The Effects of Shi-Quan-Dai-Bu-Tang and Its Ingredients on the Survival of Jejunal Crypt Cells and Hematopoietic Cells in Irradiated Mice (방사선 피폭 마우스에서 소장움세포 및 조혈세포 생존에 미치는 십전대보탕 및 구성단미의 영향)

  • We performed this study to determine the radioprotective effects of Shi-Quan-Dai-Bu-Tang, as a prescription of traditional Oriental medicine, and its major ingredients. The jejunal crypt survival, endogenous spleen colony formation, and apoptosis in jejunal crypt cells were investigated in mice irradiated with high and low dose of ${\gamma}$-rays. The administration of danggui, baishaoyao, rensan, gancao or baifuling before irradiation protected the jejunal crypts (p<0.005). Shoudehuang, danggui, baishaoyao, rensan and huangqui increased the formation of endogenous spleen colony (p<0.05). Chuanxiong, baishaoyao, rensan and baifuling reduced the frequency of radiation-induced apotosis (p<0.05). The results indicated that the extracts of danggui, baishaoyao, rensan and baifuling may have radoprotective effects in mice irradiated with high and low dose of ${\gamma}$-rays. The radioprotective effect of the prescription, Shi-Qaun-Da-Bu-Tang, was not significant.

  • This experiment was designed to study the change of cytotoxic and antitumor effects according to the prebrewed method of Semen Tiglii and Rhizoma Coptidis. The cytotoxic and antitumor effects were evaluated on human cell lines(A 549, Caki-1, LL2, Sarcoma 180, NIH/3T3) after exposure to prebrewed Semen Tiglii and Rhizoma Coptidis water extract 0.1, 0.2, 0.4, 0.8, 1.6 mg/ml using in MTT assay, LDH, colony forming efficency and SRB assay which were regarded as a valuable method for cytotoxic and antitumor effects of unknown compound on tumor cell lines. The results obtained in this studies were as follows. 1. The cytotoxicity from the result of MTT assay was low slightly in the ST II(炒巴豆霜), high in the ST III(醋炒巴豆). The cytotoxicity of ST I + RC(生巴豆霜加黃連) was similar to that of STI(生巴豆霜). 2. The cytotoxicity from the result of LDH was low slightly in the ST Ⅱ (炒巴豆霜), high in the ST III(醋炒巴豆). The cytotoxicity of ST I + RC(生巴豆霜加黃連) was similar to that of ST I(生巴豆霜). 3. The antitumor affect on A 549 tumor cell from the result of colony forming efficiency was low slightly in the ST II (炒巴豆霜) and ST I + RC(生巴豆霜加黃連). 4. The antitumor effect on Caki-1 tumor cell from the result of SRB assay was low slightly in the ST II (炒巴豆霜). 5. Median survival time and Increased life span increased slightly in the ST I RC(生巴豆霜加黃連) and ST II (炒巴豆霜). 6. The inhibitory effect on the growth of Sarcoma 180 and Lewis lung carcinoma tumor cell increased slightly in the ST I + RC(生巴豆霜加黃連) and ST II (炒巴豆霜).

  • The purpose of this study is to investigate the relationship between oral health behavior and dietary habits and caries occurrence test and to provide basic data necessary for prevention of dental caries. The average salivary salivary fraction was 8.35ml and the non - amorphous salivary fraction was 2.41ml. The salivary buffering ability was 9.29 and the mean glucose retention time was 15.81 minutes. In the Streptococcus mutans colony count test, low risk group <105 was 80.5% and low risk group <105 was 84.4% in Lactobacillus colony count test. Correlation between caries test scores showed a positive correlation between salivary ratio and non - salivary ratio, saliva buffering ability, non - salivary fraction and glucose retention time, and Lactobacillus. Factors affecting the irritant saliva fraction were alcohol as a factor affecting the ratio of nuts, pork, and non - irritant saliva. In order to prevent dental caries, proper dietary control education should be combined with dental caries activity test and oral health education for proper eating habits.

Comparison of Standard Culture Method and Real-time PCR Assay for Detection of Staphylococcus aureus in Processed and Unprocessed Foods (가공식품과 비가공식품에서의 황색포도상구균 검출을 위한 배지법과 Real-time PCR법의 비교)

  • Staphylococcus aureus is one of the major pathogens that can cause staphylococcal infection and food poisoning. In this study, we compared conventional culture methods and real-time PCR for detection of S. aureus in artificially inoculated milk, sausage, raw pork, and vegetable salad. The performance of a coagulase test for confirming S. aureus was also compared with a colony PCR test. Bulk food samples (500 g each) were artificially inoculated with S. aureus and divided into 20 samples (25 g or mL each). All samples were added to tryptic soy broth (225 mL/sample) with 10% NaCl and incubated at $37^{\circ}C$ for 24 h. After the enrichment, broth cultures were streaked onto Baird-Parker (BP) agar with egg yolk tellulite, and incubated at $37^{\circ}C$ for 24 h. In addition, 1 mL of broth cultures was collected to perform real-time PCR. Two suspicious colonies from the BP agar were picked up and plated on nutrient agar and incubated at $37^{\circ}C$ for 24 h followed, by a coagulase confirmation test and a colony PCR analysis. There were no statistical differences between culture methods and realtime PCR in food samples with low background microflora, such as milk and sausage. However, a significant statistical difference was found between the culture methods and real-time PCR for raw pork and vegetable salad. Furthermore, the colony PCR test of the presumptive colonies on BP agar for confirming S. aureus is more accurate and efficient than the coagulase test for unprocessed foods.

