The Influence of Dietary Calcium and Phosphorus Imbalance on Intestinal NaPi-IIb and Calbindin mRNA Expression and Tibia Parameters of Broilers

  • A $2{\times}2$ factorial experiment was conducted to study the effect of dietary calcium and non-phytate phosphorus (nPP) imbalance on calbindin and NaPi-IIb mRNA levels in the small intestine and tibia parameters of broiler chicks. One hundred and forty four 1-d-old Arbor Acres male broiler chicks were divided into four treatments consisted of six replicates with six chicks each. The two dietary calcium levels were 1.10% and 0.60%, and two dietary nPP levels were 0.50% and 0.27%. Results showed that a high Ca/nPP ratio diet (4.07:1) significantly depressed feed intake and weight gain of broilers (p<0.05), but a lower Ca:nPP ratio (1.2:1) had no influence (p>0.05). Low-Ca with low-P diet resulted in low tibia minerals and tibia breaking strength of broilers, and all the tibia parameters were further decreased when the dietary ratio of Ca to P was relative higher. Low dietary Ca or P up-regulated the calbindin and NaPi-IIb mRNA expression levels. Low Ca with normal P diet up-regulated duodenal calbindin mRNA expression level to the greatest extent. Low P with a normal Ca diet significantly enhanced NaPi-IIb mRNA expression level to the highest extent. These results suggest that the calbindin and NaPi-IIb mRNA expression were enhanced by the imbalance between dietary Ca and nPP, and their expression were not only influenced by Ca or nPP level, but also the ratio of Ca:nPP.

Different Phosphate Transport in the Duodenum and Jejunum of Chicken Response to Dietary Phosphate Adaptation

  • Intestinal phosphate (Pi) absorption across the apical membrane of small intestinal epithelial cells is mainly mediated by the type IIb Na-coupled phosphate co-transporter (NaPi-IIb), but its expression and regulation in the chicken remain unclear. In the present study, we investigated the mRNA and protein levels of NaPi-IIb in three regions of chicken small intestine, and related their expression levels to the rate of net phosphate absorption. Our results showed that maximal phosphate absorption occurs in the jejunum, however the highest expression levels of NaPi-IIb mRNA and protein occurs in the duodenum. In response to a low-Pi diet (TP 0.2%), there is an adaptive response restricted to the duodenum, with increased brush border membrane (BBM) Na-Pi transport activity and NaPi-IIb protein and mRNA abundance. However, when switched from a low-(TP 0.2%) to a normal diet (TP 0.6%) for 4 h, there is an increase in BBM NaPi-IIb protein abundance in the jejunum, but no changes in BBM NaPi-IIb mRNA. Therefore, our study indicates that Na-Pi transport activity and NaPi-IIb protein expression are differentially regulated in the duodenum vs the jejunum in the chicken.

Effect of Dietary Ca and Na Levels on Blood Pressure and Mineral Metabolism in Spontaneously Hypertensive Rats Fed High Fat Diet (고지방식을 섭취한 본태성 고혈압쥐에서 칼슘과 소디움 섭취가 혈압과 무기질 대사에 미치는 영향)

  • This study explored the effect of dietary levels of Na and Ca on spontaneously hypertensive rats (SHR). SHR were randomly divided into 5 groups and fed a high fat/cholesterol diet containing three levels of Na (0.05, 0.1, 1.5%) and Ca (0.1, 0.5, 1.5%) for 9 weeks. Body weight gain was not influenced by dietary intake but water intake significantly increased in high Na supplementation. Systolic blood pressure was not influenced by dietary Na and Ca levels but was decreased by dietary low Na/high Ca levels at 9 weeks. Angiotensin-II level was affected by dietary Na level but not by Ca levels. Plasma Ca, Mg, K and Na levels were in the normal range regardless of dietary Na and Ca levels. Weight, and K and Na contents of the heart and kidney were not significantly different among those with different dietary Na and Ca levels. Ca and Mg contents of the heart and kidney were significantly higher in the normal Na/normal Ca group. Ca and Mg in the feces were higher in those with high Ca intake. Na in the feces was higher in those with high Na intake. Therefore, Na and Ca had different mechanisms in the hypertension/hyperlipidemia models, respectively. And we suggested that Mg must be supplemented when Ca intake was high because Mg excretion was increased by Ca supplementation.

