황칠·미강 발효 추출물이 tyrosinase 활성 저해 및 항산화에 미치는 효과 (Effects of Vinegar Fermentation of Korean Dendorpanax and Rice Bran Mixture on the Activity of Tyrosinase and Anti-Oxidant in B16F10 Cell Line)

  • In this study, we used the mixture made from the Rice bran 45 ㎏, Dendropanax 5 ㎏, the sugar of the 10% of the total weight, and the enzyme of the 0.1% of the total weight. After the mixture were fermented for 90 days under 20 $^{\circ}C$, we measured the cell viability and the inhibition rate of the melanin biosynthesis, the activity of tyrosinase and superoxide dismutase (SOD) in malignant melanoma, B16F10 cells, in order to survey the whitening effect and the mechanism of the effects on the sample. As a result, the samples significantly suppressed the cell viability of B16F10 in more than 500 ${{\mu}g}$/㎖ and significantly inhibited the generation of melanin induced by ${\alpha}$-MSH in more than 1,000 ${{\mu}g}$/㎖. Sample decreased the activity of tyrosinase while increased the activity of SOD in dose dependent manner. Therefore, we considered that the fermentation extract made from a Rice bran and Dendropanax will be able to produce high value-added products, if used as a commercial.

동아시아에서 황칠수(黃漆樹)의 활용에 대한 연구(硏究) (A study of the application of Hwangchil Tree (黃漆樹 Dendropanax morbiferus H.Lév.) in East Asia)

  • Essence of the Hwangchil tree (黃漆樹 Dendropanax morbiferus H.Lév.) has been used for various purposes, like waxing emperor's armors, covering an astrology board, or, during the Silla Dynasty, burying it to block something bad in the soil symbolically. Essence of the Hwangchil tree was known to have remarkable preservation effects such as waterproofing, damp-proofing, rust-proofing, and moth-proofing as well as not being easily peeled off from even soft surfaces like paper. There is a record in Prescriptions for Epidemic diseases of Cows, Horses, Sheep, and Pigs (牛馬羊猪染疫病治療方), published in 1541, of Hwangchil that is local to Jeju Island being used instead of benzoin (安息香), of burning Hwangchil, and of making cows inhale its smoke to prevent plague among them. Along the same lines, there are records in the Local Chronicle of Tamra (耽羅志) and the Book of Earth Geography (輿地圖書) that identify Hwangchil with benzoin. In Seonghosaseol (星湖僿說), a book written by Lee Yik in around 1760, it is acknowledged that Hwangchil could be medicinal herb. In 2000, Ahn Duk-Kyun registered the roots and branches of Hwangchil tree as 'Boncho' (本草 herbal medicine) in the Pictorial Book of Korean Medicinals (韓國本草圖鑑) and presented the method of taking it for medical purpose. Researchers have suggested that Hwangchil essence as well as diverse parts of the plant such as its roots, branches, leaves, flowers, fruits, and gum have various meaningful medicinal properties. Regarding the history and recent researches of using Hwangchil tree, it has various medicinal probabilities such as, 'dispersing miasma' (辟邪), 'opening holes' (開竅), 'waking the heart' (醒心), 'smoothing spirits' (安神), 'piercing the block' (疎泄), 'removing the old and welcoming the new' (去故生新). This paper contributes ideas about how to expand the uses of Hwangchil Tree.

황칠나무의 揷木發根力 增進 (Promotion of Rootability of the Cuttings of Dendropanax morbifera Lev)

  • This study was conducted to examine the effects of media, crown position, clone ,a nd plant growth substances upon the rootability of cuttings for the establishment of the method of mass production by cutting of Dendropanax morbifera Lev. the artifical soil mixtureof vermiculite, perlite and peatmoss (1 : 1 : 1 v/v/v) with IBA 100mg /l treatment was most effective on the rooting rate (85%) was obtained with semihardwood cuttings, which were collected early-August after the first growing seasons. The effect of crown position on rooting of cuttings were obsreved. Average percentage rooting of cuttings on crown position showed 78% , 72% and 65% in middle , upper and lower part with IBA 100mg/l treatment , respectively. Among the four treatment levels of IBA, the highest rooting rate was observed in 100mg/l IBA treatment.

