Lactobacillus amylovorus CACC736 was originated from swine feces in Korea. The complete genome sequences of the strain contained one circular chromosome (2,057,809 base pair [bp]) with 38.2% guanine-cytosine (GC) content and two circular plasmids, namely, pCACC736-1 and pCACC736-2. The predicted protein-coding genes, which are encoding the clustered regularly interspaced short palindromic repeats (CRISPR)-associated proteins, biosynthesis of bacteriocin (helveticin J), and the related proteins of the bile, acid tolerance. Notably, the genes related to vitamin B-group biosynthesis (riboflavin and cobalamin) were also found in L. amylovorus CACC736. Collectively, the complete genome sequence of the L. amylovorus CACC736 will aid in the development of functional probiotics in the animal industry.

Lactobacillus amylovorus are known to exist in the intestinal flora of healthy cattle or pigs. The L. amylovorus strain 1394N20 was isolated from the feces of the Hanwoo calf (Bos taurus coreanae). The genome of strain 1394N20 consists of a single circular chromosome (2,176,326 bp) with overall guanine + cytosine content of 37.8 mol%. Moreover, 2,281 protein-coding sequences, 15 rRNAs, and 65 tRNAs genes were identified in the chromosome based on the results of annotation. The bacterium has a gene encoding endoglucanase, an enzyme that hydrolyzes the 1,4-β-D-glycosidic linkages in cellulose, hemicellulose, lichenin, and cereal β-D-glucans. Genomic sequencing of L. amylovorus strain 1394N20 reveals the immense potential of the strain as a probiotic with nutrient digestibility.
본 연구에서는 발효 식품의 제조에 유용한 항균성 물질을 생산하는 유산균 starter의 개발의 일환으로 장내 상존균으로 알려진 Lactobacillus acidophilus group유산균인 Lactobacillus amylovorus IMC-1 균주가 생산한 항균성 물질의 식품오염세균에 대한 항균 활성을 검토하였다. 내몽골 원산 치즈에서 분리된 L. amylovorus IMC-1 균주는 탈지유배지에서 37$^{\circ}C$에서 72시간 배양하였을 때 최대로 항균성 물질을 생산하였으며, 더 이상의 배양은 항균성 물질의 생산에 영향을 미치지 않았다. 겔 여과후의 항균성 물질은 식품오염세균인 Bacillus subtilis IFO 3025, staphylococcus aureus IAM 1011, Listeria monocytogenes VTU 206, Escherichia coli RB. 및 Pseudomonas fragi IFO 3458등과 같은 모든 피검균에 대하여 20units/ml 첨가로 항균활성을 나타내었다. 그리고 이 물질은 B. subtilis, E. coli및 Ps. fragi에 대해서는 살균 작용을 나타내었으며, Staphy. aureus와 L. monocytogenes에 대해서는 정균 작용을 보였다. 그 살균 작용은 용균 작용에 기인한 것임이 밝혀졌다. 또한 이 물질은 유기산, 과산화수소 및 단백질성 물질이 아님이 밝혀졌다.
Lactic acid production parameter optimization using Lactobacillus amylovorus NRRL B-4542 was performed using the design of experiments (DOE) available in the form of an orthogonal array and a software for automatic design and analysis of the experiments, both based on Taguchi protocol. Optimal levels of physical parameters and key media components namely temperature, pH, inoculum size, moisture, yeast extract, $MgSO_4{\cdot}7H_20$, Tween 80, and corn steep liquor (CSL) were determined. Among the physical parameters, temperature contributed higher influence, and among media components, yeast extract, $MgSO_4{\cdot}7H_20$, and Tween 80 played important roles in the conversion of starch to lactic acid. The expected yield of lactic acid under these optimal conditions was 95.80% and the actual yield at optimum conditions was 93.50%.
