자생 생강나무의 조경적 특성 및 번식방법에 관한 연구 (A Study on the Landscape Characteristics and Propagation Methods of Korean native Lindera obtusiloba Blume)

  • This study was carried out to investigate the growth characteristics and propagation methods of L. obtusiloba as a woody landscape plant. The results are obtained as follows: L. obtusiloba was dioecious shrub and shaped with bush type. Leaves were ovate, obtuse, cordate, 7.05 cm long, 7.20 cm wide, and petiole 2.0 cm long. Flowers of L. obtusiloba were diclinous. Soil acidity ranged from pH 4.06 to 5.53 with the lowest at the Mt. Soo-Ri. Mt. Soo-Ri located near factory district, which was considered to damaged by environmental deteoration. While soil organic matter was highest at Mt. Soo-Ri, inorganic nutrients were low. L. obtusiloba grows in the area with low soil acidity and low content of inorganic. Therefore it seemed to be tolerant to air pollution. L. Obtusiloba was high seed germination rate in the plug box and its shoots were longer than seeding box and softwood cutting of L. obtusiloba showed the rooting rate of 50% at 5,000ppm on June 23. To develop a mass propagation method of Korean native L. obtusiloba through an axillary bud culture as a woody landscape plant, about 2∼3 cm shoots induced from explant were subcultured to new media contained different growth regulators. Shoots multiplied most effectively on a WPM containing 1.0 mg/l BA, producing 5.5 shoots with a shoot length of 2.5 cm per shoot explant.

생강나무 잎 및 가지 추출물의 생리활성 (Physiological Activities of Leaf and Twig Extracts from Lindera obtusiloba Blume)

  • We investigated the physiological activities of extracts from Lindera obtusiloba Blume leaf and twig (LLW: water extract from Lindera obtusiloba Blume leaf, LLE: 50% ethanol extract from Lindera obtusiloba Blume leaf, LTW: water extract from Lindera obtusiloba Blume twig, LTE: 50% ethanol extract from Lindera obtusiloba Blume twig). Total polyphenol and total flavonoid contents of LTE were 445.38 mg/g and 302.09 mg/g, respectively. The electron donating ability (95.38%) of LTE was higher than that of the LLE (93.76%), LTW (88.09%), and LLW (82.06%). The oxygen radical absorbance capacity of extracts were improved with 50% ethanol condition, rather than hot water. Superoxide radical scavenging activity and FRAP activity of the extracts were improved with an increase of treatment concentration. All the extracts($1,000{\mu}g/mL$) stimulated a production of nitric oxide (NO) in macrophage RAW264.7 cells. In particular, the NO stimulating activity of LTE was superior to that of LLE, LTW, and LLW. The antitumor activity of LTE ($500{\mu}g/mL$) in A549, HeLa and SNU719 was 55.63%, 83.87% and 68.11%, respectively. The UVB-induced MMP-1 production in HS68 cells was suppressed by the treatment of LTE (88.28%), LLE (83.96%), LTW (80.59%) and LLW (76.08%).

Analysis of the genetic diversity and population structure of Lindera obtusiloba (Lauraceae), a dioecious tree in Korea

  • Lindera obtusiloba (Lauraceae) is a dioecious tree that is widely distributed in the low-altitude montane forests of East Asia, including Korea. Despite its various pharmacological properties and ornamental value, the genetic diversity and population structure of this species in Korea have not been explored. In this study, we selected 6 nuclear and 6 chloroplast microsatellite markers with polymorphism or clean cross-amplification and used these markers to perform genetic diversity and population structure analyses of L. obtusiloba samples collected from 20 geographical regions. Using these 12 markers, we identified a total of 44 alleles, ranging from 1 to 8 per locus, and the average observed and expected heterozygosity values were 0.11 and 0.44, respectively. The average polymorphism information content was 0.39. Genetic relationship and population structure analyses revealed that the natural L. obtusiloba population in Korea is composed of 2 clusters, possibly due to two different plastid genotypes. The same clustering patterns have also been observed in Lindera species in mainland China and Japan.

