A Study on the Lipid Components of Silkworn Pupae (잠용의 지질성분(脂質成分)에 관(關)한 연구(硏究))

  • To study the composition of silkworm pupae oil, lipid of silkworm pupae was separated into two fractions, ether extractable and 85% methanol extractable, and the lipid components of each fraction were analyzed by using silicic acid column, thin layer chromatography and gas chromatography. Silkworm pupae contains 35.4% crude fat (dry-basis) of which consists 34.4% diethyl ether-extract and 0.9% of 85% methanolextract. The diethyl ether-extract contained 96.1% of neutral lipid, 2.9% of glycolipid and 1.0% of phospholipid while methanol-extract was consisted of 47.4% of neutral lipid, 14.6% of glycolipid and 38.1% of phospholipid. The major components of phospholipid were phosphatidyl glycol(41.0%), and phosphatidyl choline(28.2%) and phosphatidyl ethanolamine(21.2%) in the diethyl ether-extract and phosphatidyl glycol(48.4%), phosphatidyl inositol(22.8%) and phosphatidyl choline(17.9%) in the methanol-extract. The major fatty acids of the total lipid were oleic acid(33.5%), linolenic acid(31.0%) and palmitic acid(23.1%).

Fatty Acid and Carotenoid Production by Sporobolomyces ruberrimus when Using Technical Glycerol and Ammonium Sulfate

  • The production of carotenoids, lipid content, and fatty acid composition were all studied in a strain of Sporobolomyces ruberrimus when using different concentrations of technical glycerol as the carbon source and ammonium sulfate as the nitrogen source. The total lipids represented an average of 13% of the dry weight, and the maximum lipids were obtained when using 65.5 g/l technical glycerol (133.63 mg/g). The optimal conditions for fatty acid production were at $27^{\circ}C$ using 20 g of ammonium sulfate and a pH range from 6 to 7, which produced a fatty acid yield of $32.5{\pm}1\;mg/g$, including $1.27{\pm}0.15\;mg$ of linolenic acid (LNA), $7.50{\pm}0.45\;mg$ of linoleic acid (LLA), $5.50{\pm}0.35\;mg$ of palmitic acid (PA), $0.60{\pm}0.03\;mg$ of palmitoleic acid (PAL), $1.28{\pm}0.11\;mg$ of stearic acid (SA), $9.09{\pm}0.22\;mg$ of oleic acid, $2.50{\pm}0.10\;mg$ of erucic acid (EA), and $4.25{\pm}0.20\;mg$ of lignoceric acid (LCA), where the palmitic, oleic, and linoleic acids combined formed about 37% of the total fatty acids. The concentration of total carotenoids was 2.80 mg/g when using 20 g of ammonium sulfate, and consisted of torularhodin (2.70 mg/g) and $\beta$-carotene (0.10 mg/g), at $23^{\circ}C$ and pH 6. However, the highest amount with the maximum specific growth rate was obtained (${\mu}_{max}=0.096\;h^{-1}$) with an ammonium sulfate concentration of 30 g/l.

Reduced Leptin and Raised Glycerol Secretions in Mouse 3T3-L1 Adipocytes by Garlic-added Kochujang

  • In order to develop the improved kochujang witb an anti-adipogenic effect, garlic-added kochujang was prepared and followed by fermentation at $30^{\circ}C$ for 120 days. Anti-adipogenic effects of the garlic-added kochujang in cultured 3T3-L1 adipocytes were investigated by measuring leptin and glycerol levels as indicators of lipid accumulation associated with lipolysis, respectively. Additional levels of garlic powder in the preparation of kochujang was determined from the result of a sensory evaluation test. Garlic-added kochujang showed the highest points when the added content was 3%. Fermentation of kochujang led to the decreased leptin secretion and increased glycerol release in the 3T3-L1 adipocytes. The addition of garlic to the kochujang also resulted in reduced leptin secretion and induced lipolysis. Since these results suggested that addition of garlic into kochujang can improve the anti-adipogenic effects of kochujang, it might be possible to develop garlic-added kochujang as an antiobesity-functional kochujang.

