A report of six unrecorded radiation-resistant bacterial species isolated from soil in Korea in 2018

  • Six bacterial strains 18JY42-3, 18SH, 18JY76-11, 17J11-11, 18JY14-14, and 18JY15-11 assigned to the phylum Proteobacteria, Firmicutes, and Actinobacteria were isolated from soil samples in Korea. The Cohnella species, strain 18JY42-3 was Gram-stain-positive, short rod-shaped and beige-colored. The Methylobacterium species, strains 18SH and 18JY76-11 were Gram-stain-negative, short rod-shaped and pink-colored. The Microterricola species, strain 17J11-11 was Gram-stain-positive, short rod-shaped and yellow-colored. The Paenarthrobacter species, strains 18JY14-14 and 18JY15-11 were Gram-stain-positive, short rod-shaped and white-colored. Phylogenetic analysis based on 16S rRNA gene sequence showed that strains 18JY42-3, 18SH, 18JY76-11, 17J11-11, 18JY14-14, and 18JY15-11 were most closely related Cohnella rhizosphaerae (MH497628; 98.8%), Methylobacterium goesingense (MH497632; 99.1%), Methylobacterium populi (MH497635; 99.9%), Microterricolagilva (MH504108; 98.4%), Paenarthrobacter nicotinovorans (MH497641; 100%), and Paenarthrobacter nitroguajacolicus (MH497646; 99.2%), respectively. All the six unrecorded strains showed resistance to UV radiation. This is the first report of these six species in Korea.

  • 열에 불안정하면서 -OH기를 함유하는 미지의 화합물은 LC/TSP/MS에서 [${MNH_4}^+$] 이온, [$MH^+$] 이온, 그리고 [$MH^+-OH$] 이온의 생성이 가능하며, 이들 이온의 m/z값의 차이가 17로서 같기 때문에 이들의 생성 정도에 따라 분자량 추정에 혼란이 일어날 수가 있었다. 따라서 다른 보완 가능한 실험결과가 요구되며 $CF_3COOD+NH_4OH$ 이온화 용액을 사용하여 그 유용성을 검토하였다. 또한 LC/TSP/MS에서 생성되는 토막이온들이 필라멘트의 전자살에 의해 생성되는 것인지 아니면 다른 어떤 생성기전이 존재하는지를 검토하였다.

Molecular Cloning and Characterization of Trehalose Biosynthesis Genes from Hyperthermophilic Archaebacterium Metallosphaera hakonesis

  • The trehalose $({\alpha}-D-glucopyranosyl-[1,1]-{\alpha}-D-glucopyranose)$ biosynthesis genes MhMTS and MhMTH, encoding a maltooligosyltrehalose synthase (MhMTS) and a maltooligosyltrehalose trehalohydrolase (MhMTH), respectively, have been cloned from the hyperthermophilic archaebacterium Metallosphaera hakonesis. The ORF of MhMTS is 2,142 bp long, and encodes 713 amino acid residues constituting a 83.8 kDa protein. MhMTH is 1,677 bp long, and encodes 558 amino acid residues constituting a 63.7 kDa protein. The deduced amino acid sequences of MhMTS and MhMTH contain four regions highly conserved for MTSs and three for MTHs that are known to constitute substrate-binding sites of starch-hydrolyzing enzymes. Recombinant proteins obtained by expressing the MhMTS and MhMTH genes in E. coli catalyzed a sequential reaction converting maltooligosaccharides to produce trehalose. Optimum pH of the MhMTS/MhMTH enzyme reaction was around 5.0 and optimum temperature was around 70 C. Trehalose-producing activity of the MhMTS/ MhMTH was notably stable, retaining 80% of the activity after preincubation of the enzyme mixture at $70^{\circ}C$ for 48 h, but was gradually abolished by incubating at above $85^{\circ}C$. Addition of thermostable $4-{\alpha}-glucanotransferase$ increased the yield of trehalose production from maltopentaose by 10%. The substrate specificity of the MhMTS/MhMTH-catalyzed reaction was extended to soluble starch, the most abundant maltodextrin in nature.

  • 몇 가지 종류의 corticosteroid에 대하여 액체 크로마토그래피-질량분석법으로 양이온 질량 스펙트럼을 얻었다. 화학구조에 따라 수소 첨가된 분자이온 [$MH^+$], 암모늄 첨가이온 [${MNH_4}^+$], 또는 ($MH^+-60$) 이온이 base peak였고 [$MH^+-18$] 또는 [${MNH_4}^+-18$] 이온 등이 특성적으로 나타났다. Methylprednisolone acetate를 male Sprague-Dawley rat에 경구투여한 다음 24시간 동안 배설된 뇨로부터 유리상태 또는 접합상태의 대사물질들을 가수분해, 추출 및 농축하고, thermospray LC/MS를 사용하여 양이온과 음이온 질량토막이온을 분석하였다. Methylprednisolone acetate의 C-21 위치에서의 탈아세틸화(deacetylation), C-20 위치에서 C=0의 -CHOH로의 환원, C-11 위치에서 CHOH의 C=0로의 산화 또는 C-17과 C-20 사이의 bond cleavage등에 의해 생성되는 것으로 추정되는 10여종의 대사물질을 검출하였다. 그 중에 20-hydroxymethylprednisolone(20-HMP), methylprednisolone(MP), methylprednisone(11-KMP)등은 표준물질과 비교 확인하였다.

