Chemical Composition and Size of Floating and Sunken Eggs of Olive Flounder Paralichthys olivaceus

  • Olive flounder (Paralichthys olivaceus) is one of the most commercially important species of farmed marine fish in Korea. Although techniques for rearing the larva of this species are improving, production costs are also increasing due to microbial influences and genetic degradation caused by successive culturing. Since the demand for healthy seed, which requires eggs of good quality of, is high, we examined the effects of nutrient composition on the size of P. olivaceus eggs. We analyzed floating (live) and sunken (dead) eggs of P. olivaceus from five different hatcheries for their size and amino and fatty acid composition. The sizes of eggs and oil globules from floating vs. sunken eggs were significantly different at p<0.05. No significant relationships were observed, however, among larval length, hatching percentage, and egg and oil globule size. The dry weight and amino acid levels of floating eggs were greater than those of sunken eggs (p<0.05), but no difference in fatty acid content was observed.

Growth of Larval Sweetfish Plecoglossus altivelis Fed Moina macrocopa (물벼룩 Moina macrocopa 급이가 은어 Plecoglossus altivelis 자어의 성장에 미치는 영향)

  • The effect of live food and commercial diet on sweetfish Plecoglossus altivelis culture was assessed by determining the daily growth rate and feed conversion ratio. After feeding for 8 weeks, the Moina macrocopa diet resulted in a higher growth rate of 5.101 compared to commercial (3.441) and Artemia (2.729) diets. In addition fish fed the M. macrocopa diet had the weight gain, averaging 41.29 g at the end of the experiment. The content of n-3 highly unsaturated fatty acids (HUFA) was highest in the commercial diet, while free fatty acids were not detected in the M. macrocopa diet. Triglycerides, the energy source in fishes, constituted 40.0% of the M. macrocopa diet, which was significantly higher than in the commercial and Artemia diets at 20.0 and 12.0%, respectively These results indicate that freshwater M. macrocopa can be used as feed for sweetfish larvae, even without docosahexaenoic acid (DHA) enrichment from Artemia.

Larval Morphology of Gnathagnus elongatus (Uranoscopidae) (푸렁통구멍 Gnathagnus elongatus 자치어의 형태 발달)

  • A total of 46 Gnathagnus elongatus larvae, 2.7~8.7 mm BL (Body length), were collected by bongo net and trawl-shaped fish larvae net in the coastal area of Cheju Island during 1997~1999. This species was found in the coastal area of Cheju Island and the Korea Strait. This paper describes morphological features of these specimens throughout their development. 1) The soft ray of fin appeared at about 5 mm BL and attained to a fixed number at about 8.7 mm BL. 2) Pigments pattern changed suddenly about 3 mm BL. 3) A series of pigments in the upper and the lower part of the urostyle were found less than 4.3 mm BL. 4) The pigments of caudal peduncle were found more than 6 mm BL.

Ontogeny of the Digestive Organ during Early Life Stages of the Black Sea Bream, Acanthopagrus schlegeli(Teleostei: Sparidae) (감성돔(Acanthopagrus schiegeli)의 초기생활사 동안 소화기관 발달)

  • Differentiation and development of the digestive organ of the black sea bream, AcanHepagus schlegeli were studied by means of histological methods. The hatched lawn if TL(total length) $2.0 mm (n=10)$ had a yolk sac of $1,000{\times}590 {\mu}m$ and simple straight digestive tract, which was composed of cuboidal epithelium. In the pre-larval stage of TL $3.5 mm$, digestive tract could be distinguished into esophagus, stomach and intestine, and the exocrine glands were appeared in the pancreas. In this stage mucosal folds, eosinophilic granule cells and brush border were observed in the posterior intestine. Yolky materials were completely absorbed and the brush border was recognized in the free surface of anterior intestine in TL $3.7 mm$. In the stomach mucosal folds began to appear from TL $4.0 mm$. In this time the zymogen granules were recognized in the cytoplasm of pancreatic exocrine cells. In the post-larval stage ranged from $4.5 to 5.0 mm$ in TL, hepatic cords started to develop, and the mucous secretory cells of PAS positivewere observed at esophagus and intestine. In the post-larval stage ranged from $6.3 to 7.0 mm$ in TL, histological layer of esophagus and intestine could be distinguished into serous membrane, muscular layer, submucosal layer and mucosal layer. From over TL $9.0 mm$, stomach could be distinguished into cardiac, fundic and pyloric portion, and the gastric gland began to appear at mucosal fold of fundic stomach. In the juvenile stage ranged from $10.0 to 11.0 mm$ in TL, histological structures of esophagus and intestine were similar to those of adult. From over TL $15.0 mm$, histological structures of stomach were similar to those of adult. Structural and functional digestive organ of black sea bream was present from the juvenile stage ranged from $15.0 to 17.0 mm$ in TL.