채취시기(採取時期)에 따른 황칠(黃漆)나무 잎의 화학성분(化學性分) 변화(變化) (Seasonal Changes in Chemical Components of the Leaves of Dendropanax morbifera Lev.)

  • 채취시기에 따른 황칠나무 잎중의 화학성분의 변화를 분석한 결과 수분 함량은 계절이 지남에 따라 점점 감소하였으나 지방, 회분 및 섬유 함량은 오히려 점점 증가하였고 단백질 함량은 봄부터 여름까지는 감소하다가 가을에서 겨울로 감에 따라 다시 증가하는 경향을 보였다. 유리당은 검출된 세 종류의 당이 모두 봄으로부터 겨울로 감에 따라 점정 증가하는 것으로 나타났고 지방산은 일반적으로 포화지방산보다 불포화지방산이 더 많이 함유되어 있었으며, 가장 많이 함유된 포화지방산 및 불포화지방산은 계절에 관계없이 각각 arachidic acid 및 cis-13,16-docosadienoic acid였다. 유리 아미노산은 15종이 검출되었는데 계절에 관계없이 arginine의 함량이 가장 높았고 계절이 지남에 따라 점차 감소하는 것으로 나타났다. 무기성분은 채취시기에 따라 그 함량이 약간씩 차이가 있어서 칼슘 또는 칼륨이 가장 많이 함유되어 있었고 다른 무기성분의 함량은 거의 동일한 경향을 보였다. 총 비타민 C 함량은 계절이 점차 겨울로 감에 따라 감소하는 경향이었으나 수용성 탄닌 함량은 오히려 증가하는 경향을 보여 새잎보다 성숙된 겨울에 채취한 잎에 훨씬 더 높은 것으로 밝혀졌다.

완도(莞島)지역 황칠나무의 자생지 환경 및 생육특성 (Growth Characteristics and Native Environment of Dendropanax morbifera LEV. in Wando, Korea)

  • 황칠나무 재배의 기초 자료를 얻고자 우리나라 남부해안 지대인 완도 지역의 황칠나무 자생지 환경조사와 재배지의 수령별 생육특성을 조사한 결과는 다음과 같다. 1. 황칠나무는 완도 지역에 광범위 하게 자생하고 있었으며, 대부분 해발 $30{\sim}280m$의 산에 분포하고 있었고, 특히 해발 $50{\sim}150m$ 부근에서 빈도(頻度)가 가장 높게 나타났으며, 자생지의 방향은 주로 동남향에 분포되었고, 지형은 사면에 자생하고 있었다. 2. 자생지의 토양은 $PH\;5.2{\sim}5.4$로 약산성이며, 유기물 함량도 $8.4{\sim}9.8%$로 일반재배 토양보다 높은 경향이었고, 토양수분은 $18.2{\sim}21.3%$로 비교적 습기가 많은 지역에 자생하고 있었다. 3. 황칠나무의 개화는 6년생묘에서 최초로 시작되었고 개화시기는 7월 중순경이었다. 6년생묘의 생육상은 수고 129.8cm, 경직경 34.1mm 잎수는 15.4매, 엽병장은 12.9cm가 생장하였다.

황칠나무의 집단구조와 치수의 발생과 생육동태 및 공간분포 (Population Structure, and Emergence and Growth Dynamics of Seedling, and Spatial Distribution of Dendropanax morbifera Lev.(Araliaceae))

