  • In the engineering barriers of high-level radioactive waste disposal, gases could be generated through a number of processes. If the gas production rate exceeds the gas diffusion rate, the pressure of the gas increases and gases could migrate through the bentonite buffer. Because people and the environment can be exposed to radioactivity, it is very important to clarify gas migration in terms of long-term integrity of the engineered barrier system. In particular, it is necessary to identify the hydro-mechanical mechanism for the dilation flow, which is a very important gas flow phenomenon only in medium containing large amounts of clay materials such as bentonite buffer, and to develop and validate new numerical approach for the quantitative evaluation of the gas migration phenomenon. Therefore, in this study, we developed a two-phase flow model considering the mechanical damage model in order to simulate the gas migration in the engineered barrier system, and validated with 1D gas flow modelling through saturated bentonite under constant volume boundary conditions. As a result of numerical analysis, the rapid increase in pore water pressure, stress, and gas outflow could be simulated when the dilation flow was occurred.

Introduction to Researches on the Characteristics of Gas Migration Behavior in Bentonite Buffer (벤토나이트 완충재 내 기체 이동의 거동 특성 관련 연구 동향 소개)

  • Gases such as hydrogen and radon can be generated around the canister in high-level radioactive waste disposal systems due to several reasons including the corrosion of metal materials. When the gas generation rate exceeds the gas diffusion rate in the low-permeability bentonite buffer, the gas phase will form and accumulate in the engineered barrier system. If the gas pressure exceeds the gas entry pressure, gas can migrate into the bentonite buffer, resulting in pathway dilation flow and advective flow. Because a sudden occurrence of dilation flow can cause radionuclide leakage out of the engineered barrier of the radioactive waste disposal system, it is necessary to understand the gas migration behavior in the bentonite buffer to quantitatively evaluate the long-term safety of the engineered barrier. Experimental research investigating the characteristics of gas migration in saturated bentonite and research developing numerical models capable of simulating such behaviors are being actively conducted worldwide. In this technical note, previous gas injection experiments and the numerical models proposed to verify such behaviors are introduced, and the future challenges necessary for the investigation of gas migration are summarized.

A pilot study of high flux membrane process for responding to influent turbidity changes in reservoir water (호소수 탁도변화 대응을 위한 고플럭스 막여과공정의 Pilot 연구)

  • In the membrane process, it is important to improve water treatment efficiency to ensure water quality and minimize membrane fouling. In this study, a pilot study of membrane process using reservoir water was conducted for a long time to secure high flux operation technology capable of responding to influent turbidity changes. The raw water and DAF(Dissolved Air Flotation) treated water were used for influent water of membrane to analyze the effect of water quality on the TMP (Trans Membrane Pressure) and to optimize the membrane operation. When the membrane flux were operated at 70 LMH and 80 LMH under stable water quality conditions with an inlet turbidity of 10 NTU or less, the TMP increase rates were 0.28 and 0.24 kPa/d, respectively, with minor difference. When the membrane with high flux of 80 LMH was operated for a long time under inlet turbidity of 10 NTU or more, the TMP increase rate showed the maximum of 43.5 kPa/d. However, when the CEB(Chemically Enhanced Backwash) cycle was changed from 7 to 1 day, it was confirmed that the TMP increase rate was stable to 0.23 kPa/d. As a result of applying pre-treatment process(DAF) on unstability water quality conditions, it was confirmed that the TMP rise rates differed by 0.17 and 0.64 kPa/d according to the optimization of the coagulant injection. When combined with coagulation pretreatment, it was thought that the balance with the membrane process was more important than the emphasis on efficiency of the pretreatment process. It was considered that stable TMP can be maintained by optimizing the cleaning conditions when the stable or unstable water quality even in the high flux operation on membrane process.

