Characteristics of Biorefinery Process Using Laminaria japonica for the Production of Carbohydrate and Reducing Sugar (Laminaria japonica를 이용하여 탄수화물과 환원당 생산을 위한 바이오리파이너리 공정 특성)

  • In this study, Laminaria japonica was used as a substrate for a mixed aerobic microbial consortium. Laminaria japonica is well-known as a representative brown algal biomass possessing advantages of cheap cost, and high productivity and carbohydrate content. A biological saccharification system was established by inoculating and enriching the mixed aerobic microbial consortium. Production of the soluble carbohydrate and reducing sugar at different hydraulic retention times (HRT) was observed. The efficiency of saccharification increased according to the decrease of HRT. The maximum saccharification yield in a continuous biological pretreatment process was 17.96 and 4.30 g/L/day for the soluble carbohydrate and reducing sugar, respectively at the HRT of 1 day. In contrast, the staccharification yield decreased drastically at the HRT of 0.5 day. Experimental results indicate that Laminaria japonica is a promising material for the production of useful products, in particular for the saccharification through a biorefinery process. It can thus be concluded that a continuous biological pretreatment process using a mixed cultivation system can be successfully employed for the biorefinery technology.

Removal of Off-flavor from Laminaria Japonica by Treatment Process of Supercritical Carbon Dioxide (초임계 이산화탄소 처리 공정에 의한 다시마 유래 이취성분 제거)

  • In order to reduce or remove off-flavor and volatile organic compounds (VOCs) from Laminaria japonica effectively, continuous treatment process by supercritical carbon dioxide (SC-$CO_2$) was applied. After freeze-drying, Laminaria japonica powdered with $710{\mu}m$ was used. Experiments were carried out at temperature range from 35 to $55^{\circ}C$, and pressure range from 10 to 25 MPa for evaluation of SC-$CO_2$ treatment effect. Flow rate of carbon dioxide used in this reseach was constantly fixed at 26.81 g/min. Before and after treatment of SC-$CO_2$, off-flavor and VOCs from Laminaria japonica were analyzed by gas chromatography-mass spectrometry detector (GC-MSD). Total 47 VOCs emitted from Laminaria japonica were identified before treatment of SC-$CO_2$, major components of seaweed smell (ordor) in Laminaria japonica were identified as alcohols, aldehydes, ester and acids, ketone, halogenated compounds and hydrocarbon. Off-flavor and VOCs in all experimental conditions was reduced or removed after SC-$CO_2$ treatment. Among the experimental conditions, the highest removal yield was at 25 MPa and $55^{\circ}C$.

Effects of Dialysis and Various Drying Methods on Physical Properties of Alginates Prepared from Sea Tangle, Laminaria japonica (투석과 건조방법이 다시마(Laminaria japonica) 알긴산의 물성에 미치는 영향)

  • To investigate the physical properties of sea tangle (Laminaria japonica) alginates, extracted by the Mexican process, the effects of extracting time and drying methods on intrinsic viscosities, MWs and DPs of alginates were examined. The MWs of alginates before and after dialysis decreased with increase of extracting time. The MW s before dialysis were lower than those after dialysis. The ash contents before dialysis decreased with increase of extracting time. After dialysis the ash contents of the alginates showed 10.4-10.9% of those, which were little affected by extracting time. In the ash composition after dialysis, the sodium content was 4.4 g/100 g, $90\%$ of total ash content. The ash contents, the intrinsic viscosities, the average molecular weight and the DPs of the alginates dried by AD before dialysis were higher than those by VF. The ash and uronic acid contents of alginates after dialysis showed $10.6-10.9\%\;and\l88.1-88.9\%$, respectively. But the intrinsic viscosities, the MWs and DPs of the alginates after dialysis gradually decreased by following dry methods. The decreased order was ADAD, ADVF, VFAD, VFVF. The coefficient of determination between MWs and DPs in the alginates having more than 300 kDa was 0.999.

Biosorption of Lead ions onto Laminaria japonica and Kjellmaniella crassifolia : Equilibrium and Kinetic Modelling (Laminaria japonica와 Kjellmaniella crassifolia를 이용한 Pb의 생체흡착 : 흡착속도 및 흡착평형 모델링)

  • The batch experiments of biosorption were carried out for the removal of lead ion from metal solution using Laminaria japonica and Kjellmaniella crassifolia, two species of marine algaes as biosorbent. We have investigated biosorption kinetics and equilibrium of lead by using marine algaes. We observed that biosorption of lead occurred very rapidly by marine algaes ; the biosorption reached equilibrium less than 2 hr. These experimental data could be accurately described by a pseudo-second-order rate equation, obtaining values between $0.883{\times}10^{-3}$ and $0.628{\times}10^{-3}\;g/mg/min$ for the biosorption rate constant $k_{2,ad}$. It could be described with Langmuir, Redlich-Peterson, and Koble-Corrigan(Langmuir-Freundlich) equation. The biosorption capacity by L. japonica and K. crassifolia were in the sequence of Pb>Cd>Cr>Cu and Pb>Cu>Cd>Cr, respectively. The biosorption capacity of L. japonica were increased with pH increasing.

