  • KOPRI(Korea Polar Research Institute) have researhed Arctic sea ice by using satellite remote sensing data since 2017 as a mission of KOPRI. The title of the reseach is "Development of Satellite Observation and Analysis for Arctc sea-ice". This project has three major aims; 1) development of prototype satellite data archive/manage system for Arctic sea ice monitoring, 2) development of sea ice remote sensing data processing and analysis technique, and 3) development of international satellite observing network for Arcitc. This reseach will give us that 1) deveolpment of sea ice observing system for northern sea route, 2) development of optimal remote sensing data processing technique for sea ice and selected satelite sensors, 3) development of international satellite onbservation network. I hope that this letter of introducton KOPRI satellite program for Arctic will help to understand Arctic remote sensing and will introduce you to step into the Arctic remote sensing, which Iis like a blue ocean of remote sensing.

Axenic purification and cultivation of an Arctic cyanobacterium, Nodularia spumigena KNUA005, with cold tolerance potential for sustainable production of algae-based biofuel

  • A psychrotolerant cyanobacterium, Nodularia spumigena KNUA005, was isolated from a cyanobacterial bloom sample collected near Dasan Station in Ny-${\AA}lesund$, Svalbard Islands during the Arctic summer season. To generate an axenic culture, the isolate was subjected to three purification steps: centrifugation, antibiotic treatment and streaking. The broad antibacterial spectrum of imipenem killed a wide range of heterotrophic bacteria, while the cyanobacterium was capable of enduring both antibiotics, the remaining contaminants that survived after treatment with imipenem were eliminated by the application of an aminoglycoside antibiotic, kanamycin. Physical separation by centrifugation and streaking techniques also aided axenic culture production. According to the cold-tolerance test, this mat-forming cyanobacterium was able to proliferate at low temperatures ranging between 15 and $20^{\circ}C$ which indicates the presence of cold-tolerance related genes in N. spumigena KNUA005. This suggests the possibility of incorporating cold-resistance genes into indigenous cyanobacterial strains for the consistent production of algae-based biofuel during the low-temperature seasons. Therefore, it is needed to determine the cold-tolerance mechanisms in the Arctic cyanobacterium in the next research stage.

Estimation of Energy Use in Residential and Commercial Sectors Attributable to Future Climate Change (미래 기후변화에 따른 가정 및 상업 부문 에너지수요 변화 추정)

  • In this study it is attempted to estimate the possible change in energy use for residential and commercial sector in Korea under a future climate change senario. Based on the national energy use and observed temperature data during the period 1991~2010, the optimal base temperature for determining heating and cooling degree days (HDD and CDD) is calculated. Then, net changes in fossil fuel and electricity uses that are statistically linked with a temperature variation are quantified through regression analyses of HDD and CDD against the energy use. Finally, the future projection of energy use is estimated by applying the regression model and future temperature projections by the CMIP5 results under the RCP8.5 scenario. The results indicate that, overall, the net annual energy use will decrease mostly due to a large decrease in the fossil fuel use for heating. However, a clear seasonal contrast in energy use is anticipated in the electricity use; there will be an increase in a warm-season demand for cooling but a decrease in a cold-season demand for heating.

Morphology and Molecular Data for Antarctic Cryophilic Microalga, Porosira pseudodenticulata (남극 호냉성 미세조 Porosira pseudodenticulata의 형태와 분자적 자료)

  • We have cultured more than 100 Arctic and Antarctic cryophilic microalgal strains in KOPRI culture collections of polar microorganisms (KCCPM). Among them, we tried to identify an Antarctic strain, KOPRI AnM0008 by morphological and molecular analysis. Nuclear SSU rDNA and plastid rbcL sequences were used to identify the strain. It was identified as Porosira pseudodenticulata based on SSU sequence data showing 99% identity with GenBank X85398. This result was supported by morphological features like solitary labiate process, external foramina and internal cribra by optical and scanning electron microscope. Morphological identification and molecular analysis on polar cryophilic microalgae will be accomplished to construct the databases for KCCPM.

