In this study, we investigated the effects of Undaria pinnatifida (UP), Petalonia binghamiae (PB) and Punctaria latifolia (PL) extracts on the inhibition of proliferation and apoptosis in human gastric and breast cancer cells. AGS, MDA-MB-231 and SK-BR-3 cells were treated with 0, 50, 100, and 200 μg/ml concentrations of the extracts to determine their anti-proliferative effects, using the MTT assay. The UP, PB and PL extracts inhibited proliferation of AGS, MDA-MB-231 and SK-BR-3 cells in a dose-dependent manner, and the PL extract was found to be the most effective. DAPI staining was also performed to determine changes in the cell nucleus. Further, the AGS, MDA-MB-231 and SK-BR-3 cells were treated with 0, 50, 100, and 200 μg/ml of only the PL extract. DAPI staining showed increased chromatin condensation, which is indicative of apoptosis, in the 200 μg/ml group. The expression of the Bax, Bcl-2, and PARP proteins in AGS, MDA-MB-231 and SK-BR-3 cells treated with the PL extract was also determined by western blot analysis. The expression of Bax (a pro-apoptotic protein) and cleaved-PARP was increased, whereas the expression of Bcl-2 (an anti-apoptotic protein) was decreased compared with the control. These findings indicate that the PL extract may have potential as an alternative anticancer drug and nutraceutical.
Background: Predicting the important role of intercellular adhesion molecule-1 expression on the acute ischemia-reperfusion injury, we set out to demonstrate it by assessing the degree of expression of ICAM-1 after warm ischemia-reperfusion in canine unilateral lung ischemia model. Material and Method: Left unilateral lung ischemia was induced by clamping the left hilum for 100 minutes in seven adult mongrel dogs. After reperfusion, various hemodynamic pararmeters and blood gases were analyzed for 4 hours, while intermittently clamping the right hilum in order to allow observation of the injured Ieft lung function. The pulmonary venous blood was collected serially to measure TNF- and cICAM-1 level. After 4 hours of reperfusion, the lung tissue was biopsied to assess cICAM-1 expression, and to measure tissue malondialdehyde(MDA) and ATP level. Result: The parameters including arterial oxygen partial pressure, pulmonary vascular resistance and tissue MDA and ATP level suggested severe lung damage. Serum TNF-$\alpha$ level was 8.76$\pm$2.37 ng/ml at 60 minutes after reperfusion and decreased thereafter. The cICAM-1 level showed no change after the reperfusion during the experiment. The tissue cICAM-1 expression was confirmed in 5 dogs. Conclusion: The increase of TNF-$\alpha$ Ievel and expression of tissue ICAM-1 were demonstrated after ischemia reperfusion injury in canine lung model.
This study was conducted to investigate the effects of Opuntia humifusa supplementation on lipid peroxidation and superoxide dismutase (SOD) protein expression at resting state in various organs of rats fed a high-fat diet. Sixteen Sprague-Dawley male rats, 6 weeks of age, were randomly divided into two groups: a control diet group (CG, n=8) and an experimental diet group (EG, n=8). They were given a high-fat diet (CG) or a diet supplemented with 5% of O. humifusa (EG) for 8 weeks. The results showed that the malondialdehyde (MDA) levels of the kidney and the liver were significantly lower in the EG group than in the CG group (p<0.01). In addition, the MDA levels in the skeletal muscle of the EG group tended to be lower than those in the CG group, but this difference was not significant. The Cu, Zn-SOD protein expression in the kidney of the EG group was significantly increased compared with that of the CG group (p<0.01). The Mn-SOD protein expression in the skeletal muscle of the EG group was significantly increased compared with that of the CG group (p<0.01). These results suggest that O. humifusa supplementation has antioxidative properties, which are exerted in a specific organ manner, and that it inhibits the action of lipid peroxidation and the expression of SOD in rats fed a high-fat diet.

