게이트 심근 SPECT를 이용한 좌심실의 국소탄성률 평가방법 개발 및 재현성 평가 (Development of Evaluation Method of Regional Contractility of Left Ventricle Using Gated Myocardial SPECT and Assessment of Reproducibility)

  • 목적: 게이트 심근 SPECT 영상에서 국소부피변화를 얻으면, 요골동맥 긴장도를 측정하여 얻은 중심 동맥의 압력 곡선으로부터 최대탄성률을 얻을 수 있다. 이 연구에서는 좌심실의 국소탄성률을 평가하기 위한 방법을 개발하고 국소탄성률 측정의 재현성을 평가하였다. 대상 및 방법: 게이트 Tc-99m MIBI 심근 관류 SPECT를 두 번 연속으로 시행한 환자 7명(남:여=5:2, $58{\pm}11.9$세)을 대상으로 하였다. 국소적 부피변화를 측정하기 위하여 개발한 CSA(Cardiac SPECT Analyzer) 소프트웨어를 이용하여 좌심실의 부피변화를 측정하였으며, 시간-압력곡선과 국소 시간-부피 곡선을 이용하여 반복연산을 통하여 구한 국소 시간-탄성률 곡선에서 국소탄성률을 얻었다. 같은 SPECT 영상에 대해서 두 번 측정한 국소탄성률의 재현성과, 같은 환자에서 연속하여 두 번 얻은 SPECT 영상에 대해서 측정한 국소탄성률의 재현성을 평가하였다. 결과: 평균 국소탄성률은 15분할모델에서 $3.36{\pm}3.38mmHg/mL$이었으며, 7분할모델과 5분할모델에서 각각 $3.16{\pm}2.25mmHg/mL,\;3.11{\pm}2.52mmHg/mL$이었다. 국소탄성률의 조화평균은 전체탄성률과 일치하였다. 동일한 데이터에서 두 번 측정한 국소탄성률 값의 상관계수는 모든 모델에서 0.97이상이었고, Bland Altman 도표에서 차이의 2-표준편차는 각각 1.5%, 1.0%, 0.9%였다. 동일 환자에서 두 번 연속 촬영한 SPECT 데이터로부터 측정한 국소탄성률의 상관계수는 모든 모델에서 0.95이상이었으며, Bland Altman 도표에서 차이의 2-표준편차는 각각 2.2%, 1.0%, 1.2%였다. 결론: 게이트 심근 SPECT를 이용한 좌심실의 국소탄성률을 측정하였으며 재현성 있는 결과를 얻었다. 심근 SPECT를 이용하여 얻은 국소탄성률의 평가방법은 향후 임상적인 데이터를 바탕으로 새로운 심기능 분석지표로 활용될 수 있을 것이다.

몸통 운동시 지향각(Orientation angles)을 이용한 허리 근육의 3차원 위치 좌표 추정 기법 (The method to estimate 3-D coordinates of lower trunk muscles using orientation angles during a motion)

  • The purpose of this study was to develop a method for estimating 3-D coordinates of lower trunk muscles using orientation angles during a motion. Traditional 3-D motion analysis system with DLT technique was used to track down the locations of eight reference markers which were attached on the back of the subject. In order to estimate the orientations of individual lumbar vertebrae and musculoskeletal parameters of the lower trunk muscle, the rotation matrix of the middle trunk reference frame relative to the lower trunk reference frame was determined and the angular locations of individual lumbar vertebrae were estimated by partitioning the orientation angles (Cardan angles) that represent the relative angles between the rotations of the middle and lower trunks. When the orientation angles of individual intervertebral joints were known at a given instant, the instantaneous coordinates of the origin and insertion for all selected muscles relative to the L5 local reference frame were obtained by applying the transformation matrix to the original coordinates which were relative to a local reference frame (S1, L4, L3, L2, or L1) in a rotation sequence about the Z-, X- and Y-axes. The multiplication of transformation matrices was performed to estimate the geometry and kinematics of all selected muscles. The time histories of the 3-D coordinates of the origin and insertion of all selected muscles relative to the center of the L4-L5 motion segment were determined for each trial.

요추유합술에서 응력방패 현상 감소를 위한 케이지의 유한요소해석 : CFRP 케이지와 티타늄 케이지 비교 연구 (Finite Element Analysis of Instrumented Posterior Lumbar Interbody Fusion Cages for Reducing Stress Shielding Effects: Comparison of the CFRP cage and Titanium cage)

  • 본 논문에서는 척추체 간 유합용 케이지의 응력방패현상을 감소시키기 위하여 강성 차이에 대한 연구를 수행하였다. 최근 의료 임플란트 분야에서는 탄소섬유강화 폴리머를 이용하여 좋은 결과를 보여 왔다. 그러나 생체 역학적으로 이 재료에 대하여 요추체의 안정성과 골 이식재가 받는 응력에 관련한 연구는 없었다. 따라서 이전에 유효화한 요추체 (L2-L5) 비선형 유한 요소 모델을 이용하여 L4-L5 분절의 케이지의 강성 차이에 따른 효과를 알아보기 위하여 탄소섬유강화 폴리머와 티타늄 케이지를 이용한 후방 요추체 유합 모델을 만들었다. 자가골 보다 강성이 작은 탄소 섬유강화폴리머 케이지는 인접 분절의 하종판에 응력이 적게 걸리며, 골 이식재에 응력은 증가시켰다. 위의 결과로 탄소섬유강화 폴리머 케이지는 응력 방패 현상을 감소시킬 수 있을 뿐만 아니라 골 유합률을 증가시킬 수 있다.