Water Temperature and Food on Growth and Survival of Parrot Fish Larvae, Oplegnathus fasciatus (수온 및 먹이계열에 따른 돌돔, Oplegnathus fasciatus의 초기 성장과 생존율)

  • This study was to investigate the effects of water temperature and food on the mass seed production of larval parrot fish, Oplegnathus fasciatus. Growth of the larvae reared at heated water temperature ($25.3^{\circ}C$) was significantly higher than that of the larvae reared at natural water temperature ($20.5^{\circ}C$). In addition, survival rate of the larvae at heated and natural sea water temperature were $24.0\%$ and $12.3\%$, respectively (P<0.05). Growth and survival rate of the larvae fed mixed diets of Nannochloropsis oculata, rotifer (Brachionus rotundiformis). Artemia nauplii, Tigriopus japonicus and artificial diets were significantly higher than that of the larvae fed only either rotifer or artificial diets (P<0.05). The average survival rate and total length of the larvae reared for 50 days after hatching were $22.5\%$ and 62.0$\pm$4.0 mm, respectively. Amount of consumed rotifers at one time feeding by a larva was rapidly increased with growth from 10.3 individuals per larva 5 day old to 65.5 individuals per larva 20 day old.

Morphological Development of the Larvae and Juveniles of Halfbeak Fish, Hyporhampus sajori (Temminck et Schlegel) (학공치 Hyporhampus sajori (Temminck et Schlegel) 자치어(仔稚漁)의 형태 발달)

  • Observation of morphological changes during larval and juvenile stages of the halfbeak fish, Hyporhampus sajori (Temminck et Schlegel), was made based on samples of fertilized eggs collected on the shores of Youngil Bay, Phohang-shi, Korea, on May 27, 1991, and incubated in the laboratory. During the incubation period, water temperature fluctuated between $17.6^{\circ}C$ and $23.2^{\circ}C$. The newly hatched larvae were elongated and 7.0~8.50 mm in total length (TL) with 40~42+17~18 = 59~60 myotomes. Numerous melanophores were on the head, mid-dorsal part of the body, intestine, and the tail part of the notochord. Three days after hatching the larvae had attained 8.60~10.90 m in TL, had completely absorbed the yolk, and the caudal notochord was flexed $45^{\circ}$ upward. At this time, it was attained postlarvae stage. Thirty-seven days after hatching the postlarvae were 32.37~44.95 mm in TL and had reached the juvenile stage. All fins were formed with a complete set of fin rays with the following counts: dorsal fin rays 16~17; pectoral f in rays 6; ventral fin rays 12~14; caudal fin rays 11~12+11~12 = 22~24.

The infestation states and changing patterns of human infecting metacercariae in freshwater fish in Kyongang-do and Kyonggi-do, Korea (식이성 윤충류질환의 관리전략수립을 위한 감염원의 역학 및 병원체의 생물학적 특성에 관한 조사연구 - 경상도내 3개 강 및 경기도내 4개 하천에서 채집한 민물어류의 인체기생 흡충류 피낭유충 감염상 및 변동)

