Immunogenicity of a DNA and Recombinant Protein Vaccine Combining LipL32 and Loa22 for Leptospirosis Using Chitosan as a Delivery System

  • Leptospirosis is a worldwide zoonotic disease caused by pathogenic Leptospira, a genus of which more than 250 serovars have been identified. Commercial bacterin vaccines are limited in that they lack both cross-protection against heterologous serovars and long-term protection. This study investigated in mice the immunogenicity of an anti-leptospirosis vaccine, using the outer membrane proteins LipL32 and Loa22 as antigens. The immunogenicity of this vaccine formulation was compared with those induced by vaccines based on LipL32 or Loa22 alone. A DNA-encapsulated chitosan nanoparticle was used for in vivo DNA delivery. Using a unique DNA plasmid expressing both lipL32 and loa22 for vaccination, higher antibody responses were induced than when combining plasmids harboring each gene separately. Therefore, this formulation was used to test the immunogenicity when administered by a heterologous prime (DNA)-boost (protein) immunization regimen. The specific antibody responses against LipL32 (total IgG and IgG1) and Loa22 (IgG1) were higher in mice receiving two antigens in combination than in those vaccinated with a single antigen alone. Although no significant difference in splenic CD4+ T cell proliferation was observed among all groups of vaccinated mice, splenocytes from mice vaccinated with two antigens exhibited higher interferon-γ and IL-2 production than when using single antigens alone upon in vitro restimulation. Taken together, the immunogenicity induced by LipL32 and Loa22 antigens in a heterologous primeboost immunization regimen using chitosan as a DNA delivery system induces higher immune response, and may be useful for developing a better vaccine for leptospirosis.

Sequence Analysis of Hypothetical Proteins from Helicobacter pylori 26695 to Identify Potential Virulence Factors

  • Helicobacter pylori is a Gram-negative bacteria that is responsible for gastritis in human. Its spiral flagellated body helps in locomotion and colonization in the host environment. It is capable of living in the highly acidic environment of the stomach with the help of acid adaptive genes. The genome of H. pylori 26695 strain contains 1,555 coding genes that encode 1,445 proteins. Out of these, 340 proteins are characterized as hypothetical proteins (HP). This study involves extensive analysis of the HPs using an established pipeline which comprises various bioinformatics tools and databases to find out probable functions of the HPs and identification of virulence factors. After extensive analysis of all the 340 HPs, we found that 104 HPs are showing characteristic similarities with the proteins with known functions. Thus, on the basis of such similarities, we assigned probable functions to 104 HPs with high confidence and precision. All the predicted HPs contain representative members of diverse functional classes of proteins such as enzymes, transporters, binding proteins, regulatory proteins, proteins involved in cellular processes and other proteins with miscellaneous functions. Therefore, we classified 104 HPs into aforementioned functional groups. During the virulence factors analysis of the HPs, we found 11 HPs are showing significant virulence. The identification of virulence proteins with the help their predicted functions may pave the way for drug target estimation and development of effective drug to counter the activity of that protein.

The extension of marine lantern protocol for Dual LED marine lantern control system (이중 LED 등명기 제어 시스템을 위한 확장된 해상 등명기 프로토콜)

  • Dual LED lantern, which is composed of two light sources in one lantern, can be utilized as an auxiliary light source when the main light source is in trouble. Nevertheless, there are no way to obtain status information of the system and control the light source if the current domestic standard protocol is applied, since the standards for dual LED lantern are not defined yet. Regarding to this problem, we suggested expanded lantern protocol for management system of dual LED lantern based on current domestic standard. Also, we confirmed the normal operation of dual LED lantern after implementing suggested protocol on the developed management system. Control system suggested and implemented in this study is tested not only its function of controlling and acquiring the operating status of dual LED lantern, but also its capability to utilize as a control system of currently used LED lanterns.

Effect of diluent variation on cryopreservation of large yellow croaker Larimichthys crocea

  • The aim of this research was to investigate different factors, including cryoprotective agents (CPAs), diluents, dilution ratios, equilibrium times, freezing rates, and thawing methods to optimize cryopreservation protocols for large yellow croaker (Larimichthys crocea). The parameters evaluated were sperm motility, sperm activity index (SAI), survival rate, and DNA damage. Different types of CPAs, such as dimethyl sulfoxide (DMSO), propylene glycol (PG), ethylene glycol (EG), methanol, and glycerol, were tested for sperm preservation. The highest motility, SAI, and survival rate were observed when EG was used. Different diluents such as Stein's solution, Hank's balanced salt solution, marine fish Ringer's solution, artificial seminal plasma (ASP) of small yellow croaker, and Cortland solution were investigated. The highest post-thaw motility was observed upon using ASP as the diluent. Different concentrations of EG were then mixed with ASP to identify the optimal EG concentration. Experimental results showed that the motility (70.33 ± 1.20%), SAI (5), and survival rate (78.30 ± 0.42%) of post-thaw sperm were optimum when 10% EG and ASP were used as the CPA and diluent of cryopreservation, respectively. Post-thaw sperm motility was high at equilibration times below 150 s and at an optimum dilution ratio of 1:1 (sperm: CPA + diluent) and was not significantly different compared with fresh sperm motility. The freezing rate was found to be slow below -10℃/min. The thawing temperature of 45℃ was identified as ideal. The percentage of tail DNA in post-thaw sperm at 10% EG and ASP was also investigated and was found to have more significant DNA damage than that in fresh sperm but significantly lower damage than that in post-thaw sperm at EG concentrations of 5%, 15%, and 20% (p < 0.05). The cryopreservation protocols obtained in this study will be useful in large yellow croaker hatcheries.