Optimal Cold Temperature for the Artificial Hibernation of Bombus ignitus Queen Bumblebees

  • Bumblebees are widely used to pollinate various greenhouse crops. Among the different bumblebee species, Bombus ignitus is indigenous to Korea, China, Japan and Russia. B. ignitus undergoes one generation per year, and artificial hibernation is essential for year-round rearing of the bumblebee. Keeping the queens under low-temperature conditions for several months is an effective method for terminating their diapause and promoting colony development. In the present study, we investigated how cold temperature affects the artificial hibernation of B. ignitus queens. Under chilling temperatures of $-2.5^{\circ}C$, $0^{\circ}C$, $2.5^{\circ}C$ and $5^{\circ}C$ with constant humidity >80%, the queens stored at $2.5^{\circ}C$ exhibited the highest survival rates, which were 74.0% at one month, 67.0% at two months, 60.0% at three months, 46.0% at 4 months, 33.0% at 5 months and 24.0% at 6 months. Lower survival rates were observed at $0^{\circ}C$, $5^{\circ}C$, $7.5^{\circ}C$ and $12.5^{\circ}C$. At $2.5^{\circ}C$ the colony developmental characteristics after diapause were 1.2- to 1.5-fold greater than those when queens were stored at $5^{\circ}C$. Thus, $2.5^{\circ}C$ and 70% R.H. were the most favorable chilling temperature and humidity conditions for terminating the diapause of B. ignitus queens.

Establishment of In-Vitro Culture System for Enhancing Production of Somatic Cell Nuclear Transfer (SCNT) Blastocysts with High Performance in the Colony Formation and Formation of Colonies Derived from SCNT Blastocysts in Pigs

  • Although somatic cell nuclear transfer (SCNT)-derived embryonic stem cells (ESCs) in pigs have great potential, their use is limited because the establishment efficiency of ESCs is extremely low. Accordingly, we tried to develop in-vitro culture system stimulating production of SCNT blastocysts with high performance in the colony formation and formation of colonies derived from SCNT blastocysts for enhancing production efficiency of porcine ESCs. For these, SCNT blastocysts produced in various types of embryo culture medium were cultured in different ESC culture medium and optimal culture medium was determined by comparing colony formation efficiency. As the results, ICM of porcine SCNT blastocysts produced through sequential culture of porcine SCNT embryos in the modified porcine zygote medium (PZM)-5 and the PZM-5F showed the best formation efficiency of colonies in α-MEM-based medium. In conclusion, appropriate combination of the embryo culture medium and ESC culture medium will greatly contribute to successful establishment of ESCs derived from SCNT embryos.

  • This study was performed to determine the effect of Phyllostachys nigra var. henenis Strapf leaf extract on jejunal crypt survival, endogenous spleen colony formation and apoptosis in jejunal crypt cells of mice irradiated with high and low dose of gamma-radiation. Phyllostachys nigra var. henenis Strapf administration before irradiation (I.P.: 125 mg/kg of body weight, at 24 hours before irradiation) resulted in an increase of the formation of endogenous spleen colony (p<0.01). The frequency of radiation-induced apoptosis was also reduced by pretreatment of Phyllostachys nigra var. henenis Strapf (I.P.: 280 mg/kg or 28 mg/kg of body weight, at 24 hours before irradiation, p<0.01). These results indicated that Phyllostachys nigra var. henenis Strapf might be a useful radioprotector, especially since it is a relatively nontoxic natural product. Further studies are needed to characterize better the promotion nature of Phyllostachys nigra var. henenis Strapf and its components.

Radioprotective Effect of Red Ginseng in Irradiated Mice with High and Low Dose of Radiation (고선량 및 저선량 방사선 피폭에 대한 홍삼의 방사선 방호효과)

  • Studies were performed to determine the effect of Korean red ginseng (extract powder, spray-dried), it is made of choice 6-year-old raw ginseng roots, and processed by steaming and drying, on jejunal crypt survival, endogenous spleen colony formation, and apoptosis in jejunal crypt cells of irradiated mice. Jejunal crypts were protected by pretreatment of red ginseng (1 mg/head, single I.P. at 24hours before irradiation, p<0.05). Red ginseng administration before irradiation (1 mg/head, single I.P at 24hours before irradiation) resulted in an increase of the formation of endogenous spleen colony (p<0.05). The frequency of radiation-Induced apoptosis in intestinal crypt cells was also reduced by treatment of red ginseng both pretreatment (P.O.: 2 mg/ml of drinking water for 7 days, p<0.005, I.P.: 1 mg/head, single I.P. at 24 hours before irradiation, p<0.005) and post-treatment (1 mg/head, single I.P at 30 minutes after irradiation, p<0.05). These results indicated that Korean red ginseng might be a useful radio-protector, especially since it is a relatively nontoxic natural product. Further studies are needed to characterize better the promotion nature of red ginseng and its fractions.