A lactic acid bacterium producing an antimicrobial substance against Escherichia coli O157:H7 was isolated from raw milk and identified as Lactobacillus amylovorus ME-1. In addition to E. coli O157 :H7, the antimicrobial substance also inhibited the growth of Bacillus cereus, Listeria monocytogenes, Pseudomonas aeruginosa, Salmonella typhimurium, Staphylococcus aureus, Streptococcus agalactiae, Streptococcus pyrogenes, and Yersinia enterocolitica. The antimicrobial substance was stable at pH 2-12 and $121^{\circ}C$ for 15 min and insensitive to proteinase K, protease, amylase, and catalase. Purification of the antimicrobial substance was conducted through methanol and acetonitrile/ethylacetate extraction, ultrafiltration with a 500 Da cutoff, thin layer chromatography (TLC) with silicagel 60, and high performance liquid chromatography (HPLC) with a $C_{18}$ reverse phase column. The ${\lambda}_{max}$ of the purified antimicrobial substance was determined as 192 nm by ultra violet (UV) scanning, while the molecular weight was estimated as 453 Da based on the mass spectrum. Accordingly, the current results suggest that the antimicrobial substance from the L. amylovorus ME-1 was not a bacteriocin, but rather a new non-proteinaceous substance distinct from acidophilin, acidolin, diacetyl, and reuterin.
A 4.6-kb HindIII fragment encompassing the complete 140-kDa ${\alpha}$-amylase gene of Lactobacillus amylovorus B 4540 was cloned into pBR322 by the shot gun method. Southern blotting and restriction mapping for the insert were performed. The recombinant 9.0-kb plasmid, pFML1, conferred ${\alpha}$-amylase activity to E. coli and Lactococcus lactis hosts when introduced by electroporation. SDS-PAGE and zymography confirmed the production of 140-kDa ${\alpha}$-amylase and its proteolytic degradation products with enzyme activity in transformants. Total ${\alpha}$-amylase activity of E. coli $DH5{\alpha}$ cells harboring pFML1 was 1.8 units and most activity was detected from cell pellets. Total enzyme activity of L. lactis subsp. lactis MG1363 transformant was five to ten-fold lower than that of E. coli cell but more than half of the activity was detected in the culture supernatant.
Clostridium butyricum, Lactobacillus amylophillus, Lactobacillus amylovorus, Lactobacillus acidophillus, AI-9 produced hydrogen and /or organic acids using glucose, lactose and starch at the anaerobic culture conditions. Cl. butyricum NCIB 9576 evolved 1,700 ml H2/L-culture broth and accumulated butyric acid, acetic acid, propionic acid and ethanol in its culture broth when lactose was used as a carbon source during 24 hrs of fermentation. L. amylovorus ATCC 33620 accumulated lactic and acetic acids and some reducing sugars when starch was used as a carbon source without hydrogen production. Instead of starch as a carbon source, L. amylovorus ATCC 33620 produced lactic acid from algal biomass during fermentation and the acid-heat or freeze-thaw pretreatment of algal biomass accelerate the lactic acid fermentation.
Bacteria from the genus Lactobacillus are important for the production of fermented food and dairy products, and as symbionts in human and animals. Lactobacillus acidophilus has widely been used in the production of yogurt, health foods, and even medicines. The efficacy of L. acidophilus has been proven with regards to the reduction of cholesterol, prevention and treatment of diarrhea, modulation of the immune system, suppression of cancer, etc. Using molecular biology tools, Lactobacillus acidophilus has now been reclassified into six species: L. acidophilus, L. amylovorus, L. crispatus, L gallinarium, L. gasseri, and L. johnsonii. Thus, since L. acidophilus has now been marked as a newly defined species, caution is advised when reading future publications regarding this bacterium. In this article, the results of the reclassification of L. acidophilus are mentioned after an analysis of its field inheritance was performed by my research team. Especially, L. amylovorus KU4 (formerly named as L. acidophilus KU4; KCCM 10975P) is a novel probiotic strain that is isolated from humans; it has the ability to reduce cholesterol. It has also been reported as a microorganism that effectively inhibits the growth of pathogenic E. coli. However, this Korean patent (No 10-1541280) refers to a strain obtained from calves; the origin of this strain was incorrectly labeled. Furthermore, after the discovery of L. acidophilus in 1900, its role in intestinal microbiological research was described and its utilization as a probiotic was presented.