  • The treatment of plated wastewater is subject to various and complex processes depending on the pH, heavy metal, and cyanide content of the wastewater. Alkali chlorine treatment using NaOCl is commonly used for cyanide treatment. However, if ammonia and cyanide are present simultaneously, NaOCl is consumed excessively to treat ammonia. To solve this problem, this study investigated 1) the consumption of NaOCl according to ammonia concentration in the alkaline chlorine method and 2) whether ferrate (VI) could selectively treat the cyanide. Experiments using simulated wastewater showed that the higher the ammonia concentration, the lower the cyanide removal rate, and the linear increase in NaOCl consumption according to the ammonia concentration. Removal of cyanide using ferrate (VI) confirmed the removal of cyanide regardless of ammonia concentration. Moreover, the removal rate of ammonia was low, so it was confirmed that the ferrate (VI) selectively eliminated the cyanide. The cyanide removal efficiency of ferrate (VI) was higher with lower pH and showed more than 99% regardless of the ferrate (VI) injection amount. The actual application to plated wastewater showed a high removal ratio of over 99% when the input mole ratio of ferrate (VI) and cyanide was 1:1, consistent with the molarity of the stoichiometry reaction method, which selectively removes cyanide from actual wastewater containing ammonia and other pollutants like the result of simulated wastewater.

Variation in Ploidy Level of Rice Plants Derived from Anther Culture (벼 약배양에서 유기된 식물체의 배수성)

  • Variation in ploidy level of regenerated plants from rice anthers and effective diploidization methods of haploid plants were studied to obtain basic information in rice breeding through anther culture. In a total of 574 plants derived from anther culture using 14F$_1$ hybrids as materials, there were 49.7% haploids, 48.6% diploids and 1.7% polyploids, respectively. The frequency of haploids in Japonica/Indica crosses was 60.6%, and that of Japonica/Japonica crosses was 43.0% in average. Inclusion of 2.4-D or NAA as phytohormone may increase the frequency of haploids, but kinetin may increase the frequency of diploids. The rate of auto-diploidization by tiller separation of haploid plants showed 8.2% in average. The rate of diploidization by leaf-sheath injection of colchicine showed 18.8% in average. Morphological characters of haploids plants showed that 64.6% in culm length, 63.4% in panicle length, 68% in flag leaf length, and 74.4% in flag leaf width compared to diploid plants. These apparent morphological differences will contribute to identify the ploidy of plants derived from rice anther culture.

Improvement of Reproductive Efficiency of Artificial Insemination Following Estrus Induction in Dog II. Semen Freezing and Artificial Insemination in Dog

  • Considerable attention has been focused on the cryopreservation of semen and estrus induction in dog, as consequence of poor productivity caused by long anestrus period, in order to enhance the productivity of youngs and to preserve the breeds. The objectives of this study were to evaluate semen quality after cryopreservation and to evaluate the Pregnancy rate after insemination (AI). Fifty infertilie dogs (age 2∼3 years) were selected for the study and divided into three different estrus induction treatment groups. Group 1: dogs (n=15) were given clomifene (0.1 mg/kg) orally for five days at 12 hr intervals. Group 2: dogs (n=15) were given bromocriptine (50 $\mu\textrm{g}$/kg) orally for five days at 12 hr intervals, followed by single injection intravenously of 500 IU GnRH (Group 3, n=20) when pro-estrus occurred. The rates of pregnancy in estrus inducted dogs mated naturally compared to those inseminated artificially with ejaculated fresh semen and frozen-thawed semen. Estrus detection was performed using the method of vaginal smear and confirmed by the plasma progesterone assay. The ejaculated semen to freeze was exposed to a mixture of Tris extender with cryoprotectant (Trisma, 81 mM: TES, 209 mM: citric acid, 6 mM; glucose, 5 mM; glycerol, 8%) and cryopreserved gradually by slow-cooling at 17 cm above the surface of liquid nitrogen (LN$_2$) for 23 min. The motility of frozen-thawed spermatozoa was assessed by phase-contrast microscopy. To assess their viability and acrosome content, spermatozoa were stained with a vital stain and Fluorescence conjugated lectin Pisum Savitum Agglutinin (FITC/PAS), respectively. Pregnancy was confirmed by ultrasonograpy on day 25, 35 and 55 post insemination. The use of fresh semen, the pregnancy rates were observed 66.6, 66.6, 75.0 and 83.3% in natural estrus, clomifene induced, bromocriptine induced and a combination of GnRH and bromocriptine, respectively. The use of frozen-thawed semen, the pregnancy rates were observed 66.6, 33.3, 50.0 and 60.0% in natural estrus, clomifene induced, bromocriptine induced and a combination of GnRH and bromocriptine, respectively. No difference was observed in the number of offspring produced among natural estrus and treated groups inseminated with fresh or frozen-thawed semen. In conclusion, the pregnancy rate of dogs treated with a combination of GnRH and bromocriptine was more effective than use of clomifene or bromocriptine only. In addition, frozen-thawed semen can be used successfully far artificial insemination in dog.

