증가되어 있었고 $06^{**}(32.4\%),\;04^{**}(28.7\%),\;05^{**}(20.1\%),\;02^{**}(8.6\%)$순이 었다. DQ7의 HLA-DQB1*0301 이 전체 환자의 20.1$\%$(28/139)로 가장 흔한 대립유전자였으나 두군간에 유의한 차이는 없었다. 대립유전자중 HㄴA-$DQBI^*03^{**}과\;DQB1^*05^{**}$가 이미 말기신부전으로 진행된 Group B에서 유의하게 증가되어 있었으며(P<0.05), $03^{**}$ 아형중 $DQB1^{*}0302와\;05^{**}중\;DQB1^{*}05031$이 신기능이 정상인 A군에 비해 유의하게 증가되어 있었다(P<0.05). DR 유전자형의 low-frequency type상 HLA-$DRB1^*03과\;HLA-DRB1^*15$가 Group B에서 유의 있게 증가되어 있었으나, high-frequency type상에서는 양군 사이에 유의한 차이가 없었다. A군의 경우 신조직 검사의 소견에 따라 4등급으로 분류하여 대립유전자의 발현빈도를 분석하였으나 의미 있는 관계가 없었다. 곁 론 : IgA신병증의 예후를 예측하기 위한 여러 조사에서 발병 초기의 심한 단백뇨, 고혈압, 조직 병리학상의 심한 변화 등이 있을 경우 예후가 나쁘다고 하였으며 이런 위험 인자외에 유전적 요인에 대해 알아보고자 하였다. 본 연구에서 신부전으로 진행하여 신대체요법을 받은 IgA신병증환자군에서 HLA-$DQB1^*0302와\;^*05031$ 대립유전자의 발현빈도가 유의하게 증가되어 있어 이 결과를 바탕으로 치료후의 경과 및 예후를 진단초기에 예측하여 환자의 관리에 적용할수 있는 중요한 자료로 이용될 수 있을 것으로 사료된다.

한국 재래 닭 품종 특성 및 초기성장 개량을 위한 분자표지 개발 (Characteristics and Improving Breed of Economic Traits of Korea Native Chicken)

  • 본 연구는 한국 재래 닭의 유전적 특성을 분자표지를 이용하여 그 차이를 규명하고 초기성장에 미치는 영향을 분석하여 이를 이용한 재래닭의 개량을 목적으로 실시하였다. MHC class II B-LB 유전자 내의 염기변이체가 경제형질에 미치는 영향에 대하여 연구하였다. MHC class II B-LB유전자 내 400 bp 크기의 유전자를 증폭하여 염기서열 분석과 제한효소 처리를 이용한 다형성 분석을 실시하였다. 연구결과 두 개의 제한효소 절단지역이 발견되었으며 427 지역을 Type I 으로 651 지역은 Type II로 정하여 RFLP 분석을 실시하였다. Type I지역의 유전자형은 TT, TC, CC로 나타났으며, TypeII 지역의 유전자형은 MM, Mm, mm으로 나타났다. TC와 Mm 유전자형이 다른 유전자형과 비교하였을 때 한국재래 닭에서 높은 출현빈도를 보였다(0.8, 0.88). 유전자형이 한국 재래 닭의 150일령 체중에 미치는 영향을 분석한 결과 CC와 Mm 유전자형에서 통계적 유의성이 도출되었다 (P<0.05). 따라서 본 연구의 결과를 이용하여 한국 재래 닭의 유전적 특성을 규명할 수 있으며 초기 성장이 높은 성적을 나타내는 CC, Mm 유전자형을 개량에 이용하게 된다면 큰 효과를 얻을 수 있을 것으로 사료되어진다. 본 연구의 결과는 차후 한국 재래 닭의 과학적이고 지속적인 유전자원의 보존과 육종 전략에 있어 매우 유용하게 활용될 것으로 기대된다.

