Component Analysis and Antioxidant Activity of Kalopanax pictus Leaf (개두릅의 함유성분 분석과 항산화 활성)

  • The purpose of this study is to determine the possibility of using Kalopanax pictus leaf as natural health food source. The contents of proximate and antioxidative nutrients of Kalopanax pictus leaf were measured. The contents of carbohydrate, crude protein, crude lipid and ash were 41.42%, 45.23%, 3.29% and 10.07%, respectively. The calories of Kalopanax pictus leaf was 401.52 kcal/100 g and total dietary fiber was 30.37%. The percentages of water soluble dietary fiber to insoluble dietary fiber were 9.16% and 21.21%, respectively. The protein contained a total of 18 different kinds of amino acids. The contents of essential and non-essential amino acids were 10.51 g/100 g and 17.69 g/100 g. The K was the largest mineral followed by P, Ca, and Mg, which means Kalopanax pictus leaf is alkali material. The contents of saturated fatty acids, monounsaturated fatty acids and polyunsaturated fatty acids were 1.56 g/100 g, 0.11 g/100 g and 1.84 g/100 g, respectively. The antioxidant activity of 70% ethanol extract and fractions of the Kalopanax pictus leaf has been determined by the scavenging of the stable radical DPPH; the result showed that the ethyl acetate fraction was the most active, as the amount required for 50% reduction of DPPH after 30 mins ($RC_{50}$) was $105.7\;{\mu}g$, followed by 70% ethanol extract ($247.3\;{\mu}g$), hexane fraction ($120.7\;{\mu}g$), chloroform ($107.3\;{\mu}g$), butanol fraction ($110.1\;{\mu}g$) and aqueous fraction ($491.9\;{\mu}g$).

  • We previously reported the antimutagenicity, anti-lipid peroxidation and antiinflammtory properties of the three Chinese herb medicine, Kalopanax pictus, Pueraria thunbergiana and Rhus verniciflua, which have been used as therapeutics for diabetes mellitus and inflammation in Korea. Since these drugs have been used for the prescriptions consisted of several crude drugs, we investigated whether the extract of the three drugs could increase the individual drugs or not. The three extracts were independently prepared from K. pictus, P. thunbergiana, and R. verniciflua, which extracts were named the K-1, P-1 and R-1 in this order and the extract (KPR-1) was also prepared from the combined plant materials with each same weight of the three drugs. Synergistic Effect of Methanol Extract from Kalopanax pictus and Ascorbic Acid on Antioxidant, Anticancer and Immunomodulatory Activities (음나무(Kalopanax pictus) 추출물과 비타민 C의 항산화, 항암 및 면역활성 상승효과)

  • The 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid (ABTS), nitric oxide (NO) scavenging activities and ferric- reducing/antioxidant power (FRAP) assay against extracts of Kalopanax pictus (KP) were measured. Radical scavenging and antioxidant activities were increased depend on the concentration and the effects were enhanced by ascorbic acid (AA). KP extracts and AA had a good anti-proliferating activity against HepG2 cells by MTT assay and induces cells apoptosis, which was demonstrated by flow cytometric analysis. KP extracts and AA caused the arrest of cell-cycle progression at either G0-G1-phase or G2/M-phase, which might be depending upon the KP extracts concentration. In addition, KP extracts and AA are effective in enhancing immunity and nitric oxide production by RAW 264.7 macrophages cells. KP extracts and AA inhibited tumor cell growth and exerted antioxidant effects as compared to controls. These results demonstrate that simultaneous AA and KP extracts treatment could be useful in preventing the oxidative damage and anti-proliferating HepG2 cells, and are effective in enhancing immunomodulatory and antioxidant activity.

Effects of Dietary Supplementation of Castor Aralia (Kalopanax pictus Nakai) Leaf Powder on Blood Characteristics of Chicken (육계 사료 내 엄나무 잎 분말 첨가 급여가 육계의 혈액 조성에 미치는 영향)

  • The objective of this study was to investigate the effects of supplementation diets with Kalopanax pictus leaf powder on performance, proximate composition, carcass characteristics and blood characteristics of chicken meat. Broiler chicks were fed diets containing Kalopanax pictus leaves with concentration of 0% (Control), 0.5% (T1), 1.0% (T2), and 2.0% (T3) for five weeks. There were no significant differences in performance among treatment, however there were significant decrease in mortality within treatment. The crude fat of chicken meat was increased in diets by addition of Kalopanax pictus leaf powder than control, but moisture, crude protein, and crude ash were no significant differences. The abdominal fat was significantly increased by the supplementation of Kalopanax pictus leaf powder compare to the control (P<0.05). The total cholesterol, LDL-cholesterol and triglyceride of control were higher than treatment groups, and the HDL-cholesterol and glucose of control were lower than treatment groups (P<0.05). Especially, T2 and T3 were significantly (P<0.05) more effective in improving cholesterol compare to other treatment groups. In conclusion, these data indicate that supplementation of 1.0% Kalopanax pictus leaf powder (T2) were most effective in decreasing total cholesterol, LDL-cholesterol and triglyceride and increasing HDL-cholesterol.