Effects of Diet with Laminaria religiosa on Egg Quality (다시마 첨가 사료가 계란의 품질에 미치는 영향)

  • The effect of diet with Laminaria religiosa on egg quality were investigated. Isa Brown chicken (28 weeks old) were raised for 10 week. The addition level of Laminaria religiosa to each treatment was 0, 10 and 20%, respectively. No difference in egg yolk and white color was found among treatment. The contents of Ca, Fe, K and Mg were increased but of P decreased with the feeding of Laminaria religiosa. Total mineral content were increased from 860.9 mg% to 936.8 mg%. Tensile strength of egg shell was improved from $63g/cm^2$ to $77g/cm^2$. Total cholesterol content were reduced from 1,760 mg% to 1,113 mg%. Among the analyzed amino acids, the content of each amino acid was reduced except histidine increased from 2,024.0 mg% to 2,119.6 mg%. Essential amino acids in the total amino acids, 42.1% to 42.6%, were hardly changed among treatments.

Influence of Water Temperature on Growth and Body Composition of Juvenile Abalone (Haliotis discus hannai) Fed an Artificial Formulated Diet and Macroalgae (Laminaria japonica) (배합사료 및 다시마 공급이 참전복 (Halioitis discus hannai) 치패의 성장 및 체조성에 미치는 수온의 영향)

  • The effect of water temperature and feed on the growth, survival, and body composition of juvenile abalone (Haliotis discus hannai) fed an artificial formulated diet and a macroalgae (Laminaria japonica) for 12 weeks was investigated. Rearing temperature was maintained at $13{\circ}C,\;16{\circ}C,\;19{\circ}C$ by heated and natural sea water temperatures $(7.5-10.2{\circ}C).$ Shell growth, weight gain, and survival rate of the abalone were affected by water temperature and diet (P<0.05). Survival and growth rate of the abalone fed with the formulated diet and Laminaria at $19{\circ}C$ was significantly higher than those of the abalone fed the formulated diet or Laminaria at $13{\circ}C,\;16{\circ}C,$ and ambient temperatures (P<0.05). Moisture, crude protein, crude lipid, and ash contents of the soft whole body were affected mainly by diet than rearing water temperature (P<0.05). These indicate that a mixture of formulated diet and macroalgae could improve the growth of the abalone independent of water temperature and heated water could improve the growth of abalone during the winter season.

Organic Acid and Enzyme Pretreatment of Laminaria japonica for Bio-ethanol Production (유기산 및 효소적 전처리를 이용한 다시마에서 바이오 에탄올 생산)

  • We investigated for the production of biological bio-ethanol from Laminaria japonica using the hydrolysis reaction of enzymes and organic acids and the polysaccharide content was also analyzed. The composition of the polysaccharide was characterized as 65.99% alginate, 6.24% laminaran and 27.77% mannitol. The optimum concentration for reducing the sugar conversion by Laminaria japonica was found to be 1.874 g/L at an acetic acid concentration of 1.5%, $121^{\circ}C$ for 60 min, and for an ascorbic acid of 2.0%, 4.291 g/L was produced in the same condition. The enzyme hydrolysis such as alginate lyase and laminarinase contained the maximum 2.219 g/L reducing sugar. In the result of ethanol fermentation using hydrolysate of Laminaria japonica, the organic acid treatment showed a high of reducing sugar yield, but decreased the ethanol yield, and then the maximum ethanol production obtained was 1.26 g/L using the mixed treated of enzyme.

  • Primary structure of purified fucoidans was analysed by methanolysis, methylation and HT-IR measurement. Basic linkage in the fucoidans of Laminaria religiosa, sporophylls of Undaria pinnatifida, Hizikia fusiforme and Sargassum fuivellum from Korea was $(1\rightarrow3)$ linkage of fucose, but a considerable amount of $(1\rightarrow3)$ linked galactose was also found in case of the purified fucoidan from sporophylls of U. pinnatifida. Ester sulfate was substituted mainly to C-4 position of fucose and galactose.

Ultrasonic Extraction of Phenolic Compounds from Laminaria japonica Aresch Using Ionic Liquid as Extraction Solvent

  • An ionic liquid-based ultrasonic-assisted extraction method has been successfully applied to the effective extraction of phenolic compounds from Laminaria japonica Aresch. Three kinds of 1-alkyl-3-methyl-imidazolium with different cations and anions were evaluated for extraction efficiency. The results showed that both the characteristics of anions and cations have remarkable effects on the extraction efficiency. In addition, the ionic liquid-based ultrasonic-assisted extraction procedure was also optimized on some extraction parameters, such as ultrasonic power, extraction time and solid-liquid ratio. Compared with the conventional solvent, the optimum approach gained the highest extraction efficiency within the shortest extraction time. Average recoveries of phenolic compounds were from 75.5% to 88.3% at three concentration levels.

  • Eight unusual individuals of a laminariaceaen species were collected from the Sea of Okhotsk in 1974 and described as a new species, Laminaria multiplicata Petrov et Suchovejeva in 1976. Since that time no new information, including pictures and numerical data, has been provided, although the species was cited in floristic lists of the Sea of Okhotsk based on the first record in 1976. We investigated a type and 3 paratypes of L. multiplicata and strongly believe that they were wrongfully identified abnormal plants of L. gurjanovae A. Zinova. Therefore, the species L. multiplicata needs to be closed.
