Trigonella foenum in graecum (Fenugreek) is a traditional herbal plant used to treat disorders like diabetes, high cholesterol, wounds, inflammation, gastrointestinal ailments, and it is believed to have anti-tumor properties, although the mechanisms for the activity remain to be elucidated. In this study, we prepared a methanol extract from Fenugreek whole plants and investigated the mechanism involved in its growth-inhibitory effect on MCF-7 human breast cancer cells. Apoptosis of MCF-7 cells was evidenced by investigating trypan blue exclusion, TUNEL and Caspase 3, 8, 9, p53, FADD, Bax and Bak by real-time PCR assays inducing activities, in the presence of FME at $65{\mu}g/mL$ for 24 and 48 hours. FME induced apoptosis was mediated by the death receptor pathway as demonstrated by the increased level of Fas receptor expression after FME treatment. However, such change was found to be absent in Caspase 3, 8, 9, p53, FADD, Bax and Bak, which was confirmed by a time-dependent and dose-dependent manner. In summary, these data demonstrate that at least 90% of FME induced apoptosis in breast cell is mediated by Fas receptor-independently of either FADD, Caspase 8 or 3, as well as p53 interdependently.
The present study was carried out to assess the dietary exposure and the risk for arsenic (As) and mercury (Hg) through mushrooms intake. Various samples (n = 536) covering 17 kinds of mushrooms were collected from retail outlets and markets in Korea. The contents of As were 0.003 (King oyster) ~ 1.952 (Matsutake) mg/kg in raw mushrooms and 0.015 (Fuscoporia obliqua) ~ 16.95 (Matsutake) mg/kg in dry mushrooms, respectively. The contents of Hg were 0.001 (King oyster) ~ 0.030 (Matsutake) mg/kg in raw mushrooms and 0.004 (Oyster) ~ 0.588 (Matsutake) mg/kg in dry mushrooms, respectively. The mean dietary exposures of As was $0.151{\mu}g$/day, taking 0.005% of provisional tolerable weekly intake (PTWI). The mean dietary exposures of Hg was $0.022{\mu}g$/day, corresponding to 0.071% of PTWI. Therefore, the level of overall dietary exposure to As and Hg for Korean population through mushroom was far below the levels recommended by Joint FAO/WHO Expert Committee on Food Additivies, indicating little possibility of concern.
Two endoxylanases from an alkaliphilic bacterium, Bacillus halodurans C-1, were purified 3.8- and 7.9- fold with specific activities of 9.4 and 19.8U/mg protein, respectively. The molecular masses of both purified enzymes were 23 and 47 kDa, respectively, and 23 kDa xylanase I (Xyl I) exhibited an optimum pH at 7.0, whereas 47 kDa xylanase II (Xyl II) showed a broad pH range of 5.0 to 9.0. The temperature optima of both xylanases were $60^{\circ}C\;and\;70^{\circ}C$, respectively. Both were stable in the pH range of 6.0 to 9.0 and 5.0 to 10.0, respectively, and they were stable up to $60^{\circ}C\;and\;70^{\circ}C$, respectively. The $K_m\;and\;V_{max}$ of Xyl I were 4.33mg/ml and $63.5{\mu}mol/min/mg$, respectively, whereas Xyl II had a $K_m$ value of 0.30 mg/ml and $V_{max}$ of $210{\mu}mol/min/mg$. Both xylanases hydrolyzed xylans from birchwood, oat spelt, and larchwood. However, they showed different modes of action; a series of xylooligosaccharides larger than xylotriose were released as the major products by Xyl I, whereas xylobiose and xylotriose were the main products by Xyl II. The maximum synergistic action of the two enzymes on hydrolysis of xylan was 2.16 with the ratio of Xyl I to Xyl II at 1:9.

