토끼의 관절병증에 미치는 알긴산올리고당의 치료효과 (Effect of Alginic Acid on Experimentally Induced Arthropathy in Rabbit Model)

  • For the induction of arthropathy, 5% hydrogen peroxide($H_2O_2$) was injected for 5 weeks into the intraarticular space of the New Zealand white rabbits to damage articular cartilage. Alginic acid of low molecular weight (2%) made from macromolecular alginate treated with enzyme was administered into articular space at the dose of 5 mg/kg twice a week for 3 and 6 weeks using 1 ml syringe and 26 G needle. Saline was injected for the control. Tissues surrounding the articulation were obtained for the measurements of superoxide dismutase(SOD) activity as a major antioxidant enzyme and malondialdehyde (MDA) as a lipid peroxidation level. Histopathologic examination on the surface of articular cartilage was carried out. Data showed that injection of hydrogen peroxide for 5 weeks had led to the induction of free radical damage and of articular cartilage change as confirmed by microscopic observation. The application of hydrogen peroxide caused a gradual increase in the SODs and MDA. These patterns were similar after 3 and 6 weeks of alginate treatment. Furthermore, microscopic examinations revealed that hydrogen peroxide caused flaking, fibrillation, fissuring, denudation, and hypocellularity in the articular surfaces. In conclusion, lipid peroxidation was demonstrated in the articular cartilage by the administration of hydrogen peroxide in the rabbit model. This lipid peroxidation could be caused by oxygen free radicals. The histologic and enzymatic correlations on lipid peroxidation in the articulation have provided a better understanding of arthropathy. It is possible to take advantage of these findings to evaluate effective alginate dosage more efficiently.

노화과정(老化過程)의 흰쥐에서 성심산(醒心散)이 심장(心臟)의 대사효소계(代謝酵素系)에 미치는 영향(影響) (A Study on the Effects of Sungshimsan on the Heart Lipid Peroxide and Metabolic Enzyme System in Senescence Induced Rats)

  • Aging occurs as a part of maturation as the time progresses which manifests in the human body causing morphological and functional degeneration, eventually leading to death. This experimental study was conducted to investigate a herbal formula to fortify the heart with easy clinical applications. Sungshimsan was chosen to study its effects in heart lipid peroxide and metabolic enzyme system in senescence induced rats. After pre-treatment of Sungshimsan for 2 weeks at the dosage of A (100mg/kg), B (250mg/kg), C (350mg/kg), and D (500mg/kg), a lipid peroxide and metabolic enzyme system changes of the heart were meaured in 32 weeks old rats. The following results were obtained in this study: 1. The contents of lipid peroxide was significantly reduced in the experimental groups treated with greater than 2 weeks at 250mg/kg. 2. The enzymatic activity of cytochrome P-450, cytochrome b5, and NADPH-cytochrome P450 reductase were significantly decreased in the 250mg/kg, 350mg/kg, and 500mg/kg experimental groups. 3. The activity of glutathione and glutathione S-transferase were significantly increased in the 250mg/kg, 350mg/kg, and 500mg/kg experimental groups. 4. The activity of glutathione reductase and glutathione peroxidase were not influenced compared to the control group. 5. The activity of ${\gamma}$-glutamylcystein synthetase was significantly increased in the 250mg/kg, 350mg/kg, and 500mg/kg experimental groups. 6. The activity of enzymes detoxificatioon superoxide dismutase and catalase were not influenced compared to the control group. Summarizing above results suggest that the Sungshimsan has profound effects in the heart lipid peroxide, free radicals, and delaying the heart aging process. Further clinical researches and application can be anticipated on the topic of senility and gerontology.

동충하초(冬蟲夏草)가 Hydrocortisone을 투여한 흰쥐의 Nitric Oxide Synthase 활성 및 Testosterone 함량에 미치는 영향 (Effects of Cordyceps militalis on the penile nitric oxide synthase activity and the level of blood testosterone in hydrocortisone acetate-treated rats)

  • The fallowing are the results of the experimental studies of Cordyceps militaris (CM) on the penile nitric oxide synthase (NOS) activity and the level of blood testosterone in hydrocortisone acetate-treated rats. CM was tested for the effects on activity of xanthine oxidase and lipid peroxidation in penis of hydrocortisone acetate-treated rats. In vitro, CM didn't effect the levels of lipid peroxide and the activity of NOS. In the penis of hydrocortisone acetate-treated rats, lipid peroxide, the activities and ratio of type conversion of xanthine oxidase were increased but activity of NOS and content of nitrite were decreased. In vivo, after administration of CM to hydrocortisone acetate-treated rats, levels of lipid peroxide in penis was decreased. Also, the activities and ratio of type conversion of xanthine oxidase were decreased, too. The body weight and concentration of testosterone in the blood were increased. The effects of Cordyceps militalis Broth did better than the effects of Cordyceps militalls Mycelia, These results suggest that CM decrease the activities of free radical generating enzymes such as xanthine oxidase which form lipid peroxide and increase the penile NOS activity and the level of blood testosterone in hydrocortisone acetate-treated rats. Conclusively, CM is capable of improving of sexual ability in hydrocortisone acetate-treated rats.

