Purification and Characterization of an $\alpha$ -L-Arabinofuranosidase from Bacillus sp. DSNC 101 (Bacillus sp. DSNC 101이 생산하는 $\alpha$-L-Arabinofuranosidase의 정제 및 특성)

  • ${\alpha}$-L-Arabinofuranosidase was purified from the culture supernatant of Bacillus sp. DSNC 101. The enzyme had a molecular weight of 56 kDa. Optimum temperature and pH for ${\alpha}$-L-arabinofuranosidase activity were 55$^{\circ}C$ and 7.0 respectively. The Michaelis constant(Km) and maximal reaction velo-city(Vmax) for p-nitrophenyl-${\alpha}$-L-arabinofuranoside were 1.0 mM and 113.6 U/mg protein, respe-ctively. ${\alpha}$-L-Arabinofuranosidase was completely inhibited by HgCl$_2$ and CuSO$_4$. The enzyme was spe-cific for the ${\alpha}$-linked arabinoside in the furanoside configuration. The enzyme was produced during growth on agricultural residue such as rice straw, but not during growth on spelt xylan, glucose or cellobiose.

Effect of Nepalese Pseudo Ginseng Components on Lipolytic Action of Toxohormone-L from Cancerous Ascites Fluid (Nepal산 Pseudo Ginseng 성분이 암독소 호르몬-난의 체지방 분해작용에 미치는 영향)

  • This study was divised to observe an Inhibitory effect toward a lipolytic action of toxohormone-L from large root and small root Nepal pseudo ginseng (NPG ; Nepal products) components by water extract and ethanol precipitate in vitro. Toxohormone-L is known to be a lipolytic factor that was partially purified from the ascites fluid of Sarcoma 180-bearing mice and of patients with hepatoma. The inhibitory effect that inhibited the lipolytic action of toxohormone-L by ethanol Precipitate component of large root NPG (mean 55.5%) was higher (mean 1.37 times) than that of water extract component in final reaction concentration of 500 and 1, 000ug/ml, on the other side inhibitory effect of water extract component in small root NPG (mean 55.5%) was higer (mean 1.14 times) than that of ethanol precipitate component. In a way inhibitory effect of precipitate component In large root NPG(47.6%), when final reaction concentration of sample were 1, 000ug/ml, was about 40% lower than that of Korean red ginseng.

SED modeling of the Class 0 protostar L1527 IRS

  • We model the spectral energy distribution (SED) of the Class 0 protostar L1527 IRS using a dust continuum radiative transfer code RADMC-3D to study the initial condition of gravitational collapse. To constrain the envelope structure, we use the data obtained by Herschel /PACS, which covers the far-infrared regime ($55-190{\mu}m$) where the SED of L1527 IRS peaks. According to our modeling, a more flattened density profile fits the far-infrared SED of L1527 IRS better than the density profile of a rotating and infalling envelope. Thus, we employ the density structure of a Bonnor-Ebert sphere, which consists of the inner flat-topped and the outer power-law regions and is often used for describing the density structure of the youngest sources in the low mass star formation process. A Bonnor-Ebert sphere fits very well the observed SED at ${\lambda}$ > $10{\mu}m$, suggesting that L1527 IRS might collapse from an unstable Bonnor-Ebert sphere rather than a singular isothermal sphere.

  • Probiotic 활성이 높은 L. acidophilus 30SC의 생존성을 증진시키기 위한 기초자료를 얻고자, 열처리 동안 새로이 발현되는 단백질을 일차원 및 이차원 전기영동을 이용하여 확인하였으며, 세포 모양을 주사전자현미경을 사용하여 관찰하였다. $55^{\circ}C$의 heat shock에는 L. acidphilus 30SC의 사멸이 있는 것으로 나타났다. 나머지 처리구는 $37^{\circ}C$에서 계속 배양한 것과 별다른 차이를 나타내지 않았다. $45^{\circ}C$로 heat shock을 준 경우 $37^{\circ}C$에서 배양한 것과 거의 동일하였다. $55^{\circ}C$에서 15분 heat shock을 준 경우 약 22kDa와 25kDa의 단백질들이 새로이 발현된 것으로 나타났으나, 24 kDa와 27kDa로 추정되는 단백질의 발현정도는 낮았음을 확인하였다. 이차원 전기영동을 실시한 결과, $37^{\circ}C$와 비교할 때 $55^{\circ}C$로 heat shock을 준 경우 새로이 5개의 protein spot을 발견할 수 있었다. 그러나 6개의 단백질 spot은 $55^{\circ}C$ heat shock에서 소실된 것으로 확인되어 추가적인 단백질의 분석이 필요한 것으로 생각되었다.

