Objective: The purpose of this study is to verify the usefulness of the rabbit model for disc degeneration study. Materials: The L1-L2, L2-L3, L3-L4. or L4-L5 lumbar intervertebral disc (IVD) of 9 mature male New Zealand White rabbits were injured by inserting a 16-gauge needle to a depth of 5 mm in the left anterolateral annulus fibrosus while leaving L5-L6 IVD uninjured. Three other rabbits also received intradiscal injections of rabbit disc cells transfected with adenovirus and bone morphogenetic protein-2 (ad-BMP-2) at L4-L5 in addition to injury by 16-gauge needle at the L1-L2 level. Using digitized radiographs, measurements of IVD height were made and analyzed by using the disc height index (DHI). Magnetic resonance imaging (MRI) scans of the injured discs, injected discs, and uninjured L5-L6 discs were performed at 15 weeks post surgery and compared with preoperative MRI scans. Results: All twelve rabbits showed consistent results of disc degeneration within 15 weeks following annular puncture. DHIs of injured discs were significantly lower than that of the uninjured L5-L6 discs (p<0.05). The mean value of disc degeneration grade of injured discs was significantly higher than that of uninjured discs (p<0.05). The injection of disc cell transfected with ad-BMP-2 did not induce disc regeneration at 15 weeks after injection. Conclusion: This study showed that the injured disc had a significant change in DHI on simple lateral radiograph and disc degeneration grade on MRI scans within 15 weeks in all rabbits. Rabbit annular puncture model can be useful as a disc degeneration model in vivo.
We investigated the action of NOS inhibitors on NOS in rats. Both of nitric oxide synthase inhibitors, $N^G$-monomethyl-L-arginine $(L-NMMA,\;3\;{\mu}M)$ or $N^G$-nitro-L-arginine methylester $(L-NAME,\;30\;{\mu}M),$ augmented phenylephrine $(PE,\;10^{-7}\;M)-induced$ contraction which was inhibited by acetylcholine (ACh) in rat thoracic aorta. This augmentation by L-NAME or L-NMMA was attenuated with the treatment of NO precursor, arginine. ACh, however, decreased the augmentation induced by L-NMMA, but not by L-NAME. Superoxide dismutase (SOD, 50 u/ml) potentiated an inhibitory effect of ACh on the PE $(10^{-7}\;M)-induced$ contraction. It has been known that platelet activating factor itself induces iNOS. Platelet activating factor $(PAF,\;10^{-7}\;M)$ inhibited PE $(10^{-7}\;M)-induced$ contraction. Pretreatment with L-NMMA (30 mM) or L-NAME (30 mM) significantly blocked the inhibitory action of PAF on PE-induced contraction. L-NMMA (100 mM) or L-NAME (100 mM) reduced nerve conduction velocity (NCV) relevant to nNOS in rat sciatic nerve. ACh attenuated the reduction of NCV by L-NMMA-, but not by L-NAME-induced reduction of NCV. These results suggest that L-NMMA and/or L-NAME have different action on three types of NOS in rats.
This experiment was conducted to propagate Zantedeschia spp. in vitro. The frequency of adventitious bud clusters (ABC) formation from shoot tips in Z. 'Best Gold' was high at more than 65% on media with 2.0∼5.0 mg/L BA or 0.1∼1.0 mg/L thidiazuron. The highest formation rate of ABC (75%) was obtained on medium containing 2.0 mg/L BA. Comparing to treatment of BA alone, combined one of BA and NAA did not stimulate the formation of ABC and the shoot regeneration from shoot tips. The proliferation of ABC from sections (0.7∼1.0 cm) of ABC occurred effective on medium with 2.0 mg/L BA. Shoots developed from the sections (0.7∼1.0 cm) of ABC grew and rooted favorably on media containing 1.0∼2,0 mg/L IBA. The shoots were multiplicated effectively on medium with 0.5 mg/L thidiazuron in Z. 'Childsiana', on medium with 3.0 mg/L BA in 2. 'Golden Affair', and on medium with 5.0∼10.0 mg/L BA in Z. 'Pacific Pink'.

