The Oogenesis of Chinese minnow, Leuciscinae, Teleostei (경골어류 황어아과 버들치의 난자형성과정)

  • Chinese minnow, Rhynchocypris oxycephalus is a teleost belonging to Leuciscinae, Cyprinidae. The oogenesis and ultrastructure of egg envelope in Chinese minnow were investigated by light and electron microscopes. The ovary was of white yellowish and ellipsoidal shape with the major axis 30 mm and the minor axis 7mm. Cytoplasm of oogonia was basophilic and many nucleoli were located at inside of nuclear membrane. In primary oocytes, yolk vesicles were distributed only in the marginal area and egg envelope was not formed on the outside of an egg. In secondary oocytes, the egg envelope was formed and yolk vesicles in the cytoplasm were increased than the earlier stage. The basophilic substance of cytoplasm was changed to acidic. In case of matured egg, thickness of egg envelope and size of egg were increased. The yolk vesicles were changed to yolk mass in accordance with development. The outer surface of egg envelope was covered by microvilli-structures, and had a micropyle on the area of animal pole. Egg envelope consisted with 2 layers, an adhesive outer layer with microvilli-structures and fibrillar inner layer. In conclusion, the oogenesis of Chinese minnow was characterized by the increase in cell size, the formation and accumulation of yolk, and the decrease of basophilic substance in the cytoplasm. The oogenesis of Chinese minnow seems to share common patterns in Cyprinidae, but these ultrastructural unique characters of egg envelope can be utilized in taxonomy of teleost.

Chemical and Morphological Change and Discoloration of Cedar Wood Stored Indoor (실내 보관 삼나무 목재의 재색 및 화학적·현미경적 변화 특성)

  • The modification of wood color occurs rapidly during the service period at indoor. It is crucial to investigate the characteristics of color change, chemical and microscopical modification of wood at indoor. Wood products made of Japanese cedar at different years were used for this work. The tests were performed in order to evaluate the degree of color change of wood surface, breakpoint of brightness from surface to inside of wood, chemical analysis with FT-IR, and microscopical characteristics using the LM and TEM. Surface color of cedar wood stored indoor were rapidly changed at early stage, particularly ${\Delta}a$ (yellow), and ${\Delta}b$ (red) values were steeply decreased for one year old indoor wood, ${\Delta}L$ (white) value was dropped until 5 years old indoor wood compared with control sample. Decrease of peaks related to polysaccharide and lignin was noticed, especially, lignin was severely degraded. Although degradation of cell wall limited only to surface layers of indoor wood, degradation pattern of indoor wood showed similar degradation pattern to natural weathering of wood during outdoor weathering or wood behavior under artificial UV irradiation.

Integrated RT-PCR Microdevice with an Immunochromatographic Strip for Colorimetric Influenza H1N1 virus detection

  • Recently, Point-of-care (POC) testing microdevices enable to do the patient monitoring, drug screening, pathogen detection in the outside of hospital. Immunochromatographic strip (ICS) is one of the diagnostic technologies which are widely applied to POC detection. Relatively low cost, simplicity to use, easy interpretations of the diagnostic results and high stability under any circumstances are representative advantages of POC diagnosis. It would provide colorimetric results more conveniently, if the genetic analysis microsystem incorporates the ICS as a detector part. In this work, we develop a reverse transcriptase-polymerase chain reaction (RT-PCR) microfluidic device integrated with a ROSGENE strip for colorimetric influenza H1N1 virus detection. The integrated RT-PCR- ROSGENE device is consist of four functional units which are a pneumatic micropump for sample loading, 2 ${\mu}L$ volume RT-PCR chamber for target gene amplification, a resistance temperature detector (RTD) electrode for temperature control, and a ROSGENE strip for target gene detection. The device was fabricated by combining four layers: First wafer is for RTD microfabrication, the second wafer is for PCR chamber at the bottom and micropump channel on the top, the third is the monolithic PDMS, and the fourth is the manifold for micropump operation. The RT-PCR was performed with subtype specific forward and reverse primers which were labeled with Texas-red, serving as a fluorescent hapten. A biotin-dUTP was used to insert biotin moieties in the PCR amplicons, during the RT-PCR. The RT-PCR amplicons were loaded in the sample application area, and they were conjugated with Au NP-labeled hapten-antibody. The test band embedded with streptavidins captures the biotin labeled amplicons and we can see violet colorimetric signals if the target gene was amplified with the control line. The off-chip RT-PCR amplicons of the influenza H1N1 virus were analyzed with a ROSGENE strip in comparison with an agarose gel electrophoresis. The intensities of test line was proportional to the template quantity and the detection sensitivity of the strip was better than that of the agarose gel. The test band of the ROSGENE strip could be observed with only 10 copies of a RNA template by the naked eyes. For the on-chip RT-PCR-ROSGENE experiments, a RT-PCR cocktail was injected into the chamber from the inlet reservoir to the waste outlet by the micro-pump actuation. After filling without bubbles inside the chamber, a RT-PCR thermal cycling was executed for 2 hours with all the microvalves closed to isolate the PCR chamber. After thermal cycling, the RT-PCR product was delivered to the attached ROSGENE strip through the outlet reservoir. After dropping 40 ${\mu}L$ of an eluant buffer at the end of the strip, the violet test line was detected as a H1N1 virus indicator, while the negative experiment only revealed a control line and while the positive experiment a control and a test line was appeared.