  • In the previous study. we isolated kalopanaxsaponin A and pictoside A from the EtOAc fraction of Kalopanax pictus extract. In the present study, the BuOH fraction of K pictus extract was hydrolyzed by alkali and antirheumatic effect of the fraction was evaluated. It was found that the hydrolysate of the BuOH fraction showed inhibition of adjuvant-induced arthritis in rats. Of the EtOAc and BuOH fractions of the hydrolysate, only the former exhibited anti-arthritic activity. (omitted)

Monthly Variation of the Kalopanaxsaponin Content in the Leaves of Kalopanax pictus (개두릅에서 칼로파낙스사포닌의 월별 함량 변화의 추이)

  • To find the monthly variation of kalopanaxsaponin contents in the leaves of Kalopanax pictus with thorns (KPT) and with no thorns (KPN), the leaves monthly collected from May to September were extracted with MeOH and then the kalopanaksaponin fractions were prepared. KPT collected on May showed the lowest saponin content of the KPTs whereas KPT on August exhibited the highest saponin content. From September, the saponin content in the loaves decreased. The highest saponin content was shown to be 7.3% in KPN collected on August. Evaluation of six kalopanaxsaponins A, I, J, B, H and K (KPA, KPI, KPJ, KPB, KPH, and KPK) were performed using TLC densitometer. In this measurement, considerably higher KPB and KPH, both hederagenin bisdesmosides, were found whereas very low contents In monodesrnosides KPI and KPJ were observed. In conclusion, it was shown that the leaves of KPN of August could be a biomaterial source for the kalopanaxsaponin fraction. It was also suggested that measurement of the weight of kalopanaxsaponin fraction and the content of KPB as the representative compound for kalopanaxsaponins will be used for the quantitative evaluation of the kalopanaxsaponins of K. pictus.

  • An experiment was conducted to examine the effects of Kalopanax pictus Nakai (Kalopanax) on in vitro rumen fermentation and methane (CH4) reduction. Kalopanax trunk was extracted with 70% ethanol and 70% methanol. Rumen fluid, alfalfa hay and buffer (control: C) supplemented with 0.3% Kalopanx juice (T1), 0.3% ethanol extract (T2) and 0.3% methanol extract (T3) in the total volume of culture medium were incubated at $38^{\circ}C$ for 24h and 48h. Rumen pH was lower in all Kalopanax treatments during all incubations than that in control (p<0.05). Total VFA and total gas production in T2 and T3 was significantly higher than that in C at 48h incubation (p<0.05). Ammonia-N was decreased in all treatments compared with C during the incubation periods (p<0.05). At 24h incubation, $CH_4$ contents were significantly reduced by both alcohol extracts. It is concluded that supplementing Kalopanax extracts can stimulate ruminal fermentation of rumen microorganisms and inhibit methanogenesis.

Oxidative Stress in the Cell and Antioxidant Activity of Kalopanax Pictus Extracts (음나무 추출물의 세포 내 산화 스트레스와 항산화 활성)

  • This study reviewed the application of an extract from Kalopanax pictus stem bark and root bark as natural antioxidants. To investigate the effect on cell toxic level against transformed mouse fibroblast L929 in formula added with various extracts from Kalopanax pictus stem bark and root bark, this experiment was carried out by in vitro cytotoxicity method. Using DCFA-DA method, oxidative stress in cell was measured with other antioxidant activity methods including DPPH assay and NBT assay. Active oxygen inhibition rate for root bark insoluble hot water extracts showed the highest with 57.9% for 15 min treatment. In DPPH and NBT test, antioxidant activities of methanol extract from stem bark and insoluble hot water extract from stem bark were 96% (at 0.1%) and 95% (at 0.5%), respectively.

Growth Factors Affecting to Kalosaponins Contents of Kalopanax pictus Nakai (한국산 음나무의 Kalosaponins 함량과 이에 영향하는 몇 가지 생장요인)

  • Four kinds of kalosaponins from inner bark of Kalopanax pictus Nakai as a medicinal and edible timber tree species growing in Korea was analyzed by HPLC. A considerable range of variation in the contents was observed among population. The kalosaponins contents in inner bark from each population were the highest in the Mt. Barwang (30.37 mg/g on the dry weight basis) followed by Mt. Gariwang, Hanra II, Mangun, Paltan, and Hanra II population. Growth factors that might have influenced the contents of the kalosaponins were also examined. As both analysis results of partial correlation and multiple regression, the weight of inner bark and tree age were influenced the kalosaponin O contents. The kalosaponin B contents was influenced by both the weight of inner layer bark and annual mean DBH increment. The contents of kalosaponins appeared to be closely related to the inner bark characteristics.