Dasania marina gen. nov., sp. nov., of the Order Pseudomonadales, Isolated from Arctic Marine Sediment

  • An obligately aerobic bacterium, strain KOPRI $20902^T$, was isolated from a marine sediment in Ny-${\AA}$lesund, Spitsbergen Islands, Norway. Cells were irregular rods and motile with polar monotrichous flagellum. The optimum growth temperature was $17-22^{\circ}C$. Cells grew best in pH 7.0-10.0 and 3-4% sea salts (corresponding to 2.3-3.1% NaCl). The novel strain required $Ca^{2+}$ or $Mg^{2+}$ in addition to NaCl for growth. Sequence analysis of 16S rRNA gene revealed that the Arctic isolate is distantly related with established species (<92.4% sequence similarity) and formed a monophyletic group with Cellvibrio, which formed a distinct phylogenetic lineage in the order Pseudomonadales. Predominant cellular fatty acids [$C_{16:1}\;{\omega}7c/15:0$ iso 2OH (45.3%), $C_{16:0}$ (18.4%), ECL 11.799 (11.2%), $C_{10:0}$ 3OH (10.4%)]; DNA G+C content (37.0 mol%); nitrate reduction to nitrogen; absence of aesculin hydrolysis, N-acetyl-${\beta}$-glucosaminidase and esterase; no assimilation of arabinose, galactose, glucose, lactose, maltose, and trehalose differentiated the strain from the genus Cellvibrio. Based on the phylogenetic and phenotypic characteristics, Dasania marina gen. nov., sp. nov. is proposed in the order Pseudomonadales. Strain KOPRI $20902^T$ (=KCTC $12566^T$=JCM $13441^T$) is the type strain of Dasania marina.

A Review on the Application of Stable Water Vapor Isotope Data to the Water Cycle Interpretation (수증기안정동위원소의 물순환 해석에의 적용에 대한 고찰)

  • Studies using stable water vapor isotopes have been recently conducted over the past two decades because of difficulties in analysis and sample collection in the past. Stable water vapor isotope data provide information of the moisture transport from ocean to continent, which are also used to validate an isotope enabled general circulation model for paleoclimate reconstructions. The isotopic compositions of groundwater and water vapor also provide a clue to how moisture moves from soil to atmosphere by evapotranspiration. International Atomic Energy Agency designates the stations over the world to observe the water vapor isotopes. To analyze the water vapor isotopes, a cryogenic sampling method has been used over the past two decades. Recently, two types of laser-based spectroscopy have been developed and remotely sensed data from satellites have the global coverage. In this review, measurements of isotopic compositions of water vapor will be introduced and some studies using the water vapor isotopes will also be introduced. Finally, we will suggest the future study in Korea.

Ln-resin and HIBA Method for La-Ce and Sm-Nd Isotope Measurement (La-Ce 및 Sm-Nd 동위원소계 측정을 위한 란탄-레진법과 HIBA(Hydroxy Isobutyric Acid)분리법의 상호비교)

  • A column chemistry is the most useful tools for isolating the elements of interest in isotope geochemistry. Here we introduce the chemical experimental procedure for Sm, Nd, La and Ce separation such as Teflon powder or Ln-resin method using HDEHP of KIGAM, KBSI, KOPRI and ${\alpha}$-HIBA(${\alpha}$-Hydroxy Isobutyric acid) method of Nagoya University, Japan. This technical report will provide an useful information in selecting the experiment method for rare earth element isotope system study such as Sm-Nd and La-Ce isotope system.

Isolation of Protease-Producing Arctic Marine Bacteria

  • We isolated and identified three protease-producing bacteria that had inhabited the region around the Korean Arctic Research Station Dasan located at Ny-Alesund, Svalbard, Norway $(79^{\circ}N,\;12^{\circ}E)$. Biofilms were collected from the surface of a floating pier and from dead brown algae in a tide pool near the seashore. The biofilm samples were transported to the Korea Polar Research Institute (KOPRI) under frozen conditions, diluted in sterilized seawater, and cultured on Zobell agar plates with 1% skim milk at $10^{\circ}C$. Three clear zone forming colonies were selected as protease-producing bacteria. Phylogenetic analysis based on 16S rDNA sequences showed that these three stains shared high sequence similarities with Pseudoalteromonas elyakovii, Exiguobacterium oxidotofewm Pseudomonas jessenii, respectively. We expect these Arctic bacteria may be used to develop new varieties of protease that are active at low temperatures.