발효식품의 제조에 유용한 starter 개발 및 probiotics로서 이용 가능성을 타진하기 위하여 항균성 물질을 생산하는 Lactobacillus amylovorus IMC-1 균주를 이용한 skim milk medium에서의 산생성, 단백질 분해활성, lactose 분해활성 및 유사구균과 혼합발효 그리고 병원성 세균에 대한 살균력 등을 검토하였다. L. amylovorus IMC-1은 10% skim milk에 yeast extract를 첨가하여 12시간 배양하였을 때 산도 0.8%이었고, 72시간째에는 2.7%로 우수한 산 생성력을 보였다. IMC-1 균주의 ${\beta}-galactosidase$ 활성은 균체 1 mg당 약 $39{\mu}M/minute$로 매우 강한 반면, $phospho-{\beta}-galactosidase$의 활성은 매우 낮았다. 또한 이 균주는 10% skim milk에서 배양 12시간째 $6\;{\mu}g/mL$, 72시간째에 $69\;{\mu}g/mL$의 유리 tyrosine을 생산하여 단백질 분해활성이 낮았다. 전기영동상의 band pattern으로부터 casein중 ${\alpha}-casein$을 주로 이용하며, ${\beta}-casein$은 거의 이용하지 못함을 알 수 있었다. 10% skim milk에서 IMC-1 균주 및 St. thermophilus NIAI 510의 단독 또는 혼합배양을 실시한 결과, 혼합배양, IMC-1 균주 및 St. thermophilus 순으로 산 생성력이 높았으며, 유산구균 단독배양 및 혼합배양에서는 시간이 경과함에 따라 치즈 향기가 증가하는 것을 확인할 수 있었다. 또한 10% skim milk medium에서 혼합배양에 의한 최적의 유산발효 시간은 산 생성력 및 공존 균수 등을 고려할 때, yeast extract 0.1% 첨가시에는 16시간, 0.5% 첨가시에는 12시간 배양하는 것이었다. IMC-1이 생산한 항균성 물질은 Escherichia coli O157을 배양 24시간째 2 log정도 감소시켰고, 48시간 배양 후에는 피검균이 검출되지 않았다. 또한 Shigella flexneri는 배양 4시간째 2 log이상 감소하였고, 6시간 배양 후에는 검출되지 않아 항균성 물질은 강한 살균작용을 나타냄을 알 수 있었다.
본 연구는 육계 및 산란계 맹장에서 분리하여 유기산 생성 능력, 내산성, 내담즙성 및 성장특성을 토대로 선발된 유산균 6종(Lacto- bacillus reuteri BLA5, Lactobacillus crispatus avibro1, Lactobacillus reuteri avibro2, Lacto- bacillus amylovorus LLA7, Lactobacillus crispatus avihen1, Lactobacillus vaginalis avihen2)의 효소, 항균 활성 및 항생제 감수성을 조사하여 생균제로서의 이용 가치를 구명하고자 실시하였다. 육계 및 산란계 맹장 유래 유산균은 효소 활성에 있어서는 균주마다 차이가 있었으나, 일반적으로 amylase와 lipase 활성이 강한 것으로 나타났고 반면, cellulase 및 protease의 활성은 약한 것으로 나타났다. 유산균 배양액은 병원성 E. coli에 대하여 강한 항균 활성을 보였으며, 주요 억제 요인으로는 유기산 분비에 의한 pH 저하에 의한 것으로 나타났다. 병원성 Salmo- nella에 대해서는 유산균 배양액의 억제성이 약한 것으로 나타났다. 유산균은 치료용 항생제로 이용되는 ampicillin과 amoxicillin에 대해서는 높은 감수성을 보인 반면 사료용 항생제(virginiamycin 및 salinomycin)의 경우 정상적인 첨가 수준에서는 영향을 받지 않았다.
