광질(光質), 생장조절물질(生長調節物質) (GA, BA, ABA) 및 Sodium Hypochlorite의 처리가 상치 (Lactuca sativa L.) 종자의 발아(發芽)에 미치는 영향(影響) (Effects of Light Source, Plant Growth Regulators (GA, BA, ABA) and Sodium Hypochlorite on 'Grand Rapid' Lettuce (Lactuca Sativa L.) Seed Germination)

  • 상치종자(種子)에 광질(光質), 생장조절물질(生長調節物質), soduim hypochlorite침지처리하여 발아시험(發芽試驗)한 결과(結果)는 다음과 같다. 1. 자연광(自然光) 및 적색광하(赤色光下)에서 GA 및 BA는 발아(發芽)를 촉진(促進)시켰으며, 청색광(靑色光)이나 ABA는 발아(發芽)를 억제(抑制)시켰다. 2. 생장조절물질(生長調節物質)의 복합처리시(複合處理時) GA 농도(濃度)가 높아질수록 발아율(發芽率)은 상승(上乘)하며 암흑하(暗黑下)에서는 BA농도(濃度) 10ppm에서 발아율상승효과(發芽率上乘效果)가 컸다. 3. Sodium hypochlorite 20분(分) 이상(以上) 침지처리로 암흑하(暗黑下)에서도 85%이상(以上)의 발아율(發芽率)을 보였다. 4. ABA하(下)에서도 sodium hypochlorite 침지처리로 발아율상승효과(發芽率上乘效果)를 볼 수 있었다.

상추 유식물체 절편의 조직배양에 의한 고빈도 기관발생과 식물체 재분화 (High Frequency Organogenesis and Plant Regeneration in Tissue Cultures of Lettuce Seedling Explants)

  • 부정아를 유도하기 위하여 발아 7∼10일된 일본 결구상추 2품종과 국내 잎상추 (Lactuca sativa L.)4품종의 하배축 및 자엽절편으로부터 식물체 재분화시스템을 확립하였다. 하배축 및 자엽절편을 BA와 NAA가 조합 처리된 MS 및 SH배지에 치상하여 5주 동안 명배양하였다. 일반적으로 하배축보다는 자엽절편에서, SH 배지보다는 MS 배지에서 배양하였을 때에 부정아 형성률이 높았다. MS배지에 BA는 0.5 ㎎/L, NAA는 0.1∼l ㎎/L로 처리하였을 때에 최고의 부정아 형성률을 보였으며, 품종에 따라 30% ( '만추대청치마')에서 86% 까지의 부정아 형성률을 나타내었다. 형성된 shoot의 95% 이상이 MS 기본배지에서 발근하여 소식물체로 발달하였다.

  • 상추에 제초제 저항성을 도입하기 위해 상추 자엽 조직을 NPTII-GUS 및 제초제 저항성 유전자 (PAT)가 삽입된 A. tumefaciens MP 90과 2일간 공동배양한 다음 0.1 mg.L$^{-1}$ NAA, 1.0 mg.L$^{-1}$ 2ip, 50 mg.L$^{-1}$ kanamycin, 500 mg.L$^{-1}$ carbenicillin을 첨가한 MS배지에 배양하여 식물체를 재분화시켰다. 재분화 식물체는 kanamycin 내성 검정을 통해 NPTII 유전자의 도입과, PCR, Northern blot 분석을 통해 제초제 저항성 유전자가 식물체의 게놈상에 삽입된 형질전환체임이 확인되었다. 또한 1500배액의 농도로 Basta를 살포한 결과 살포 10일 후 대조 식물체는 완전히 고사하는 데 반해 형질전환체는 지속적인 생육을 보여 얻어진 형질전환 상추가 제초제 저항성 개체임이 확인되었다.