Recently, consume for functional cosmetics containing natural products has been greatly increased. In order to develop it as a natural cosmetic material, we selected Ixeridium dentatum (Thunb.) Tzvelev, Plantago asiatica L. and Rumex crispus L. that have antioxidant and anti-inflammatory effects. In this study, simultaneous quantitative analysis of the isolated compounds and natural product complexes (Mix.) were validated using high performance liquid chromatography (HPLC). The isolated six compounds were shown in a large linearity with a correlation coefficient (R2) of 0.99. The limit of detection (LOD) of chlorogenic acid, plantamajoside, acteoside, emodin chrysophanol and physcion were 0.36 ㎍/mL, 0.36 ㎍/mL, 0.37 ㎍/mL, 0.30 ㎍/mL, 0.22 ㎍/mL and 0.12 ㎍/mL, respectively. The limit of quantification (LOQ) of chlorogenic acid, plantamajoside, acteoside, emodin chrysophanol and physcion were 1.10 ㎍/mL, 1.08 ㎍/mL, 1.12 ㎍/mL, 0.99 ㎍/mL, 0.66 ㎍/mL and 0.35 ㎍/mL, respectively. Content analysis showed chlorogenic acid (0.19 ± 0.02%), plantamajoside (0.48 ± 0.01%), acteoside (0.65 ± 0.01%), emodin (1.15 ± 0.11%), chrysophanol (0.73 ± 0.01%) and physcion (0.69 ± 0.09%). Therefore, the results of this study may provide for basic data of standardization research natural cosmetic material development on the I. dentatum, P. asiatica and R. cripus.
Four strains of Sporobolomyces genera were isolated from soy sauce mashes at the fermentating and aging periods and identified. Their salt-resistance and color formation were examined in order to improve the color density and nutrition of red pepper paste preparations. The results obtained were as follows; a) The strain L1, L2, L3 and L4 isolated were identified as Sporobolomyces gracilis, Sporobolomyces roseus, Sporobolomyes gracilis and Sporobolomyces roseus respectively. b) The yeasts isolated were grown on the media containing $0{\sim}10$ percent of sodium chloride but their growth were almost restrained on the media containing $15{\sim}18$ per cent of sodium chloride. And the strain L2 and L3 were more salt-resistant as compared with L1 and L4. c) The strain L3 were the best among the 4 strains of yeasts comparison with their salt-resistance, color and flavour formation.
The effect of auxins and cytokinins on the formation of adventitious shoots, adventitious roots, trichomes, and calli in MS basal medium was investigated in leaf segments from ecotype Columbia of Arabidopsis thaliana. Adventitious shoots, adventitious roots, trichome, and calli were formed from leaf segments by a wide range of hormone concentrations and combinations. Adventitious shoots were formed respectively in treatment with 0.1mg/L IAA and 10 mg/L BA. Adventitious roots were formed in treatments with low concentration of IAA and NAA. Trichomes and calli were formed by increasing the concentration of IAA and NAA. The optimal combination was 0.5mg/L NAA and 0.1mg/L BA for trichome formation, 10mg/L NAA and 10mg/L BA for calli formation. When NAA was treated alone in culture media, adventitious roots were formed in 0.1mg/L, trichomes were formed in 2.0mg/L, and calli were formed in 10mg/L. Inductive time for formation of adventitious roots, trichomes and calli were determined at 6,7 and 18 days respectively by periodical transfer of leaf segments from NAA containing medium to NAA free medium.
