Comparison of Genetic Diversity and Population Structure of Kalopanax pictus (Araliaceae) and its Thornless Variant Using RAPD

  • Kalopanax pictus is a long-lived woody species mostly distributed in East Asia. K. pictus has been regarded as medically and ecologically important species in Korea. Thornless castor aralia variant, local name 'Cheongsong' is an endemic to Cheongsong province in Korea. Random amplified polymorphic DNA (RAPD) was used to investigate the genetic variation and structure of Korean populations of two species. A high level of genetic variation was found in six K. pictus populations. Twelve primers revealed 49 loci, of which 29 were polymorphic (59.2%). Nei's gene diversity for K.pictus and K. pictus variant were 0.119 and 0.098, respectively. Mean of genetic diversity in K. pictus was higher than average values for species with similar life history traits. The asexual and sexual reproduction, perennial habitat, and longevity are proposed as possible factors contributing to high genetic diversity. An indirect estimate of the number of migrants per generation (Nm=0.857) indicated that gene flow was not extensive among Korean populations of K.pictus. It is suggested that the isolation of geographical distance and reproductive isolation between K.pictus and K.pictus variant populations may have played roles in shaping the population structure of this species.

Comparison of Genetic Diversity and Relationships of Genus Kalopanax Using ISSR Markers (ISSR을 이용한 음나무속 분류군의 유전적 다양성과 관련성 비교)

  • Inter simple sequence repeat (ISSR) markers were performed in order to analyse the phylogenetic relationships of four taxa of Castor-aralia (Kalopanax pictus): K. pictus, K. pictus var. magnificus, K. pictus var. maximowiczii, and thornless K. pictus. The 11 primers were produced 64 reproducible ISSR bands. Analysis of ISSR from individual plants of Korean K. pictus resulted in 41 polymorphic bands with 64.1%. When species were grouped by four taxa, within group diversity was 0.115 $(H_S)$, while among group diversity was 0.467 $(G_{ST})$ on a per locus basis. The estimated gene flow (Nm) for K. pictus var. maximowiczii and K. pictus var. magnificus were very higher than K. pictus. It is suggested that the isolation of geographical distance and reproductive isolation among K. pictus populations may have played roles in shaping the population structure of this species. In phenetic tree, ISSR markers are very effective in classifying natural populations as well as taxon levels of genus Kalopanax in Korea.

Genetic diversity of Kalopanax pictus populations in Korea based on the nrDNA ITS sequence

  • $Kalopanax$ $pictus$ is a long-lived deciduous perennial tree in the family Araliaceae mainly distributed in the East Asia. In Korea, this species is of ecological and medical importance. Because typical populations of this species are small and distributed in patches, $K.$ $pictus$ has been considered as a narrow habitat species. To understand the genetic diversity and population structure of this species, the sequence variation of the nuclear ribosomal DNA (nrDNA) internal transcribed spacer (ITS) region was analyzed among 18 different $K.$ $pictus$ populations in the present investigation. The nrDNA ITS sequences of Korean populations investigated in this study showed identical of 616 bp in length, and no any nucleotide variation was found in the entire nrDNA ITS region sequence. This result suggested that the $K.$ $pictus$ populations in Korea might belong to the same isolate, and no mutation was found in the nrDNA ITS region. Compared with other known ITS sequence sources from $K.$ $pictus$ populations, only four variable nucleotide sites were found within the entire ITS region. Very narrow genetic diversity appearing in the population level of $K.$ $pictus$ makes us hypothesize that their relatively isolated habitats. The long-lived traits might be one main reason. However, another probability was that the nr-DNA ITS region might be noneffective in classifying populations of $K.$ $pictus$. Thus, to further understand the phylogenetic relationship of $K.$ $pictus$ populations, more samplings should be performed based on more DNA sequences.