Effect of Ginseng on $Na^+$, $K^{+}-ATPase$ Activities of Potassium Deficient Rat Intestinal Mucosa (인삼이 칼륨결핍랫트 장점막의 $Na^+$,$K^{+}-ATPase$ 활성에 미치는 영향)

  • We have studied the effect of fasting on $Na^+$, $K^{+}-ATPase$ activities in the rat intestinal mucosa. Rats were fasted for $18{\sim}48hr$. Intestinal microsomal fraction was prepared by the method of Robinson and ATPase activities were determined by the modified method of Fiske and Subbarow. $Na^+$, $K^{+}-ATPase$ activity was not changed after fasting for 18 and 24 hr but significantly decreased after fasting for 48 hr. Fasting over 18 to 48 hr period had no effect on the $Mg^{++}-ATPase$. Thus, it may be concluded that 48 hr fasting has inhibitory effect on rat intestinal absorptive capabilities. In order to study the effect of Ginseng on the $Na^+$, $K^{+}-ATPase$ activities of the small intestine in chronic $K^{+}-depleted$ rats, rats were fed $K^{+}-depleted$ diets for 3 weeks and Ginseng ethanol extracts were administered orally for 3 weeks concomitantly. ATPase activity was measured by the same method as fasting group. $Na^+$, $K^{+}-ATPase$ activity in the $K^{+}-depleted$ diet group was increased and Ginseng ethanol extracts inhibited the increase of enzyme activity induced by $K^{+}-depleted$ diet. Thus, it may be suggested that increase in the intestinal $Na^+$, $K^{+}-ATPase$ activity of chronic $K^{+}-depleted$ group may be due to the compensatory mechanism and administration of Ginseng with $K^{+}-depleted$ diet may be associated with inhibition of increase in the enzyme activity of the $K^{+}-depleted$ group due to the prevention of the $K^+$ loss in the $K^{+}-depletion$.

Discrimination of Astaxanthin Fed Laying Hens and Their Peroxidated Carcasses by Electronic Nose

  • The applicability of electronic nose was tested to detect lipid peroxidation in chickens and to measure antioxidant effect of astaxanthin in chicken carcasses. Two sources of astaxanthin were fed to 62-wk-old spent laying hens to improve meat quality: natural astaxanthin (NA) from the red yeast, Phaffia rhodozyma, and synthetic astaxanthin (SA) from chemical synthesis. One hundred forty four ISA Brown laying hens were used in a 6-wk feeding trial. Three treatments consisted of the basal diet (control), SA (100 mg astaxanthin/kg basal diet) and NA (50 mg astaxanthin/kg basal diet). The astaxanthin levels of SA and NA were set to give a similar degree of skin pigmentation. After 6-wk feeding of astaxanthin, the skins from NA and SA were discriminated from the control by electronic nose. However, electronic nose failed to distinguish between SA and NA skins after 6-wk feeding. The astaxanthin level differences between skins of SA and NA were not remarkable during the 6-wk trial. The lipid peroxide formation in skin was significantly decreased by SA but not by NA. The antioxidation effect of SA was detected by electronic nose because SA skin was discriminated from others. NA was a better pigmentation agent than SA, but the reverse was true in antioxidation. Electronic nose is applicable for detecting astaxanthin in chicken, and meat off-flavor caused by lipid peroxidation during storage.

UTILIZATION OF EGYPTIAN MALLOW IN FEEDING COMMON CARP (Cyprinus carpio L.)

