Preparation and Characteristics of Functional Sauce from Shrimp Byproducts (새우 부산물을 이용한 기능성 소스의 제조)

  • The functional sauce from shrimp byproducts (heads, shells and tails) was prepared and examined for its characterization. The results of volatile basic nitrogen (VBN) suggested that shrimp byproducts were suitable materials for preparing functional sauce. The shrimp hydrolysate, which was incubated with Alcalase for 30 min, showed excellent yield and ACE inhibitory activity. The concentrated sauce from shrimp byproduct was high in crude protein, while low in VBN content and salinity when compared to commercial shrimp sauce. The total amino acid content (23,095.2 mg/100 mL) of concentrated sauce from shrimp byproduct was higher than that (4,582.5 mg/100 smL) of commercial shrimp sauce; also, the major amino acids were glutamic acid, aspartic acid, arginine and lysine. The free amino acid content and taste value of concentrated sauce from shrimp byproduct were 2,705.5 mg/100 mL and 81.0, respectively. The results on the taste value of concentrated sauce from shrimp byproducts suggested that the major taste active compounds among free amino acids were glutamic acid and aspartic acid.

Improvement on the Functional Properties of Gomtang-like Product from Salmon Frame Using Commercial Enzymes (상업적 효소를 이용한 연어 Frame 유래 곰탕 유사 제품의 기능성 개선)

  • This study was conducted to improve functional properties of salmon frame extracts using various commercial enzymes (Alkalase 2.4 L FG, Flavourzyme 500 MG, Neutrase 0.8 L and Protamex 1.5 MG). The ACE (angiotensin I converting enzyme) inhibitory activity was the highest ($IC_{50}=0.67mg/mL$) in the product incubated with Neutrase for 4 hrs (N4-treated hydrolysates) among the various extracts incubated with commercial enzymes for different times. However, antioxidant activities of all salmon frame extracts were less than 15%. There were no significant differences in the proximate composition and sensory evaluation of the fish odor and taste. However, N4-treated hydrolysate was improved in the extractive-nitrogen content and transmission compared to the other enzymatic hydrolysates. When compared to commercial Gomtang products, N4-treated hydrolysate was also high in protein, extractive-nitrogen, total amino acid, and calcium contents, while low in taste sensory score. There were no differences in transmission and sensory score on the fish odor between N4-treated hydrolysates and commercial Gomtang.

Antioxidant and Antihypertensive Activities of Styela plicata according to Harvesting Time and Size (채취시기와 크기에 따른 주름 미더덕의 항산화 및 항고혈압 활성)

  • Styela plicata of large (9.82~11.66 g) and small (0.93~2.21 g) sizes harvested at different times was extracted with 4 different solvents (methanol, ethanol, acetone, and water). DPPH radical scavenging and reducing power was the highest in acetone extracts of small ones (38.98% and 1.724, respectively) harvested in November. The lowest radical scavenging activity was found in water extracts of large ones (12.03% and 0.114) in December. On the other hand, large ones harvested in September showed significantly higher inhibition rate of DNA damage (water, 56.54%; methanol, 55.83%; ethanol, 48.63%) than others. Overall, the water extraction of S. plicata tended to show a higher antigenotoxic effect. In addition, water extracts of large S. plicata from November showed the highest angiotensin converting enzyme (ACE) inhibitory activity (64.38%), while that of small ones from December exhibited the lowest (51.33%). Overall the results indicate that antioxidant and antihypertensive activities S. plicata are variable depending on harvesting time, size, and extraction solvent.