오징어 채낚기어선용 LED 집어등의 어획성능 (Fishing efficiency of LED fishing lamp for squid jigging vessels)

  • This study has conducted a comparative analysis on the fishing efficiency of LED fishing lamps by squid jigging vessels, the Yeongrak-ho (16 tons) and Somang-ho (9.77 tons), which operated during September and October 2010 and during October 2011, comparing with MH (Metal Halide) fishing lamp-equipped fishing vessels. This study has also examined vessel's fuel consumption level. The light powers of LED fishing lamps of the Yeongrak-ho and Somang-ho were 25.8kW and 32kW, respectively. Those of the MH fishing vessels, that is, the MH fishing lamp-equipped fishing vessels, were 105kW and 81kW, respectively. The average squid catch in number of an LED vessel, Yeongrak-ho, was 39.2% of the MH fishing lamp-equipped fishing vessels; however, that of the Somang-ho improved to 78.7% of the MH fishing lamp-equipped vessels. Average catch in number by Yeongrak-ho crew was 2.6 times more than catch in number by automatic jigging machines. Average catch in number by MH fishing vessel crew was 1.8 times more than that by automatic jigging machines. An LED vessel, Yeongrak-ho's fishing rate was 17.5%~152.2% of the MH fishing vessels, that is, 61.1% on average, in comparison of combined catch in number per automatic jigging machine and per crewmember. Somang-ho's fishing rate was 6.7%~127.6% of the MH fishing vessels, that is, 73.1% on average. The average fuel consumption level of the Somang-ho, throughout its departure from to arrival at the port, was 475.7l, and that during fishing hours was 109.6l, or 23.0% of the total fuel consumption level. Somang-ho's fuel consumption level per fishing hour was 9.7l on average.

Ni-MH 전극용 $AB_2$계 수소저장합금의 볼밀링 처리에 의한 표면개질 연구 (Surface Modification of $AB_2$ Type Hydrogen Storage Alloys by Ball Milling for Ni-MH Battery)

  • In order to improve the activation properties of the $AB_2$ type hydrogen storage alloys for Ni-MH battery, the alloy surface was modified by employing high energy ball milling. The $Zr_{0.54}Ti_{0.45}V_{0.54}Ni_{0.87}Cr_{0.15}Co_{0.21}Mn_{0.24}$ alloy powder was ball milled for various period by using the high energy ball mill. As the ball milling time increased, activation of the $AB_2$ type composite powder electrodes were enhanced regardless of additives. When the ball milling time was small discharge capacities of the $AB_2$ type composite powder electrodes increased with the milling time. On the other hand for large milling time it decreased with increasing milling time. The maximum discharge capacity was obtained by ball milling for 3-4 min.

생장조정제(生長調整劑) 씨엠처리(處理)가 시호(柴胡)의 생육(生育) 및 근(根) 수량(收量)에 미치는 영향(影響) (Effect of Growth Regulator C-MH Application on Growth and Root Yield in Bupleurum falcatum L.)

  • 본(本) 연구(硏究)는 1994년부터 1995년까지 전남농촌진흥원(全南農材振興院) 특용작물(特用作物) 시험포장(試驗圃場)에서 삼도(三島) 시호(柴胡)을 공시(供試)하여 생장조정제(生長調整劑) C-MH 처리(處理)가 시호(柴胡)의 생육(生育) 및 근수량(根收量)에 미치는 영향(影響)을 구명(究明)하기위해 실시(實施)한 시험결과를 요약(要約)하면 다음과 같다. 1. C-MH 액제(液劑) 살포(撒布)가 처리농도(處理濃度)가 높을수록, 살포회수(撒布回數)가 많을수록 개화기(開花期)는 점차 늦어졌으며 무처리(無處理)에 비하여 개화(開花)가 $17{\sim}21$일 지연(遲延)되었고 화아분화(花芽分化)가 억제(抑制)되어 주당(株當) 화경수(花莖數)의 감소효과(減少效果)가 있었다. 2. C-MH 액제(液劑) 2,000ppm 살포농도(撒布濃度)로 하여 처리시기(處理時期)는 초장(草長)이 30cm 및 40cm일때 2회(回) 살포(撒布)하면 무처리(無處理)에 비해 초장(草長)은 15.2cm 짧아졌으나 주당(株當) 분기수(分枝數)가 3.0개로 많아졌으며, 규근장(圭根長) 3.8cm, 규근경(圭根徑) 0.65mm 및 주당(株當) 기근수(支根數) 3.0개(個) 정도로 지하부(地下部)의 생장량(生長量)이 증가(增加)되었다. 3. 건근(乾根) 수량(收量)은 초장(草長)이 30cm및 40cm인 시기에 C-MH액제(液劑) 2,000ppm 농도(濃度)로 2회살포(回撒布)하였을 때 상근중(上根重) 비율(比率)이 10% 높아 전체적인 개체(個體) 생육량(生育量)이 많아 근부(根部)의 비대(肥大)로 무처리(無處理) 65kg/10a에 비하여 27% 증수(增收)를 가져왔다.