PURPOSE. To compare the accuracy of marginal and internal adaptation of zirconia (Zr) copings fabricated on anatomic (A), semi-anatomic (SA) and non-anatomic (NA) occlusal surface preparations. MATERIALS AND METHODS. 45 extracted bicuspid teeth were prepared for receiving zirconia crowns, with different occlusal preparation designs A=15, SA=15 & NA=15. The Zr copings were fabricated by using CAD4DENT, CAD/CAM. The copings were adjusted, cemented and were cross sectioned centrally from buccal cusp tip to lingual cusp tip into mesial and distal halves. The copings were examined under electron microscope at ${\times}200$ magnification and the measurements were recorded at 9 predetermined areas in micrometers. RESULTS. Overall mean gap values for the three groups was found to be $155.93{\pm}33.98{\mu}m$ with Anatomical Occlusal preparation design having the least gap value of $139.23{\pm}30.85{\mu}m$ showing the best adaptation among the groups. Post Hoc Tukey's test showed a statistically significant difference (P=.007) between the means of gap for A & NA preparation designs. Measurements recorded at 9 predetermined points showed variations for the three groups. CONCLUSION. Anatomical occlusal preparation designs resulted in better marginal and internal adaptation of Zr copings. There is a considerable variation between the measured marginal and internal gap values for the Zr copings fabricated by the (CAD4DENT-CAD/CAM). This variation may be associated with the lack of standardization of the preparation of teeth, computerized designing of the coping for each tooth, cement used, uniform pressure application during the cementation of the copings, sectioning of the copings and the microscopic measurements.
We investigated seasonal variation of microalgal assemblages, sea water temperature, salinity and suspended solid and the parameters measured daily from January 1998 to October 1999 at a nearshore shallow-water in Marian Cove, Maxwell Bay, King George Island, the Antarctic. Annual mean surface water temperature was -0.3$0^{\circ}C$ and the highest water temperature was 4.53$^{\circ}C$ (22 January 1999) and the lowest water temperature was -2.07$^{\circ}C$ (23 August 1998). Annual mean salinity was 33.38 psu, ranging from 42.80 psu (6 January 1999) to 19.50 psu (6 June 1999). Annual mean suspended solid (SS) during two years was 34.14 mgㆍ1$^{-1}$, ranging from 60.62 mgㆍ1$^{-1}$(7 March 1998) to 12.90 mgㆍ1$^{-1}$ (26 December 1998). Chlorophyll $\alpha$ (Chl $\alpha$) concentrations were measured in order to know seasonal variations of microalgae in the surface seawater. Annual mean of total Chl a concentration was 0.55$\mu\textrm{g}$ㆍ1$^{-1}$, the highest Chl $\alpha$ concentration (12.16$\mu\textrm{g}$ㆍ1$^{-1}$) appeared in 4 October 1998, the lowest Chl $\alpha$ concentration appeared 0.19$\mu\textrm{g}$ㆍ1$^{-1}$, Monthly mean total Chl $\alpha$ concentration was high in October 1998 (1.32$\mu\textrm{g}$ㆍ1$^{-1}$) and low in July on 1998 (0.28$\mu\textrm{g}$ㆍ1$^{-1}$). Annual mean nano-sized Chl $\alpha$ concentration was 0.40$\mu\textrm{g}$ㆍ1$^{-1}$, monthly mean nano -sized Chl $\alpha$ concentration was high in November 1998 (0.90$\mu\textrm{g}$ㆍ1$^{-1}$), and low in July 1999 (0.22$\mu\textrm{g}$ㆍ1$^{-1}$). Annual mean micro-sized Chl $\alpha$ concentration was 0.15$\mu\textrm{g}$ㆍ1$^{-1}$ monthly mean micro-sized Chl $\alpha$ concentration was high in October 1998 (0.81$\mu\textrm{g}$ㆍ1$^{-1}$), and low July 1998, January, February and September 1999 (0.05$\mu\textrm{g}$ㆍ1$^{-1}$). More than 65% of total Chl $\alpha$ was concentrated during spring and summer time between October and March. Microalgal variation appeared to be due to physical factors of seawater in the Antarctic nearshore from 1998 to 1999. The reason why micro-sized Chl $\alpha$ did not increase during austral summer was the bay had been frozen by decrease of water temperature. We think that total microalgal abundance was decreased because the summer microalgal abundance was determined by variation of water temperature during winter season. This study was carried out to investigate proper modified atmosphere condition of king oyster mushroom (Pleurotus eryngii) during cold storage and distribution. King oyster mushrooms were divided into two treatments: $1{\mu}L/L$ 1-methylcyclopropene for 20 hours at $0^{\circ}C$ (1-MCP), while the other treatment was left at $0^{\circ}C$ (control). They were packaged with $30{\mu}m$ oriented polypropylene (OPP, $1238.0cc/m^2{\cdot}day{\cdot}atm\;O_2$), and microperforated (MP3, $3179.9cc/m^2{\cdot}day{\cdot}atm\;O_2$) film. Quality and sensory evaluation parameters of weight loss, stem firmness, skin color, off-flavor, overall quality were monitored after 3, and 4 weeks storage at $0^{\circ}C$ and plus 3 days on the shelf at room temperature. 1-MCP treated mushroom packaged with MP3 film kept 3.6~10.9% $O_2$ and 9.0~13.3% $CO_2$ concentration in the bag during storage, and showed high overall quality at 4 weeks storage at $0^{\circ}C$ + 3 days on the shelf at room temperature because of the lowest development of off-flavor, stem discoloration, and cap softening among the treatments.