  • The infestation rates and abundances of human infecting metacercariae (Clonorchis sinensis, Metcgonimus spry. , Centrocestus crmatus, Echinostoma hortense, Echinochusnus japonicw, Clinostomum complanctum) in freshwater fish were investigated at the three river areas - Taewhagang (river), Hyongsangang (river), Nakdonggang (river) - in Kyongsang-do and at four streams - Yonpungchon, Munsanchon, Kyonganchon, and Konjiamchon- in Kyonggi-do, Korea in 1994-1995. The fish caught at Taewhagang were heavily infested with metacercariae of Clonorchis sinensis and Centrocestw armctus. At Hyongsangang, Zncco platypus and Z. temmincki were moderately infested with metacercariae of C. crmctus. Chomanpo, at the basin of Nakdonggang, was still endemic for C. sinensis. In the fish caught at four streams of Kyonggi-do, metacercariae of C. sinensis exhibited the highest infestation rate and intensity out of 6 species of metacercariae. The infestation intensity of C. sinensis metacercariae in fish flesh was markedly different according to each division of flesh. The cause of this difference was conjectured as a result of larval behavior. The metacercariae of C. omnt5 were found in almost all parts, except scales and fins, of fish. The infestation rates and intensities of C. sinensis and C. armntus metacercariae in Taewhagang greatly increased as compared with those of previous reports . RhinoBobius bmnneus and Aconthorhone5 macropterus are newly recorded intermediate hosts of Echinostonn hortense. The reason of large differences from previous data was discussed and the standard method of metacercaria examination was proposed.

Studies on Anisakis Type Larvae (Anisakis 형(型) 유충(幼蟲)에 관한 연구(硏究))

  • As it has been known recently that anisakis type larvae harbouring in marine fishes are a causal agent of zoonosis to human and probably to land living mammal animals, attention was focused on the study on the larvae in an aspect of epidemiology or epizootiology. The present work was conducted from 1966 to 1975 for i) survey on the harbouring status of anisakis type larvae in marine fishes of this country, ii) observation on the response to the experimental infestation of the larvae to the pigs, in the reason that they could well fetid raw fish viscera occasionally containing the larvae as a high protein source of swine food, and iii) observation on the larval resistance and response to vermicidal agents for the purpose of prevention of the larval infection to the mammal animals. The data obtained in the studies were summarized as follows: 1. In the survey on the status of larvae harbouring in main species of marine fishes of this country, 15 species, a total of 1,940 fishes, were observed and the result was summarized in table 2. Average number of larvae, in upper rank of 5 out of all 15 species of fishes, were as highest as 156 larvae ranging 74 to 450 in Pseudosciaena manchurica (chamjogi), 54.5 ranging 15 to 240 in Trichiurus haumela (kalchi), 35.6 ranging 8 to 112 in Trachurus japonica (junggengi), 30.6 ranging 4 to 65 in Parapristipama trilineatum (benjari) and 20.5 ranging 3 to 48 in Nibea argentata (boguchi) respectively. In morphological observation, size of the larvae in the fishes were varied, ranging from 2 to 32mm long, and a tendency to larger size and number of larvae in the fishes, which were wider sea migration, higher age and lager bodily size, was observed The favorite places harbouring the larvae in fishes were mainly around the intraperitoneal viscera such as mesentery, omentum, liver, pyloric suspensory, fat tissue and cloaca, and rarely in body muscles of fish. Fishes heartily infested with the larvae showed stunted growth decreased egg formation and severe damage of liver. 2. In the experimental infestation of the larvae to normal pigs, as illustrated in table 3, a group with large dose of larvae (a total of 1,800 larvae, 300 larvae Per dose, twice in a dart for 3 days) showed acute clinical syndrome terminatine death with a week course, whereas two groups with less dose of larvae (a total of 180~360 larvae, 10 larvae per dose, at 5 days interval for 70~180 days) showed subclinical syndrome with remarkably stunted growth as. much as approximately one half of body size in contest to the control pigs. In the pathological findings, a group with large dose of larvae showed macroscopically larvae penetrating to the gastric wall with severe gastroenteritis, and histopathologically various acute lesions caused by active larvae penetration into the wall of stomach and interstine, whereas two groups with less dose of larvae showed chronic lesions such as hypertrophy and verminous granulomatous swelling of gastric wall, suggesting strongly the possibility of natural infestation of larvae to swine. 3. In the resistance of the larvae to the chemical solutions, the larvae tolerated for 2 days in 15 percent solution of sodium chloride and acetic acid, and for 7 days in 70 percent solution of ethyl alcohol. In the resistance to the temperature, the larvae died within 1 second at $62^{\circ}C$ and tolerated for 24 hours at $-3^{\circ}C$, 12 hours $-5^{\circ}C$ respectively. 4. For the experiment on the vermicidal effect to larvae, general vermicidal drugs such as Neguvon, Combantrin, antimony Potassium, piperazine adipate and piperazine dihydrochloride, oxidizer such as potassium permanganate and potassium chlorate, and dyes such as gentian violet and crystal violet were used, and among them, as illustrated in table 6, potassium permanganate was proved as the best. In the successive test for the practical use of potassium permanganate, vermicidal effect in seawater solution of potassium permanganate and common-water solution of potassium permanganate were compared, and then retested by dipping the fish viscera including the larvae into the two different solutions of potassium permanganate. The result through these tests indicated that 0.01 percent common water and sea-water solution of potassium permanganate could be apparently recommended as a preventive vermicidal solution, having 90 to 100 percent vermicidal effect by dipping for 12 to 24 hours even though sea-water solution of potassium permanganate had a tendency to slightly less effect than the common-water solution of potassium permanganate (Table 8).