  • A Korean endemic and evergreen small tree ' Dendropanax morbifera $L_{EV}$.(Araliaceae)' is a component of evergreen forest and mainly idstributein sourthern region and islands in Korea. A local population of D. morbifera which is located between evergreen and deciduous forest within 50m x 50m quadrate was investigated to ascertain the change of population structure, emergence and growth dynamics of seedlings and saplings, and pattern of spatial distribution by the temproal and spatial expansion of population . The result of analysis of population structure by Importnace Value(IV), evergreen forest showed a high species diversity of evergreen tree species such as Cinnamomum japonicum, Machilus japonica, Neolitsea serica, Daphniphyllum macropodum, Ligustrum japonicum, and etc, in middle and under story than in upper story where Camelia japonica and Quercus acuta were dominant. And in conterminous deciduous fores, the major component of evergreen forest in this region, Camellia japonica, Quercus acuta, evergreen tree of Lauraceae and etc. were abundant in only under story. IV of D. morbifera differed from among three story. In comparative analysis of emergence and growth dynamics of D. morbifera seedlings and saplings between evergreen and deciduous forest, emergece and density of seedlings were significantly greater in evergreen than in deciduous forest, and growth of height and basal diameter of seedlings and saplings were slightly larger in evergreen than in deciduous forest. The spatial distribution patterns by Moristia's index mapping of indivuduals using a lattice method of XY axis within this population showed that seedlings(age up to 2 years) and saplings (age>2 years and height<1m) both evergreen and deciduous forest were more or less aggregated apart from mature trees, and thougth intermediate trees(height>1m and dbh<10cm) had a aggregated distribution pattern, mature trees(dbh>10cm were uniform. In conclusion , the expansion of D. morbfera population from evergreen to deciduous forest accompanied with a mumber of evergreen woody species, and also, emergence and recruitment, and growth of seedlings were greatly influenced moisture and canopy by around community structure.

한라산 황칠나무 집단의 구조 및 생육동태 (Population Structure and Growth Dynamics of Dendropanax morbifera Lev.(Araliaceae) in Mt. Halla)

  • 천연도료 황칠자원을 보전하고 육성하기 위한 육종계획의 일환으로 한라산 황칠나무 천연집단의 식생구조, 공간분포 및 생육동태 등 생태적 특성을 조사한 결과, 중요치(Ⅳ)가 높은 수종은 구실잣밤나무, 서어나무, 황칠나무, 종가시나무, 가시나무, 동백나무, 당단풍 등인 것으로 조사되었으며 , 모든 조사구에서 공통적으로 출현한 수종은 구실잣밤나무와 서어나무 2종이었다. 층위별 황칠나무의 적산중요치는 상층에서 17.2%, 중층 12.9%, 하층 10.3%로 비교적 높은 우점도를 나타내고 있었으며, 분포형은 상층, 중층, 하층에서 확률분포하는 것으로 나타났다. 황칠나무 임분은 낙엽활엽수인 서어나무를 우점으로 하는 군집에서 종가시나무, 구실잣밤나무 등의 상록활엽수를 우점으로 하는 군집으로 진행되는 천이단계에 있었으며, 황칠나무는 이들 임분에서 주로 상층의 아우점종이거나 중층의 우점종으로 나타났다. Morisita's index를 통한 공간분포는 치수(seedling)와 어린개체 (sapling)들은 대체로 모수로서 추정되는 성숙목으로부터 떨어진 곳에 집중분포하는 경향을 보였으나, 성숙목(Mature tree)은 규칙 분포하는 양상이었다. 결론적으로, 황칠나무는 다른 상록활엽 수들과 동반하여 집단 확산이 일어나며 , 집단의 확산을 위한 치수의 발생과 생장에 있어서 주변의 임분구조에 큰 영향을 미치는 것으로 나타났다. 또한, 개체군의 동태를 예측한 결과, 낮은 직경급의 밀도가 높고 높은 직경 급의 밀도가 낮은 전형적인 역J자형 분포를 보였다.