생강나무 추출물의 항산화 효과 및 타이로시네이즈 저해 활성 (Antioxidative Effect and Tyrosinase Inhibitory Activity of Lindera obtusiloba Blume Extracts)

  • 본 연구의 목적은 생강나무 추출물의 항산화 활성과 타이로시네이즈 저해 활성을 살펴봄으로써 화장품 원료로서의 응용 가능성을 확인하는 것이다. 모든 실험은 생강나무의 50 % 에탄올 추출물, 에틸아세테이트(ethyl acetate) 분획, 아글리콘(aglycone) 분획을 이용하여 진행하였다. 에틸아세테이트 분획의 DPPH (1,1-diphenyl-2-picrylhydrazyl) 소거활성은 기존에 잘 알려져 있는 항산화제인 (+)-${\alpha}$-tocopherol 보다 높은 것으로 나타났다. $Fe^{3+}$-EDTA/ $H_2O_2$ 계에서 생성된 활성산소종에 대한 세 분획의 소거활성(총항산화능)은 대표적인 항산화제인 L-ascorbic acid와 비슷한 것으로 나타났다. Rose-bengal로 증감된 $^1O_2$에 의한 적혈구 파괴에서, 50 % 에탄올 추출물과 에틸아세테이트 분획의 세포보호 효과는 농도 의존적($1{\sim}25{\mu}g/mL$)으로 증가하였다. $10{\mu}g/mL$ 농도를 기준으로 비교하였을 때 에틸아세테이트 분획의 ${\tau}_{50}$은 361.0 min으로 가장 높은 세포 보호 활성을 나타내었다. 타이로시네이즈 저해활성에서 에틸아세티이트 분획과 아글리콘 분획은 알부틴보다 높은 저해 효과를 나타내었다. 이상의 결과들로부터 생각나무 추출물은 활성산소종을 소거하는 항산화제로 여러 산업 분야에 응용 가능할 것이라 생각된다. 또한 알부틴을 대체하는 미백 기능성 소재로서 응용될 수 있는 가능성을 확인하였다.

  • 생강나무 (Libdera obtusiloba BL.)의 페놀성 화합물들을 분리하기 위하여 잎고 줄기의 MeOH 엑스로부터 계통 분획한 EtOAc 분획물은 silica gel column chro-matography로 분리하여 2종의 화합물을 얻었다. IR, UV,$^1H-NMR$, $^13C-NMR$ 등의 분석을 행하여 화학구조를 밝힌 결과 이들 화합물의 화학구조는 잎에서 $quer-cetin-3-O-\alpha-L-rhamnopyranoside(quercitrin),$ 줄기에서는 quercetin $3-O-\beta-D-galactoyranoside(hyperoside)로$ 각각 결정하였다. 이 화합물들은 생강나무에서 처음으로 분리하였다.

GC-MS를 이용한 계절에 따른 생강나무(Lindera obtusiloba BL.)의 부위별 향기 성분 비교 분석 (Comparison Analysis of Essential Oils Composition in Difference Parts from Lindera obtusiloba BL. according to the Season by Gas Chromatography-Mass Spectrometry (GC-MS))

  • The essential oils of stems, roots, fruits and leaves of Lindera obtusiloba BL. were collected in the winter and summer extracted by simultaneous distillation extraction (SDE) apparatus and analyzed by gas chromatography-mass spectrometry (GC-MS). In present study, 58 kinds of volatile components in the winter stems (WS), 70 in the winter roots (WR), 77 in the summer stems (SS), 78 in the summer roots (SR), 70 in the summer fruits (SF) and 76 in the summer leaves (SL) were identified. The results showed that, the major components were monoterpenes including ${\alpha}$-thujene (1.22~13.80%) camphene (1.56~18.40%), ${\beta}$-mycrene (1.75~9.27%), limonene (5.57~12.83%), ${\beta}$-phellandrene (3.03~7.72%), linalyl acetate (2.29~12.55%), dihydromycrene (0~11.15%), germacrene B (0~7.54%) of which the contents had major fluctuations in different seasons and parts. In general, monoterpenes were the major constituent of SF in L. obtusiloba BL. that have presented possibilities for industrial applications.