Effects of Polyacetylene Compounds from Panax Ginseng C.A. Meyer on $CCl_4$-Induced Lipid Peroxidation in Mouse Liver

  • The inhibitory effect of three polyacetylene compounds, panaxydol, panaxynol and panaxytriol isolated from Panax ginseng C.A. Meyer on $CCl_4$induced lipid peroxidation in vivo and in vitro hepatic microsomal lipid peroxidation induced by ADP-$Fe^{3+}$, NADPH and NADPH-cytochrome P-450 reductase were investigated. Their effects on lowering the lipid peroxide levels both in serum and liver and lowering the serum enzyme (GOT, GPT, LDH) activities without the $CCl_4$-induction were also determined. Male ICR mice were pretreated i.p. with polyacetylene compounds or DL-${\alpha}$-tocopherol before administration of $CCl_4$ i.p. and 20 hr after the administration of $CCl_4,$ serum and liver were analyzed. Hepatic microsome was isolated and used for the in vitro NADPH-dependent lipid peroxidation system. Except for panaxynol, treatment with polyacetylenes to control mice did not reduce the levels of lipid peroxides and serum enzyme activities. Panaxynol itself inhibited lipid peroxidation in the liver of normal mice. Polyacetylene compounds protected from the $CCl_4$-induced hepatic lipid peroxidation and lowered serum lipid peroxide levels. Polyacetylenes also inhibited the in virto hepatic microsomal lipid peroxidation in a dose-dependent manner. The results suggest that panaxydol, panaxynol and panaxytriol seem to be the antioxidant components which contribute the anti-aging activities of Panax ginseng C.A. Meyer.

  • The fruits of Cornus officinalis have been used in traditional Oriental medicine for treatment of inner ear diseases, such as tinnitus and hearing loss. In the present study, we showed that the ursolic acid obtained from Corni fructus protected HEI-OC1 auditory cells from hydrogen peroxide cytotoxicity in a dose-dependent fashion. In addition, to investigate the protection mechanism of ursolic acid on hydrogen peroxide cytotoxicity toward HEI-OC1, we measured the effects of ursolic acid on lipid peroxidation and activities of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPX) in hydrogen peroxide treated cells. Ursolic acid (0.05 - 2 ${\mu}g/ml$) had protective effect against the hydrogen peroxide-induced HEI-OC1 cell damage and reduced lipid peroxidation in a dose-dependent manner. Pre-treatment with ursolic acid significantly attenuated the decrease in activities of CAT and GPX, but SOD activity was not affected by the ursolic acid or hydrogen peroxide. These results indicate that ursolic acid protects hydrogen peroxide-induced HEI-OC1 cell damage through inhibition of lipid peroxidation and induce the antioxidant enzymes CAT and GPX.

꽁치 자연동결건조(과메기)중 지방함량과 지방산 조성 변화 (The Change in Content of Constitutive Lipid and Fatty Acid of Pacific Saury during Natural Freezing Dry(Kwa Mae Kee))

  • The results which were examined changes In contents of constitutive lipid and fatty acid during the natural drying of Pacific Saury were as follows. During the natural drying of Pacific Saury, offal lipid and moisture contents gradually decreased. In the survey of rancidity degree extracted lipid of Pacific Saury, peroxide value was gradually Increased, TBA and acid value were radically increased thereafter 9 days of drying. Neutral lipid, phospholipid and glycolipid contents of total lipid were 95%, 4% and 2%, respectively, and in the neutral lipid, content of TG was more than 90%, and in the phospholipid, contents of PC and PE were 45∼85%, 27∼34%, respectively. Fatty acid composition of total lipid was substantially higher in C18:0 and C20:1, C22:1, C22:6 contents and fatty acid composition of neutral lipid was higher In C22:1, C22:1, contents and fatty acid composition of phospholipid was the highest in C22:6 content. In the fatty acid composition of glycolipid, monounsaturated fatty acid content has substantially higher, C16:0 content was gradually Increased In saturated fatty acid and C22:1 content was gradually decreased in monounsaturated fatty acid. It showed that fatty acid composition of triglyceride has higher in monounsaturated fatty acid content and contents of C20:1, C22:1 was gradually decreased.