Development of the Digestive Tract in Tiger Puffer (Takifugu rubripes) (자주복(Takifugu rubripes)의 소화관 발달)

  • Anatomical and histological observations were used to examine the morphological differentiation and development of the digestive tract during ontogenesis in tiger puffer, Taklyugu rubripes. Before hatching, the digestive tract was located between yolk sac and notochord of embryo. Newly hatched larvae had a straight tubular digestive tract. The larval mouth opened at 2 days after hatching (DAH). At 4 DAH, sphincters separated the digestive tract into the esophagus, anterior intestine, mid-intestine and rectum, and the anus was opened. At 5 DAH, the larval intestine bent between the anterior and intermediate parts and mucous cells developed in the esophageal epithelium. At this time yolk sac was absorbed completely. At 6 DAM, the digestive tract was differentiated into the U type with expansion of the anterior intestine toward the head of the larvae. At 10 DAH, the expanding anterior intestine became wider and a elliptical. At 15 DAH, the anterior intestine was divided into two portions by invagination of dorsal epithelium and the mid-intestine was convoluted dorsally. At 21 DAM, the frontal portion of the anterior intestine formed a differentiated expansion sac. At 24 DAH, the second convolution was observed in the anterior part of the posterior intestine. At 30 DAH the expansion sac was extended longitudinally and the digestive tract took on the form seen in adult fish.

Water Temperature, Rearing Density and Feeding Rate on Growth and Survival Rate of Red Marbled Rockfish, Sebastiscus tertius Larvae (수온, 자어밀도 및 먹이공급량에 따른 붉은쏨뱅이, Sebastiscus tertius 자어의 성장과 생존율)

  • This study was performed to investigate the effects of water temperature and stocking density and rotifer density on the mass production of larval red marbled rockfish, Sebastiscus tertius. Total length of the marbled rockfish larva grown in $23^{\circ}C$ of water temperature were 10.80mm, which was the best, while those grown in $29^{\circ}C$ of water temperature were 6.28mm, which was the lowest. The survival rates of red marbled rockfish larvae grown in $20^{\circ}C$ and $23^{\circ}C$ were 24.0% and 18.5%, respectively. However, the survival rate of red marbled rockfish larvae grown in $26^{\circ}C$ was $7^{\circ}C$ and no larvae survived in $29^{\circ}C$. The total length of red marvled rockfish larva stocked at the density of 5 and 10 larvaper liter of water were high, which were 11.52 mm and 11.22 mm, respectively, but those stocked at the density of 30 larva per liter of water were 7.55mm, which was the lowest. The survival rate of red marbled rockfish larva stocked at the density of 2.5 larva per liter of water was 52.0%, which was the best, but the lowest, 18.0% for the red marbled rockfish larva stocked at the density of 30 larva per liter. There was a trend toward decrease in survival rate of red marbled rockfish larva as their stocking density increased. The survival rates of red marbled rockfish larva fed rotifers at the density of 10 and 5 individuals per ml were high, 48.5% and 48.0%, and their total lengths were 11.92 and 11.89mm, respectively, which grew relatively fast. The survival rate and the total length of red marbled rockfish larva fed rotifers at the density of 30 individuals per ml were the lowest, which were 8.40mm and 21.5%, respectively. Also, red marbled rockfish larva fed rotifers at the density of more than 20 individuals per ml achieved poor survival and growth. These results indicated that the proper conditions for the mass production of larval red marbled rockfish were $23^{\circ}C$ of water temperature, fish larva stocking density of 5 to 10 larva per liter of water and rotifers density of 5 to 10 individuals per ml as live feed.