  • The immunogenetic analysis was performed to characterize the Korean native chickens (KNC) determined by monoclonal antibodies specific to chicken leukocyte differentiation antigens and flow cytometry. A total of 174 chickens including 58 KNC (black, brown and darkbrown colored), 77 foreign breed (Nagoya, White Reghorn, Rhode Island and Cornish) and 39 mixed breed (19 KNC with Nagoya and 20 KNC with Rhode Island) separately growing at Animal Science and Technology Institute were examined. The proportion of cells expressing MHC class II molecule (B-L in chicken) was significantly high in KNC. Proportion of CD4+ T helper cells was also higher in KNC and two mixed breed than that in foreign breed. However, proportion of CD8+ cells and TCR1 + (${\gamma}^{\delta}$ T cell receptor) cells was the lowest among the breed examined. Otherwise, those proportions were significantly high in White leghorn and two mixed breeds with two exclusive subpopulations. The two subpopulations were also typically shown in MHC class $II^+$ cells in KNC and one mixed breed, black-colored KNC with Nagoya. Although genotypic analysis was not pursued to characterize the immunogenetic properties of KNC, difference of phenotypic expression based on leukocyte differentiation molecules could be elucidated in KNC in this study.

Vitamin C Up-regulates Expression of CD80, CD86 and MHC Class II on Dendritic Cell Line, DC-1 Via the Activation of p38 MAPK

  • Vitamin C is an essential water-soluble nutrient which primarily exerts its effect on host defense mechanisms and immune homeostasis, but the mechanism related to immune-potentiation is poorly understood. Since dendritic cells (DCs) are known as a potent antigen presenting cell (APC) that could enhance the antigen specific immune responses, we investigate the effects of vitamin C on activation of DCs and its related mechanism by using dendritic cell lines, DC-1. First, we found that there was no damage on DC-1 by 2.5 mM of vitamin C. In the presence of vitamin C, the expression of CD80, CD86, and MHC molecules was increased, but it was decreased by the pre-treatment of SB203580, p38 MAPK-specific inhibitor. We confirmed the phosphorylation of p38 MAPK was increased by the treatment of vitamin C. Taken together, these results suggest that vitamin C could enhance the activity of dendritic cells via the up-regulation of the expression of CD80, CD86, and MHC molecules and the activation of p38 MAPK is related to this process.

Cyclooxygenase Inhibitors, Aspirin and Ibuprofen, Inhibit MHC-restricted Antigen Presentation in Dendritic Cells

  • Background: Nonsteroidal anti-inflammatory drugs (NSAIDs) are widely used to relieve pain, reduce fever and inhibit inflammation. NSAIDs function mainly through inhibition of cyclooxygenase (COX). Growing evidence suggests that NSAIDs also have immunomodulatory effects on T and B cells. Here we examined the effects of NSAIDs on the antigen presenting function of dendritic cells (DCs). Methods: DCs were cultured in the presence of aspirin or ibuprofen, and then allowed to phagocytose biodegradable microspheres containing ovalbumin (OVA). After washing and fixing, the efficacy of OVA peptide presentation by DCs was evaluated using OVA-specific CD8 and CD4 T cells. Results: Aspirin and ibuprofen at high concentrations inhibited both MHC class I and class II-restricted presentation of OVA in DCs. In addition, the DCs generated in the presence of low concentrations of the drugs exhibit a profoundly suppressed capability to present MHC-restricted antigens. Aspirin and ibuprofen did not inhibit the phagocytic activity of DCs, the expression level of total MHC molecules and co-stimulatory molecules on DCs. Ibuprofen rather increased the expression level of total MHC molecules and co-stimulatory molecules on DCs. Conclusion: These results demonstrate that aspirin and ibuprofen inhibit the intracellular processing event of the phagocytosed antigen, and further suggest that prolonged administration of NSAIDs in high doses may impair the capability of DCs to present antigens in asiociation with MHC molecules.

