Structural Characteristics of Immunostimulating Polysaccharides from Lentinus edodes

  • There is a significant amount of experimental evidence suggesting that polysaccharides from mushrooms enhance the host immune system by activating various mechanisms in immune cells, including macrophages. In this study, polysaccharides from Lentinus edodes were found to stimulate the functional activation of macrophages to secrete inflammatory mediators and cytokines and increase the phagocytotic uptake. The chemical properties of the stimulatory polysaccharides, CPFN-G-I, CPBN-G, and CPBA-G, were determined based on their monosaccharide composition, which mainly consisted of glucose and mannose. According to FT-IR and GC/MS, the structure of CPFN-G-I, purified from the fruiting body of L. edodes, was found to consist of a $\beta$-1,6-branched-$\beta$-1,4-glucan, whereas CPBN-G and CPBA-G, purified from the liquid culture broth, were found to be composed of a heteromannan. The configuration of the p-linkage and triple helical conformation of each polysaccharide were confirmed using a Fungi-Fluor kit and Congo red, respectively.

Prevalence of Listeria monocytogenes and Related Species in Minimally Processed Vegetables

  • Listeria spp. were isolated from a total of 402 naturally contaminated domestic ready-to-eat (RTE) vegetable samples by the conventional Food and Drug Administration protocol and confinned by API-Listeria kit. Also, the susceptibility to 12 antibiotics, polymerase chain reaction (PCR) assay for virulence gene of pathogenic Listeria monocytogenes isolates, and in vitro virulence assay using myeloma and hybridoma cells from murine and human sources were tested. Among the samples, 17 samples (4.2%) were found to be contaminated with Listeria species. Among the 17 strains of Listeria spp. isolates, only 2 strains (11.8%) of L. monocytogenes and 15 strains (88.2%) of L. innocua were identified. Antibiotic susceptibility test showed that the Listeria spp. isolates were very susceptible to the antibiotics tested, except for nalidixic acid. Among 17 strains of Listeria spp., PCR analysis showed that 2 strains of L. monocytogenes isolates proved to have a virulence hly gene, but none of L. innocua had the hly gene. Also, hybridoma Ped-2E9 cells assay showed that only L. monocytogenes isolates killed approximately 95-99% hybridoma cells after 6 h, but L. innocua isolates had about 0-5% lethal effect. These results indicate that PCR assay with hly primer or hybridoma Ped-2E9 cells assay could be used as a good monitoring tool or in vitro virulence test for L. monocytogenes.

Adult Dirofilaria immitis in the Aorta of a Dog (개에서 심장사상충증의 대동맥 이소기생)

  • Unknown age, spayed female mixed dog was presented with a severe lethargy. Radiography, ultrasonography, and blood screen test were performed to make a diagnosis. There were no specific radiographic findings. On the ultrasonography, small amount of ascites was found around gallbladder and hepatic vein was dilated approximately 6.1 mm. Blood screen test revealed a severe anemia. Result of heartworm ELISA kit was positive. At necropsy, parasites were in the left atrium, right ventricles, aorta, pulmonary artery, right renal artery.

Changes in Median Ages at Death from Selected Cancer Types in Relation to HLA-DRB1/DQB1

  • The median ages at death from cancers between 1985 and 2005 were calculated to demonstrate that inherent anticancer mechanisms may be a common pathway for different cancers. Seventy-eight patients with gastric, liver and lung cancers, were recruited in the solid cancer group. The leukemia group consisted of 31 patients with three main types of leukemia. The controls were 100 healthy individuals. The samples were typed using an HLA-DR/DQ PCR-SSP typing kit. The results showed that the median ages at death from all causes were 64.7 years in 1985 and 70.1 years in 2005. The range of the median ages at death from all cancers was similar to the corresponding value for deaths attributed to all causes. The frequency of $DRB1^*03$ was 9.6% in the solid cancer group and 3.0% in the control group (p<0.05). The frequency of $DRB1^*04$ in the leukemia group were significantly lower than that of the control (p<0.05). $DRB1^*13$ and $DRB1^*06$ frequencies in the leukemia group were significantly higher than those of the controls (p<0.05). It is suggested that inherent anti-cancer mechanisms may be a common pathway for different cancers and are associated with the immune system and HLA.

  • Rabies causes the highest mortality of all viral diseases in the world unless the victim has been protected either by active immunization or post-exposure immunoprophylaxis. Infected stray dogs, raccoons, skunks, foxes and bats are the demonstrated carriers of most cases of rabies. It is difficult to diagnose a rabid animal in the field unless characteristic clinical signs are evident. However, this study used a commercial fast check kit comprised of immunochromatographic test (ICT) strips (ICTS) to diagnose rabies infection in clinically suspected samples obtained from a wildebeest. A 10-year old male wildebeest (approximate weight, 150 kg) died at Bangabandhu Sheikh Mujib Safari (BSMS) Park, Cox's Bazar, Bangladesh with a clinical history of severe excitation and abundant oral secretions. A gross pathological examination revealed no specific lesions indicating any fatal diseases. The entire brain was collected within 6 hours of death, and the brain sample was tested using the ICT strips following the manufacturer's directions. The rabies viral antibody was detected within the brain stem and medulla of the brain tissue of the dead wildebeest.