Functionalities of Squid Liver Hydrolysates (오징어 간 가수분해물의 기능성)

  • The autolysate and hydrolysate of a common squid liver, Todarodes pacificus, were prepared. Autolysis (liver ratio, pH, temperature) and Protamex-treated hydrolysis (pH, temperature, ratio of protease to liver) conditions were optimized by response surface methodology using central composite design for under 1 hr of hydrolysis time. The desirability profile indicated that maximum DH could be achieved at a squid liver of 93.5%, pH 6.4, and $47^{\circ}C$ in autolysis, while that of Protamex-treated hydrolysis did at a Protamex-to-squid liver level of 0.33%, pH 6.0, and $55^{\circ}C$. Three amino acids, proline, cysteine, and methionine, were not detected in the total amino acid composition of the Protamex-treated hydrolysate, while they were detected in the free amino acid composition. Cadmium was $8.32{\pm}0.03$ mg/100 g-powder for raw, $3.56{\pm}0.02$ mg/100 g-powder for the autolysate, and $13.26{\pm}0.04$ mg/100 g powder for the Protamex-treated hydrolysate. The major molecular weight ranged from 1.0 to 1.5 kDa for the autolysate and from 210 to 470 Da for the Protamex-treated hydrolysate. Food functionalities of the autolysate, such as surface hydrolphobicity, emulsion activity index, emulsion stability, water, and fat adsorption, were similar to the Protamex-treated hydrolysate. Both the autolysate and Protamex-hydrolysate showed high inhibitory activities on the angiotensin-I converting enzyme. Cell toxicity against the HepG2 cell line was not detected in the autolysate or the Protamex-treated hydrolysate by 200 ${\mu}g/mL$.

Physiological activity of Pholiota nameko sp. ethanol extract (맛버섯 에탄올 추출물의 생리활성)

  • This study was conducted to investigate the physiological activities of the ethanol extracts from 10 different strains of Pholiota nameko. In addition, the ${\beta}$-glucan and polyphenol contents from these strains were also measured. Total polyphenol contents from all the strains were more than 40 mg% with the highest content of $61.50{\pm}0.59$ mg% and ${\beta}$-glucan contents were 30% with the highest content of $37.20{\pm}1.12%$. The highest inhibitory activities on ${\alpha}$-amyloglucosidase activity and nitric oxide production were $13.78{\pm}0.56%$ and $56.59{\pm}7.11%$, respectively. However, ethanol extracts from all the strains have little effects on DPPH radical and nitrite scavenging rates and the activity of angiotensin I converting enzyme. The cytotoxic activity of ethanol extracts from all the strains on A549 cell was shown upto 30% with the highest effect of $47.96{\pm}8.46%$ treated at 1 mg/mL concentration.

  • Effect of electron beam irradiation on various physiological property changes of the ethanol extract from the cooking drips of Enteroctopus dofleini was investigated. The Hunter's color values of extract were increased with increasing irradiation doses. The radical scavenging activity of the extract was increased depending on the irradiation dose. The inhibitory activities against tyrosinase and angiotensin I converting enzyme were also increased with irradiation dose. It was also shown that the contents of proteins and polyphenols were increased by the irradiation. It was reasoned that the antioxidant activities were increased by proteins modified by the irradiation.

Physiological Properties of Sarcodon aspratus Extracts by Ethanol Concentration (능이버섯 추출물의 생리활성 효과)

  • Physiological properties of Sarcodon aspratus extracts were investigated. Yields of water, 30, 60, and 90% ethanolic extracts were 52.10, 46.90, 41.50, and 32.20%, respectively. The 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity of 60% ethanolic extract was 40.40% at a concentration of 4 mg/mL, which was highest of all the extracts. The polyphenol content of 60% ethanolic extract was the highest at 16.23 mg/g at a concentration of 1 mg/mL. Superoxide anion radical scavenging activities were 56.30~77.05% at concentrations of 1, 2, and 4 mg/mL, which were better than the ascorbic acid activity (56.19% at 10 mg/mL) (P<0.05). Nitrite-scavenging ability was the most effective in 30% ethanolic extract. A pH level of 3.0 was the most effective for all extracts. Angiotensin-converting enzyme inhibitory activities of Sarcodon aspratus extracts were from 52.67 to 89.52%.

