A Study on the Distribution of Wild Edible Herb Species in Mt. Kariwang (가리왕산의 산채(山菜) 분포(分布)에 관(關)한 연구(硏究))

  • This study was conducted to maintain the productivity of forest production and byproducts production of natural forest. Distribution of wild edible herb species and correlations between wild edible herb species and forest-type, and site factors were surveyed on national forest in Changjeon-ni, Pyungchang-gun, Kang-won-do. Ligularia fischeri was distributed mainly on cutting sites or meadow or Quercus mongolica forest, Pimpinella brachycarpa did on broadleaved mixed forest, and Synurus deltoides did cutting sites or meadow. On conifer plantations major wild edible herb species were not distributed. Pimpinella brachycarpa and Ligularia fischera were distributed mainly on the site of wet soil-moisture condition, Codonopsis lanceolata, Saussurea grandifolia and Synurus deltoides were distributed randomly. The distribution of Ligularia fischeri and Pimpinella brachycarpa were significantly different between slope directions, but those of Codonopsis lanceolata, Saussurea grandifolia and Synurus deltoides were not significant. Ligularia fischeri and Pimpinella brachycarpa were distributed more frequently in north directed slope than the other. Highly positive correlations were proved Ligularia fischeri and Pimpinella brachycarpa, Aconitum jaluense and Veratrum patulum; Pimpinella brachycarpa and Smilacina japonica, Aconitum jaluense and Veratrum patulum.

Basic Performance Evaluation of the Ecotoxicity Detection Device for Heavy Metals (중금속류 생태독성 검출장치의 기초성능 평가)

  • The ecotoxicity detection device for preliminary test (Test jig) was manufactured to develop the biological early warning system using Vibrio fischeri. In this study, the ecotoxicity detection charateristics of the Test jig was investigated for 6 heavy metals (Cr, Zn, Pb, Cd, Cu and Hg). It was observed that relative luminescence unit (RLU) of Vibrio fischeri constantly decreased by the concentrations of the tested heavy metals. In contrast with other heavy metals, RLUs of Pb and Hg constantly decreased even at low concentrations. RLU of Hg drastically decreased when its concentration increased from 0.13 mg/L to 0.25 mg/L. $EC_{50}$ values of Cr, Zn, Pb and Cd gradually decreased with exposure time, whereas there was no significant change in $EC_{50}$ values of Cu and Hg with time. On the other hand, $EC_{50}$ values between the Test jig and Reference device were compared to evaluate the ecotoxicity detection performance of the Test jig. No big difference was found in $EC_{50}$ vlaues between the two devices, indicating that the Test jig could be applied as the ecotoxicity detection device for heavy metals.

Protective Effect of 3,5-Dicaffeoylquinic Acid Isolated from Ligularia fischeri against Oxidative Damage in HepG2 Cells (HepG2 세포에서 산화적 손상에 대한 곰취 유래 3,5-Dicaffeoylquinic Acid의 보호 효과)

  • This study was conducted to investigate the hepatoprotective effects of 3,5-dicaffeoylquinic acid (3,5-DCQA) isolated from Ligularia fischeri against hydrogen peroxide-induced oxidative stress in HepG2 cells. Antioxidative effects of 3,5-DCQA were determined by measuring antioxidant enzyme [superoxide dismutase (SOD), catalase (CAT) glutathione peroxidase (GPx)] expression levels against hydrogen peroxide-induced oxidative stress using real-time PCR analysis. 3,5-DCQA treatment significantly increased gene expression levels of SOD, CAT, and GPx in a dose-dependent manner ($10{\sim}30{\mu}g/mL$) in HepG2 cells. Hepatoprotective effects were analyzed by measuring glutamic oxaloacetic transaminase (GOT), lactate dehydrogenase (LDH), and gamma-glutamyl transferase (GGT) activities using a biochemistry analyzer in hydrogen peroxide-treated HepG2 cells. 3,5-DCQA treatment significantly reduced GOT, LDH, and GGT activities in a dose-dependent manner ($10{\sim}30{\mu}g/mL$) against increased liver function index enzyme activities induced by hydrogen peroxide oxidative stress in HepG2 cells. The results reveal that 3,5-DCQA compound isolated from Ligularia fischeri can be useful for the development of an effective hepatoprotective agent.

