  • Anaerobic reductive dechlorination of tetrachloroethylene(PCE) to ethylene was investigated by performing laboratory experiments using semi-continuous flow two-in-series soil columns. The columns were packed with soils obtained from TCE-contaminated site in Korea. Site ground water containing lactate(as electron donor and/or carbon source) and PCE was pumped into the soil columns. During the first operation with a period of 50 days, injected mass ratio of lactate and PCE was 620:1 and incomplete reductive dechlorination of PCE to cis-DCE was observed in the columns. However, complete dechlorination of PCE to ethylene was observed when the mass ratio increased to 5,050:1 in the second operation, suggesting that the electron donor might be limited during the first operation period. During the degradation of cis-DCE to ethylene, the concentration of hydrogen was $22{\sim}29mM$. These positive results indicate that the TCE-contaminated groundwater investigated in this study could be remediated through biological anaerobic reductive dechlorination processes.

  • TLC condition was developed for its simple separation and quantitative analysis of lactic acid. Rapid and clear separation of lactic acid by silica gel TLC plate was obtained by using nitromethane : 1-propanol : $H_2O$ (2 : 5 : 1.5, v/v/v) and a suitable dipping solution of 40 mg bromocresol purple in 100 mL 5% ethanol (pH 10.0). The lactic acid was shown as a bright yellow spot on a light cinnabar background. The quantitatively detectable concentration range of lactic acid was between 0.5 and 4% with 99.4%, confidence. Quantitative TLC analysis result was confirmed with HPLC and with enzymatic Quantitative analysis methods (by using lactate dehydrogenase).

  • [Purpose] To date, there have been no studies on the response of wrist tremor to increased muscle activity during exercise. This study aimed to evaluate the wrist tremor response with increasing muscle activity during bench press exercise. [Methods] Triceps muscle activity and wrist tremor response were measured by electromyography and an accelerometer, respectively, during bench press exercise in 11 healthy men without weight-training experience. Subjects performed bench press at 30% repetition maximum (RM), and the rating of perceived exertion (RPE) and lactate concentration were measured before and after exercise. One week later, an equivalent number of bench presses at 30% RM was performed without weight load as a control trial (CT). [Results] RPEs and lactate concentrations significantly increased after resistance exercise (30% RM) from 7.4 to 14.3 and 1.7 to 4.9, respectively (P<.01), but no such difference was observed in the CT. Muscle activity linearly increased during the 30% RM exercise, and wrist tremors were shown to linearly decrease. A strong negative correlation was observed between the two variables (r=-0.88, P<.001). [Conclusion] We found that wrist tremors during resistance exercise, as measured using an accelerometer, can be used to predict muscle activity.

The Effect of Sport Drink Ingestion on the Physical Fatigue Level and Heart Rate during Prolonged Exposure in Hot Ambient Temperature in Professional Golfers (장시간 고온 환경 노출시 수분의 섭취가 프로골프선수들의 신체피로도 및 심박수에 미치는 영향)