  • AB -type block copolymers of poly($\tau$-benzyl L-glutamate) (PBLG, a segment) and polyoxypropylene (POP, Bsegment) were synthesized and they were reacted with polyoxypropylene his(6-aminohexyl) (POE) to enhance the hydrophilicity of the poly- mers. The degree of swelling of the POE -crosslinked block copolymers (CPB -2) was higher than that of the block dopolymers (PB). The amount of 5-fluorouracil re- leased from CPB-2 block copolymers was greater than that from the PB block copol- ymers when they were incubated in the same period. These results indicate that the amount of 5-fluorouracil released from the matrices is proportional to the hydrophilicity of the polymers. Observing polymer surfaces by scanning electron mi- croscope, pores (diameter ; $0.5-1.5{\mu}$m) were appeared on the surface of CPB-2

Plant Regeneration from Turnip (Brassica rapa ssp. rapifera) Organs

  • Shoot induction system was developed in the recalcitrant plant species, Brassica rapa ssp. rapifera by using optimum selection of profit organ, phytohormone combination, seedling age and kind of culture container. Out of in vitro cultured leaf segment, petiole, hypocotyl, and cotyledon with petiole, only cotyledon with petiole derived from 4 day-old seedlings induced multiple shoot. The optimum combination of auxin and cytokinin for the multiple shoot induction was MS medium containing 5mg/L BA and 0.5mg/L NAA. The major factors for multiple shoot propagation were part of plant organ, age of seedling, and ratio of auxin and cytokinin. In addition, shoot regeneration was promoted in the 100ml Erlenmeyer flask compared with the $90mm{\times}20mm$ Petri-dish. The induced shoots formed roots easy on MS medium containing 0.1mg/L IBA and the whole plants were successfully cultivated in soil.

In vitro shoot proliferation of Alnus japonica (Thunberg) Steudel

  • In vitro proliferation system was achieved by using nodal segment excised from greenhouse grown juvenile stock plants of Alnus japonica. Stem explants were cultured on MS medium supplemented with different plant growth regulators of cytokinin and/or their combinations. The most effective cytokinin source was the combination of zeatin 2.0 mg/L and TDZ 0.05 mg/L producing the average number of shoots (16.8 $\pm$ 3.6). In addition, healthy roots were formed after small clumps of shoots were transferred to half strength of MS medium containing IBA 0.02 mg/L with optimal rooting capacity. Soil acclimatization was successfully conducted in cell tray containing artificially mixed soil with 92 % survival rate.

MdMADS2 - transgenic chrysanthemum (Dendranthema grandiflorum (Ramat.) Kitamura) showing the reduction of the days to flowering

  • This study was conducted to develop new lines expressing the characteristic of early flowering by introducing MdMADS2 gene in chrysanthemum (Dendranthema grandiflorum (Ramat.) Kitamura) ‘Zinba'. Transformation of chrysanthemum was conducted by Agrobacterium tumefaciens LBA4404 harboring the binary vector containing MdMADS2 controlled by double CaMV 35S promoters. Ninety three shoots were regenerated from 1,463 leaf segment explants cultured on the first selection medium (MS basal salts + 1.0 mg/L BA + 0.5 mg/L IAA + 10 mg/L kanamycin + 400 mg/L cefotaxime, pH 5.8) after co-cultivation, and 20 out of the 93 shoots rooted on the second selection medium containing 20 mg/L kanamycin and 400 mg/L cefotaxime. Many escapes (98.6%) were removed on the selection stage for rooting. Nineteen lines were confirmed as transgenic plant with transgene by PCR analysis. Six transgenic plants flowered 2-11 days earlier than non-transgenic plant without big change of phenotype, and especially, 3 (Mo-7, Mo-11, Mo-17) out of 6 transgenic lines showed a significant reduction in days to flowering compared to non-transgenic plant. Introduction and expression of MdMADS2 gene in them were confirmed by Southern and real-time PCR analyses, respectively.

한지형 마늘에 있어서 기내뿌리로부터 식물체 재분화 (Plant regeneration from callus derived root of northen type in garlic (Allium sativum L.))

  • This study was conducted to develop an effective production of callus induction and plant regeneration system for garlic transformation. The best callus production occurred on in vitro root segment initially cultured on MS medium with 1.0 mg/L 2,4-D and 0.2 mg/L IAA in both ‘Danyang' and ‘Euseong'. The frequency of callus formation were 81.2% ‘Danyang' and 76.1% ‘Euseong'. Eight weeks after callus induction, callus lines were transferred to regeneration medium during 7 weeks. The best shoot regeneration medium was MS supplemented with 5 mg/L Kinetin and 1 mg/L NAA for ‘Danyang' and MS supplemented with 10 mg/L BAP for ‘Euseong'. The frequency of shoot regeneration were 51.5% ‘Danyang' and 56.6% ‘Euseong' The plantlets were acclimatized and transferred to the greenhouse with almost survival. This in vitro regeneration system should be useful for garlic transformation.