Analysis of Swine Leukocyte Antigen Haplotypes in Yucatan Miniature Pigs Used as Biomedical Model Animal

  • The porcine major histocompatibility complex (MHC) is called swine leukocyte antigen (SLA), which controls immune responses and transplantation reactions. The SLA is mapped on pig chromosome 7 (SSC7) near the centromere. In this study, 3 class I (SLA-1, SLA-3, and SLA-2) and 3 class II (DRB1, DQB1, and DQA) genes were used for investigation of SLA haplotypes in Yucatan miniature pigs in Korea. This pig breed is a well-known model organism for biomedical research worldwide. The current study indicated that Korean Yucatan pig population had 3 Class I haplotypes (Lr-4.0, Lr-6.0, and Lr-25.0) and 3 class II haplotypes (Lr-0.5, Lr-0.7, and Lr-0.25). The combinations of SLA class I and II haplotype together, 2 homozygous (Lr-4.5/4.5 and Lr-6.7/6.7) and 3 heterozygous (Lr-4.5/6.7, Lr-4.5/25.25, and Lr-6.7/25.25) haplotypes were identified, including previously unidentified new heterozygous haplotypes (Lr-4.5/4.7). In addition, a new SLA allele typing method using Agilent 2100 bioanalyzer was developed that permitted more rapid identification of SLA haplotypes. These results will facilitate the breeding of SLA homozygous Yucatan pigs and will expedite the possible use of these pigs for the biomedical research, especially xenotransplantation research.

Use of Cell-Penetrating Peptides in Dendritic Cell-Based Vaccination

  • Cell-penetrating peptides (CPPs) are short amino acids that have been widely used to deliver macromolecules such as proteins, peptides, DNA, or RNA, to control cellular behavior for therapeutic purposes. CPPs have been used to treat immunological diseases through the delivery of immune modulatory molecules in vivo. Their intracellular delivery efficiency is highly synergistic with the cellular characteristics of the dendritic cells (DCs), which actively uptake foreign antigens. DC-based vaccines are primarily generated by pulsing DCs ex vivo with various immunomodulatory antigens. CPP conjugation to antigens would increase DC uptake as well as antigen processing and presentation on both MHC class II and MHC class I molecules, leading to antigen specific CD4+ and CD8+ T cell responses. CPP-antigen based DC vaccination is considered a promising tool for cancer immunotherapy due to the enhanced CTL response. In this review, we discuss the various applications of CPPs in immune modulation and DC vaccination, and highlight the advantages and limitations of the current CPP-based DC vaccination.

Recombinant Adeno-Associated Virus Expressing Truncated IK Cytokine Diminishes the Symptoms of Inflammatory Arthritis

  • IK can downregulate interferon-gamma-induced major histocompatibility complex (MHC) class II expression through the MHC class II transactivator, which suggests that IK can inhibit the interactions between immune cells. We delivered adeno-associated virus serotype 2 (AAV2) encoding the genes for truncated IK (tIK) or green fluorescent protein (GFP) to DBA1/J mice via intravenous injection. Seven weeks after injection, collagen-induced arthritis was induced in the AAV2-treated mice. AAV2-tIK injection reduced the severity of arthritis and the percentage of pathogenic Th17 cells compared with AAV2-GFP injection. These results suggest a novel gene therapy strategy for treatment of inflammatory arthritis.

아카바네 생 바이러스 백신 접종에 의한 한우 백혈구 아군의 변화 (Leukocyte subpopulations of peripheral blood in Korean indigenous cattle vaccinated with attenuated live Akabane virus vaccine)

  • To identify immune response of leukocytes in peripheral blood of cattle vaccinated with an attenuated live Akabane virus vaccine, leukocytes were reacted with monoclonal antibodies which are specific to bovine lymphocyte surface antigens and assayed by the flow cytometry. Serum neutralizing (SN) test was used to measure antibody titers after vaccination, SN antibody was appeared to 7 days post-vaccination (PV) and 2-8 antibody titers were observed in 14 days PV. Proportion of $CD8^-$ MHC $class II^+$ expressing cells were rapidly increased at 3 days PV. $CD8^+$ MHC $class II^-$ cells were increased at 7 days PV. $CD4^+CD8^-,\;WC^+CD4^-,\;CD4^+CD8^+,\;WC1^-CD4^+, \;WC1^-CD8^+$, and $CD4^-CD8^+$ cells were highly increased at 3, 3, 7, 7, 14, 14 days PV, respectively.