Quality Characteristics of Noodles Containing Shoots and Leaves of Aralia elata and Kalopanax pictus (참두릅과 개두릅을 첨가한 국수의 제조특성)

  • The objective of this study was to evaluate the quality characteristics of noodles made with a dried powder of the shoots and leaves from $Aralia$ $elata$and $Kalopanax$ $pictus$. Ash, protein, and dietary fiber contents of $A.$ $elata$ and $K.$ $pictus$ were higher than those of wheat flour. The protein and fat contents of $K.$ $pictus$ powder were higher than those of the $A.$ $elata$ powder, but ash and dietary fiber contents were less than those of $A.$ $elata$. When the amount of $A.$ $elata$ powder increased, brightness (L) values of dough decreased, and redness (a) and yellowness (b) values of dough increased. The weight and volume of the cooked noodles also increased, as did the turbidity of the soup. Addition of $A.$ $elata$ and $K.$ $pictus$ powders in noodles reduced the cutting forces needed to cut the cooked noodles. The sensory evaluation indicates that noodles with 5% $A.$ $elata$ and $K.$ $pictus$ powder added were the most preferable.

Effects of Dietary Supplementation of Castor Aralia (Kalopanax pictus Nakai) on Physico-chemical Properties and Quality of Chicken Thigh Meat (사료에 엄나무 잎 분말을 첨가 급여한 닭다리 육의 이화학적 특성 및 품질에 미치는 영향)

  • This study were investigated the effects of dietary supplementation of ground Kalopanax pictus leaves powder on pH, total phenol contents, DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity, TBARS, WHC, shear force, meat color, sensory evaluation, and fatty acid composition of chicken meat. Broiler chicks were fed diets for five weeks containing 0% Kalopanax pictus leaves powder (Control), 0.5% Kalopanax pictus leaves powder (T1), 1.0% Kalopanax pictus leaves powder (T2), and 2.0% Kalopanax pictus leaves powder (T3). The pH and TBARS were significantly decreased by the supplementation of Kalopanax pictus leaves powder compared to the control (P<0.05). The total phenol contents and DPPH radical scavenging activity were significantly increased by the supplementation of Kalopanax pictus leaves powder compared to those of the control group (P<0.05), and especially, T3 was significantly (P<0.05) more effective in improving freshness compared to other treatment groups. The WHC and shear force were not significantly different. CIE $L^*$ and $a^*$ value of treatment groups (especially T3) showed significantly higher value compare to the control, however, no difference in the CIE $b^*$ values was observed among treatment groups. In fatty acid composition, linolenic acid of chicken meat was increased by the supplementation of Kalopanax pictus leaves powder than compared to those of the non-supplementation group. In conclusion, a supplementation of Kalopanax pictus leaves powder was effective in decreasing pH and TBARS, and increasing total phenol contents and DPPH radical scavenging activity.

Activation of the Chemosensory Ion Channels TRPA1 and TRPV1 by Hydroalcohol Extract of Kalopanax pictus Leaves

  • TRPA1 and TRPV1 are members of the TRP superfamily of structurally related, nonselective cation channels. TRPA1 and TRPV1 are often co-expressed in sensory neurons and play an important role in somatosense such as cold, pain, and irritants. The first leaves of Kalopanax pictus Nakai (Araliaceae) have long been used as a culinary ingredient in Korea because of their unique chemesthetic flavor. In this study, we observed the intracellular $Ca^{2+}$ response to cultured cells expressing human TRPA1 (hTRPA1) and human TRPV1 (hTRPV1) by $Ca^{2+}$ imaging analysis to investigate the ability of the first leaves of K. pictus to activate the hTRPA1 and hTRPV1. An 80% ethanol extract of K. pictus (KPEx) increased intracellular $Ca^{2+}$ influx in a response time- and concentration-dependent manner via either hTRPA1 or hTRPV1. KPEx-induced response to hTRPA1 was markedly attenuated by ruthenium red, a general blocker of TRP channels, and HC-030031, a specific antagonist of TRPA1. In addition, the intracellular $Ca^{2+}$ influx attained with KPEx to hTRPV1 was mostly blocked by ruthenium red, and capsazepine, a specific antagonist of TRPV1. These results indicate that KPEx selectively activates both hTRPA1 and hTRPV1, which may provide evidence that the first leaves of K. pictus primarily activate TRPA1 and TRPV1 to induce their unique chemesthetic sense.

