  • Cobrotoxin의 항염증 치료 기전에 대해 연구하기 위하여 성상세포에 LPS 및 SNP로 염증을 유도한 후 NF-${\kappa}B$와 DNA의 결합 능력, NF-kB Dependent Luciferase 발현, Astrocyte의 세포활성, NF-${\kappa}B$ 구성 단백질인 P50, P-$1{\kappa}B$, $1{\kappa}BB$ 및 염증(炎症)관련 유전자(遺傳子)인 Cox-2, iNOS, cPLA2의 발현과 GSH와 DTT로 sulf-hydryl기를 환원시 NF-${\kappa}B$와 DNA의 결합 능력, NF-${\kappa}B$ 구성 단백질인 P50 발현에 미치는 영향과 Cobrotoxin의 성상세포 내 유입 등을 관찰하여 다음과 같은 결론을 얻었다. 1. LPS 로 염증을 유발한 후 NF-${\kappa}B$와 DNA의 결합 능력을 관찰한 결과 Cobrotoxin 0.1${\mu}g/m{\ell}$ 처리군, Astrocyte 내에서의 Cobrotoxin 0.1, 0.5${\mu}g/m{\ell}$ 처리군에서 모두 대조군에 비하여 유의한 억제를 나타내었다. 2. LPS로 염증을 유발한 후 Astrocyte 내에서 NF-${\kappa}B$ Dependent Luciferase 발현을 살펴본 결과 Cobrotoxin 모든 처리군에서 대조군에 비하여 유의한 억제를 나타내었다. 3. SNP로 염증을 유발한 후 Cobrotoxin이 NF-${\kappa}B$ 구성 단백질인 P50, P-$1{\kappa}B$, $1{\kappa}B$ 발현에 미치는 영향을 살펴본 결과 P50와 $1{\kappa}B$는 Cobrotoxin 0.1, 0.5 및 $1{\mu}g/m{\ell}$ 모든 처리군에서 대조군에 비하여 유의한 억제를 나타내었고, P-$1{\kappa}B$는 Cobrotoxin $0.1{\mu}g/m{\ell}$ 처리군에서 대조군에 비하여 억제를, Cobrotoxin 0.5, $1{\mu}g/m{\ell}$ 처리군에서 각각 대조군에 비하여 유의한 억제를 나타내었다. 4. LPS로 염증을 유발한 후 Cobrotoxin이 NF-${\kappa}B$ 구성 단백질인 P50, P-$1{\kappa}B$, $1{\kappa}B$ 발현에 미치는 영향을 살펴본 결과 P50와 $1{\kappa}B$는 Cobrotoxin 0.5, $1{\mu}g/m{\ell}$ 처리군에서 각각 대조군에 비하여 유의한 억제를 나타내었다. 5. SNP로 염증을 유발한 후 Cobrotoxin이 염증(炎症) 관련 유전자(遺傳子)인 Cox-2, iNOS, cPLA2 발현에 미치는 영향을 살펴본 결과 Cox-2, iNOS, cPLA2 모두 Cobrotoxin $1{\mu}g/m{\ell}$ 처리군에서 대조군에 비하여 유의한 억제를 나타내었다. 6. LPS로 염증을 유발한 후 Cobrotoxin이 염증(炎症) 관련 견전자(遣傳子)인 Cox-2, iNOS, cPLA2 발현에 미치는 영향을 살펴본 결과 Cox-2와 cPLA2의 경우 Cobrotoxin 0.1, 0.5 및 $1{\mu}g/m{\ell}$ 모든 처리군에서, iNOS의 경우 Cobrotoxin 0.5, $1{\mu}g/m{\ell}$ 처리군에서 대조군에 비하여 유의한 억제를 나타내었다. 7. Astrocyte내에서 SNP로 염증을 유발한 후 GSH와 DTT로 sulf-hydryl기를 환원하여 NF-${\kappa}B$와 BNA의 결합 능력을 관찰한 결과 Cobrotoxin $0.5{\mu}g/m{\ell}$과 DTT 1mM Cobrotoxin $0.5{\mu}g/m{\ell}$과 DTT 5mM의 동시처리군은 각각 Cobrotoxin $0.5{\mu}g/m{\ell}$ 처리군에 비하여 유의한 증가를 나타내었다. 8. Astrocyte 내에서 LPS로 염증을 유발한 후 GSH와 DTT로 sulf-hydryl기를 환원하여 NF-${\kappa}B$와 DNA의 결합 능력을 관찰한 결과 cobrotoxin $0.5{\mu}g/m{\ell}$과 DTT 1mM Cobrotoxin $0.5{\mu}g/m{\ell}$과 DTT 5mM의 동시처리군과 Cobrotoxin $0.5{\mu}g/m{\ell}$과 GSH 1mM Cobrotoxin $0.5{\mu}g/m{\ell}$과 GSH 5mM의 동시처리군 모두에서 Cobrotoxin $0.5{\mu}g/m{\ell}$ 처리군에 비하여 유의한 증가를 나타내었다. 9. Astrocyte 내에서 SNP로 염증을 유발한 후 GSH와 DTT로 sulf-hydryl기 환원시 NF-${\kappa}B$ 구성 단백질인 P50 발현에 미치는 영향을 관찰한 결과 SNP, Cobrotoxin $1{\mu}g/m{\ell}$와 DTT 1 또는 5mM 동시처리군과 SNP, Cobrotoxin $1{\mu}g/m{\ell}$와 GSH 1 또는 5mM 동시처리군 모두에서 Cobrotoxin $1{\mu}g/m{\ell}$ 처리군에 비하여 발현의 유의한 증가를 나타내었다. 10. Cobrotoxin의 세포 내 유입 확인을 살펴본 결과 Astrocyte 내에서 Cobrotoxin이 세포내로 유입되는 것으로 나타났다. 이상의 결과로 보아 Cobrotoxin 처리는 성상 세포 들을 대상을 LPS 및 SNP로 유도된 NF-${\kappa}B$ 관련 염증 기전과 iNOS, COX-2, cPLA2와 같은 염증관련 유전자의 발현 및 NO, PGE2,에 유의한 변동을 나타내었고, 이들 결과는 Cobrotoxin의 항염증 효과 및 그 치료기전에 대하여 입증한 것이며, 향후 안전성 연구를 바탕으로 신경계 및 심혈관계 염증치료에 약침개발과 같은 적극적인 활용이 기대된다.