NADPH Oxidases Are Required for Appressorium-Mediated Penetration in Colletotrichum scovillei-Pepper Fruit Pathosystem

  • NADPH oxidase (Nox) complexes are known to play essential roles in differentiation and proliferation of many filamentous fungi. However, the functions of Noxs have not been elucidated in Colletotrichum species. Therefore, we set out to characterize the roles of Nox enzymes and their regulators in Colletotrichum scovillei, which causes serious anthracnose disease on pepper fruits in temperate and subtropical and temperate region. In this study, we generated targeted deletion mutants for CsNox1, CsNox2, CsNoxR, and CsNoxD via homologous recombination. All deletion mutants were normal in mycelial growth, conidiation, conidial germination, and appressorium formation, suggesting that CsNox1, CsNox2, CsNoxR, and CsNoxD are not involved in those developmental processes. Notably, conidia of 𝜟Csnox2 and 𝜟Csnoxr, other than 𝜟Csnox1 and 𝜟Csnoxd, failed to cause anthracnose on intact pepper fruits. However, they still caused normal disease on wounded pepper fruits, suggesting that Csnox2 and CsnoxR are essential for penetration-related morphogenesis in C. scovillei. Further observation proved that 𝜟Csnox2 and 𝜟Csnoxr were unable to form penetration peg, while they fully developed appressoria, revealing that defect of anthracnose development by 𝜟Csnox2 and 𝜟Csnoxr resulted from failure in penetration peg formation. Our results suggest that CsNox2 and CsNoxR are critical for appressorium-mediated penetration in C. scovillei-pepper fruit pathosystem, which provides insight into understanding roles of Nox genes in anthracnose disease development.

  • The article aims at studying the institution of human rights in an ever-evolving world in the context of the interdisciplinary approach. The main scientific method was deduction that allowed examining the specific interdisciplinary approach in relation to the institution of human rights on the global scale. To solve the issue set, it is necessary to study legal foundations and features of the interdisciplinary approach to the institution of human rights in the modern world. The article proves there is no theoretical anthropological understanding of the institution of human rights. It has been concluded that the appeal to anthropological jurisprudence requires the identification of the initial theoretical and methodological principles, parameters and axioms of cognition, the integration of a person into the subject field of legal science, linking jurisprudence with the chosen external environment (philosophy, sociology, theology, etc.), predetermining the existence (understanding) of a person, causing qualitative differences and the structure of subject-methodological phenomena. In addition to the identification of such hypotheses, prerequisites and axioms, the basic method (principle) of cognition and its heuristic potential are also being searched (defined). The terminological designation of the formed subject-methodological phenomenon (legal anthropology, anthropology of law, anthropological approach, etc.) reveals its role in the system of interdisciplinary relations of legal science.

  • 일본 우주항공연구개발기구(Japan Aerospace Exploration Agency, JAXA)는 2007년부터 2017년까지 달 탐사선 셀레네(Selenological and Engineering Explorer, SelEnE)가 관측한 데이터를 수집하고, 연구했다. JAXA는 지구 상층 대기에 존재하는 산소가 자기장의 꼬리 부분에 실려 달로 이동한다는 사실을 발견했다. 하지만 이 연구는 아직 진행 중이며 달의 산화 과정 규명에 추가 연구가 필요하다. 본 논문에서는 생성적 적대 신경망(Generative Adversarial Networks, GAN)으로 달 분화구의 영구 그림자 영역을 제거하고, 물과 얼음을 발견하여 선행 연구의 완성도를 향상하고자 한다. 실험에 사용할 모델은 CIPS(Conditionally Independent Pixel Synthesis)다. CIPS는 실제 같은 영상을 고해상도로 합성한다. 합성할 데이터의 최적인 가중치 초기화 및 파라미터 갱신 방법, 활성 함수 조합은 실험을 통해 확인한다. 필요에 따라 앙상블 학습을 할 수도 있다. 성능평가는 FID(Frechet Inception Distance), 정밀도, 재현율을 사용한다. 제안한 방법은 진행 중인 연구의 시간과 비용을 절약하고, 인과관계를 더욱 명확히 밝히는 데 도움 될 수 있다고 사료된다.