This study was conducted to determine the volume of pig wastewater and It's characteristics from 6 pig farms. The results obtained in this study was summarized as fellow; The volume of pig waste water was 4.15 l/head/day and 4.04 l in spring, 5.20 l in summer, 4.11 l in fall, and 3.44 $\ell$ in winter. The average moisture content of faces and urine was 74.1%, 98.4%, respectively. Water pollutant concentration, $BOD_5,\;COD_{Mn}$, SS, T-N and T-P, excreted from pig was 56,847mg/l, 50,658mg/l, 119,750mg/l, 2,683mg/l, 139mg/l in feces and 2.951mg/l 2,002mg/l, 1,983mg/l, 2,921mg/l, 35mg/l wastewater of swine farms, respectively.
This study was carried out to examine the effects of fermentation conditions on the production of erythritol by osmophilic yeast Candida magnoliae. It was found that sucrose was superior to glucose as carbon source and 109 g/L erythritol was produced from 400 g/L sucrose. When yeast extract was used as nitrogen source, maximum values of yield and productivity for erythritol were obtained at 15 and 20 g/L of yeast extract, respectively. A mixture of 15 g/L yeast extract and 3 g/L ammonium phosphate allowed more efficient utilization of sucrose and hence resulted in 149 g/L of erythritol, 0.37 g erythritol/g sucrose of erythritol yield and $0.78\;g/L{\cdot}hr$ of erythritol productivity. A batch fermentation supplemented with 40 g/L KCl resulted in an erythritol concentration of 167 g/L and an erythritol yield of 0.42 g erythritol/g sucrose.
This study was to investigate the changes of shape of the lower limb surface, the rate of the measurement of expansion and contraction and correlation coefficient between variables caused by hip joint and knee joint movements. The results of the investigation are as follows; 1. According to the development figure of shell when the leg was raised $45^{\circ}$forward($M_{2}$), total length of F.L shortened while B.L lengthened. This result is contarary to $M_{3}$raising the leg $15^{\circ}$ backward. In both $M_{2}$, $M_{3}$movements, the rate of expansion and contraction to the course direction was insignificant. When hip joint was bent $15^{\circ}$ with knee joint $120^{\circ}$bent ($M_{4}$) and hip joint was bent $30^{\circ}$ with knee joint $90^{\circ}$ bent($M_{5}$), upper section of back hip expanded while the front hip section contracted slightly. In the Movement of sitting on the chair($M_{6}$), abdomen, front hip section and upper thight section contracted to the wale direction remarkably while the back hip section expanded conspicuously. 2. According to the rate of expansion and contraction of skin (surface) by the somatometry. In $M_{2}$, C.F.L. upper and middle thight girth contracted and B.L, C.L, L.L expanded. This fact is contarary to M3. In M4, M5, C.F.L showed remarkable contraction and C.B.L expanded remarkably. In $M_{6}$, C.B.L contracted most of all the items measured and knee girth, F.L, L.L, C.B.L, hip girth expanded conspicuously. 3. According to the correlation coefficient between variables. In various movements, the correlation among girth items commonly showed a high or middle grade, the correlation among length items also commonly showed a low grade and that girth and length items showed a very low grade commonly. Waist girth, hip grith, F.L, B.L, L.L items showed that there were significant correlation.
Heterogeneous nuclear ribonucleoprotein L (hnRNP L) is a major pre-mRNA binding protein and it is an abundant nuclear protein that shuttles between the nucleus and the cytoplasm. hnRNP L is known to be related to many cellular processes, including chromatin modification, pre-mRNA splicing, mRNA export of intronless genes, internal ribosomal entry site (IRES)-mediated translation, mRNA stability, and spermatogenesis. In order to identify the cellular proteins interacting with hnRNP L, this study performed a yeast two-hybrid screening, using a human liver cDNA library. The study identified insulin-like growth factor binding protein-4 (IGFBP-4) as a novel interaction partner of hnRNP L in the human liver. It then discovered, for the first time, that hnRNP L interacts specifically with IGFBP-4 in a yeast two-hybrid system. The authenticity of this two-hybrid interaction of hnRNP L and IGFBP-4 was confirmed by an in vitro pull-down assay.
