Galangin and Kaempferol Suppress Phorbol-12-Myristate-13-Acetate-Induced Matrix Metalloproteinase-9 Expression in Human Fibrosarcoma HT-1080 Cells

  • Matrix metalloproteinase (MMP)-9 degrades type IV collagen in the basement membrane and plays crucial roles in several pathological implications, including tumorigenesis and inflammation. In this study, we analyzed the effect of flavonols on MMP-9 expression in phorbol-12-myristate-13-acetate (PMA)-induced human fibrosarcoma HT-1080 cells. Galangin and kaempferol efficiently decreased MMP-9 secretion, whereas fisetin only weakly decreased its secretion. Galangin and kaempferol did not affect cell viability at concentrations up to $30{\mu}M$. Luciferase reporter assays showed that galangin and kaempferol decrease transcription of MMP-9 mRNA. Moreover, galangin and kaempferol strongly reduce $I{\kappa}B{\alpha}$ phosphorylation and significantly decrease JNK phosphorylation. These results indicate that galangin and kaempferol suppress PMA-induced MMP-9 expression by blocking activation of NF-${\kappa}B$ and AP-1. Therefore, these flavonols could be used as chemopreventive agents to lower the risk of diseases involving MMP-9.

Kaempferol Regulates the Expression of Airway MUC5AC Mucin Gene via IκBα-NF-κB p65 and p38-p44/42-Sp1 Signaling Pathways

  • In the present study, kaempferol, a flavonoidal natural compound found in Polygonati Rhizoma, was investigated for its potential effect on the gene expression and production of airway MUC5AC mucin. A human respiratory epithelial NCI-H292 cells was pretreated with kaempferol for 30 min and stimulated with epidermal growth factor (EGF) or phorbol 12-myristate 13-acetate (PMA), for the following 24 h. The effect on PMA-induced nuclear factor kappa B (NF-κB) signaling pathway or EGF-induced mitogen-activated protein kinase (MAPK) signaling pathway was investigated. Kaempferol suppressed the production and gene expression of MUC5AC mucins, induced by PMA through the inhibition of degradation of inhibitory kappa Bα (IκBα), and NF-κB p65 nuclear translocation. Also, kaempferol inhibited EGF-induced gene expression and production of MUC5AC mucin through regulating the phosphorylation of EGFR, phosphorylation of p38 MAPK and extracellular signal-regulated kinase (ERK) 1/2 (p44/42), and the nuclear expression of specificity protein-1 (Sp1). These results suggest kaempferol regulates the gene expression and production of mucin through regulation of NF-κB and MAPK signaling pathways, in human airway epithelial cells.

그라비올라로부터 분리된 Kaempferol 및 Nicotiflorin의 1O2으로 유도된 세포손상에 대한 보호 효과와 그 메커니즘 (Cellular Protective Effects and Mechanisms of Kaempferol and Nicotiflorin Isolated from Annona muricata against 1O2-induced Damage)

  • 본 연구에서는 그라비올라의 주성분인 nicotiflorin을 분리하고 그 아글리콘 성분인 kaempferol을 얻어 세포 보호 효과 및 그 보호 메커니즘을 규명하였다. L-Ascorbic acid 및 (+)-${\alpha}$-tocopherol을 대조군으로 하여, $^1O_2$로 유도된 세포 손상에 대해 nicotiflorin 및 kaempferol의 보호 효과를 측정한 결과 nicotiflorin < (+)-${\alpha}$-tocopherol < kaempferol 순으로 보호 효과가 증가하였다. L-Ascorbic acid는 세포 보호 효과를 보이지 않았다. 이들의 세포 보호 효과 메커니즘을 밝히기 위해 singlet oxygen 소광 속도 상수, 자유라디칼 소거 활성, ROS 소거 활성 및 적혈구 세포 침투율을 측정하였다. 실험 결과, kaempferol과 그 배당체인 nicotiflorin의 세포 보호 효과에 있어서 큰 차이는 세포막에의 침투가 가장 큰 요인으로 확인되었다. 대조군 L-ascorbic acid가 항산화능은 크지만 실험 조건에서 세포막에 침투가 잘 안되어 세포 보호 효과가 나타나지 않은 것으로 확인되었다. Kaempferol과 (+)-${\alpha}$-tocopherol의 비교를 통해 세포 침투뿐만 아니라 라디칼 소거활성 및 ROS 소거 활성도 세포 보호 효과에 기여하는 것으로 나타났다. 결론적으로, 광증감 반응으로 유도된 세포막 파괴에 대한 보호작용은 항산화제들의 세포 침투, 자유라디칼 및 ROS 소거 활성이 큰 영향을 미치는 것으로 확인되었다.