A Study on Location and Space Layout of Traditional House of Jeong Dong-Ho - Focused on the Hyungsei-ron of Pungsu(Fengshui) - (예산 정동호가옥의 입지와 공간배치에 대한 연구 - 풍수 형세론을 중심으로 -)

  • Pungsu theory is important one in the site selection and lay-out of traditional Korean village and house. In this aspect, Study with a view of Pungsu theory might be used as a proper method for understanding the traditional architecture in Korea. In this context, this study analyzes the Jeong, Dong-Ho's house which is designated no. 19 as an important folk cultural heritage, located in Jigok Ochu-gil 133-62, Godeok-myon, Yesan-gun, Chungnam province. The analysis of the site and space lay-out is carried out by Yong(Dragon), Hyoel(Auspicious Spot), Sa(Sands), Su(Water) aspects of the Pungsu(Fengshui) Hyungsei-ron. The house is depending upon soft dragon vein connecting with a earth type rear mountain. It looks southeast direction, Gonjwasonhyang(乾坐巽向). The hyeolseong(穴星) has classical venus shape, and there is no faults relatively. The Sasinsa condition is almost perfect that right blue dragon and left white tiger surround the hyeol with 3-4 layers and the facing mountain covers the front open area. The water flowing from inside of left and right mountain is joined in front of the house so it could stop flowing out of vital energy. Bibo forest(裨補樹) is placed properly to protect the easy disclose of water outlet. The house is well organized western house(西四宅) by analysis of Dongseosataek-ron(東西四宅論). Through the analysis, I found that the house has good Pungsu(Fengshui) environment fitted with Pungsu Hyungsei-ron. The house composed of small thatched roof is enclosed several times by low hills of back, left and right side. So it is believed that the site might be carefully evaluated by Yong, Hyoel, Sa, Su of Hyeongsei-ron for knowing whether the site condition could minimize the demage by strong winds and protect from the winter cold wind and secure water for drinking and farming and then selected. The method of Pungsu for evaluating the geographical condition of surrounding of a site is used as a traditional site analysis method for evaluating the suitability of long-term well and safe residence.

Developing the District Unit Plan Simulation using Procedural Modeling (절차적 모델링을 활용한 지구단위계획 시뮬레이션 개발)

  • This research aimed to develop the district unit plan simulation using procedural modeling based on shape grammar. For this, Esri's CityEngine 2020.0 was selected as a main development tool, and Inside Commercial Area in Bangi-dong, Songpa-gu, Seoul as the research site where about 25% of the total area was developed over the past five years. Specifically, the research developed the simulation through the following three phases of Data-Information-Knowledge after selecting necessary parameters. In the Data phase, 2 and 3 dimensional data were obtained by utilizing data sharing platforms. In the next Information phase, the acquired data were generated into various procedural models according to the shape grammar, and the 2D and 3D layers were then integrated using relevant applications. In the final Knowledge phase, three-dimensional spatial analysis and storytelling contents were produced based on the integrated layer. As a result, the research suggests the following three implications for the simulation development. First, data accuracy and improvement of sharing platforms are needed in order to effectively carry out the simulation development. Second, the guidelines for district unit plans could be utilized and developed into shape grammar for procedural modeling. Third, procedural modeling is expected to be used as an alternative tool for communication and information delivery.

Enhanced Cycle Performance of Bi-layer Structured LMO-NCM Positive Electrode at Elevated Temperature (겹층구조의 LMO-NCM 복합양극을 통한 고온 사이클 수명개선 연구)

  • Spinel LiMn2O4 (LMO) and layered LiNi0.5Co0.2Mn0.3O2 (NCM) are widely used as positive electrode materials for lithium-ion batteries. LMO and NCM positive electrode materials have a complementary properties. LMO has low cost and high safety and NCM materials show a relatively high specific capacity and better cycle life even at elevated temperature. Therefore, the LMO and NCM active materials are blended and used as a positive electrode in large-size batteries for electric vehicles (xEV). In this study, the cycle performance of a blended electrode prepared by simply mixing LMO and NCM and a bi-layer electrode in which two electrode layers aree sequentially coated are compared. The bi-layer electrode prepared by composing the same ratio of both active materials has similar capacity and cycle performance to the blend electrode. However, the LN electrode coated with LMO first and then NCM is the best in the full cell cycle performance at elevated temperature, and the NL electrode, in which NCM is first coated with LMO has a faster capacity degradation than the blended electrode because LMO is mainly located on the top of the electrode adjacent to electrolyte and graphite negative electrode. Also, the LSTA (linear sweep thermmametry) analysis results show that the LN bi-layer electrode in which the LMO is located inside the electrode has good thermal stability.