  • Two experiments were conducted to study the effect of using Egyptian mallow leaf meal (EMLM) on growth performance and feed utilization of common carp (Cyprinus carpio L.) in experiment 1. Four diets containing 0, 5, 15 and 25% EMLM were included at the expense of berseem leaf meal and fed to fingerlings of common carp for 98 days. The results showed that the average daily gain, feed intake and feed coefficient ratio (FCR) were improved (p<0.05) with increasing the level of EMLM in the diet. Fish fed 25% EMLM were similar to control fish (30% berseem leaf meal) in the productive value (PPV%). In experiment 2, five diets were prepared to contain: 1) 30% berseem leaf meal, 2) 25% untreated EMLM, 3) 25% cooked EMLM 4) 25% treated EMLM with 0.5% NaOH and 5) 25% treated EMLM with 1% NaOH. The results showed that diet containing EMLM gave the best growth performance and feed utilization. However, diet containing 1% NaOH treated EMLM superior to the other diets in PPV% and energy utilization. Diets containing 0.5% NaOH-treated EMLM or cooked EMLM decreased the protein utilization compared to those containing EMLM.

Effect of coenzyme Q10 and Ardisia japonica Blume on plasma and liver lipids, platelet aggregation, and erythrocyte Na efflux channels in simvastatin-treated guinea pigs

  • Forty guinea pigs were divided into four groups and fed 0.04% cholesterol based control diet, plus 0.05% simvastatin, and statin plus 0.1% CoQ10 or 10% Ardisia Japonica Blume (AJB) leave powder for 4 weeks. Plasma total cholesterol levels decreased significantly in all groups fed the statin-containing diet compared with that in guinea pigs fed the control diet (P < 0.01). Plasma and liver triglycerides decreased significantly in the statin plus CoQ10 group compared with those in the control (both P < 0.05). Maximum platelet aggregation was significantly higher in the statin plus CoQ10 group than that in the other groups (P < 0.05). Na-K ATPase activity increased in the statin group and decreased in the statin plus CoQ10 group (P < 0.01). Na-K co-transport and Na passive transport decreased significantly in the control group compared with those in the other groups (both P < 0.05). Intracellular Na was highest in the statin group and lowest in the statin plus CoQ10 group and was correlated with Na-K ATPase activity. Thiobarbituric acid reactive substance production in platelet-rich plasma and liver tended to decrease in the statin plus CoQ10 group compared with those in the other groups. Plasma glutamic-pyruvic transaminase and glutamic-oxaloacetic transaminase increased significantly in the statin group compared with those in the control (P < 0.05). These result suggest that antioxidant rich AJB did not have positive effects on cardiovascular disease parameters. The statin plus CoQ10 seemed to decrease cholesterol more efficiently than that of statin alone.

Elevation of heart-femoral pulse wave velocity by short-term low sodium diet followed by high sodium diet in hypertensive patients with sodium sensitivity

  • BACKGROUND/OBJECTIVES: We compared changes in heart-femoral pulse wave velocity (hfPWV) in response to low sodium and high sodium diet between individuals with sodium sensitivity (SS) and resistance (SR) to evaluate the influence of sodium intake on arterial stiffness. SUBJECTS/METHODS: Thirty-one hypertensive and 70 normotensive individuals were given 7 days of low sodium dietary approach to stop hypertension (DASH) diet (LSD, 100 mmol NaCl/day) followed by 7 days of high sodium DASH diet (HSD, 300 mmol NaCl/day) during 2 weeks of hospitalization. The hfPWV was measured and compared after the LSD and HSD. RESULTS: The hfPWV was significantly elevated from LSD to HSD in individuals with SS (P = 0.001) independently of changes in mean arterial pressure (P = 0.037). Conversely, there was no significant elevation of hfPWV from LSD to HSD in individuals with SR. The percent change in hfPWV from the LSD to the HSD in individuals with SS was higher than that in individuals with SR. Subgroup analysis revealed that individuals with both SS and hypertension showed significant elevation of hfPWV from LSD to HSD upon adjusted analysis using changes of the means arterial pressure (P = 0.040). However, there was no significant elevation of hfPWV in individuals with SS and normotension. CONCLUSION: High sodium intake elevated hfPWV in hypertensive individuals with SS, suggesting that high sodium intake increases aortic stiffness, and may contribute to enhanced cardiovascular risk in hypertensive individuals with SS.