The Effect of Wood Extract as a Water-Soluble Fertilizer in the Growth of Lactuca sativa

  • Recently, due to environmental and toxicity issues, there has been increasing attention on research regarding natural products that can reduce the use of chemical fertilizers. Wood extracts derived from the biorefining process contain various fertilizer ingredients. HPLC analysis revealed that wood extract contains approximately 5.2% hemicellulosic sugar. The growth of lettuce (Lactuca sativa) upon treatment with wood extract (extract obtained from steam-exploded pine) or water-soluble fertilizers containing different nutrients was analyzed in this study. After two weeks, the growth characteristics of lettuce as affected by wood extract or water-soluble fertilizers were significantly different. The effect of water-soluble fertilizers containing ascorbic acid, magnesium sulfate, citric acid, potassium nitrate, amino acids, or seaweed extract was less desirable than that of wood extracts regarding plant height (18.6 cm), number of leaves (10), leaf length (14.1 cm), shoot fresh wight (9.8 g/plant), root fresh weight (0.8 g/plant) and shoot dry weight (0.6 g/plant). The plant height, number of leaves, leaf length, shoot fresh wight, root fresh weight, shoot dry weight of water-soluble fertilizers containing wood extract were significantly different compared to the control (plant height :13.5 cm, number of leaves : 7, leaf length : 9.4 cm, shoot fresh wight : 5.3 g/plant, root fresh weight : 0.7 g/plant, shoot dry weight : 0.4 g/plant, root dry weight : 0.07 g/plant). From these results, it was concluded that wood extract can be used as a potential water-soluble fertilizer to increase the yield of leafy vegetables.

  • Lettuce (Lactuca sativa L., Family Asteraceae) is highly ranked in production and economic value and is consumed either fresh or in salad mixes because of its important dietary source of bioactive phytochemicals. The world collection of Lactuca spp. leafy crops, maintained in NAC, includes 2,464 accessions from 71 countries around the world, of which 2,411 belong to L. sativa species, nineteen to L. saligna, and fifteen to L. serriolar. We aimed to investigate the lettuce germplasm with morphological and biochemical analyses and provide new material for breeding. The lettuce crop comprises seven main groups of cultivars (including oilseed lettuce) differing phenotypically. Agricultural characteristcs were investigated including time to bolting, time to flowering, seed color, flower color, leaf attitude, leaf color, leaf anthocyanin coloration, type of incision of margin, depth of incisions of margins, and leaf venation. Screening of the health beneficial metabolites like anthocyanin and bitter sesquiterpene lactones (lactucin and lactucopicrin) was also conducted. The range of anthocyanin and SLs were 0~563.78 mg/100g D.W. and 3.74~3311.66 ug/g D.W., respectively. The investigation of the degree of variation regarding phenotypic traits and biochemical revealed adaptive stable and highly variable use of trait collection.

GalUR 유전자를 이용한 비타민 C 증대 상추 (Lactuca sativa L.) 형질전환체 개발 (Transgenic lettuce (Lactuca sativa L.) with increased vitamin C levels using GalUR gene)

  • 사람은 채소를 통해 필수영양소인 L-Ascorbic acid (vitamin C)를 공급받는다. 본 실험의 목적은 비타민 C 생합성 유전자인 GalUR 유전자를 상추 (Lactuca sativa L.)에 형질전환 하고자 실시하였다. (주)농우바이오의 화홍적축면 상추의 자엽 절편체를 선발배지 (MS + 30 g/L Sucrose + 0.5 mg/L BAP + 0.1 mg/L NAA + 100 mg/L Kanamycin + 200 mg/L Lilacillin, pH 5.2.)에 치상하여 3주 경과후 자엽 절편체의 절단면에서 callus와 신초가 형성되었다. 그 결과 GalUR 유전자로 상추형질전환을 성공하였고 비타민 C 함량을 분석하였다. 대조구에 비하여 상추 형질전환체 line에서 높은 함량의 비타민 C 특히, GalUR 유전자가 삽입된 $T_1$ 중 일부는 비형질전환체에 비해 $3{\sim}4$배 높은 비타민 C 함량을 나타내었다. 이 결과는 GLOase 유전자 형질전환 상추 $T_1$ 세대에서 고함량의 비타민 C를 함유한 결과와 일치한다. 이런 결과를 기초로 하여 비타민 C 고함량 $T_2$ line을 선발하였다.