고지방식이로 비만이 유도된 C57BL/6 마우스에서 Metformin과 황금추출물의 병용 투여 시 비만관련 지표에 미치는 영향 (The Effect of Scutellariae Radix Combined with Metformin on Obesity-Relating Biomarker in High Fat Fed C57BL/6 Mice)

  • Objectives: The purpose of this study was to investigate anti-obesity effect of Scutellariae Radix extract combined with metformin. Methods: Male C57BL/6 mice, 4 weeks of age, were used to set high-fat diet induced obesity model. They were grouped NOR (normal control), HFD (high fat diet control), MET (metformin, 100 mg/kg/day), MH2 (metformin 50 mg/kg/day+Scutellariae Radix 200 mg/kg/day), and HG4 (Scutellariae Radix 400 mg/kg/day). MET, MH2, and HG4 were orally administered for 10 weeks. Body weight was measured every week. Fasting blood glucose, triglyceride, total cholesterol (TC), high density lipoprotein, glutamic oxaloacetic transaminase (GOT), and glutamic-pyruvic transaminase (GPT) were measured before sacrifice. Oral glucose tolerance test (OGTT) was also performed. Organ weight and internal fat weight were measured after sacrifice. Results: MH2 group showed a reduction of body weight when compared with HFD group. MH2 group showed stable blood level control which was calculated areas under the curves by OGTT. TC, GOT, GPT level, internal fat, and organ weight in MH2 group reduced. Conclusions: The combined treatment of Scutellariae Radix and Metformin has impact on treating obesity. Further studies are needed to evaluate the effect of Metformin and herbal medicine combination therapy.

왕불유행, 목통, 통초가 산후 생쥐의 유즙분비량과 유즙분비 관련 인자에 미치는 영향 (Effect of Melandrii Herba, Akebia Quinata Decaisne, and Tetrapanax Papyriferus on Milk Secretion and Lactation Related Factors in Postpartum Mice)

  • Objectives: The purpose of this study is to investigate the effect of Melandrii Herba (MH), Akebia Quinata Decaisne (AQ), and Tetrapanax Papyriferus (TP) on milk secretion and aquaporin (AQP) expression in lactating mice. Methods: For the experiment, the mice were divided into three groups, which were orally administered MH (2,720 mg/kg), TP (400 mg/kg) and AQ (2,800 mg/kg) extracts respectively for 3 weeks from Day 1 after the birth, compared with the control group (C group), which was administered distilled water. A group consisted of six infantile mice per postpartum mouse. For comparison with the C group, non-pregnant SKH-1 mice were used as the virgin group. Results: 1. When it comes to the immunohistochemical staining for prolactin receptors in the mammary glands, the AQ and MH groups showed a strong immune response to the secretory epithelial cells constituting the mammary alveoli, while the TP group represented a weaker immune response. 2. In the immunohistochemical staining for AQP in the mammary glands, AQP1 showed a strong immune response in the walls of capillaries and venules around the mammary alveoli, and AQP3 in the epithelial cells constituting the mammary alveoli, and AQP5 in some tissues between the mammary alveoli. AQP1 was expressed in the order of TP group>AQ group=C group>MH group, and AQP3 was MH group and AQ group>TP group=C group, and AQP5 was MH group>C group>AQ group and TP group. 3. In the Western blot, AQP1 was expressed in the order of TP group>AQ group>C group>MH group, and AQP3 was MH group>AQ group>C group>TP roup, and AQP5 was MH group>TP Group>C group>AQ group. All of AQP1, 3, 5 expression were significantly higher in the C group than in the Virgin group. Conclusions: The administration of Akebia Quinata Decaisne, Tetrapanax Papyriferus and Melandrii Herba have the effect of improving prolactin levels in postpartum mice and increasing the expression of prolactin receptor and AQPs in the mammary glands, suggesting that lactation might be enhanced by the development of the mammary glands.

  • The downing of the MH17 reminds the world that the international civil aviation is not as safety and security as people expected. Such tragedy is partly due to the risk and danger of the armed conflict zones, but is more attributed to the ignorance to the international law by the responsible parties concerned. International laws applicable to the armed conflict zones shall be strictly followed, and the reparation shall be provided to the victims, otherwise such disaster could not be avoided in the future.