The aim of this study was to investigate whether the exposure to vinyl chloride monomer(VCM) induces lipid peroxidation in workers by evaluating the concentration of malondialdehyde(MDA) in the urine in order to assess worker's oxidative stress due to exposure of vinyl chloride monomer. The subjects investigated in the study were divided into the experimental group; 18 workers exposed to VCM, and the control group; 19 workers unexposed to VCM. A gas chromatography/pulsed flame photometric detector(GC/PFPD) was utilized to analyze thiodiglycolic acid(TDGA), which was methylated with trimethylsilyldiazomethane (2.0M in diethyl ether) in urine and the urinary MDA, the product of lipid peroxidation, was determined by high-performance liquid chromatography/ultraviolet-visible detector after derivatized with 2,4-dinitrophenylhydrazine(DNPH). The concentrations of urinary TDGA in controls and VCM exposure workers were 0.13(2.01)mg/g Cr. GM(GSD) and 0.35(1.96)mg/gCr. GM(GSD), respectively. The concentrations of urinary MDA were $0.12(2.21){\mu}mol/gCr$. GM(GSD) in controls and $1.35(1.79){\mu}mol/gCr$. GM(GSD) in VCM exposure workers. As a result of simple regressions analysis between urinary concentration of TDGA and MDA in VCM exposure workers, it was found that the $R^2$ value was 0.261 (p=0.03) and the drinking and smoking did not affect their level. In conclusion, the workers exposed to VCM have a potentially to suffered by oxidative stress due to VCM exposure and the urinary MDA can be applicable to the marker of effect to assess the level of worker's VCM exposure.

Prunus mume Sieb. et Zucc is widely distributed in East Asia (Korea, Japan, and China), and its fruit is often used as a medication and food material. However, because most previous studies have only investigated the state of Prunus mume fruit extract, studies on the various ways of processing this extract are still needed to increase its utilization. In this study, we evaluated the physicochemical properties and physiological activities of spray-dried Prunus mume vinegar powder (SPP). The sugar content, pH, total acidity, and moisture content of the SPP were 8.90 °Brix, 3.19, 1.05%, and 3.07%, respectively. The SPP exhibited significantly high antioxidant activity in terms of DPPH radical scavenging activity (65.55%), reducing power (1.48), and hydrogen peroxide scavenging activity (48.07%). In addition, the SPP remarkably decreased the cell viability of human breast MDA-MB-231 and human skin cancer SK-MEL-28 in a dose-dependent manner. The morphological results of the treatment of MDA-MB-231 cells with SPP were distorted, shrunken cell masses. Furthermore, apoptotic bodies and nuclear condensation formed in the SPP-treated MDA-MB-231 cells. The total polyphenol and flavonoid contents of the SPP were 59.58 ㎍/g (gallic acid equivalent) and 57.56 ㎍/g (quercetin equivalent). The results of this study indicate that SPP, which has antioxidant activity and anticancer effects, can be useful in the development of natural medicines and functional food ingredients.
Many epidemiological and observational studies show that distorted body image of normal body weight is widespread among Korean young females. This study was performed to evaluate the prevalence of overweight and underweight of young females and, to compare nutrient intakes, body composition, serum indices (lipids, MDA: Malondialdehyde, TAS : Total Antioxidant Status) with different BMI groups. The subjects were 75 university students in Seoul. We divided the subjects into 3 groups according to their BMI by IOTF guideline (UW: underweight group, BMI < 18.5, NW: normal body weight group; 18.5 $\leq$ BMI < 23.0, OW: overweight group; BMI $\geq$ 23.0). Data on dietary intakes, body compositions and serum indices were obtained in 3 groups. Differences on all of the above variables were assessed by body weight groups. Using IOTF guidelines, the prevalence of overweight and underweight in young females were 23%, 61%, 16% respectively. $\beta$-carotene and vitamin A intake of UW were significantly higher than that of NW (p < 0.05). Fiber intakes of NW was significantly higher than that of W (p < 0.05). But intakes of energy and the other nutrients were not significantly different among BMI groups. Calcium and folate intakes were 75%, 61% of KDRIs. Serum TGs were significantly higher in OW than that of NW, UW (p < 0.05), but level of MDA and TAS were not significantly different. The association of overweight and low intake of $\beta$-carotene and vitamin A may be one of many factors predisposing obese females to a high risk of oxidative stress later in life. This requires urgent nutritional intervention programs involving enough intake of fruit and vegetables, with modification of inappropriate dietary habits.
