The LRV1-4 capsid protein possesses an endoribonuclease activity that is responsible for the single site-specific cleavage in the 5' untranslated region (UTR) of its own viral RNA genome and the formation of a conserved stem-loop structure (stem-loop IV) in the UTR is essential for the accurate RNA cleavage by the capsid protein. To delineate the nucleotide sequences, which are essential for the correct formation of the stem-loop structure for the accurate RNA cleavage by the viral capsid protein, a wildtype minimal RNA transcript (RNA 5' 249-342) and several synthetic RNA transcripts encoding point-mutations in the stem-loop region were generated in an in vitro transcription system, and used as substrates for the RNA cleavage assay and RNase mapping studies. When the RNA 5' 249-342 transcript was subjected to RNase T1 and A mapping studies, the results showed that the predicted RNA secondary structure in the stem-loop region using FOLD analysis only existed in the presence of Mg$\^$2+/ ions, suggesting that the metal ion stabilizes the stem-loop structure of the substrate RNA in solution. When point-mutated RNA substrates were used in the RNA cleavage assay and RNase T1 mapping study, the specific nucleotide sequences in the stem-loop region were not required for the accurate RNA cleavage by the viral capsid protein, but the formation of a stem-loop like structure in a region (nucleotides from 267 to 287) stabilized by Mg$\^$2+/ ions was critical for the accurate RNA cleavage. The RNase T1 mapping and EMSA studies revealed that the Ca$\^$2+/ and Mn$\^$2+/ ions, among the reagents tested, could change the mobility of the substrate RNA 5' 249-342 on a gel similarly to that of Mg$\^$2+/ ions, but only Ca$\^$2+/ ions identically showed the stabilizing effect of Mg$\^$2+/ ions on the stem-loop structure, suggesting that binding of the metal ions (Mg$\^$2+/ or Ca$\^$2+/) onto the RNA substrate in solution causes change and stabilization of the RNA stem-loop structure, and only the substrate RNA with a rigid stem-loop structure in the essential region can be accurately cleaved by the LRV1-4 viral capsid protein.

This paper deals with an improved fault location algorithm with compensation ground capacitance through distributed parameter for a long parallel T/L. For the purpose of fault locating algorithm non-influenced by source impedance and fault resistance, the loop method was used in the system modeling analysis. This algorithm uses a positive and negative sequence of the fault current for high accuracy of fault locating calculation. Power system model of 160km and 300km long parallel T/L was simulated using EMTP software. To evaluate of the proposed algorithm, we used the several different cases 64 sampled data per cycle. The test results show that the proposed algorithm was minimized the error factor and speed of fault location estimation.
A lot of recent researches have shown that a Pseudo Random Binary Signal is a quite effective test signal to measure the impulse response of a plant. Generally speaking, however, such a response itself is not satisfactory to determine the appropriate control parameters or control inputs. Here, the author intends to estimate the unknown parameters of the First Order Plant with Dead Time by means of correlation method using M-sequence signal. The time constant T and the dead time L of the plant are eatimated with one tracking loop by automatically adjusting delay time .tau. of M-sequence signal according to variations of T and L. In this paper, a three level M-sequence signal is used as a test signal in order to avoid troublesome operations to calculate partial derivatives of a given performance index with respect to the parameters which are usually required in the Model Method. Several experiments with analogue computer using low pass filters as averaging circuits showed good results as expected.
When LIM is drived by the general purpose inverter, there is no food-back of speed and thrust, etc. Therefore, through analysis of LIM by the simulation, the prediction of operating characteristics and patterns of LIM is necessary. But, the open-loop control system is used widely, because it can consist of simple and cheap. In this paper, we foaled with the dynamic characteristics of LIM drived by V/f constant during acceleration, cruising speed deceleration. And, we compared the simulation value with actual measurement value.