A Study on the Degradation of Parathion and Reduction of Acute Toxicity in Solar $TiO_2$ Photocatalysis (태양광 $TiO_2$ 광촉매를 이용한 Parathion의 분해와 독성저감에 관한 연구)

  • The photocatalytic degradation of methyl parathion was carried out using a circulating $TiO_2$/solar system. Under the photocatalytic condition, parathion was more effectively degraded than by the photolysis or $TiO_2$ only condition. The parathion degradation followed pseudo first-order kinetics. With photocatalysis, 10 mg/L parathion was completely degraded within 90 min with a TOC decrease exceeding 63% after 150 min. The nitrogen from parathion was recovered mainly as ${NO_2}^-$, ${NO_3}^-$, and ${NH_4}^+$, 80% of sulfur as ${SO_4}^{2-}$, and less than 5% of phosphorus as ${PO_4}^{3-}$ during photocatalysis. The organic intermediates 4-nitrophenol and paraoxon were also identified, and these were further degraded. Two different bioassays using V. fischeri and D. magna were employed to measure the toxicity reduction in the solutions treated by both photocatalysis and photolysis. Relative toxicity was reduced almost completely after 150 min in both organisms under the photocatalysis, whereas in photolysis, 76 and 57% reduction was achieved for V. fischeri and D. magna, respectively. The acute toxicity reduction pattern corresponded with the decrease in parathion and TOC concentrations.

  • Four types of polyphenol oxidase were isolated from the crude extract of a Ligularia fischeri by gel filtration on Sephadex G-150. Optimum pH and temperature for the activity of partially purified enzyme were 7.5 and $25^{\circ}C$, respectively. It was stable at temperature $40^{\circ}C$ when examined at pH 7.5 for 5 min, and lost 90% of its activity at $70^{\circ}C$ in 30 min at pH 7.5. The enzyme has good activity on catechol and chlorogenic acid but was inactive on dopamine.

Quality Characteristics of Tofu Added Ligularia fischeri Powder (곰취분말을 첨가한 두부의 품질특성)

  • The quality characteristics of tofu added Ligularia fischeri powder (LFP) were investigated. The proximate composition of LFP used was as follows: moisture, 7.7%; crude protein, 12.0%; crude lipid, 5.9%; crude ash, 14.1%; and carbohydrate, 60.3%. The yield of tofu added LFP increased with the addition of LFP. However, there was a significant decrease in pH (from $6.03{\pm}0.11$ in the control to $5.78{\pm}0.11$ when 0.4% LFP was added) and a significant increase (from $2.60{\pm}0.01$ in the control to $2.85{\pm}0.10$ when 0.4% LFP was added) in total acidity. In addition, the L, a, and b values of tofu decreased with the increasing addition of LFP. In terms of textural properties, the hardness, cohesiveness, and brittleness increased, whereas springiness decreased, with the increasing addition of LFP. In the sensory evaluation, the overall preference for tofu added 0.3% LFP was the highest. According to the results, the addition of LFP positively affects the overall sensory evaluation of tofu, and 0.3% is the optimal level for addition.

Quality Characteristics of Dried Noodles Added with Ligularia fischeri Powder (곰취 분말을 첨가한 국수의 품질특성)

  • This study investigated the quality of noodles containing different amounts of Ligularia fischeri powder (LFP). Noodles were prepared at ratios of 0, 1, 3, 5, and 7% LFP based on flour weight. Cooking quality, mechanical texture properties, and viscosity were measured, and a sensory evaluation was performed with the prepared noodles. Gelatinization points of the composite LFP-wheat flours increased. As measured via amylography, viscosity at $95^{\circ}C$, viscosity at $95^{\circ}C$ after 15 minutes, and maximum viscosity values of samples decreased, as the LFP content increased. As increasing amounts of LFP were added, the L, a, and b values decreased while color values, weight, and volume of cooked noodles increased, as did the turbidity of the soup. With regard to textural characteristics, LFP additive increased hardness, cohesiveness, and springiness, while decreasing adhesiveness. Sensory evaluation showed that high quality cooked noodles could be produced by inclusion of 3% LFP.

Ecotoxicity of Daphnia magna and Aliivibrio fischeri on Potentially Harmful Substances Emissionsfrom Battery Manufacturing Processes: Lithium, Nickel, and Sulfate (배터리 제조공정에서 배출되는 잠재 유해 물질에 대한 물벼룩과 발광박테리아의 생태독성: 리튬, 니켈, 황산염을 대상으로)