생강나무(Lindera obtusiloba)의 잎에서 분리한 내생균의 다양성 (Diversity of Foliar Endophytic Fungi Isolated from Lindera obtusiloba in Korea)

  • 생강나무에 공생하는 내생균의 다양성을 확인하기 위하여, 강원도 4 개 지역에서 병증이 없는 생강나무 잎을 채집하여 내생균을 분리하였다. 분리된 내생균의 rDNA의 ITS지역을 분석한 결과, Alternaria alternata, Annulohypoxylon annulatum, Creosphaeria sassafras, Diaporthe eres, Discosia sp., Epicoccum nigrum, Glomerella acutata, Glomerella cingulata, Paraconiothyrium brasiliense, Pestalotiopsis neglecta, Phomopsis amygdali, Xylaria sp.의 총 7과 11속 12종이 확인되었으며 Phomopsis amygdali가 모든 연구 지역의 생강나무 잎에서 가장 높은 빈도로 분리되었으며 채집지에 따라 내생균의 종 다양성에 차이가 있음을 확인하였다.

Sedative and Antinociceptive Properties of Lindera obtusiloba

  • The stem of Lindera obtusiloba (Lauraceae), has been widely used as a traditional medicine for the treatment of abdominal pain, bruise and hepatocirrhosis. In the present study, antinociceptive and sedative properties of the methanol extract of L. obtusiloba (MLO) were evaluated. MLO demonstrated strong and dose-dependent antinociceptive activities on various experimental pain models including thermal nociception and chemical nociception, compared to tramadol and indomethacin, reference drugs. In combination test using naloxone, the diminished analgesic activity of MLO was observed, indicating the relation with opioid receptor. Moreover, MLO also decreases pentobarbital-induced sleep latency and increases sleeping time suggesting its hypnotic and sedative action. The present results indicate that MLO could be used as valuable antinociceptive and sedative agent for the treatment of various diseases.

생강나무 추출물의 알레르기성 염증반응 억제 효과 (Inhibitory Effects of Water Extract of Lindera obtusiloba on the Mast Cell-Mediated Allergic Inflammation)

  • Lindera obtusiloba has been used for centuries as a traditional medicine in Korea and recently known to have an anti-fibrotic effect. In this report, we investigated the effect of hot water extract from L. obtusiloba (WELB) on the mast cell-mediated allergic inflammation and studied its possible mechanisms of action. WELB inhibited phorbol-12-myristate 13-acetate and calcium ionophore A23187 (PMACI)-induced histamine release in HMC-1 human mast cells. WELB reduced PMACI-induced gene expression and secretion of proinflammatory cytokines such as tumor necrosis factor-$\alpha$, interleukin (IL)-$1{\beta}$, IL-6, and IL-8. The inhibitory effect of WELB on the expression of proinflammatory cytokines was c-jun N-terminal kinase and nuclear factor-${\kappa}B$ dependent. These results indicate that WELB may be beneficial in the treatment of mast cell-mediated allergic inflammation.

Two New Lignans from Lindera obtusiloba Blume

  • Two new furanolignans (3, 5), together with three known lignans (1, 2, 4,), were isolated from the stem of Lindera obtusiloba (Lauraceae). The structures of the compounds were determined as actifolin (1), pluviatilol (2), 5,6-dihydroxymatairesinol (3), (+)-syringaresinol (4), and $(+)-9^{l}$-O-trans-feruloyl-5,51-dimethoxylariciresinol (5) on the basis of physicochemical and spectroscopic evidences. Compounds 1, 2, 3, and 5 showed cytotoxicity against a small panel of human tumor cell lines with values of $3.40{\sim}19.27 {\mu}g/ml$.
