Comparative studies of the five edible mountain vegetables by DNA marker fingerprinting (DNA marker 지문법에 의한 취나물 5종 (청옥취 , 개미취 , 참취 , 수리취 , 곰취)의 비교연구)

  • Five edible mountain vegetables(Saussurea sp. Aster tataricus, A. scaber. Synurus deltoides, Ligularia fischeri) were investigated on the basis of amplified DNA polymorphisms resulted from PCR (polymerase chain reaction) analysis. The sampled plants consisted of 38 individuals in 5 taxa. Only 10 primers out of 62 primers (60 random [10-mer] primers, two 15-mer [M13 core sequence, and (GGAT) sequence]) tested gave rise to polymorphisms in all of the tested plants, producing 176 DAN fragments amplified. Intraspecific polymorphisms found in each taxa showed intraspecies constancy (31.1-61.1%) in the banding patterns of individual plants: Saussurea sp. 31.1%, 15 bands, Aster tataricus, 40.9%, 18 bands, A. scaber. 38.5%, 15 bands. Synurus deltoides, 34.7%, 17 bands, and Ligularia fischeri, 61.1%, 22 brands, respectively. All five species were well classified from each other at the 0.93 level of similarity index value. Intraspecific and interspecific variations were appeared at the levels ranging from 0.62 to 0.99. Based on these results, our PCR analyses support the previous data derived from external morphology of the 5 edible mountain vegetables, but very low levels o intraspecific variations were detected in all of these taxa.

Community Classification and Distribution of the Vascular Plants in the Daemisaem Region (데미샘 주변의 식물 분포와 군락 분류)

  • The vascular plants of the study area in the Daemisaem region were listed as 302 taxa; 81 families, 196 genera, 251 species, 1 subspecies, 42 varieties, 7 forms and 1 hybrid. Based on the list of Korean endemic plants, 7 taxa were recorded; Carex okamotoi, Salix purpurea var. japonica, Thalictrum uchiyamai, Lespedeza ${\times}$ tomentella, Weigela subsessilis, Aster koraiensis, Cirsium chanroenicum. Specific plants by floral region were total 21 taxa; Silene jenisseensis, Cimicifuga heracleifolia in class IV, 3 taxa (Lilium leichtlinii var. tigrinum, Betula davurica, Lonicera subhispida) in class III, 3 taxa (Viola tokubuchiana var. takedana, Ligularia fischeri, Cirsium chanroenicum) in class II, 13 taxa (Glyceria leptolepis, Meliosma oldhamii, Fraxinus mandshurica, etc.) in class I. The naturalized plants in this site were 8 families, 20 genera, 23 species, 1 varieties, 24 taxa and naturalization rate was 7.9% of all 302 taxa vascular plants. The vegetation community of the selected Daemisaem region were growth of configuration to slope, ridge and valley; Quercus variabilis community, Quercus serrata community, Quercus mongolica community, Salix koreensis community in the slope, Quercus mongolica community, Carpinus tschonoskii community in the ridge, Fraxinus mandshurica community, Carpinus laxiflora community, Acer mono community, Cornus controversa community, Morus bombycis community in the valley.

Vascular Plants of Distribution and Characteristics of the Mt. Bangjang (방장산일대의 관속식물 분포 특성)

  • The vascular plants of the studied area in the Mt. Bangjang was listed 422 taxa : 106 families, 299 genera, 372 species, 1 subspecies, 46 varieties and 3 forms. Based on the list of the rare plants by the Forest Research Institute, 5 taxa were recorded in the studied areas; Lilium distichum(Preservation priority order : No. 159), Tricyrtis dilatata (No. 97), Aristolochia contorta (No. 151), Prunus yedoensis (No. 110), Patrinia saniculaefolia (No. 122) and based on the list of Korean endemic plants, 9 taxa were recorded; Cephalotaxus koreana, Carex okamotoi, Populus tomentiglandulosa, Salix purpurea var. japonica, Prunus yedoensis, Forsythia koreana, Paulownia coreana, Weigela subsessilis, Patrinia saniculaefolia. Specific plant species by floral region were total 29 taxa; Prunus yedoensis in class V, 3 taxa(Acer palmatum, Asperula lasiantha, Lonicera subhispida ) in class III, 5 taxa(Lilium distichum, Potentilla dickinsii, Caryopteris incana, Patrinia saniculaefolia, Ligularia fischeri) in class II, 20 taxa(Ulmus parvifolia, Clematis patens, Aconitum jaluense, Ilex macropoda, Grewia biloba var. parviflora etc.) in class I. The naturalized plants in this site were 13 families, 31 genera, 36 species, 2 varieties, 38 taxa and naturalization rate was 9.0% of all 422 taxa vascular plants. Because of recent rapid increase of cars and people around here, analysis shows many naturalized plants and high naturalized ratio. And wild plants disturbing ecosystem like Ambrosia artemisiifolia var. elatior have been increasing. So, it needs continuing control and conservation measures on the plant ecosystem.