Kaempferol, quercetin 및 그 배당체들의 apoptosis 조절을 통한 신경세포 보호 효과 (Neuroprotective Effects of Kaempferol, Quercetin, and Its Glycosides by Regulation of Apoptosis)

  • 알츠하이머 질환은 대표적인 신경퇴행성 질환으로, 뇌 내에서 $A{\beta}$ 단백질 축적은 알츠하이머 질환의 원인으로 알려져 있다. 본 연구에서는 amyloid beta ($A{\beta}$)로 손상을 유도한 SH-SY5Y 신경세포에서 kaempferol, kaempferol-3-O-glucoside, quercetin, quercetin-3-${\beta}$-D-glucoside의 신경세포 보호 효과에 대해 검토하였다. SH-SY5Y 신경세포에 $A{\beta}_{25-35}$ ($25{\mu}M$)를 처리하였을 때, 처리하지 않은 normal군에 비해 세포생존율이 유의적으로 감소하였다. 반면, kaempferol, quercetin 및 그 배당체들을 각각 처리하였을 때 $A{\beta}_{25-35}$만을 처리한 control군에 비해 유의적으로 세포생존율의 증가를 나타내었다. 또한, apoptosis에 관여하는 cleaved caspase9, Bcl-2-associated X protein (Bax) 단백질 발현을 측정한 결과, normal군에 비해 control군에서 유의적으로 cleaved caspase9 및 Bax 단백질 발현의 증가를 나타내어 $A{\beta}$ 유도 신경세포 손상으로 인한 apoptosis가 유발됨을 확인 하였으며, kaempferol, quercetin 및 그 배당체들의 처리 시 apoptosis 관련 단백질 발현이 감소함으로써 신경세포 보호 효능이 나타냄을 확인하였다. 이러한 결과는 kaempferol, quercetin 및 그 배당체들이 apoptosis 조절을 통해 신경세포 보호 효과를 나타내며, 신경세포 손상으로 인한 알츠하이머 질환을 예방하는 유용한 소재로써 사용 가능성이 있음을 보여준다.

Assessment of MMP-1, MMP-8 and TIMP-2 in experimental periodontitis treated with kaempferol

  • Purpose: The objective of this study was to investigate the effect of a dietary flavonoid, kaempferol, which has been shown to possess antiallergic, anti-inflammatory, anticarcinogenic, and antioxidant activities on the periodontium by histomorphometric analysis and on gingival tissue matrix metalloproteinase-1 (MMP-1), MMP-8, and tissue inhibitor of metalloproteinase-2 (TIMP-2) by biochemical analysis of rats after experimental periodontitis induction. Methods: Sixty Wistar rats were randomly divided into six groups of ten rats each, and silk ligatures were placed around the cervical area of the mandibular first molars for 15 days, except in the healthy control rats. In the experimental periodontitis groups, systemic kaempferol (10 mg/kg/2d) and saline were administered by oral gavage at two different periods (with and without the presence of dental biofilm) to all rats except for the ten non-medicated rats. Alveolar bone area, alveolar bone level, and attachment level were determined by histomorphometric analysis, and gingival tissue levels of MMP-1, MMP-8, and TIMP-2 were detected by biochemical analysis. Results: Significantly greater bone area and significantly less alveolar bone and attachment loss were observed in the kaempferol application groups compared to the control groups (P<0.05). In addition, gingival tissue MMP-1 and -8 levels were significantly lower in the kaempferol application groups compared to the control groups and the periodontitis group (P<0.001). There were no statistically significant differences in TIMP-2 levels between the kaempferol and saline application groups (P>0.05). Conclusions: Kaempferol application may be useful in decreasing alveolar bone resorption, attachment loss, and MMP-1 and -8 production in experimental periodontitis.

  • 회화나무잎에서 3종의 화합물을 단리하였으며, 기기분석 결과 hypogallic acid, kaempferol, quercetin으로 각각 동정하였다. 이들 단리 물질의 항산화 활성은 라디칼 소거능으로 측정하였으며, 항산화 활성은 100 ㎍/ml의 농도에서 각각 98.2%, 96.2% 그리고 90.2%를 나타내었고 이 중 화합물 1 (hypogallic acid)과 화합물 2 (kaempferol)의 경우는 합성 항산화제인 BHT보다 높은 항산화 활성을 나타내었다.