A study of the redesign fashion of cultural sustainability - Development of women's clothing using men's Hanbok - (문화적 지속가능성의 리디자인 패션 연구 - 남성 한복을 이용한 여성복 개발 -)

  • The emergence of environmental and social issues has led to global discussions for the realization of sustainable development. The purpose of this study is to propose a method of sustainable fashion design using waste Hanbok, focusing on men's Hanbok with cultural values. The study utilizes literature research and fashion design development. From the literature study, reform was judged to be a higher concept encompassing reduction, recycling, and upcycling. Reduction is a design method through removal, and recycling design transforms from its original form into a completely different product. Upcycling design focuses on improvement and change in functionality. Accordingly, nine redesigns using men's Hanbok were developed from which three were produced. Consequently, the reduction design demonstrated a small range of variation without changing the item, and cultural sustainability was confirmed through the design that removed the components and recombined the Hanbok. Second, recycling redesign can be reconstituted into a different item. Third, upcycling enables various designs through module assembly, which prolongs the lifespan of the product and confirms its value as a raw material for waste Hanbok. This study is meaningful in realizing sustainable fashion and suggesting practical measures for the sustainability of Korean traditional culture and creative fashion design planning.

A Study on Standardization of Measuring Time for Heart Rate Variability (심박변이도(心搏變移度)의 측정시간(測定時間) 표준화(標準化) 연구(硏究))

  • Objectives : We performed this study to search the best time for HRV measuring and which index has the validity in a short term measuring. Methods : At first, three examiner measured electrocardiogram(ECG) of 5 people(4 male + 1 female) for 20 minutes in two times. and we devided ten regions (0-5, 5-10, 10-15, 15-20, 0-10, 0-15, 0-20, 5-15, 10-20, 5-20 minutes). After that we finded the HRV index for each regions. To evaluate the validity, we used Gage Repeatability and Reproducibility Study by MINITAB(ver. 13.20). If the '%Study Var' is under 30 and 'Number of Distinct Categories' is over 4, we could tell it has validity(repeatability and reproducibility). Results : 1) The region and HRV index has repeatability and reproducibility, was HF index in 10-15 minutes. The HF index in 15-20 minutes gained 27.41(%Study Var) and 5(Number of Distinct Categories). 2) The HF index in 10-15 minutes has the better reproducibility than repeatability. The value of measurement is under the control. There is consistency among the examiner. The distribution of measurement is not large. There is no interaction between examiner and each people. 3) The HF index in 10-15, 0-10, 0-15, 0-20, 10-20 minutes was not enough to gain the repeatability and reproducibility. But it had some meanings ; %Study Var, Number of Distinct Categories is 32.68, 4 for 10-15 minutes, 34.43, 4 for 0-10 minutes, 34.44, 4 for 0-15 minutes, 33.56, 4 for 0-20 minutes, 32.07, 4 for 10-20 minutes. Conclusions : This study shows the best index of measuring HRV is the HF index in 15-20 minutes. So, from now on we should measure the HRV for 20 minutes and extract the HF index in 15-20 minutes. It must be the standard for HRV measuring.

Narrative Inquiry about Practical Experience of an Interdisciplinary Undergraduate Class of Design and Engineering - Focusing on an improvement of the class - (학부과정의 다학제 융합 교과 수업경험에 관한 내러티브 탐구 - 수업 개선을 중심으로 -)

  • The study applied a narrative inquiry to the undergraduate interdisciplinary course taught for three years(2012~2014). Narratively I have described interactions among teachers' and students' performing the interdisciplinary consilience course with various situations such as place, curriculum and class interaction. I have specifically illuminated a three-dimensional narrative inquiry space embracing the dilemma of the consilience curriculum, PBL(Problem Based Learning) and TBL(Team Based Learning), and an effective presentation. The result of the study is as follows: First, the ratio of giving lecture is high. The burden of doing project will be reduce if the teacher decreases the lecture up to one fourth, gives students enough time to finish their tasks, and gives them advice for a better project. Second, for opening the interdisciplinary consilience class maintaining previous college curriculum, each department has to put a 'consilience course' on the existing curriculum that allows teachers open the consilience course whenever they need to teach with other department teachers. Third, an effective teaching method using the PBL(Problem Based Learning) in Team Based Learning environment is making students to consider projects as problem situations to solve and to apply the problem situations to PBL method. Lastly, teachers should give students lots of opportunities for presenting what they have learned in the class. Three times of presentation as a team member is ideal in a semester but the way of presenting has to be changed. The students in next year will relieve the tension of 30 minute instead of 15 minute, two days instead of one day for the team presentation.