Physicochemical Properties, Lactic Acid Bacteria Content and Physiological Functionalities of Korean Commercial Makgeolli (시판 막걸리의 이화학적 특성과 젖산균 함량 및 생리기능성)

  • The goal of this study was to characterize the physicochemical properties, lactic acid bacteria and physiological functionality of Korean commercial Makgeolli. We collected 60 kinds of raw and pasteurized Makgeolli commercially available in Korea. Their physicochemical properties, lactic acid bacteria content and physiological functionalities were determined. In the commercial raw Makgeolli, ethanol and residual sugar content ranged from 3.6% to 9.6% and 0.20% to 4.52%, respectively. Furthermore, the raw Makgeolli contained 3.50 to 101.9 mg% of amino nitrogen. In the pasteurized Makgeolli, ethanol content ranged from 4.0% to 7.0% and their residual sugar content were 0.98% to 7.57%. The raw Makgeolli contained 2.0 $ent^3$~1.2 $ine^8$ CFU/mL of lactic acid bacteria. Among several functionalities, the antihypertensive angiotensin I-converting enzyme inhibitory activities of EDS-14 raw Makgeolli and PWR-12 pasteurized Makgeolli were very high, at 89.9% and 87.0% respectively. The other functionalities were seen to be below 30% or not detectable. The ${\beta}$-Glucan contents of the raw and pasteurized Makgeolli were noted as average, at 14.1% and 14.6%, respectively.

Physiological Activities of Mulberry Leaf and Fruit Extracts with Different Extraction Conditions (용매에 따른 뽕잎과 오디의 생리활성 효과)

  • Physiological activities in mulberry leaf and fruit were examined. Electron-donating ability (EDA), tyrosinase activity, SOD-like action (SOD), angiotensin I-converting enzyme-(ACE) inhibitory activity,and nitrite-scavenging ability of mulberry leaf and fruitextracted with water, with 50% (v/v) or with 100% ethanol, were measured. The EDA of mulberry leaf and fruit extracted with water or 50% (v/v) ethanol were greater (by 65.72-81.30%) than that with the 100% ethanol extract, whereas the activities of both former extracts were lower than those with 1.0% and 0.1% (both w/v) L-ascorbate solutions. The SOD-like activities of water, 50% (v/v) and 100% ethanol extracts of all samples were 24.13.26.80% lower than those of 1.0% and 0.1% (both w/v) L-ascorbate solutions. Nitrite-scavenging activity at pH 1.2 was observed in all extracts. The results further our understanding of the physiological activities of mulberry leaf and fruit extracts.

Antioxidative and Antimicrobial Activities of Persimmon Leaf Tea and Green Tea (감잎차와 녹차의 항산화 및 항균 활성 비교)

  • In this study, the antioxidative and antimicrobial activities of persimmon leaf tea and green tea were assessed. The total polyphenol contents of green tea (0.5875 mg/mL) were slightly higher than those of persimmon leaf tea (0.3938 mg/mL). The electron-donating abilities (EDA) of persimmon leaf tea and green tea toward $\alpha$, $\alpha$-diphenyl-$\beta$-picyryl hydrazyl (DPPH) radical were $65.7{\pm}3.7%$ and $68.4{\pm}0.3%$, respectively, as compared to $68.6{\pm}1.1%$ for L-ascorbic acid (1%). The SOD-like activity of persimmon leaf tea ($29.7{\pm}0.9%$) was higher than that of green tea ($28.1{\pm}1.0%$). The nitrite scavenging ability was pH-dependent, highest at pH 1.2, and lowest at pH 6.0, in and persimmon leaf tea was found to exhibit more effective nitrite scavenging ability than green tea. The inhibitory effects of persimmon leaf tea and green tea against angiotensin I converting enzyme were $56.7{\pm}1.9%$ and $67.1{\pm}1.7%$, respectively. Persimmon leaf tea evidenced profound antimicrobial activities against Listeria monocytogenes, Staphylococcus aureus, Escherichia coil, and Salmonella typhimurium. These results clearly established the antioxidative and antimicrobial potency of persimmon leaf tea.