Plant Diversity and Conservation in Oruem of Jeju City (제주시 일대 오름의 식물다양성과 보전방안)

  • Vascular plants of Oruem in region of Jeju-City were investigated 454 taxa totally; 116 families, 301 genera, 359 species, 3 subspecies, 78 varieties and 14 forms. In the studied Oruems in region of Jeju-City within altitude 800m, evergreen tree forests does not range and a lot of Pines thunbergii forest and Cryptomeria japonica and Chamaecyparis obtusa plantation forest were. The endemic species of Oruem in region of Jeju-City were 14 taxa such as Asarum maculatum, Hepatica insularis, Cardamine glechomifolia etc. The floristics degree categorized by the specific distribution of plant species were total 116 Taxa, that is, V grade species Magnolia kobus, Aconitum japonicum subsp. napiforme, Ligularia taquetii etc, 6 kinds be, IV grade species Rubus ribisoideus, Ligustrum salicinum, Carex macrandrolepis etc, 16 kinds be, III grade species Illicium anisatum, Hydrangea petiolaris, Rubus schizostylus etc, 37 kinds be, II grade species Pseudostellaria coreana, potentilla dickinsii, Euonymus pauciflorus etc, 6 kinds and I grade species Sphenomeris chinensis, Pteris multifida, Machilus thunbergii etc, be. 51 kinds were confirmed. Rare and Endemic Plant Species of Korea Forest Service were 11 taxa such as Asarum maculatum, Aconitum japonicum subsp. napiforme and Magnolia kobus etc. Naturalized plant were confirmed 31 taxons with Rumex acetosella, Hypochaeris radicata and Sisyrinchium atlanticum etc.

Floral Characteristics of Asteraceae Flowers and Insect Pollinators in Korea (우리나라 국화과 식물의 화기구조와 방화 곤충 연구)

  • To search for the co-relationships between insect-pollinators and the plant species of Asteraceae, insects visiting in flower and the flowers in Korea, were studied from April 2010 to October 2011. The sum of flower visiting degrees are shown 38 in Lepidoptera, 38 in Diptera, 36 in Hymenoptera, and the lowest 6 in Coleoptera, respectively. 65 insect species are identified pollinators, Hymenoptera 13 species(Apidae 11 sp., Formicidae 2 sp.), Lepidoptera 29 species(Pieridae 5sp., Nymphalidae 12 sp., Satyridae 3 sp., Hesperirdae 3 sp., Lycaenidae 2 sp., Danaidae 1 sp., Moth 2 sp.), Diptera 16 species(Tachinidae 1 sp., Syrphidae 12 sp., Muscidae 1sp., Others 2 sp.), and Coleoptera 6 species(Cetoniidae 1 sp., Cermbycidae 3 sp., Chrysomelidae 1 sp., Mordellidae 1 sp.). 31 pollinator species visits the flower of $Erigeron$ $annuus$, next 15 pollinator species does the flower of $Eupatorium$ $japonicum$, and then 13 pollinator species does the flower of $Aster$ $ageratoides$. Only 2 pollinator species visit the flower of $Tephroseris$ $kirilowii$, $Ixeridium$ $dentatum$, $Inula$ $britannica$ var. $japonica$, $Carduus$ $crispus$, $Ligularia$ $fischeri$, $Ainsliaea$ $acerifolia$, $Synurus$ $deltoides$, $Cirsium$ $setidens$, $Crepidiastrum$ $enticulatum$ and $Dendranthema$ $boreale$. Pollinators of Lepidoptera visit more frequently white flower than yellow or purple one. This study found out that mutualisic relations between plants and insect pollinators is carried out in Korea.