  • We investigated the effects of prolonged exposure in hot environmental condition and ingestion of fluid on various physiological variables including plasma glucose, lactate, the rating of perceived exertion (RPE), and heart rate as well as golf putting performance. Six male professional golfers were voluntarily participated in three different putting trials which were separated by seven days of time interval period. Three different putting trials were conducted at either 20℃ or 32℃, or 32℃ + Fluid ingestion. Performing 32℃ + Fluid ingestion trial, all subject ingested sport drink as much as their body mass was decreased. For each experiment, all subjects were undertaken total 48 putting, which separated by four x 12 putting in four different time points (i.e., Rest, 1 hr, 2 hr, and 3 hr). Plasma glucose concentration was significantly decreased with hot ambient condition but it was almost fully recovered by fluid ingestion. Plasma lactate concentration was significantly higher when subjects were exposed in hot environmental condition, and it did not change with fluid ingestion. There was a no different in putting performance and psychological fatigue level (performed by GRID test) at any environmental conditions. The RPE, commonly used for evaluating of physical fatigue level, was significantly dropped by fluid ingestion which indicates lower physical fatigue level. In addition to this, heart rate (HR) was also significantly decreased after fluid ingestion. Based on these results, it was concluded that the ingestion of fluid during prolonged exposure in hot ambient condition decrease the degree of physical fatigue levels and heart rate, which will possibly improve the golf performance when exposed in extreme weather condition in summer.   • This study was focused on investigating the growth properties of a sulfate reducing bacterium Deslfovibrio sp. B5 which has metabolic ability for desulfurization of petroleum. The optimal temperature and pH for growth of Desulfovibiro sp. B5 were $38^{\circ}C$ and 6.6-7.0, respectively. Addition of 10% corn steep liquor to the Postgate medium C resulted in 0.79 g/L cell concentration, corresponding to a 1.8-fold increase in dry cell mass. Acetate concentrations above 10g/$\ell$ inhibited cell growth significantly. $H_2S$ generated from the sulfate reduction also inhibited the growth of Desulfovibrio sp. B5 at a concentration of 10mM total sulfide. But $N_2$ gassing relieved the growth inhibition by $H_2$S and thereby resulted in a 1.75-fold enhancement in specific growth and lactate consumption pattern of Desulfovibrio sp. B5.

Effect of Microbial and Chemical Combo Additives on Nutritive Value and Fermentation Characteristic of Whole Crop Barley Silage

  • This study was conducted to assess the effects of microbial and chemical combo additives on nutritive values, fermentation indices and aerobic stability of whole crop barley silage. Barley forage (Youngyang) was harvested at about 30% dry matter (DM) by treatments, chopped to 5 cm length and treated with distilled water only (CON), Lactobacillus plantarum (INO), propionic acid (PRO) or an equal mixture of INO and PRO (MIX). Barley forages were ensiled in 4 replications for 0, 2, 7, and 100 days. On 100 days of ensiling, MIX silage had higher (p<0.05) in vitro DM digestibility than CON silage, but lower (p<0.05) acid detergent fiber concentration. The pH in all treated silages was lower (p<0.05) than CON silage. The MIX silage had higher (p<0.05) lactate concentration and lactate to acetate ratio than in CON, but lower (p<0.05) yeast count. Aerobic stability in CON, PRO, and MIX silages were higher (p<0.05) than in INO silage. It is concluded that microbial and chemical combo additives using L. plantarum and propionic acid could efficiently improve nutritive values of barley silage in terms of increased in vitro DM digestibility compared to other treatments. In addition, all treatments except CON reduced yeast count which is the initiate microorganism of aerobic spoilage.

Changes of serum cortisol concentration and stress responses in cohe salmon(Oncorhynchus kisutch) to netting (그물작업에 의한 은연어의 스트레스 반응)

  • The production of cultured coho salmon (Oncorhpchus kisutoh) in Korea has being increased year after year. Smolt being reared in freshwater suffer transferring into seawater and are farmed in cages for fattening. This handling processes including transportation, confinement into cages are unavoidable stress to fish in salmon farming and often end up to mass mortality, This study aimed to investigate the impact of handling process on the stress responses of coho salmon. The indicator of stress was measured by cortisol to be a first response, and for the second response test, glucose, triBlyceride, cholesterol, lactate and electrolyte of $K^+, Na^+, Cl^-$ in serum and the activities of alanine aminotrtnferase (ALT), aspartate aminotransferase(AST) and lactate dehydrogenase (LDH) were analyzed. As a result, the concentration of cortisol, glucose as well as LDH activity were significantly increased, whereas others showed no difference comparing with control group. It obviously demonstrated that handling process made fish stressful.