Effect of AlF3 addition to the plasma resistance behavior of YOF coating deposited by plasma-spraying method (플라즈마-스프레이법에 의해 코팅한 옥시불화이트륨(YOF) 증착층의 플라즈마 내식성에 미치는 불화알루미늄(AlF3) 첨가 효과)

  • In order to manufacture a semiconductor circuit, etching, cleaning, and deposition processes are repeated. During these processes, the inside of the processing chamber is exposed to corrosive plasma. Therefore, the coating of the inner wall of the semiconductor equipment with a plasma-resistant material has been attempted to minimize the etching of the coating and particle contaminant generation. In this study, we mixed AlF3 powder with the solid-state reacted yttrium oxyfluoride (YOF) in order to increase plasma-etching resistance of the plasma spray coated YOF layer. Effects of the mixing ratio of AlF3 with YOF powder on crystal structure, microstructure and chemical composition were investigated using XRD and FE-SEM. The plasma-etching ratios of the plasma-spray coated layers were calculated and correlation with AlF3 mixing ratio was analyzed.

Preparation and Characterization of Sodium Caseinate Coated Papers with Bentonite (벤토나이트를 첨가한 카제인나트륨 기반 코팅지 제조 및 특성 연구)

  • This study reports on the preparation of sodium caseinate-cardanol (CasNa/CL)-based papers coated with different amounts of bentonite (BN) for use as a sustainable packaging material. Their chemical and morphological structures, mechanical properties, water vapor permeability, surface properties, and antioxidant activity of coated papers was assessed as a function of the BN content. The drying of the CasNa/CL coated papers led to the formation of pinholes on their surfaces owing to the presence of trapped water resulting from the difference in the drying rate between the external surface and the inside of the coated layers. Increasing the BN content reduced the pinholes on surface of CasNa/CL/BN coated papers and highly decreased the water vapor transmittance rate of the papers from 387.3±1.9 g/m2·day to 269.25±4.5 g/m2·day. Free radical scavenging assays indicated the addition of CL to the CasNa exhibited the antioxidant activity and antioxidant activity of CasNa/CL/BN did not changed as increase of BN contents. The improved water vapor barrier property and antioxidant activity of CasNa/CL/BN coated papers can be promised for various packaging applications.

Prediction of the remaining time and time interval of pebbles in pebble bed HTGRs aided by CNN via DEM datasets

  • Prediction of the time-related traits of pebble flow inside pebble-bed HTGRs is of great significance for reactor operation and design. In this work, an image-driven approach with the aid of a convolutional neural network (CNN) is proposed to predict the remaining time of initially loaded pebbles and the time interval of paired flow images of the pebble bed. Two types of strategies are put forward: one is adding FC layers to the classic classification CNN models and using regression training, and the other is CNN-based deep expectation (DEX) by regarding the time prediction as a deep classification task followed by softmax expected value refinements. The current dataset is obtained from the discrete element method (DEM) simulations. Results show that the CNN-aided models generally make satisfactory predictions on the remaining time with the determination coefficient larger than 0.99. Among these models, the VGG19+DEX performs the best and its CumScore (proportion of test set with prediction error within 0.5s) can reach 0.939. Besides, the remaining time of additional test sets and new cases can also be well predicted, indicating good generalization ability of the model. In the task of predicting the time interval of image pairs, the VGG19+DEX model has also generated satisfactory results. Particularly, the trained model, with promising generalization ability, has demonstrated great potential in accurately and instantaneously predicting the traits of interest, without the need for additional computational intensive DEM simulations. Nevertheless, the issues of data diversity and model optimization need to be improved to achieve the full potential of the CNN-aided prediction tool.

Effects of Dielectric Curing Temperature and T/H Treatment on the Interfacial Adhesion Energies of Ti/PBO for Cu RDL Applications of FOWLP (FOWLP Cu 재배선 적용을 위한 절연층 경화 온도 및 고온/고습 처리가 Ti/PBO 계면접착에너지에 미치는 영향)

  • The effects of dielectric curing temperature and temperature/humidity treatment conditions on the interfacial adhesion energies between Ti diffusion barrier/polybenzoxazole (PBO) dielectric layers were systematically investigated for Cu redistribution layer applications of fan-out wafer level package. The initial interfacial adhesion energies were 16.63, 25.95, 16.58 J/m2 for PBO curing temperatures at 175, 200, and 225 ℃, respectively. X-ray photoelectron spectroscopy analysis showed that there exists a good correlation between the interfacial adhesion energy and the C-O peak area fractions at PBO delaminated surfaces. And the interfacial adhesion energies of samples cured at 200 ℃ decreased to 3.99 J/m2 after 500 h at 85 ℃/85 % relative humidity, possibly due to the weak boundary layer formation inside PBO near Ti/PBO interface.