Study on the Effects of the Extracts from Sophora Japonica L. Flowers, Fruits and Branches on Improvements in Skin Condition (회화나무 꽃, 열매, 가지 추출물의 피부개선 효과에 관한 연구)

  • The purpose of this research was to examine the effect of the ehanol extracts of Sophora japonica L. (S. japonica). flowers, fruits and branches on skin enhancement with assessing anti-oxidative, whitening, and wrinkle enhancement effects. Results showed that l,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activities were $17.68{\pm}1.59{\sim}51.40{\pm}1.04$, $27.48{\pm}0.22{\sim}50.89{\pm}0.13$ and $30.79{\pm}0.55{\sim}45.17{\pm}0.83%$, respectively, in 50~1,000 mg/L of concentrations. The capacities of inhibiting tyrosinase of ethanol extracts from S. japonica. flowers, fruits and branches were $0.27{\pm}0.12{\sim}11.38{\pm}0.57$, $0.27{\pm}0.02{\sim}0.82{\pm}0.27$ and $0.09{\pm}0.16{\sim}0.55{\pm}0.27%$, respectively. The capacities of preventing porcine pancreatic elastase (PPE) were $3.70{\pm}1.23{\sim}7.28{\pm}1.01$, $3.06{\pm}2.13{\sim}13.03{\pm}2.99$ and $6.00{\pm}0.96{\sim}9.71{\pm}0.44%$, respectively, in the case of 50~1,000 mg/L of concentrations. It is concluded that the effects of S. japonica. flowers, fruits and branches on skin improvement are varied significantly.

Validation of ICP-MS method for trace level analysis of Pb in plasma (혈장 중 극미량 납 분석을 위한 ICP-MS 분석법 검증)

  • The analytical method of lead in plasma by ICP-MS was validated after securing environment within class 1,000 classification. We tested specificity and accuracy of within-run and between-run. According to measurement of the amount of suspended particulates in a clean room, 0.3~62 particles were detected in 0.3 µm size while 0.0~28.3 particles were observed in 0.5 µm size. Total suspended particulates met required environment with up to 90.3 particles. The MDL (Method detection limit) of the sample which has been fabricated using fetal bovine serum (FBS) blank was 1.77 ng/L, and LOQ (Limit of quantification) was 5.55 ng/L. The slope, intercept and correlation coefficient of the calibration curve were y=1.09×10−3x+4.88×10−2 and r=0.9999, which showed good correlation. The specificity, within-run and between-run accuracy satisfied the standard at more than 50 ng/L. The average lead concentration in plasma of the general people, current workers and retired workers was 55.4 ng/L, 440 ng/L, and 132 ng/L.

Effects of Media and Temperatures on Micro Stem Cutting of Dendrobium nobile 'Hamana Lake Dream' × 'No. 55' in Vitro (Dendrobium nobile 'Hamana Lake Dream' × 'No.55'의 기내 줄기삽목에 미치는 배지의 종류와 온도의 영향)

  • The optimal growth condition of in vitro stem cutting of Dendrobium nobile 'Hamana Lake Dream' ${\times}$ 'No. 55' was investigated. Among various media and their concentrations, MS media had better effect on the growth of micro stem cutting than Hyponex media in all concentration levels except stem length. The activated charcoal concentration in MS media showed different effects on number of stem and root, leaf length, and fresh weight: the most effective in the range of 0.1 to 1.0 g/L and barely effective above 2.0 g/L. Addition of agar 5 g/L, sucrose at 40 g/L, and peptone at 1 g/L to MS media increased significantly stem length, leaf width, and fresh weight, internode length and number of roots, and the number of stem and leaves. On the other hand, addition of gelite with any concentration had no effect on the growth of micro stem cutting compare to that of control. The optimal temperature for growth of micro stem cutting was $28^{\circ}C$. Under the same temperature, MS medium was better than Hyponex medium for the growth of stem. In addition, sucrose at 40 g/L was the most effective on growth at $28^{\circ}C$.

Kinetic Properties of $\alpha$-Galactosidase from Aspergillus niger ATCC 16513 and Soybean(Glycine max. L) (Aspergillus niger ATCC 16513과 대두(Glycine max. L) $\alpha$-galactosidase의 kinetic 성질)

  • This experiment was carried out to elucidate some kinetic properties of the $\alpha$-galactosidase which produced and purified from Aspergillus niger ATCC 16513 and soybean(Glycine max. L). The Km value of Asp. niger and soybean $\alpha$-galactosidase were 37.0mM and 50.0mM for raffinose and55.5mM and 55.5mM for stachyose, respectively. The activity of Asp. niger and soybean $\alpha$-galactosidase were inhibited by galactose. Among the amino acids in active sites of both Asp. niger and soybean $\alpha$-galactosidase, histidine was identified by chemical modification of diethyl pyrocarbonate. Number of amino acids residues per mole of Asp. niger and soybean $\alpha$-galactosidase were 902 and 286, respectively.
