Isolation and Identification of Antimicrobial Compounds against Helicobacter pylori from Rosemary (Rosmarinus officinalis L.) Extracts (로즈마리(Rosmarinus officinalis L.) 추출물로부터 Helicobacter pylori에 대한 항균물질 분리 및 동정)

  • Rosmarinus officinalis extracts had a significant antimicrobial activity against Helicobacter pyori. Total phenolic contents and inhibition zone of rosemary extracts were estimated to be 25.7 mg/g and 14 mm at $200{\mu}g/mL$ of phenolic contents, respectively. The Sephadex LH-20 and MCI-gel CHP-20 column chromatographic separations for the phenolic extracts from R. officinalis leaves led to isolation of five acids, whose structures were determined as protocatechuic acid (A), coumaric acid (B), caffeic acid (C), chlorogenic acid (D), and rosmarinic acid (E), from interpretation of spectroscopic data including nagative fast atom bombardment (FAB)-mass, $^1H$-NMR, $^{13}C$-NMR, and IR. All isolated compounds were tested for antimicrobial activity against H. pyori. The purified single compound showed less antimicrobial activity against H. pylori than the mixed purified compounds, which generate A+B, A+E, C+D, C+E (each $200{\mu}g/disc$) excellent as large clear zone by synergy effect. These results indicate rosemary extracts are preventive agents against H. pyori.

  • We screened the methanol extracts from 133 plant species growing in Korea for antimicrobial activity against enterohemorrhagic Escherichia coli 0157 : H7. Those are selected from three plant grouping ; traditional medicinal herbs, edible plants, and flowers. They were tested by disk diffusion assay. From evaluation of the inhibition zone diameter of microbial growth, we found that the flower extract of Rhododendron Schilpenbachii Max had the most significant antimicrobial activity against this bacteria. Extracts from most of the vegetables and plants did not show antimicrobial activity except for the leaves of Ginkgo biloba L. and seeds of Prunus Dallicina L. did not show antimicrobial activity except for the leaves of Ginkgo biloba L. and the seeds of Prunus sallicina L.

Inhibitory effect of sakuranetin on (1,3)-β-glucan synthase

  • An examination of the kinetic properties of UDP-glucose, (1,3)-$\beta$-glucans (callose) synthase, from mung bean seedlings (Sorbus commixta cortex) shows that these enzymes have a complex relationship with UDP-glucose and various effectors. Fluorescence assay showed that deoxynojirimycin increased the inhibitory effect of (1,3)-$\beta$-glucan synthase in a concentration-dependent manner. The inhibitory effect of sakuranetin (34.34%) was higher than that of deoxynojirimycin (80.63%). Disk diffusion method revealed that sakuranetin inhibited the growth of Candida albicans to a 1.5 mm inhibition zone. These results suggest that sakuranetin, isolated from Sorbus commixta cortex extract, can be used as stable antifungal material.

  • This study was conducted to develop a new biomaterial to be used for skin whitening. The melanin elimination effect of chondroitin sulfate and phelinus linteus mushroom in rabbit back skin were evaluated. Rabbit dorsum was exposed to chronic UV irradiation(320nm) once daily for 30 days after initial melanin injection (100mg/kg). And then, chondroitin sulfate and phelinus linteus mushroom at dose of 0.7g for 30days were applied on the zone. The dorsal skin was histologically examined. Furthermore, we investigated free-radical extinction effect, antioxidation and tyrosinase activity inhibition effects. The histological study indicated that chondroitin sulfate and phelinus linteus mushroom decreasd melanine pigment significantly. As a result, chondroitin sulfate and phelinus linteus mushroom have a remarkable effect on the skin whitening by melanin elimination.

  • The purpose of this study was to evaluate the in vitro antifungal effect of several kinds of denture lining materials containing nystatin and silver-zeolite on Candida albicans. Three commercially available tissue conditioners (Soft-Liner, Coe-Comfort, Coe-Soft) and two direct denture relining materials (Tokuso Rebase, Durabase) were selected. In terms of the zone of inhibition and some basic physical properties, I could find the following results ; 1. Nystatin or silver-zeolite included in those relining materials had definite antifungal activities against the Candida albicans. 2. As times went on, both of the antifungal agents's activities decreased gradually. 3. Antifungal agents did not affect the relining materials' basic physical properties. 4 Direct chairside relining materials showed unfavorable dark discoloration with response to silver-zeolite.