  • The compositions of the polar lipids in the free and bound lipids from four varieties of experimentally cultivated potatoes were identified and quantified by thin layer- and gas-liquid chromatographies. The results were summarized as follows : 1. The glycolipids contained in the free and bound lipids were fractionated and identified as esterified sterol glycoside, monogalactosyl diglyceride, sterol glycoside, digalactosyl diglyceride, and trigalactosyl diglyceride, of which the highest content to the total lipid quantity were 5.8% of esterified sterol glycoside in the free lipid and 6.1% of trigalactosyl diglyceride in the bound lipid. The content of monogalactosyl diglyceride in the free and bound lipids was almost the same, whereas the content of esterified sterol glycoside was higher in the free lipid than in the bound lipid, and the contents of sterol glycoside, digalactosyl diglyceride, and trigalactosyl diglyceride were higher in the bound lipid than in the free lipid on the contrary. 2. The phospholipid contained in the free and bound lipids were fractionated and identified as phosphatidyl ethanolamine, phosphatidyl glyceride, phosphatidyl choline, and phosphatidyl inositol, but the phosphatidyl glyceride was not detected in the free lipid. The highest content in the total lipid quantity was 3.3 % of phosophatidyl choline in the case of the free lipid, while 14.9 % of the phosphatidyl ethanolamine contained in the bound lipid was the highest. All other constituents of phospholipid were contained in larger quantity in the bound lipid than in the free lipid. 3. The fatty acid composition of glycolipid in the free and bound lipids was also the same as that of the total free and bound lipids. The differences were that the content of palmitic acid was higher in the glycolipid of the free lipid than in the total free lipid and the content of linoleic acid was lower in the glycolipid.

Effects of Castration on Expression of Lipid Metabolism Genes in the Liver of Korean Cattle

  • Castration induces the accumulation of body fat and deposition of intramuscular fat in Korean cattle, resulting in improved beef quality. However, little is known about the metabolic adaptations in the liver following castration. To understand changes in lipid metabolism following castration, hepatic expression levels of lipid metabolism genes were compared between Korean bulls and steers. Steers had higher (p<0.001) hepatic lipids contents and higher (p<0.01) mRNA levels of lipogenic acetyl-CoA carboxylase. This differential gene expression may, in part, contribute to increased hepatic lipid content following the castration of bulls. However, we found no differences in the hepatic expression levels of genes related to triglyceride synthesis (mitochondrial glycerol-3-phosphate acyltransferase, diacylglycerol O-acyltransferase 1 and 2) and fatty acid (FA) oxidation (carnitine palmitoyltransferase 1A, C-4 to C-12 straight chain acyl-CoA dehydrogenase, very long chain acyl-CoA dehydrogenase) between bulls and steers. No differences in gene expression for very-low-density lipoprotein (VLDL) secretion, including apolipoprotein B mRNA and microsomal triglyceride transfer protein (MTTP) protein, were observed in the liver although MTTP mRNA levels were higher in steers compared to bulls. In conclusion, FA synthesis may contribute to increased hepatic lipid deposition in steers following castration. However, hepatic lipid metabolism, including triglyceride synthesis, FA oxidation, and VLDL secretion, was not significantly altered by castration. Our results suggest that hepatic lipid metabolism does not significantly contribute to increased body fat deposition in steers following castration.

항암제 Ara - C유도체의 합성에 관한 연구

  • Thioglycerol과 Glycerol을 출발물질로 1번 탄소(C) 위치의 산소(O),또는 황(S)에 octadecyl기로 alkylation한 후, 2번 탄소(C)위치 산소(O)를 palmitoyl기로acetylation시켜 만든 thioether lipid 및 ether lipid유도체를 인산화시킨 rac-1-S-octa-decyl-2-O-palmitoyl-1-S-thioglycerol-3-phosphate (DL-PTBA-P)와 rac-1-O-octadecyl-2-O-palmitoylglycerol-3-phosphate (DL-PBA-P)등을 합성하였다. 이들 thioether lipid 및 ether lipid유도체는 그 자체로도 in vitro와 in vivo에서anti-neoplastic property를 가지고 있는데, 이들 중간체를 핵산 화합물로써 역시 강한 항암 작용에도 불구하고 독성등 부작용이 문제가 되고있는 Ara-C (일명 Cytarabine)의 인산화물인 ara-CMP morpholidate와 conjugation시켜 최종물질로서 ara-CDP-DL-PCA, ara-CDP-DL-PBA, ara-CDP-DL-PTCA 및 ara-CDP-DL-PTBA등을 합성하였다. 이들 중 ara-CDP-DL-PTBA가 본 과제와 관련하여 항암제로서 최종 개발하고자하는 주요 물질인 Cytoros이다.

Effect of Triterpenoidal Glycosides of Dammarane Series and Their Aglycones on Phase Transitions of Dipalmitoylphosphatidylcholine (DPPC의 상전이에 미치는 Dammarane Series의 Triterpenoidal Glycoside와 그 Aglycone의 영향)

  • The effect of ginseng glycosides and their aglycones on the thermodynamic characteristics of membranes from dipalmitoylphosphatidylcholine (DPPC) was investigated. Total saponins (TS) from Korean red ginseng, Panax ginseng C.A. Meyer, interacted with the Eel Phase of lipid in the Polar region and did not penetrate the deeper glycerol backbone of lipid molecule. From the all investigated components of TS (aglycons and ginsenosides), only 20-(S)-panaxadiol (PD) had an effect similar to TS. High concentration of TS penetrated in hydrophobic Cl-C8 region. The presence of cholesterol did not influence the interaction of TS with DPPC. An elimination of transition, however, took place at 10~100 $\mu\textrm{g}$/ml of TS. DPPC had a low ability to interact with cholesterol (CHL) as compared with other lecithins except ethanolamine. From our results, only TS and PD, at high concentrations (100 mol%), influenced the phase transition of mixture of DPPC:CHL.