Shikonin Isolated from Lithospermum erythrorhizon Downregulates Proinflammatory Mediators in Lipopolysaccharide-Stimulated BV2 Microglial Cells by Suppressing Crosstalk between Reactive Oxygen Species and NF-κB

  • According to the expansion of lifespan, neuronal disorder based on inflammation has been social problem. Therefore, we isolated shikonin from Lithospermum erythrorhizon and evaluated anti-inflammatory effects of shikonin in lipopolysaccharide (LSP)-stimulated BV2 microglial cells. Shikonin dose-dependently inhibits the expression of the proinflammatory mediators, nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), and tumor necrosis factor-${\kappa}B$ (TNF-${\alpha}$) as well as their main regulatory genes and products such as inducible NO synthase (iNOS), cyclooxygenase-2 (COX-2), and TNF-${\alpha}$ in LPS-stimulated BV2 microglial cells. Additionally, shikonin suppressed the LPS-induced DNA-binding activity of nuclear factor-${\kappa}B$ (NF-${\kappa}B$) to regulate the key regulatory genes of the proinflammatory mediators, such as iNOS, COX-2, and TNF-${\alpha}$, accompanied with downregulation of reactive oxygen species (ROS) generation. The results indicate that shikonin may downregulate the expression of proinflammatory genes involved in the synthesis of NO, $PGE_2$, and TNF-${\alpha}$ in LPS-treated BV2 microglial cells by suppressing ROS and NF-${\kappa}B$. Taken together, our results revealed that shikonin exerts downregulation of proinflammatory mediators by interference the ROS and NF-${\kappa}B$ signaling pathway.

패장(敗醬) 물 추출물의 LPS로 유도된 RAW 264.7 세포와 mouse 염증모델에서 cytokine 및 NF-κB의 활성에 미치는 효과 (Effects of Patrinia Scabiosaefolia Aqueous Extract on Cytokine and NF-κB Activation in LPS-induced RAW 264.7 Cells and Mouse)

  • Objectives: The object of this study was to identify the anti-inflammatory effects of Patrinia scabiosaefolia aqueous extract (PSE). Methods: RAW 264.7 cells were pre-treated with PSE and then incubated with or without lipopolysaccharide (LPS). Cell viability, production of nitric oxide (NO), secretion of pro-inflammatory cytokine, activation of mitogen-activated protein kinases (MAPKs) and nuclear factor-kappa B (NF-${\kappa}B$) were measured. In addition, we observed mice survival rate after LPS and their cytokine levels of serum. We also observed inflammatory and hemorrhagic change on the histological sections of the liver. Results: PSE inhibited LPS-induced NO production, interleukin (IL)-6 secretion, c-Jun NH2-terminal kinase (JNK) and NF-${\kappa}B$ activation. In addition, PSE reduced the death rate of LPS-induced mice and IL-6 production on the serum of mice. PSE inhibited inflammation and hemorrhage on liver tissue as well. Conclusions: The results suggest that PSE have anti-inflammatory effects by inhibited NF-${\kappa}B$ and JNK activation, IL-6 secretion, and NO production. So PSE may be effective treatment for the inflammatory disease.

  • Inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) are important inflammatory mediators implicated in pathogenesis of inflammation and certain types of human cancers. The present study was designed to determine whether Red Ginseng (RG) could modulate $I{\kappa}B$-kinase, iNOS and COX-2 gene expression and immune responses in RAW 264.7 macrophages stimulated with lipopolysaccharide (LPS). RG extract suppressed the expression of LPS-induced $I{\kappa}B$, iNOS, COX-2, and immune responses in a dose-dependent manner. It also showed an anti-inflammatory effect by inhibiting NF-${\kappa}B$ immune response induced by LPS treatment. Inhibitory effect of RG on LPS-induced inflammation was mediated by suppressed phosphorylation of ERK, JNK and p38 through the regulation of the mitogen-activated protein kinase (MAPK) pathway leading to a decreased production of NO, iNOS, COX-2 and NF-${\kappa}B$. The results implied the role of RG as an inflammation regulator and its possible application for curing inflammatory diseases.

  • Flowers of Magnolia denudata has been reported to possess a variety of pharmacological activities. In this study, we investigated the anti-inflammatory effects and mechanism of the water extract of Flowers of Magnolia denudata(MD) in lipopolysacchride (LPS)-mediated inflammatory mediators in murine peritoneal macrophages. MD itself does not have any toxic effects in murine peritoneal macrophages. MD inhibits LPS-induced nitric oxide (NO), tumor necrosis factor $(TNF)-{\alpha}$, IL-6 and IL-12 production in murine peritoneal macrophages. Furthermore, we have found that MD inhibited LPS-induced $NF-{\kappa}B$ but not c-Jun N-terminal kinase (JNK), p38 and extracellular signal-ragulated kinase (ERK) activation. These results suggested that MD inhibit LPS-induced production of $TNF-{\alpha}$, IL-6 and IL-12 via suppression of the $NF-{\kappa}B$ activation.

Aromadendrin Inhibits Lipopolysaccharide-Induced Nuclear Translocation of NF-κB and Phosphorylation of JNK in RAW 264.7 Macrophage Cells

  • Aromadendrin, a flavonol, has been reported to possess a variety of pharmacological activities such as anti-inflammatory, antioxidant, and anti-diabetic properties. However, the underlying mechanism by which aromadendrin exerts its biological activity has not been extensively demonstrated. The objective of this study is to elucidate the anti-inflammatory mechanism of aromadedrin in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophage cells. Aromadendrin significantly suppressed LPS-induced excessive production of pro-inflammatory mediators such as nitric oxide (NO) and $PGE_2$. In accordance, aromadendrin attenuated LPS-induced overexpression iNOS and COX-2. In addition, aromadendrin significantly suppressed LPS-induced degradation of $I{\kappa}B$, which sequesters NF-${\kappa}B$ in cytoplasm, consequently inhibiting the nuclear translocation of pro-inflammatory transcription factor NF-${\kappa}B$. To elucidate the underlying signaling mechanism of anti-inflammatory activity of aromadendrin, MAPK signaling pathway was examined. Aromadendrin significantly attenuated LPS-induced activation of JNK, but not ERK and p38, in a concentration-dependent manner. Taken together, the present study clearly demonstrates that aromadendrin exhibits anti-inflammatory activity through the suppression of nuclear translocation of NF-${\kappa}B$ and phosphorylation of JNK in LPS-stimulated RAW 264.7 macrophage cells.

Magnolol Inhibits LPS-induced NF-${\kappa}B$/Rel Activation by Blocking p38 Kinase in Murine Macrophages

  • This study demonstrates the ability of magnolol, a hydroxylated biphenyl compound isolated from Magnolia officinalis, to inhibit LPS-induced expression of iNOS gene and activation of NF-${\kappa}B$/Rel in RAW 264.7 cells. Immunohisto-chemical staining of iNOS and Western blot analysis showed magnolol to inhibit iNOS gene expression. Reporter gene assay and electrophoretic mobility shift assay showed that magnolol inhibited NF-${\kappa}B$/Rel transcriptional activation and DNA binding, respectively. Since p38 is important in the regulation of iNOS gene expression, we investigated the possibility that magnolol to target p38 for its anti-inflammatory effects. A molecular modeling study proposed a binding position for magnolol that targets the ATP binding site of p38 kinase (3GC7). Direct interaction of magnolol and p38 was further confirmed by pull down assay using magnolol conjugated to Sepharose 4B beads. The specific p38 inhibitor SB203580 abrogated the LPS-induced NF-${\kappa}B$/Rel activation, whereas the selective MEK-1 inhibitor PD98059 did not affect the NF-${\kappa}B$/Rel. Collectively, the results of the series of experiments indicate that magnolol inhibits iNOS gene expression by blocking NF-${\kappa}B$/Rel and p38 kinase signaling.