This paper proposed the new current feed-back amplifier for low supply voltage application. The input stage was designed to be a class-AB circuit and achieve the low supply-voltage operation down to $2V_{TH}+2V_{DS(SAT)}$. With the self-adjust bias current, the high performance can be adopted with high stability. The circuit was successfully proven by the simulation with MOSIS 0.5 ${\mu}$m MOS technology.
Background: Gold nanoparticles have recently been investigated with respect to biocompatibility according to their interactions with cells. The purpose of this study was to examine cytotoxicity and apoptosis induction by well-characterized gold nanoparticles in human breast epithelial MCF-7 cells. Methods: Apoptosis was assessed by TUNEL, cytotoxicity by MTT assay and caspase 3, 9, p53, Bax and Bcl expression by real-time PCR assays. Results: Gold nanoparticles at up to $200\;{\mu}g/mL$ for 24 hours exerted concentration-dependent cytotoxicity and significant upregulation of mRNA expression of p53, bax, caspase-3 & caspase-9, whereas expression of antiapoptotic bcl-2 was down-regulated. Conclusion: To the best of our knowledge this is the first report showing that gold nanoparticles induce apoptosis in MCF-7cells via p53, bax/bcl-2 and caspase pathways.
Previous screening of novel antibacterial agents revealed that some bacterial isolates exhibited antibiotic activity against both gram-positive and gram-negative bacteria and that they showed antibacterial activity, even against methicillin-resistant Staphylococcus aureus (MRSA). Among these isolates, one bacterial strain, BCNU 1204, was identified as Pseudomonas aeruginosa using phenetic and phylogenetic analysis, based on 16S ribosomal RNA gene sequences. The maximum productivities of antimicrobial substances of BCNU 1204 were obtained after being cultured at $35^{\circ}C$ and pH 7.0 for 4 d in King's medium B (KMB). Dichloromethane (DCM) and ethylacetate (EA) extracts of P. aeruginosa BCNU 1204 exhibited strong antimicrobial activity, particularly against gram-positive bacteria. The EA extracts exhibited broad-spectrum activity against antibiotic resistant strains. Fraction 5-2, was obtained by recycling preparative liquid chromatography (LC) and preparative thin-layer chromatography (TLC) and was identified as phenazine-1-carboxylic acid belonging to phenazines using gas chromatography and mass spectrometry (GC/MS). Its minimum inhibitory concentration (MIC) values were $25{\mu}g/ml$, $50{\mu}g/ml$, ${\geq}25{\mu}g/ml$, and ${\geq}50{\mu}g/ml$ for MRSA CCARM 3089, 3090, 3091, and 3095 strains, respectively. P. aeruginosa BCNU 1204 may be a potential resource for the development of anti-MRSA antibiotics. Additional research is required to identify the active substance from P. aeruginosa BCNU 1204.
Thailand is one of the largest citrus producers in Southeast Asia. Pathogenic infection by Phytophthora, however, has become one of major impediments to production. This study identified a pathogenic oomycete isolated from rotted roots of pomelo (Citrus maxima) in Thailand as Phytophthora nicotianae by the internal transcribed spacer ribosomal DNA sequence analysis. Then, we examined the in vitro and in vivo effects of Chaetomium globosum, Chaetomium lucknowense, Chaetomium cupreum and their crude extracts as biological control agents in controlling this P. nicotianae strain. Represent as antagonists in biculture test, the tested Chaetomium species inhibited mycelial growth by 50~56% and parasitized the hyphae, resulting in degradation of P. nicotianae mycelia after 30 days. The crude extracts of these Chaetomium species exhibited antifungal activities against mycelial growth of P. nicotianae, with effective doses of $2.6{\sim}101.4{\mu}g/mL$. Under greenhouse conditions, application of spores and methanol extracts of these Chaetomium species to pomelo seedlings inoculated with P. nicotianae reduced root rot by 66~71% and increased plant weight by 72~85% compared to that in the control. The method of application of antagonistic spores to control the disease was simple and economical, and it may thus be applicable for large-scale, highly effective biological control of this pathogen.