유기질비료와 토양 미생물제제의 시용이 배추(Brassiea campestris L.)와 상추(Lactuca sativa L.)의 수량에 미치는 영향 (Effects of Brassiea campestris L. and Lactuca sativa L. Yield by Application of Organic Fertilizers and Microorganisms)

  • 돈분발효퇴비(P)와 혼합유박비료(B, F), 부산물퇴비(D) 4종의 유기질비료와 토양 미생물제제(M)의 시용 효과를 규명하기 위하여 배추와 상치를 공시작물로 시설하우스에서 재배하면서 생육 및 수량을 조사하였다. 유기질비료의 추천 시비량 처리구에서 수량이 가장 좋았으며, 과량 처리구에서는 비해가 발생하였으나, 미생물제제의 혼용에 의해 현저한 비해 감소 현상을 볼 수 있었다. 또한 같은 비종 및 동일한 시용량 처리구에서도 미생물제제를 혼용하였을 때 생육이 안정되었으며, 수량도 높은 경향을 보였다. 결론적으로 유기질비료의 과량처리는 비해가 발생할 수 있으며, 미생물제제의 시용으로 생육 및 수량이 향상되었다.

Genetic Relationships of Lactuca spp. Revealed by RAPD, Inter-SSR, AFLP, and PCR-RFLP Analyses

  • RAPD, Inter-SSR, and AFLP markers were used to assess the genetic diversity of lettuce cultivars and the phylogenetic relationships in Lactuca spp. A total of 216 polymorphic bands from seven RAPD primers, four Inter-SSR primers, and five AFLP primer combinations were used to elucidate the genetic similarity among lettuce cultivars. Forty-four lettuce accessions were subdivided into discrete branches according to plant type: crisphead, butterhead, and stem type, with some exceptions. The leafy- and cos-type accessions were intermingled in other groups with no discrete branch indicating that these are more diverse than others. Three accessions, including the Korean cultivar 'Cheongchima', the Korean local landrace 'Jinjam', and the German cultivar 'Lolla Rossa' were classified as the most diverse accessions. Twenty bands were unique in specific cultivars. Among these, three were specific in a plant type; one in Korean leafy type, one in crisphead type, and one in cos type lettuce. In the phylogenetic analysis among Lactuca species, L. saligna, L. serriola, and L. georgica clustered in a sister branch of the L. sativa complex. Two L. virosa accessions show the highest intra-specific relationships. L. perennis outlied from all the other Lactuca species at a genetic similarity of 0.53 and clustered with two Cichorium species, C. intybus and C. endivia, with genetic similarity of 0.67. The phylogenetic tree was supported by data from polymorphism of chloroplast genome which was revealed by PCR-RFLP.

Phytochemical constituents of Lactuca serriola leaves and their content analysis by HPLC-UV

  • This study aimed to identify the phytochemical constituents of Lactuca serriola leaves and perform quantitative analysis of the methanol (MeOH) extract of L. serriola, L. indica, L. raddeana, L. sativa, and L. triangulata. Six compounds were isolated from the MeOH extracts of L. serriola using open column chromatography and identified as protocatechuic acid (1), caffeic acid (2), cynaroside (3), apigenin 7-glucuronide (4), luteolin (5), and apigenin (6) using 1H-, 13C-nuclear magnetic resonance, and mass spectrometry. Quantitative analysis of the six compounds was performed on the MeOH extract of Lactuca species using high-performance liquid chromatography (HPLC) and an ultraviolet (UV). A reverse-phased column was used, and the UV absorbance was set to 280 nm. The contents of compounds 2 and 3 were the highest (1.58 and 2.64 mg/g ext., respectively) in L. serriola extracts. However, compounds 4 and 6 were higher (1.46 and 0.40 mg/g ext., respectively) in L. triangulata. These results provide quantitative data for the application of Lactuca species in the pharmaceutical and functional food industries.