Development of a Quality Manual for a Research and Educational Centre: A Case Study

  • There have been growing concerns about whether proper measures are in place to ensure the delivery of quality services and the operations in research and educational institutions. This paper describes various quality models and discusses their relevance into the total quality concepts and quality management systems (QMS) in education. It then illustrates how a QMS and the accompanied quality manual be developed in a research and educational institution using the Centre for Enterprise Research Integration (ERI) at The University of the West Indies, Trinidad as a case. Through personal interviews with the $Centre{\square}s$ personnel, views on the operations and determinants of the QMS were acquired. Results showed that the people's resistance to changes, conflicts on academic freedom, and difficulties on measuring performance were the obstacles to QMS implementation. People involvement and proper communication are determinants of QMS implementation. The system also rests on having a tailored quality manual that entails the documentation of processes and procedures for ascertaining the delivery of quality services and operations in the Centre. The development of quality manual is briefly described in the paper.

Manufacture of High Quality Musical Steel Drums in Trinidad and Tobago

  • This paper explores innovative manufacturing processes, which can be used to manufacture the national musical instrument of Trinidad and Tobago, the Musical Steel Drum or Steel Pan. The main manufacturing process used today is the manual hand-forming technique. In order to achieve more consistent and deeper formed components while maintaining the high quality of the instrument, it is proposed that the Marforming process and the Flow-forming process, an adaptation of the Spinforming process, be used more frequently in the future to replace the traditional Hand-forming method. In the traditional Spinforming technique material is pushed from the outer circumference of the metal disc to the center in progressive passes of the former. This results in a thinning of the outer region of the formed component with thickening of the center, however the opposite is required for the musical steel drum and by adapting the process the required strain distributions were achieved. Evaluation took the form of strain analyses of pre-formed steel drums and visual inspection of the quality of the surface finish. It was found that the Marformed components had the smallest range of strain values while the Spinformed components had the largest range.

Turbulence-induced noise of a submerged cylinder using a permeable FW-H method

  • Among underwater noise sources around submerged bodies, turbulence-induced noise has not been well investigated because of the difficulty of predicting it. In computational aeroacoustics, a number of studies has been conducted using the Ffowcs Williamse-Hawkings (FW-H) acoustic analogy without consideration of quadrupole source term due to the unacceptable calculation cost. In this paper, turbulence-induced noise is predicted, including that due to quadrupole sources, using a large eddy simulation (LES) turbulence model and a developed formulation of permeable FW-H method with an open source computational fluid dynamics (CFD) tool-kit. Noise around a circular cylinder is examined and the results of using the acoustic analogy method with and without quadrupole noise are compared, i.e. the FW-H method without quadrupole noise versus the permeable FW-H method that includes quadrupole sources. The usability of the permeable FW-H method for the prediction of turbulence-noise around submerged bodies is shown.

Investigation of canine dirofilariasis and brucellosis in free roaming dogs from public animal shelters in Gwangju area (광주지역 동물보호소내 유기견의 개심장사상충과 개 브루셀라병 감염 실태조사)

  • This study was conducted to investigate the prevalence of canine heartworm infections, canine brucellosis and hematologic values from 153 free roaming dogs in the area of Gwangju city from March to November 2006. Nineteen (12.4%) of 153 samples tested with modified Knott's technique showed positive reaction for microfilariae. Polymerase chain reaction using specific primers for D immitis amplified the expected product from all samples of 19 microfilaremic canine blood samples as determined by the modified Knott's test for microfilariae. The seasonal infection rates of microfilariae were higher in the spring season (10/19, 52.6%) than in the other seasons. The major hematological findings in microfilaremic dogs were mild leukocytosis and mild monocytosis. A total of 100 dogs randomly selected from 153 free roaming dogs were negative for canine brucellosis by serological test using immunochromatographic antibody test kit.

Comparative Studies on Detecting Methods of Fecal Indicators (Coliforms) in Surface Water (지표수에서의 분변오염지표세균 (대장균군) 검출방법의 비교연구)

  • For monitoring the fecal pollution at Nak-Dong River, one of the eutrophicated rivers, the differences between total coliforms (TC) and fecal coliforms (FC) using both of membrane filtration (MF)/MPN method, and also fecal streptococcus (FS) by MF-method was investigated. To evaluate the correlation between TC, FC, and FS statistical analyses were performed by using Minitab. And a part of the presumptive TC/FC and background colonies was purified and identified using API 20E kit (Biomeriux). As results, most (89%) of presumptive FC by MF was identified as Escherichia coli while only 14% (MPN) and 11% (MF) of TC were identified as E. coli. Furthermore, FC by MF was correlated significantly with other fecal indicators (TC/FS by MF and FC by MPN), while TC by MPN was not correlated with any other indicators. Thus, the detection of FC by MF-method may be the most reasonable for monitoring the fecal pollution.