The Clinical Investigation of Necrotizing Fasciitis in Burn Center (화상센터에서 치료한 괴사성근막염의 임상적 고찰)

  • Purpose: The necrotizing fasciitis is a terrifying infectious disease that can rapidly spreads to surrounding tissues when fascia is infected and it can cause sepsis to death if not properly diagnosed and treated. The purpose of this study is to investigate the characteristics, causes, and treatment methods of necrotizing fasciitis in Korea through reviewing patients admitted to our burn center. Methods: 21 patients with necrotizing fasciitis were selected for this study among those inpatients with electronic medical records (EMR) admitted to Hallym University Hangang Sacred Heart Medical Center from Jan 1, 2008 to June 30, 2019. The medical records and wound photos of those 21 selected subjects were reviewed. Results: There were 13 male and 8 female patients and mean age was 58.76 years old. 13 of 21 subjects were survived and 8 died (38% mortality rate). The surgical treatments performed were I&D, fasciotomy, debridement, allograft, burring, STSG, flap, and amputation. The most common causes were burns in 9 subjects (6 contact burns) and cellulitis occurred on skins in 5 subjects. And other various causes were observed as fournier's gangrene, stab wound, intramuscular injection, tumor and bleu toe syndrome (toe necrosis). The infected areas were 11 feet and legs, 7 hips, 3 abdomen and trunk in 21 subjects. Of the 8 deaths, 3 were infected in feet and legs, 2 were infected in hips, and 2 were infected in abdomen and trunk. As for underlying diseases, 12 patients with hypertension or diabetes were the highest and others such as cancer and stroke were found. Conclusion: The only method to increase the survival rate is to 'suspect' the disease as much as possible and perform early extensive excision. It is advisable to treat the disease by the burn center to properly provide adequate and optimal wound management, infection control, medical care and nutritional supports.

Experiments for Pressure Drop of Scrubbing Layer in a Scrubber System (스크러버 내 충진층에서의 압력강하 특성에 관한 실험적 연구)

  • According to the regulation on the pollution of the marine environment, SOx emission from ships has to be reduced. A SOx scrubbing system installed in a funnel of a ship is considered in order to reduce SOx emission. A scrubbing layer with a porous material is present in the funnel to increase the contact area between exhaust gas and water. In this study, experiments on the pressure drop characteristics in the scrubbing layer are conducted to investigate the effect of the scrubber on the engine load. The pressure drop according to flow rate of air instead of exhaust gas was measured for fillers such as sphere, pall ring and saddle in the scrubbing layer. First of all, porosity is experimentally measured for the three types of filler and it is confirmed that the porosity of the saddle-type filler was the largest. The pressure drop according to the change in air flow rate was measured for the three types of fillers in the scrubbing layer. As a result, the pressure drop was the smallest in the scrubbing layer with the saddle-type filler which has the largest porosity. In addition, the effect of spraying water flowing counter flow against air flow is experimentally examined. It is known that the pressure drop is increased because the air flow space is reduced when water is sprayed. In the case of the saddle, the pressure drop is about 1.5 to 2 times greater than that when only air flows at the optimum exhaust gas-water injection ratio.

