Isolation and Cultivation Characteristics of Acetobacter xylinum KJ-1 Producing Bacterial Cellulose in Shaking Cultures

  • Eight strains producing bacterial cellulose (BC) were isolated from rotten fruits and traditionally fermented vinegars. One of the isolated strains from the rotten grape in Gwangju, Korea, maintained a relatively stable BC production in shaking cultures. This isolated strain proved to be Acetobacter xylinum, based on several biochemical and morphological tests. It was shown that the slant-baffled flask was more efficient than the conventional flask for the BC production in shaking cultures. To determine the most suitable carbon and nitrogen sources for the production of BC, various compounds were examined. Fructose was found to be the most effective carbon source with an optimal concentration of 2%. Mixed carbon source (glucose:fructose=1:3) was also better than glucose or fructose alone. Optimal nitrogen source, when basal medium was used, was 10% (v/v) com steep liquor (CSL). When com steep liquor was used with a mixed carbon source (glucose:fructose=1 :3),4% CSL exhibited the best BC production. Based on these results, a defined medium was developed for the BC production by Acetobacter xylinum KJ-1. When this medium was used under optimal culture conditions, the BC production was 7.2 g/1, which was approximately 3 times higher than that with the traditional HS medium.

  • The Neospora sp. was isolated from the brain of 1 calf via continuous in vitro cultivation in Vero cell. Neospora tachyzoites were observed 45 days after inoculation of the homogenized brain suspension into the Vero cell. The isolated parasite (named tentatively as NCKB-1) was morphologically and ultrastructurally similar to the previously reported Neospora sp isolated in cattle (BPA-1, JPA-1). A comparison of the antigenic reactivity of cultivated tachyzoites with polyclonal antisera to Neospora caninum and Toxoplasma gondii confirmed that this protozoal isolate was similar to N caninum. This is the first report of successful isolation of Neospora sp from cattle in Korea.

  • Microcystis aeruginosa is common form of cyanobacteria (blue-green algae) capable of producing toxic heptapeptide (microcystin) that cause illness or death. The comparison of molecular genetic method with the morphological characteristics of cyanobacteria was conducted. We have designed PCR primers (JJM98F, JJM1141R) for cyanobacterial 16S rRNA and phycocyanin intergenic spacer (PC-IGS) gene domain. To confirm the production of microcystins, PCR primers for the N-methyltransferase (NMT) domain of microcystin synthetase gene mcyA were designed using 21 cyanobacteria strains Most of isolated strains from the Nakdong River was classified as Microcystis aeruginosa and the similarities were $99\%$ with M. aeruginosa AF 139292. $38.1\%$ of isolated strains contained microcystin synthesis gene. NMT (N-methyltransferase) were not detected in isolated strain in several strains, which means non-toxic. However, the NMTs of the strains were detected during the cultivation.

  • Oxidation stability and cold fuid property are considered as the most important factors for determining biodiesel quality. Among the fatty acids, monounsaturated fatty acid satisfy both oxidation stability and cold flow property of biodiesel quality standards. Microalgae with high monounsaturated fatty acid contents is have some benefit for producing to produce biodiesels with satisfying quality standards. In this study, monounsaturated fatty acid contents of a isolated microalga from Youngheung island was analyzed. Phaeodactylum tricornutum was isolated by streaking, and growth rate and fatty acid composition of the algae were investigated. Total FAME contents were consisted of 26% of saturated fatty acids, 43% of monounsaturated fatty acids, and 18% of polyunsaturated fatty acids. The contents of monounsaturated fatty acid were especially high in the Phaeodactylum species. This result implies that the FAMEs from P. tricornutum may contribute to improve the oxidation stability and cold flow property of biodiesel.

Structure Determination of Heishuixiecaoline A from Valeriana fauriei and Its Content from Different Cultivated Regions by HPLC/PDA Analysis

  • A germacrane-type sesquiterpenoid was isolated and purified from a methanol extract of the roots of Valeriana fauriei (RVF) through open column chromatography using silica gel. This compound was verified to be heishuixiecaoline A by spectroscopic analysis. This compound was isolated for the first time from RVF. Quantitative analysis of heishuixiecaoline A from RVF cultivated from three different regions (Eumseong, Jinbu, and Jinan regions) was performed by combining high-performance liquid chromatography with a photodiode array detector. The extract of RVF cultivated in the Jinbu region showed the highest content (9.23 mg/g). In addition, a significant amount of the compound was detected in all RVF samples, which could be expected since it is a characteristic compound of RVF. The sesquiterpenoid group heishuixiecaoline A was isolated from RVF, a resource for various pharmacological substances, and quantitative analysis of RVF cultivated from three different regions was performed. As a result of these experiments, basic data on RVF that can be used in the development and application of pharmaceuticals and health functional foods in the future were obtained.