The pepsinolytic hydrolysate from Johnius belengerii frame inhibited LPS-stimulated production of pro-inflammatory mediators via the inactivating of JNK and NF-κB pathways in RAW 264.7 macrophages

  • The objective of this study was to investigate the anti-inflammatory effects of the pepsinolytic hydrolysate from the fish frame, Johnius belengerii, on lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. The J. belengerii frame hydrolysate (JFH) significantly suppressed nitric oxide (NO) secretion on LPS-stimulated RAW264.7 macrophages. Moreover, the JFH markedly inhibited the levels of protein and mRNA expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). Furthermore, the LPS-stimulated mRNA expression of pro-inflammatory cytokines, including tumor necrosis factor $(TNF)-{\alpha}$, interleukin $(IL)-1{\beta}$, and IL-6 was downregulated when cells were cultured with the JFH. The JFH significantly reduced the phosphorylation of c-Jun N-terminal kinase (JNK) and the translocation of nuclear factor-kappa B ($NF-{\kappa}B$) in macrophages. As the result, the JFH has the potential anti-inflammatory activity via blocking the JNK and $NF-{\kappa}B$ signal pathways.

The Effect of Cobrotoxin on $NF-{\kappa}B$ binding Activity in Raw264.7 cells

  • Cobrotoxin, a venom of Vipera lebetina turanica, is a group of basic peptidescomposed of 233 amino acids with six disulfide bonds formed by twelve cysteins. NF-kB is activated by subsequent release of inhibitory IkB and translocation of p50. Since sulfhydryl group is present in kinase domain of p50 subunit of NF-kB, cobrotoxin could modify NF-kB activity by protein-protein interaction. We therefore examined effect of cobrotoxin on NF-kB activities in lipopolysaccharide (LPS) and sodium nitroprusside (SNP)-stimulated Raw 264.7 mouse macrophages. Cobrotoxin suppressed the LPS and SNP-induced release of IkB and p50 translocation resulted in inhibition of DNA binding activity of NF-kB. Inhibition of NF-kB resulted in reduction of the LPS and SNP-induced production of inflammatory mediators NO and PGE2 generation. The inhibitory effect of cobrotoxin on the NF-kB activity were blocked by addition of reducing agents dithiothreitol and glutathione. These results demonstrate that cobrotoxin inhibits activation of NF-kB, and suggest that pico to nanomolar range of cobrotoxin could inhibit the expression of genes in the NF-kB signal pathway.

발아현미가 LPS로 유도된 지방세포의 염증반응에 미치는 영향 (Effect of Germinated Brown Rice on LPS-Induced Inflammation in Adipocytes)

  • Germinated brown rice (GBR, Orysa sartiva L.) has been reported to have anti-obesity and anti-inflammatory effects. However, the mechanisms underlying these effects in adipocytes are not fully understood. Therefore, this study was conducted to explore the anti-inflammatory mechanisms of GBR on lipopolysaccharide (LPS)-stimulated 3T3-L1 adipocytes. 3T3-L1 adipocytes were pretreated with GBR extracts (0-20 mg/mL) 1 h before LPS stimulation. The mRNA expression of adipokines and Toll-like receptor 4 (TLR4) were measured by RT-PCR. The protein expressions of TLR4-related molecules were detected by western blotting and nuclear factor-${\kappa}B$ ($NF-{\kappa}B$) activation was measured. Our results showed that GBR extract dose-dependently inhibited mRNA expression of LPS-induced tumor necrosis factor-${\alpha}$ ($TNF-{\alpha}$), interleukin-6 (IL-6), and monocyte chemoattractant protein-1 (MCP-1). GBR extract was found to inhibit LPS-induced mRNA expression of TLR4 and protein expression of both myeloid differentiation factor 88 (MyD88) and TNF receptor-associated factor 6 (TRAF6). Furthermore, GBR extract significantly inhibited extracellular receptor-activated kinase (ERK) phosphorylation and $NF-{\kappa}B$ activation. These results suggest that GBR extract has the anti-inflammatory effects on LPS-induced inflammation via inhibition of TLR4 signaling, includingthe ERK and $NF-{\kappa}B$ signaling pathways, in adipocytes.