  • Wastewater generated in the secondary battery production process contains lithium and high-concentration sulfate. Recently, as demand as demand for high-Ni precursors with high-energy density has surged, nickel emission is also a concern. Lithium and sulfate are not included in the current water pollutant discharge standard, so if they are not properly processed and discharged, the negative effect on future environment may be great. Therefore, in this study, the ecotoxicity of lithium, nickel, and sulfate, which are potential contaminants that can be discharged from the secondary battery production process, was evaluated using water flea (Daphnia magna) and luminescent bacteria (Aliivibrio fischeri). As a result of the ecotoxicity test, 24-hour and 48-hour D. magna EC50 values of lithium were 18.2mg/L and 14.5mg/L, nickel EC50 values were 7.2mg/L and 5.4mg/L, and sulfate EC50 values were 4,605.5mg/L and 4,345.0mg/L, respectively. In the case of D. magna, it was found that there was a difference in ecotoxicity according to the contaminants and exposure time (24 hours, 48 hours). Comparing the EC50 of D. magna for lithium, nickel, and sulfate, the EC50 of nickel at 24h and 48h was 39.6-37.2% compared to lithium and 0.1-0.2% compared to sulfate, which was the most toxic among the three substances. The difference appeared to be at a similarlevelregardless of the exposure time. The EC50 of sulfate was 253.0-299.7% and 639.5-804.6%, respectively, compared to lithium and nickel, showing the least toxicity among the three substances. The 30-minute EC50 values of luminescent bacteria forlithium, nickel, and sulfate were 2,755.8mg/L, 7.4mg/L, and 66,047.3mg/L,respectively. Unlike nickel, it was confirmed that there was a difference in sensitivity between D. magna and A. fischeri bacteria to lithium and sulfate. Studies on the mixture toxicity of these substances are needed.

A Comparison of Volatile Flavor Characteristics of Chwi-namuls by Terpenoid Analysis (Terpenoid 분석을 통한 취나물류의 향기지표물질 비교)

  • A comparison of essential oils composition of Aster tataricus L. (gaemichwi), Ligularia fischeri (gomchwi), Solidago virga-aurea var. asiatica Nakai (miyeokchwi), and Aster scaber (chamchwi) was performed by gas chromatography and mass spectrometry for the identification of volatile flavor characteristics in chwi-namuls. The essential oils were extracted by the hydro distillation extraction method. One hundred volatile flavor components were identified from gaemichwi essential oil. ${\alpha}$-Pinene (11.5%) was the most abundant compound, followed by myrcene (8.9%) and ${\beta}$-pinene (7.5%). Ninety-one volatile flavor components were identified from the essential oil of gomchwi. Aromadendrene (14.8%) was the most abundant component, followed by ${\beta}$-caryophyllene (7.6%) and 1-methyl-4-(1-methylethylidene)-cyclohexene (7.3%). Ninety-five volatile flavor constituents were detected in the essential oil of miyeokchwi, moreover, spathulenol (15.7%) was the most abundant component. Ninety-six volatile flavor constituents were detected in the essential oil of chamchwi. Epi-bicyclosesquiphellandrene (21.9%) was the most abundant component, followed by ${\beta}$-caryophyllene (9.5%) and ${\delta}$-terpinene (8.9%). The essential oil composition of gaemichwi was characterized by a higher contents of pinenes. The essential oil composition of gomchwi can be easily distinguished by the percentage of aromadendrene. Spathulenol and epi-bicyclosesquiphellandrene were regarded as the characteristic odorants of miyeokchwi and chamchwi, respectively.

Phylogenetic Relationship of Ligularia Species Based on RAPD and ITS Sequences Analyses (RAPD 및 ITS 염기서열 분석을 이용한 곰취 속(Ligularia) 식물의 유연관계 분석)

  • The genetic relationships in 5 species of $Ligularia$ were investigated using RAPD (Randomly Amplified Polymorphic DNA) and ITS (Internal Transcribed Spacer) sequences analyses. In RAPD analysis, sixty three of 196 arbitrary primers showed polymorphism. The amplified fragments ranged from 0.2 to 1.6 kb in size. The dendrogram was constructed by the UPGMA clustering algorithm based on genetic similarity of RAPD markers. A total of 16 accessions were classified into 5 major groups corresponding each species at the similarity coefficient value of 0.77. In the ITS sequence analysis, the size of ITS 1 was varied from 248 to 256 bp, while ITS 2 was varied from 220 to 222 bp. The 5.8S coding region was 164 bp in lengths. Forty nine sites (10.2%) of the 478 nucleotides were variable, and the G+C content of ITS region ranged from 49.4 to 53.5%. In the ITS tree, five species of $Ligularia$ were monophyletic, and $L.$ $taquetii$ was the first branching within the clade. $Ligularia$ $intermedia$ formed a clade with $L.$ $fischeri$ var. $spiciformis$ (BS=79), and $L.$ $stenocephala$ and $L.$ $fischeri$ were also claded. Two data sets were congruent, except of the position of $L.$ $fischeri$ var. $spiciformis$.