New Records of Fungi Isolated from Indoor Air of Greenhouse Used for Shiitake Cultivation in Korea (표고 재배사 실내 공기에서 분리한 국내 미기록 진균)

  • Mold contamination is one of the detrimental factors affecting sawdust media-based shiitake cultivation in greenhouses. During mold monitoring of indoor air of greenhouses, several fungi were isolated. Among them, Aspergillus pulverulentus and Cosmospora butyri were found to be new in Korea and Lecanicillium psalliotae and L. antillanum were known in Korea without taxonomic validation. In this study the morphological characteristics and phylogenetic analysis based on the internal transcribed spacer (ITS) rDNA region or ${\beta}$-tubulin gene of the four identified species were described.

Increase of the Treatment Efficiency of a Pharmaceutical Wastewater and a Paperboard Wastewater by the addition of Bacteria (세균첨가에 의한 제약폐수 및 판지폐수의 처리효율의 향상)

  • Some bacterial strains isolated from activated sludges and media and type cultures were cultivated in a pharmaceutical wastewater and a paperboard wastewater and added during batch treatment of those wastewaters in order for these strains to increase the treatment efficiency. Bacillus sp(PC-3) isolated from the charcoal media of the pharmaceutical wastewater plant grew remarkably over there strains in that wastewater and the viable cell count after 24hr cultivation was $1.1{\times}10^6m/L$. Bacillus subtills KCTC 1028 a type strain grew best in the paperboard wastewater and the viable cell count after 24hr cultivation was $1.1{\times}10^7m/L$. Addition of PC-3 in a batch treatment of the pharmaceutical wastewater increased COD removal by 18% after 8 day. And addition of Bacillus subtills KCTC 1028 in a batch treatment of the paperboard wastewater increased COD removal by 14% only after 24hy Bacillus subtills DCTC 1028 was though to be able to be produced economically using alcohol distillery wastewaters from starch material.

  • Fusarium wilts of strawberry, caused by Fusarium oxysporum f. sp. fragariae, is a serious soil-borne disease. Fusarium wilt causes dramatic yield losses in commercial strawberry production and it is a very stubborn disease to control. Reliable chemical control of strawberry Fusarium wilt disease is not yet available. Moreover, other well-known F. oxysporum have different genetic information from F. oxysporum f. sp. fragariae. This analysis investigates the genetic diversity of strawberry Fusairum wilt pathogen. In total, 110 pathogens were isolated from three major strawberry production regions, namely Sukok, Hadong, Sancheong in Gyeongnam province in South Korea. The isolates were confirmed using F. oxysporum f. sp. fragariae species-specific primer sets. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analyses were executed using the internal transcribed spacer, intergenic spacer, translation elongation factor1-${\alpha}$, and ${\beta}$-tubulin genes of the pathogens and four restriction enzymes: AluI, HhaI, HinP1I and HpyCH4V. Regarding results, there were diverse patterns in the three gene regions except for the ${\beta}$-tubulin gene region. Correlation analysis of strawberry cultivation region, cultivation method, variety, and phenotype of isolated pathogen, confirmed that genetic diversity depended on the classification of the cultivated region.

  • Twelve species of fungi were isolated from rice straw media for oyster mushroom cultivation. Trichoderma, Aspergillus and Rhizopus were the predominant fungi. Seven species of Trichoderma were isolated and identified from the rice straw media and the order of their frequency in the media was pseudokonigii, aureoviride, viride, harzianum and koningii. Occurrence of harmful fungi in mushroom houses become more severe as the number of cultivation times increased, and that was more severe in spring culture than in autumn culture. Mycelial growth and sporulation of Trichoderma, Aspergillus and Rhizopus were fovorable on the media appended with extracts of rice straws and oyster mushrooms. This results indicate that the rice straw media and mushrooms give favorable conditions for the occurrence of the fungi in the mushroom houses. Mycelial growth of Trichoderma spp. was favorable on saw­dust extraction media and rice bran extraction media, and the spawns inoculated at the mushroom beds present media of the fungi.

Optimization of Hydrogen Production Using the Mixed Microflora Isolated from Sewage Sludge (하수슬러지로부터 분리된 혐기세균에 의한 수소생산 최적화)

  • Fermentative $H_2$ production was studied using microbial consortia isolated from heat-treated ($90{\circ}C$, 20 min) sewage sludge. Important parameters investigated were carbon(C) and nitrogen(N)-sources, C/N ratio, phosphate concentration, pH and temperature during anaerobic cultivation in serum bottles. Starch, ribose, sucrose and glucose were good C-sources for the culture growth and $H_2$ production. Yeast extract was better N-source than $(NH_4)_2SO_4$ or peptone when individually added to the synthetic media, however the combination of above three N-sources exhibited the additional effect for cell growth and $H_2$ evolution. Addition of 100 mM phosphate as a buffering agent prevented the rapid pH drop during the cultivation. The optimum initial pH for the cell growth was at 7.0, whereas $H_2$ production was observed at pH 5.5. Optimum temperature for the cell growth and $H_2$ production was $37{\circ}C$. Initial C/N ratio of 1.22 in the media using glucose and yeast extract as the C- and N-sources, respectively, showed the $H_2$ yield 1.0 mol $H_2$/mol glucose.