Streptozotocin으로 유도된 당뇨 랫드에서 황칠나무 잎 열수추출물의 항당뇨 효과 (Anti-diabetic effects of aqueous extract of Dendropanax morbifera Lev. leaves in streptozotocin-induced diabetic Sprague-Dawley rats)

  • This study examined the anti-diabetic effects of aqueous extracts of Dendropanax morbifera leaves (DMWEs) in streptozotocin-induced diabetic Sprague-Dawley (SD) rats. Thirty male SD rats (body weight [BW], 250.4 ± 19.7 g) were divided into the following six groups: normal control rats (NC), diabetic control rats (DC), diabetic rats treated with metformin HCl 100 mg/kg BW (DT), diabetic rats treated with DMWEs 50 mg/kg BW (DM-50), diabetic rats treated with DMWEs 100 mg/kg BW (DM-100), and diabetic rats treated with DMWEs 200 mg/kg BW (DM-200). From two weeks of administration of DMWEs, the BW of all groups treated with DMWEs increased significantly compared to DC (p < 0.05). At four weeks after treatment, the blood glucose levels in DT, DM-100, and DM-200 decreased below 200 mg/dL, while the glycated hemoglobin concentrations in all groups administered DMWEs were similar to those of NC and DT. Regarding the blood biochemical parameters, the levels of aspartate transaminase, alanine transaminase, blood urea nitrogen, and creatinine in DM-100 and DM-200 were similar to those in NC and DT. Overall, these results highlight the effectiveness of DM-100 in the treatment of diabetes.

황칠나무(Dendropanax morbifera)의 부정근 유도 및 기내증식조건 (Induction and in vitro proliferation of adventitious roots in Dendropanax morbifera)

  • Dendropanax morbifera (Araliaceae) is an endemic species in Korea and distributed in the southern part of Korea. The roots and stems of this plant have been used for folk medicine for the treatment of migraine headache, dysmenorrheal, and remove wind dampness and for Vanishes production. Production of adventitious roots in D. morbifera by in vitro cultures could be used as alternatives materials. Leaf, stem, and root segments from D. morbifera seedling were cultured on Murashige and Skoog (MS) medium supplemented with 3.0 mg/L IBA and 30 g/L sucrose. After 4 weeks of culture, the highest induction of adventitious roots was obtained from the leaf segment. Frequency of adventitious root formation on medium with various kinds of auxins (IAA, NAA, 2,4-D, and IBA) and various concentrations of IBA (0, 0.1, 0.5, 1.0, 3.0, and 5.0 mg/L) was tested. The maximum induction of adventitious root was obtained on medium with 1.0 mg/L IBA. In liquid culture, growth of root was best 1/2MS medium supplemented with 1.0 mg/L IBA and 30 g/L sucrose. Adventitious roots were cultured in 5 L bioreactor containing 1/2 MS medium supplemented with 1.0 mg/L IBA and 30 g/L sucrose and mass-production of adventitious roots was successfully achieved. This study demonstrated for the first time to produce adventitious roots in D. morbifera.

Dendropanax morbifera Extract Inhibits Intimal Hyperplasia in Balloon-Injured Rat Carotid Arteries by Modulating Phenotypic Changes in Vascular Smooth Muscle Cells

  • The plant Dendropanax morbifera Léveille is effective folk medicines for the treatment of several conditions, such as infectious diseases, skin diseases, and other illnesses. Although the inhibitory effects of D. morbifera on the proliferation and migration of vascular smooth muscle cells (VSMCs) have been shown in our previous study, its effects in vivo remain to be elucidated. In this study, we aimed to investigate the protective effects of the extracts from D. morbifera (EDM) on neointimal hyperplasia of rat carotid artery and explore the underlying mechanisms. We observed that the ratio of intima to media thickness (I/M) was significantly decreased in the EDM-treated groups by ~80% compared to that of the control. The expression of Ki-67 and proliferating cell nuclear antigen was decreased by ~70% in the EDM-treated groups compared to that of the control. In addition, matrix metalloproteinase (MMP)2 and MMP9 significantly reduced in the neointimal layer of the EDM-treated groups. Moreover, the decreased levels of contractile phenotypic markers of VSMCs, such as α-smooth muscle actin, myocardin, and smooth muscle-myosin heavy chain, were successfully restored by EDM treatment. Furthermore, the levels of synthetic phenotypic markers, cellular retinal binding protein 1 and connexin 43 were also restored to normal levels. These results suggest that EDM inhibits vascular neointimal hyperplasia induced by balloon injury in rats via phenotypic modulation of VSMCs. Therefore, EDM may be a potential drug candidate for the prevention of restenosis.