Metallothionein이 지질과산화반응과 Aldehyde Oxidase활성에 미치는 영향 (Effect of Metallothionein on the Lipid Peroxidation and Aldehyde Oxidase Activity)

  • The effects of metallothionein and cadmium ion on the hepatic aldehyde oxidase activity and brain lipid peroxidation were tested in vitro. The content of brain lipid peroxide at the condition of normal or $300{\mu}M$ Fe(II)-induced was remarkably reduced by the addition of metallothionein in the incubation mixture. The induced content of lipid peroxide by cadmium $(30{\mu}g/ml)$ ion was reduced by metallothionein $(100{\mu}g/ml)$. The activity of aldehyde oxidase was not affected by metallothionein, but cadmium ion $(8.38{\mu}g/ml)$ increased the activity of aldehyde oxidase about 80% compared to the control. The cadmium-induced activity of aldehyde oxidase was restored to the control level by metallothionein or penicillamine.

Schisandra Chinensis Inhibits Oxidative DNA Damage and Lipid Peroxidation Via Antioxidant Activity

  • Schisandra chinensis have been traditionally used in Asia for the treatment of dyspnea, cough, mouth dryness, spontaneous diaphoresis, nocturnal diaphoresis, nocturnal emission, dysentery, insomnia and amnesia. The purpose of this study is to evaluate the protective effects of Schisandra chinensis on oxidative DNA damage and lipid peroxidation induced by ROS in non cellular and cellular system. DPPH radical, hydroxyl radical and hydrogen peroxide scavenging assay were used to measure the antioxidant activities. Phi X-174RF I plasmid DNA cleavage assay and intracellular DNA migration assay were used to evaluate the protective effect on oxidative DNA damage. MTT assay and lipid peroxidation assay were used for evaluating the protective effect on oxidative cell damage. It was found to scavenge DPPH radical, hydrogen peroxide and hydroxyl radical and it inhibited oxidative DNA damage, lipid peroxidation and cell death induced by hydroxyl radical. These data indicate that Schisandra chinensis possesses a spectrum of antioxidant and DNA-protective properties

흰쥐의 간(肝) 조직(組織)에서 녹용(鹿茸) 약침(藥針) 제제(製劑)의 항산화작용(抗酸化作用)에 관(關)한 연구(硏究) (Effects of Cervus elaphus for herb-acupuncture solution on Antioxidation in Rat‘s liver)

  • Cervus elaphus for herb-acupuncture solution(CEHAS) was tested for the effects of free radical generating enzyme and lipid peroxidation in rat's liver. In vitro, levels of lipid peroxide in tissues of liver were proportionally decreased to concentration of CEHAS. They were much more decreased. when lipid peroxidation was induced with ferrous iron $(Fe^{-2})$. Also, enzyme activities of xanthine oxidase were decreased. The ratio of type conversion of xanthine oxidase was lowered, too. But, here was not special changes on enzyme activities of aldehyde oxidase. These results suggest that CEHAS decrease the activities of free radical generating enzymes such as xanthine oxidase which form lipid peroxide.

Hesperidin 과 Naringin 이 흰쥐의 항산화능에 미치는 영향 (Effects of Hesperidin and Naringin on Antioxidative Capacity in the Rat)

  • This study was performed to investigate effects of hesperidin and naringin on linpid peroxide formation and antioxidative enzyme activities in rats. Thiobarbituric acid reactive substance (TBARS) concentrations were measured in plasma and liver. Catalase, superoxide dismutase, and glutathione peroxidase activities were measured in erythrocyte and liver. Forty-nine male Sprague-Dauley rats weighing 275.3$\pm$3.3g were blocked into seven groups according to body weight and were raised fro four weeks on diets containing 0.25, 0.50 or 1.00%(w/w) hesperidin or naringin . Food intake, weight gain , food efficiency ratio, and weights of liver, kidney, spleen ,and epididymal fat pad were not significantly different among groups. In 0.50 and 1.00% naringin groups , plasma TBARS concentrations were significantly decreased with a dose response patter. In 0.25, 0.50 and 1.00% hesperidin groups, liver TBARS concentrations were significantly decreased without a dose dependent patter. Antiosidative enzyme activities in erythrocyte and liver were not significantly affected by type and amountof dietary bioflavonoid, but in the 1.00% hesperidin group, catalase, superoxide dismutase, and glutahione perosidase activities in linver showed a tendency to increase. In conclusion, naringin inhibited lipid peroxide formation with a dose response pattern in plasma without changing the activities of antioxidative enzymes. Hesperidin adminstration, regardless of the level in the diet, inhibited lipid peroxide formation in liver.