  • A new flavonol $3,5-di-O-KSO_3$:kaempferol 7,4'-dimethyl ether $3,5-O-KSO_3$, was isolated and identified from the flowers of Tamarix amplexicaulis. The known compounds quercetin $3-mono-O-KSO_3$, kaempferol 4'-methyl ether $3-mono-O-KSO_3$, kaempferol 7,4'-dimethyl ether $3-O-KSO_3$, quercetin 7,4'-dimethyl ether $3-mono-O-KSO_3$, kaempferol 3-O-glucuronide and quercetin 3-O-glucuronide were also separated and identified. Structures were established by conventional methods, including electrophoretic analysis, and confirmed by negative FAB-MS, $^1H-\;and\;^{13}C-NMR$.

  • Flavonoids are widely reported to be beneficial to human health. Among flavonoids, in general, flavonoid aglycons have better biological activities than flavonoid glycosides, in that aglycons can easily penetrate through cell membrane because of their low polarity. Therefore, kaempferol, quercetin and various their glycosides were evaluated for their abilities to inhibit NO and $PGE_2$ productions in LPS-induced RAW 264.7 cells. Of these flavonoids and flavonoid glycosides, kaempferol-7-O-${\beta}$-D-glucoside(kp-7-glu) which possesses a glycoside at C-7 position of the A ring in kaempferol, potently inhibited NO, $PGE_2$ and $TNF-{\alpha}$, $IL-1{\beta}$, IL-6 productions in LPS-induced RAW 264.7 macrophages.

황정(黃精)과 Kaempferol의 지방세포 분화 억제 효과 (Anti-adipogenic Effect of Kaempferol, a Component of Polygonati Rhizoma)

  • Objective: It has been reported that Polygonati rhizoma (Pr) has anti-hyperglycemia, anti-triglycemia, anti-diabetic, and anti-tumor activity. Total extract of Pr was tested to identify anti-adipogenic activity in 3T3-L1 differentiation and molecular mechanism of Pr in 3T3-L1 differentiation. Methods: Differentiation of 3T3-L1 pre-adipocyte was induced in the presence of Pr extract and kaempferol. The level of lipid accumulation was measured by Oil Red O staining. The expression of genes associated with adipocyte differentiation was measured by RT-PCR. Results: Extract of Pr and its component kaempferol reduced lipid accumulation in 3T3-L1 during adipogenesis and also reduced mRNA levels of genes associated with adipogenesis, such as adipsin, aP2, LPL, SERBP-1c and $PPAR{\gamma}$. Conclusions: In this study, we showed that the molecular mechanism of Pr and kaempferol activity is related to regulation of $PPAR{\gamma}$ expression and activation.

Effect of Kaempferol on Modulation of Vascular Contractility Mainly through PKC and CPI-17 Inactivation

  • In this study, we investigated the efficacy of kaempferol (a flavonoid found in plants and plant-derived foods such as kale, beans, tea, spinach and broccoli) on vascular contractibility and aimed to clarify the detailed mechanism underlying the relaxation. Isometric contractions of divested muscles were stored and linked with western blot analysis which was carried out to estimate the phosphorylation of myosin phosphatase targeting subunit 1 (MYPT1) and phosphorylation-dependent inhibitory protein for myosin phosphatase (CPI-17) and to estimate the effect of kaempferol on the RhoA/ROCK/CPI-17 pathway. Kaempferol conspicuously impeded phorbol ester-, fluoride- and a thromboxane mimetic-derived contractions regardless of endothelial nitric oxide synthesis, indicating its direct effect on smooth muscles. It also conspicuously impeded the fluoride-derived elevation in phospho-MYPT1 rather than phospho-CPI-17 levels and phorbol 12,13-dibutyrate-derived increase in phospho-CPI-17 and phospho-ERK1/2 levels, suggesting the depression of PKC and MEK activities and subsequent phosphorylation of CPI-17 and ERK1/2. Taken together, these outcomes suggest that kaempferol-derived relaxation incorporates myosin phosphatase retrieval and calcium desensitization, which appear to be modulated by CPI-17 dephosphorylation mainly through PKC inactivation.