In general DGPS system, the correction message is transferred to users by wireless modem. To cover wide area, many DGPS station should be needed. And DGPS users must have a wireless modem that is not necessary in standalone GPS. But SBAS users don't need a wireless modem to receive DGPS corrections because SBAS correction message is transmitted from the GEO satellite by L1 frequency band. SBAS signal is generated in the GUS(Geo Uplink Subsystem) and uplink to the GEO satellite. This uplink transmission process causes two problems that are not existed in GPS. The one is a time delay in the uplink signal. The other is an ionospheric problem on uplink signal, code delay and carrier phase advance. These two problems cause ranging error to user. Another critical ranging error factor is clock synchronization. SBAS reference clock must be synchronized with GPS clock for an accurate ranging service. The time delay can be removed by close loop control. We propose uplink ionospheric error correcting algorithm for C/A code and carrier. As a result, the ranging accuracy increased high. To synchronize SBAS reference clock with GPS clock, I reviewed synchronization algorithm. And I modified it because the algorithm didn't consider doppler that caused by satellites' dynamics. SBAS reference clock synchronized with GPS clock in high accuracy by modified algorithm. We think that this paper will contribute to basic research for constructing satellite based DGPS system.
We present a highly effective T-DNA inserted gene screening method as part of a reverse genetics model system using the Chinese cabbage (Brassica rapa L. spp. pekinensis). Three-step two-dimensional (2D) matrix strategies are potentially accurate and useful for the identification of specific T-DNA inserted mutants from a large population. To construct our Chinese cabbage model, we utilized a forward genetics screening approach for the abnormal phenotypes that were obtained from transgenic plants of Brassica rapa generated with Agrobacteria tumefaciens containing the pRCV2 vector. From one transgenic plant with an abnormal phenotype, we observed that the st1 gene (which is related to senescence-associated process proteins) contained a T-DNA fragment, and that its expression level was decreased. This T-DNA insert was then used as a control to construct an effective screening pool. As a result, the optimum template concentration was found to be 0.1-1 ng in our PCR strategy. For other conditions, positive changes to the Gibbs free energy prevented the formation of oligo dimers and hairpin loop structures, and autosegment extension gave better results for long fragment amplification. Using this effective reverse genetics screening method, only 23 PCR reactions were necessary to select a target gene from a pool of 100 individual DNAs. Finally, we also confirmed that the sequence we obtained from the above method was identical to the flanking sequence isolated by rescue cloning.
Equine follicle stimulating hormone receptor (eFSHR) has a large extracellular domain and an intracellular domain containing approximately 10 phosphorylation sites within the G protein-coupled receptor. This study was conducted to analyze the function of phosphorylation sties at the eFSHR C-terminal region. We constructed a mutant of eFSHR, in which the C-terminal cytoplasmic tail was truncated at residue 641 (eFSHR-t641). This removed 10 potential phosphorylation sites from the C-terminal region of the intracellular loop. The eFSHR-wild type (eFSHR-wt) and eFSHR-t641 cDNAs were subcloned into the pCMV-ARMS1-PK2 expression vector. These plasmids were transfected into PathHunter CHO-K1 Parental cells expressing β-arrestin 2 enzyme acceptor fusion protein and analyzed for agonist-induced cAMP response. The cAMP response in cells expressing eFSHR-t641 was lower than the response in cells expressing eFSHR-wt. EC50 values of eFSHR-wt and eFSHR-t641 were 1079 ng/mL and 1834 ng/mL, respectively. eFSHR-t641 was approximately 0.58-fold compared with that of eFSHR-wt. The maximal response in eFSHR-wt and eFSHR-t641 was 24.7 nM and 16.7 nM, respectively. The Rmax value of phosphorylation sites in eFSHR-t641 was also decreased to approximately 68.4% of that in eFSHR-wt. The collective data implicate that the phosphorylation sites in the eFSHR C-terminal region have a pivotal role in signal transduction in PathHunter CHO-K1 cells, and indicate that β-arrestin is involved in coupling the activated receptors to the internalization system.

This paper proposes a simple MPPT control scheme of a Current- Control-Loop Error system Based that can be obtains a lot of advantage to compare with another digital control method, P&O and IncCond algorithm, that is applied mostly a PV system. An existent method is needed an expensive processor such as DSP that calculated to change the measure power of a using current and voltage sensor at the once. But, a proposed method is easy to solve the cost reduction and power unbalance problems that it is used by control scheme to limit error of a current control of common sensor. This proposed algorithm had verified through a simulation and an experiment on battery charger using PIC that is the microprocessor of a low price.
This paper deals with a robust controller for an induction motor (IM) which is represented as a linear parameter varying systems. To do so linear matrix inequality (LMI) based approach and robust Lyapunov feedback are associated. This approach is related to the fact that the synthesis of a linear parameter varying (LPV) feedback controller for the inner loop take into account rotor resistance and mechanical speed as varying parameter. An LPV flux observer is also synthesized to estimate rotor flux providing reference to cited above regulator. The induction motor is described as a polytopic LPV system because of speed and rotor resistance affine dependence. Their values can be estimated on line during systems operations. The simulation and experimental results largely confirm the effectiveness of the proposed control.
To operate the ion cyclotron resonance heating (ICRH) antennas in a better heating state and produce relatively low impurities, it is necessary to control the antenna spectrum by changing the antenna phasing. As the electrical length of the antenna feeding transmission lines is changing as a matter of the standing wave pattern at the ceramic supports, 90° elbows, T-connectors and antenna loops, we chose to measure the current at the grounding points of the antenna loops by antenna strap probe. The voltage drops along a small, several millimeter-long paths at the end of the antenna loops give a signal that is proportional to the current in the antenna loop. Through the simulation of the antenna strap probe and the actual measurement of the antenna phasing under vacuum conditions, the reliability of the antenna strap probe based diagnostic system have been successfully proved. Moreover, this system was successfully applied to the ICRH daily experiments in the spring of 2021. In the near future, the active real-time feedback control of the antenna phasing system will be developed based on this diagnostic system in the EAST tokamak.