  • Seven diterpenes, four polyacetylenes, a lipid glycerol, and two sterols were isolated from the methylene chloride fraction of the root of Aralia cordata. Their chemical structures were determined as (-)-pimara-8(14), 15-dien-19-oic acid (2), pimaric acid (3), (-)-kaur-16-en-19-oic acid (4), 17-hydroxy-ent-kaur-15-en-19-oic acid (9), $7{\alpha}$-hydroxy-(-)-pimara-8(14), 15-dien-19-oic acid (10), $16\alpha$, 17 -dihydroxy-(-)-kauran-19-oic acid (11), 16-hydroxy-17-isovaleroyloxy-ent-kauran-19­oic acid (12), falcarindiol (5), dehydrofalcarindiol (6), dehydrofalcarindiol-8-acetate (7), falcarin­diol-8-acetate (8), alpha-mono palmitin (13), stigmasterol (1), and daucosterol (14) by the spectral evidences. These compounds were tested with COX-1 and COX-2 inhibition assays. This study found that compounds 2, 4, 5, 6, 7, 8, and 10 inhibited COX-1 dependent conversion of the exogenous arachidonic acid to $PGE_2$ in a dose-dependent manner with $IC_{50}$ values of $134.2{\mu}M$, $121.6{\mu}M$, $170{\mu}M$, $50.4{\mu}M$, $11.7{\mu}M$, $99.6{\mu}M$, and $69.6{\mu}M$, respectively. But, most of these compounds weakly inhibited COX-2 dependent $PGE_2$ generation. Among them, only compound 4 showed relatively significant inhibitory activity $(IC_{50}\;:\;127.6{\mu}M)$.

Reduction of Body Weight by Capsaicin is Associated with Inhibition of Glycerol-3-Phosphate Dehydrogenase Activity and Stimulation of Uncoupling Protein 2 mRNA Expression in Diet-induced Obese Rats

  • Capsaicin is a pungent component of red pepper, which is widely consumed as food adjuncts. The present study was performed to investigate anti-obesity effects of capsaicin in diet-induced obese rats. Male Sprague-Dawley rats (n=14) were fed with a high-fat diet (Control) or high-fat diet containing 0.016% capsaicin (w/w) (Capsaicin) for 8 weeks. The final body weight and the mass of white adipose tissue were significantly lower in capsaicin supplemented group compared to control. Dietary capsaicin ameliorated lipid profiles with decrease in the plasma concentrations of triglycerides and total cholesterol, and decrease in the levels of total lipids and triglycerides in the liver. Activity of glycerol-3-phosphate dehydrogenase (GPDH), an indicator of triglyceride biosynthesis in white adipose tissue, decreased by 35% in the group supplemented with capsaicin. However, consumption of capsaicin increased the expression of uncoupling protein 2 (UCP2) in white adipose tissue, which is related to energy consumption. Our data suggests that capsaicin may reduce body weight and fat accumulation in high fat diet-induced obese rats. These effects may be mediated, at least partially, by the upregulation of UCP2 gene expression and its ability to inhibit GPDH activity.

Fumigaclavine C attenuates adipogenesis in 3T3-L1 adipocytes and ameliorates lipid accumulation in high-fat diet-induced obese mice

  • Fumigaclavine C (FC), an active indole alkaloid, is obtained from endophytic Aspergillus terreus (strain No. FC118) by the root of Rhizophora stylosa (Rhizophoraceae). This study is designed to evaluate whether FC has anti-adipogenic effects in 3T3-L1 adipocytes and whether it ameliorates lipid accumulation in high-fat diet (HFD)-induced obese mice. FC notably increased the levels of glycerol in the culture supernatants and markedly reduced lipid accumulation in 3T3-L1 adipocytes. FC differentially inhibited the expressions of adipogenesis-related genes, including the peroxisome proliferator-activated receptor proteins, CCAAT/enhancer-binding proteins, and sterol regulatory element-binding proteins. FC markedly reduced the expressions of lipid synthesis-related genes, such as the fatty acid binding protein, lipoprotein lipase, and fatty acid synthase. Furthermore, FC significantly increased the expressions of lipolysis-related genes, such as the hormone-sensitive lipase, Aquaporin-7, and adipose triglyceride lipase. In HFD-induced obese mice, intraperitoneal injections of FC decreased both the body weight and visceral adipose tissue weight. FC administration significantly reduced lipid accumulation. Moreover, FC could dose-dependently and differentially regulate the expressions of lipid metabolism-related transcription factors. All these data indicated that FC exhibited anti-obesity effects through modulating adipogenesis and lipolysis.