A Comparative Study on the Habitat of Abies koreana WILSON between Mt. Jiri and Mt. Ha1la

  • This examination of vegetation is conducted from lune to Oct. 1999 through on the spot survey, centering on the habitat of Abies koreana around Imgeolryeong, langteomok, and Jeseokbong in Mt Jiri and Yeongsil path and Seongpanak path in Mt. Halla. Species composition tables of the Abies koreana community by altitude and slope in Mt. Jiri and Mt. Halla are made based on the community composition tables examined in quadrat. The Abies koreana-Saso quelpaertensis community in a west slope of Mt. Halla is found that Abies koreana of 4-6m in height forms subtree layer and that of 8-9m in height the tree layer and it reflects a difference between community structure by slope and main composition species. While the tree layer of Abies koreana community is 12-14 in height around the area of 1, 290-1, 560m above the sea of Imgeolryong and path from Baekmudong to Jangteomok, it is 8-12m in height in the area of 1, 680-1, 780m above the sea of path from Jangteomok to Jeseokbong. It means that community structure depends on area and attitude. This study finds out that commonly appearing composition species similar to coverage and presence are Lepisorus thunberianus, Quercus mongolica, Soidago virga-aurea var. asiatica, Ligularia fischeri, Carex lanceolata, Clintonia udensis, Magnolia sieboldii, Betula ermani, Acer Pseudo-sieboldianum, As soil environment of surveyed area, pH of surveyed spots is similar in Mt. Jiri and Mt. Halla, electrical conductvity is higher in Mt Jiri, and contents of organic matter is relatively higher in a west slope of Mt. Jiri and Mt. Halla and lower in their south and west slopes. Contents of organic matter and total nitrogen show that the area of 1, 550m above the sea in a west slope of Mt. Halla is some higher. For pH and contents of total nitrogen and organic matter, Mt. Halla is higher than Mt. Jiri but for electrical conductivity , Mt. Jiri is relatively higher than Mt. Halla.

Effect of gomchwi (Ligularia fischeri) extract against high glucose- and H2O2-induced oxidative stress in PC12 cells (PC12 신경세포에서 고당 및 과산화수소로 유도된 산화적 스트레스에 대한 곰취 추출물의 효과)

  • Effects of the ethyl acetate fraction from gomchwi (Ligularia fischeri) extract against high $glucose/H_2O_2-induced$ oxidative stress and in vitro neurodegeneration were investigated to confirm the physiological property of the extract. The ethyl acetate fraction of gomchwi extract showed the highest total phenolic contents than the other solvent fractions. An anti-hyperglycemic activity of the ethyl acetate fraction was evaluated using the ${\alpha}-glucosidase$ inhibitory assay, and the half maximal inhibitory concentration ($IC_{50}$) value for ${\alpha}-glucosidase$ was found to be $727.64{\mu}g/mL$. In addition, the ethyl acetate fraction showed excellent 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) diammonium salt radical scavenging activity, and inhibition of malondialdehyde production. The ethyl acetate fraction also decreased intracellular reactive oxygen species, whereas neuronal cell viability against high glucose/$H_2O_2$-induced cytotoxicity was found to be increased. Finally, 3,5-dicaffeoylquinic acid as a main phenolic compound in the ethyl acetate fraction was analyzed by high-performance liquid chromatography. These results suggest that gomchwi might be a good natural source of functional materials to prevent diabetic neurodegeneration.

The Vegetation of Seoimal-Lighthouse area in koje Island (거제 서이말등대 주변의 식생)

  • The vegetation of Seoimal-lighthouse area was investigated from August, 2001 to July, 2002. In order to analyze the vegetation of this area, synthesis table, actual vegetation map were prepared. The main community structures of this area represent evergreen broad-leaf community(about 48%), which are very good condition(nearly last sere) to preserve and 11communities(Camellia japonica community, Castanopsis cuspidnta var. thunbergii community, Neolitsea sericea community, Quercus serrata community, Quercus variabilis community, Carpinus corenna community, Srtrax japonica community, Pinus thunbergii community, Quercus acutissima community, Zelkova serrata community, Cinnamimum japonicum community) were classified by vegetation type. And Daphne kiusiana, Asirum maculatum, Cymbidium goeringii, Chloranthus glaber, Ligularia taguetii should be worthwhile species to conserve in this area.

Intersubgeneric Protoplast Fusion of Pelargonium aridum (Ligularia) and P.zonale(Ciconium) (Pelargonium aridum과 P. zonale 아속간의 원형질체융합)

  • In an attempt to obtain intersubgeneric somatic hybrids of Pelargonium aridum and P.zonale, protoplast isolated from the two species were fused by using polyethylene glycol(PEG) and electorfusion methods. Protoplast were isolated from cotyledon and leaf tissues using MS medium containing 550 mM sucrose, 0.7% cellulase (Onozuka R-10) and 0.4% Macerozyme. The optimum number of protoplasts per mL of culture medium was 6 x 10$^4$. Protoplast fused by the electrofusion method were more active than by PEG method. Heterokaryotically fused protoplasts formed calli when cultured in MS medium containing 550 mM glucose, 1 to 2 mg/L NAA and 0.5 to 1 mg/L BA.