In this study, we investigated the antioxidative effects of So-Dang-Tang (SDT) on streptozotocin (STZ)-induced diabetic rats. Diabetes was induced by intraperitoneal injection of STZ (45 mg/kg body weight) into Sprague-Dawley rats. The SDT (200 mg/kg) and the reference drug, glibenclimide (1mg/kg), were orally administered once a day for 28 days in STZ-induced diabetic rats. The activity of antioxidant enzymes, including those of superoxide dismutase (SOD) and catalase, and the levels of glutathione (GSH) and production of malondialdehyde (MDA) were measured in the liver, kidney, and pancreas of diabetic rats. Treatment with SDT in STZ-induced diabetic rats significantly increased the activities of antioxidant enzymes and GSH levels in the liver, kidney, and pancreas when compared to those of the STZ-control group. SDT also significantly decreased lipid peroxidation product and MDA levels in STZ-induced diabetic rats. These results indicate that SDT has an antioxidative action in STZ-induced diabetic rats.

Inhibitory effects of allergic contact dermatitis of hot water extract of Duchung (Eucommia ulmoides Oliver) leaf, bark and stem growing at Sancheong-gun were investigated for female BALB/c mouse induced by repeat elicitation of DNCB (2,4-dinitro-chlorobenzene). Skin reactions, consisting of increased ear thickness and the presence of ear inflammation, were observed in mice treated with DNCB and Duchung. Weight of lymph node, spleen and thymus in mice treated with Duchung extracts were lower than that of mouse treated with DNCB. Ear weight of mouse treated with Duchung extracts was decreased by increasing the concentration of sample as compared to control group and dropped as low as control level at 1,000 mg/kg. Ear thickness became thinner as test time on Duchung extract progressed. MDA (malondialdehyde) contents in liver tissue were not different in sample group with DNCB group, but were different in ear tissue. NO (nitric oxide) contents was decreased in Duchung extract groups at serum and ear tissues as compared to 1% DNCB group. In the present study, the results suggested that Duchung extract inhibits inflammatory response and oxidative damage induced DNCB allergen.
This study was to investigate the biological activities of green tea seed methanol extract (GTSME) and compared those of green tea methanol extract (GTME) for using green tea seed as the functional food material. The hydrogen-donating activity of GTSME was over 50% at the $100 {\mu}g/mL$ concentration the activity of GTME was 21.86% at the $1000{\mu}g/mL$ concentration compared with that of control. The MDA (malondialdehyde) production was 60 Mol/g and 50 Mol/g in the mouse liver homogenate teated with GTME and GTSME of $1000{\mu}g/mL$ concentrations, respectively, and the values were lower than 86 Mol/g of control. GTME and GTSME of $1000{\mu}g/mL$ concentration inhibited the proliferation of over 50% and over 20% in A549 and SW480 human cancer cells, respectively. The morphology transformation was shown in the cancer cells treated with GTSME of $500{\mu}g/mL$ with the decrease of cell numbers lower than that of control cells numbers. The NO production was increased in a dose dependent manner in the RAW264.7 macrophage cells treated with GTME and GTSME of 1, 10, 100 and $1000{\mu}g/mL$ concentrations, and the NO production by GTSME was $2.04{\mu}M$ at $100{\mu}g/mL$ concentration, and the value was higher than $0.77{\mu}M$ by GTME.