Comparative Study of Indocyanine Green Intravenous Injection and the Inflation-Deflation Method for Assessing Resection Margins in Segmentectomy for Lung Cancer: A Single-Center Retrospective Study

  • Background: The inflation-deflation (ID) method has long been the standard for intraoperative margin assessment in segmentectomy. However, with advancements in vision technology, the use of near-infrared mapping with indocyanine green (ICG) has become increasingly common. This study was conducted to compare the perioperative outcomes and resection margins achieved using these methods. Methods: This retrospective study included patients who underwent direct segmentectomy for clinical stage I lung cancer between January 2018 and September 2022. We compared perioperative factors, including bronchial and parenchymal resection margins, according to the margin assessment method and the type of segmentectomy performed. Since the ICG approach was adopted in April 2021, we also examined a recent subgroup of patients treated from then onward. Results: A total of 319 segmentectomies were performed. ID and ICG were utilized for 261 (81.8%) and 58 (18.2%) patients, respectively. Following April 2021, 61 patients (51.3%) were treated with ID, while 58 (48.7%) received ICG. We observed no significant difference in resection margins between ID and ICG for bronchial (2.7 cm vs. 2.3 cm, p=0.07) or parenchymal (2.5 cm vs. 2.3 cm, p=0.46) margins. Additionally, the length of hospitalization and the complication rate were comparable between groups. Analysis of the recent subgroup confirmed these findings, showing no significant differences in resection margins (bronchial: 2.6 cm vs. 2.3 cm, p=0.25; parenchymal: 2.4 cm vs. 2.3 cm, p=0.75), length of hospitalization, or complication rate. Conclusion: The perioperative outcomes and resection margins achieved using ID and ICG were comparable, suggesting that both methods can safely guide segmentectomy procedures.

  • Background : Activation of neutrophil is critical for the clearance of microorganisms and toxic host mediators during sepsis. Unfortunately the activated neutrophil and its toxic byproducts can produce tissue injury and organ dysfunction. The leucocyte CD11/18 adhesion complex regulates neutrophil-endothelial cell adhesion, the first step in neutrophil migration to sites of injection and inflammation. To investigate the potential of neutrophil inhibition as a treatment strategy for sepsis, we evaluated the effects of monoclonal antibody against CD11b (MAb 1B6) in rats intrabronchial challenged with Escherichia coli. Methods : Animals were randomly assigned to receive monoclonal antibody against CD11b (1 mg/kg, sc) and bovine serum albumin(BSA, 1 mg/kg, sc) 6 hr before, at 0 and 6 hr after intrabronchial challenge of $20x10^9$ CFU/kg E. coli 0111. Animals were randomized to treat either 24, 60 or 90% oxygen after bacterial challenge and begining 4 hr after inoculation, all animals were received 100 mg/kg ceftriaxone qd for 3 days. Peripheral and alveolar neutrophil(by bronchoalveolar lavage) counts and lung injury parameters such as alveolar-arte rial $PO_2$ difference, wet to dry lung weight ratio and protein concentration of alveolar fluid were measured in survived rats at 12 hr and 96 hr. Results : Monoclonal antibody against CD11b decreased circulating and alveolar neutrophil especially more in 12 hr than in 96 hr The lung injury parameters of antibody-treated animals were not different from those of BSA-treated animals. but It was meaningless due to small number of survived animals. The early(6 hr) mortality rate was significantly increased in antibody-treated group(51%) compared to BSA-treated group(31%) (P=0.02) but late(from 12 hr to 72 hr) mortality rate was not different in antibody-treated group(44%) from BSA-treated group(36%) (P =0.089). Conclusion : Leucocyte CD11b/18 adhesion molecule is known to regulate neutrophil migration to the site of infection and inflammation. The monoclonal antibody against CD11b decreased alveolar neutrophil in rats with pulmonary sepsis and increased early mortality rate. Therefore, we can speculate that monoclonal antibody against CD11b blocks of alveolar recruitment of neutrophils, impairs host defense mechanism and increases early mortality rate of pulmonary sepsis in rat.