The effect of Hot Water-Extract and Flavor Compounds of Mugwort on Microbial Growth (쑥의 열추출물과 주요 향기성분이 세균의 생육에 미치는 영향)

  • Hot water extract from mugwort (Artemisia asiatica nakai) leaves and tea inhibited the growth of Bacillus subtilis by adding to the nutrient broth 1.0% and 0.5% concentration, respectively. Among the important compounds that contributing mugwort like flavor to the mugwort leaves and tea were considered, authenic compound of thujone, caryophyllene and farnesol showed bactericidal effect for Escherichia coli, Enterobacter aerogenes, Vibrio paraheaemolyticus, Psudomonas aeruginosa, Bacillus subtilis and Staphylococcus aureus when teste by paper disc method ( 8 mm diameter). The mixture of caryophyllene and farnesol was more bactericidal effect for various bacteria than the mixture of thujone, caryophyllene and farnesol was more bactericidal effect for various bacteria that the mixture of thujone, caryophyllene and farnesol or each compounds . Especially, the mixture of caryophyllene and farnesol showed strong bactericidal effect (diameter of inhibition zone>40mm) for Vibrio parahaemolyticus , Enterobacter aerogenes and BAcillus subtilis.

  • 4-Geranyloxy compound (1) has elucidated by spectroscopic analysis. This compound (1) inhibited the growth of the dermatophytic fungus Trichophyton mentagrophytes ATCC 28185, (2 mm inhibition zone at 15 ㎍/disc), cytotoxic to P388 murine leukaemia eel/lines ATCC CCL 46 P388D1, (IC/sub 50/ 1,125 ng/ml at 7.5㎍/disc) and BSC monkey kidney cell lines (100% of well at 15 ㎍/disc).

  • Strains showing antifungal activity against Cryphonectria parasitica were isolated from coast soil of Taean , Korea. Of 152 strains isolated, 6 strains showed antifungal activity in vitro against C. parasitica. Ta24 strain showed highest activity with 1.6 cm clean inhibition zone. For strain identification, the morphological characteristic and 16S rDNA sequences were determined. Ta24 strain showed 99% homology with Streptomyces sampsonii and was identified as Streptomyces sp.

  • During the screening of inhibitors of melanin biosynthesis from microbial secondary metabolites, a fungal strain MR-93 which was capable of producing high level of an inhibitor was selected from plant leaf. Based on taxonomic studies, the fungus could be classified as a strain of Trichoderma sp.. The active compound (MR-93D) was purified from the culture broth by Diaion HP-20 column chromatography, ethylacetate extraction, Sephadex LH-20 column chromatography and HPLC. The inhibitor was identified as 4-hydroxy-8-isocyano-l-oxaspiro[4-4]cyclonon-8-en-2- one by spectroscopic methods of UV, $^{1}$H-NMR, ESIMS and IR. MR-93D showed a strong tyrosinase inhibitory activity with 0.03 $\mu$g/m of IC$_{50}$ value. It also inhibited melanin biosynthesis with 35 mm inhibition zone at 30 $\mu$g/paper disc in Streptomyces bikiniensis, a bacterium used as an indicator organism in this work.

  • Levosulpiride is the levo-enantiomer from of racemic sulpride. abenzamide derivative selectively inhibition doparninergic D2 receptos at the trigger zone both in the central nervous system and in the gastrointestinal tract. We report a rapid and sensitive HPLC method using reverse phase C 18 column with fluorescence detection for separation and quantitation of levosulpiride in plasma. Tiapride was used as an internal standard. After adding an internal standard. levosulpiride in 800 ${\mu}l$ of plasma was extracted under basic conditions with ethyl acetate and methylene chloride.