  • The aim of this study was to investigate influence of body build on body composition, energy metabolic state and insulin concentration of blood. 29 male athletes and 36 male non-athletic students were recruited for the study. Anthropometry including chest depth and breadth, fat mass, fat fee mass, tricep skinfold thickness were measured. fasting glucose, lactate, triglyceride, fee fatty acid, and insulin concentration in serum were measured . Body build was assessed using metric index, which calculated by regression equations of Mohr and Greil. The athletic and non-athletic students were allocated to 3 body build, that is leptomorph, mesomorph, and pyknomorph. Resting metabolic rate was calculated. Respiratory quotient was determined through ratio of measured VO$_2$, and V$CO_2$. Most non-athletes have a leptomorphic body build, in contrast to athletes mesomorphic type. The body build type influenced body composition differently between non-athletic group and athletic group. Weight, body mass index, body fat mass and fat mass proportion (%), and fat-free mass increased from leptomorph to pyknormorph in non-athletic group. Pyknormorphic athletes have a significant higher body mass index, fat mass, fat free mass than other body build type. Serum glucose, triglyceride, lactate, insulin showed significant differences only in non-athletic group between leptomorph and mesomorph. RMR increased significantly from leptomorph to mesomorph in non-athletes. There was no significant difference of RQ among 3 body build types in both athletes and non-athletes. This study gives a coherent data on body build and body composition for athletes and non-athletes students. The influence of body builds on energy metabolic status of serum was different between athletes and non-athletes.

Novel Encapsulation with New Glyceryl Ester Vesicle Enhances Stability of Nanoemulsion Containing Astaxanthin (아스타잔틴을 포함하는 나노에멀젼의 안정성 향상을 위한 신규 Glyceryl Ester 이용 캡슐화)

  • Oil in water nanoemulsion of astaxanthin was prepared by high pressure homogenization. The emulsifying conditions including emulsifier type, concentration and astaxanthin concentration were optimized. Stability of nanoemulsion was measured using zetasizer, freeze-fracture scanning electron microscope (FF-SEM), particle analyzer and colorimeter. The mean diameter of the dispersed particles containing astaxanthin ranged from 160 to 190 nm. Size distribution was unimodal and extended from 40 to 200 nm. The nanoemulsion prepared by glyceryl citrate/lactate/linoleate/oleate had smaller particle size and narrow size distribution. Stable incorporation of astaxanthin in nanoemulsion was performed and checked using high performance liquid chromatography (HPLC), freeze-fracture scanning electron microscope (FF-SEM). Physical stability of nanoemulsion was not significantly changed during storage at both light and thermal condition for a month with zeta potential value of -41 mV meaning stable colloid.

Effects of Using Illite Warm Water Mats on Lactate, CRP and ACR Induced High Intensity Exercise in Adults (일라이트 온수매트 사용이 고강도 운동으로 유발된 성인의 젖산, CRP, ACR에 미치는 영향)

  • Purpose : The purpose of this study was to investigate the effective recovery method of exercise-induced fatigue and muscle pain by comparing the effect of the use of illite hot mat product and general hot mat product on the recovery of muscle pain induced by high intensity exercise. Methods : To measure and analyze the changes in lactic acid, CRP, and ACR according to the high-intensity circuit training program, this study was conducted for the healthy adult men and women, who exercise st the K-region sports center. A total of 45 subjects were studied in 15 groups of 15 patients who received an illite hot-water mat recovery group (A group), 15 general hot-water mat recovery group (B group), and 15 control group (C group). The circuit training exercise program was conducted as a one-time exercise, and each exercise time consisted of 30 minutes of warm-up exercise, 5 minutes of main exercise, 20 minutes of clean-up exercise, and 5 minutes of strength exercise. The intensity setting was high intensity of subjective exercise intensity It carried out by setting to (14-16RPE). Results : Changes in Lactic Acid Concentration There was a significant difference in the lactic acid concentrations between the groups after the high intensity circuit training program (p <.05). The illite rest group (A) decreased 7.71 mmol / L and the control group decreased 4.03 mmol / L. Significantly decreased (p <.05). Changes in ACR Concentration. There was a significant difference in the ACR concentrations Significant differences were found in CRP and ACR during the recovery period after exercise. (p <.05), the elite rest group (A) decreased 2.47 mg / mmol, and the control group increased 1.63 mg / mmol. There was a significant difference (p <.05). Conclusion: The static rest on a heated mat after high-intensity exercise has an effect on changes in blood lactate and ACR levels.