Food Nutritional Composition of Castor Aralia (Kalopanax pictus N) Sprouts (음나무(Kalopanax pictus N) 순의 식품학적 성분 분석)

  • This study was carried out to investigate the food value of K. pictus sprouts. The total ginsenoside content was 67.4 mg/kg. Including Ginsenoside Rg1, a total of five different types were detected by the experiment. The total phenol content was 33.77 mg/100 g. Including seven different type of essential amino acids, the total amino acid content of K. pictus sprouts was 33,744 mg/100 g. The results of the analysis of the free amino acid content showed that the total bitterness was much higher than the total sweetness. From the above results, the food characteristics of K. pictus sprouts could be determined. To improve their consumption, their bitterness must be reduced.

Sensory Properties of $Kalopanax$ $pictus$ and $Cedrela$ $sinensis$ Shoots under Different Blanching Conditions and with Different Thawing Methods (엄나무와 참죽 햇순의 blanching 조건 및 해동방법에 따른 관능품질 특성)

  • The sensory properties of $Kalopanax$ $pictus$ and $Cedrela$ $sinensis$ shoots were analyzed according to their blanching conditions and the thawing methods used. For the Hunter colors, the $K.$ $pictus$ that was blanched without adding salt for 6 min had the highest L value, and the $C.$ $sinensis$ that was blanched with 1% salt for 6 min had the highest L value. The chlorophyll content of $K.$ $pictus$ was 141.1 mg% when it was blanched for 3 min with 2% salt, and the chlorophyll content of $C.$ $sinensis$ that was blanched without adding salt for 6 min was highest (124.8 mg%). When $K.$ $pictus$ and $C.$ $sinensis$ were blanched without salt addition for 3 min, their texture showed a higher value than that of the other samples. The sensory test result showed the highest overall preference for the sample that was blanched without adding salt for 3 min, among all the samples. The L value of the $K.$ $pictus$ that was thawed in a microwave oven was significantly higher than that of the other samples, but the L value of the $C.$ $sinensis$ that was thawed at $4^{\circ}C$ was higher than that of the other samples. As for the overall preference for the samples according to the thawing method used, $K.$ $pictus$ and $C.$ $sinensis$ scored highest in the case of thawing in a microwave oven.

First Record of the Frogfishes Antennarius pictus (Antennariidae, Lophiiformes) from Korea (한국산 씬벵이과 어류 1미기록종 Antennarius pictus)

  • Two specimens of Antennarius pictus, belonging to the family Antennariidae, were collected from the coastal waters of Jeju Island, Korea. They were characterized by having 7 anal fin rays, 10 pectoral fin rays, 12 dorsal fin rays, 9 caudal fin rays, and second spine of dorsal fin diminishing and curved backwards distally. Since this species has various sizes of round black spots on head, body and fins, we propose a new Korean fish name "Heuk-jum-bak-ssin-beng-i" for A. pictus.

Insulin Sensitizing and Insulin-like Effects of Water Extracts from Kalopanax pictus NAKAI in 3T3-L1 Adipocyte (엄나무 추출물이 3T3-L1 지방세포에서 인슐린 민감성과 인슐린 유사성 작용에 미치는 영향)

  • Effects of the water extract from Kalopanax pictus NAKAI on insulin-like action and glucose uptake in 3T3-L1 cells were investigated. The bark of K. pictus NAKAI was treated with hot water and the extract was freeze-dried. Total extract of K. pictus NAKAI was fractionated into 6 fractions with increasing gradients from 0 to 100% MeOH on Amberlite XDA-4. Treatment of 3T3-L1 fibroblasts with 1 ${\mu}g/ml$ and 10 ${\mu}g/ml$ of K. pictus NAKAI total extracts significantly increased the differentiation of the cells. When co-treated with inducers such a dexamethasone, 1-methyl-3-isobutylxanthine and insulin, the differentiation was increased at 1 ${\mu}g/ml$ of total extract, but not at 10 ${\mu}g/ml$. In 3T3-L1 adipocytes, glucose uptake was increased by 3.3 times with addition of 0.3 and 3 ${\mu}g/ml$ of Fr. 1 (0-10% MeOH) and Fr. 3 (30% MeOH) at 3 ng/ml insulin. In conclusion, K. pictus NAKAI contains such compounds that play a role of insulin-like action and insulin sensitizer.