Comparison of Dietary Intakes by 24-hr Dietary Recall, Dietary Record and Food Frequency Questionnaire among Elderly People (회상법, 기록법 및 식품섭취빈도조사법을 이용한 노인의 영양소 섭취 수준의 비교)

  • Nutrient intakes estimated using a 24-hr recall, a dirt record and a food frequency questionnaire(FFQ) were compared in a group of ninety-four elderly people(21 males, 73 females) in Cheongju, a city in Chung-Buk province. Mean intakes for energy, protein, Ca, p, Na, K thiamin and niacin obtained from the diet record were higher than those from the 24-hr recall. Mean intakes for energy, protein, Ca, P, Na, K thiamin, niacin and vitamin C from the FFQ were higher than those obtained from the 24-hr recall or the diet record. Correlation coefficients between the nutrient intake values from the 24-hr recall and those from the diet record ranged from 0.84 to 0.95 and were significantly correlated(p < 0.001). About 80% of the subjects in the lowest quintile by the 24-hr recall were also in the lowest two quintiles by the diet record. While the percentage fallen into the opposite category ranged from 0% to 15%. For most nutrients, at least 65% of the subjects when classified by the 24-hr recall fell into the same quintile when classified by the diet record, and the mean kappa value was 0.7. About 52% of the subjects in the lowest category by the 24-hr recall fell into the lowest two categories by FFQ. The mean percentage of the subjects in the lowest quintile by the diet record or in the lowest two quintiles by the FFQ was 51%. For most nutrients, at least 24% of each of the subjects when classified by both the 24-hr recall and the diet record fell into the same category when classified by the FFQ. The kappa values between the 24-hr recall or the diet recall and the FFQ were 0.17. These data indicate that in elderly subjects the 24-hr recall can provide very similar information to that obtained from the diet record but the FFQ can not provide good information if the right FFQ method is not used for only elderly subjects.

Effects of Fiber and Vitamin Food Sources on Mineral Balance (무기질 출납에 있어서의 섬유질 및 Vitamin 급원 식품의 영향)

  • Present study was to investigate the effect of dietary vegetable sources such as carrot, peahull and squash on mineral balance of Na, K, Ca, Fe, Cu, Zn and Mg in mice. It would also determine if the kind, level and feeding period of vegetables can affect the mineral balance. One hundred and thirty mice weighing $29{\sim}30g$(8 weeks of age) were randomly selected. Ten mice were fed their standard diet, and the others were divided into three experimental groups that were fed standard diet supplemented with carrot, peahull and squash. Mineral intake and urinary and fecal excretion of minerals were measured to investigate the effect of the kind, level and feeding period of vegetables on absorption rate and balance of minerals. Body-weight gain was increased in high peahull diet group compared to the control. No significant difference was observed for feed efficiency ratios between different experimental groups and urine volume was increased by vegetable fed group. Na, Ca, Fe, and Cu intake were lower in high squash-diet fed soup. K intake was higher in carrot fed mice, and Zn, and Mg intake were higher in peahull fed mice. Absorption rate and balance of mineral in control group resulted in positive balance while experimental groups showed negative. The kind of vegetable, feeding level and feeding period affected the intake of Na, K, Ca, Cu and absorption rate of Cu, Zn and balance of K, Cu, Zn to negative. In spite of the higher intake of t Zn and Mg, in experimental groups the absorption rate showed negative balance.
