Recent Advances in Anti-inflammatory Flavonoid Research since 2004

  • Certain flavonoids possess anti-inflammatory activity. Besides their antioxidative property, the cellular action mechanisms of flavonoids include an inhibition of arachidonate metabolizing enzymes such as cyclooxygenases and lipoxygenases, and a down-regulation of proinflammatory gene expression such as cyclooxygenase-2, inducible nitric oxide synthase and tumor necrosis factor-$\alpha$. In this review, the recent findings of anti-inflammatory flavonoid research since 2004 were summarized. And the cellular mechanisms on signal transduction pathways were also discussed.

토복령(土茯笭)의 Raw 264.7 세포에 대한 항염효과 (Anti-inflammatory Effects of Smilacis Glabrae Rhizoma in Raw 264.7 Cells)

  • Objective : Inflammatory cytokines have a close relationship to insulin dependent diabetes mellitus (IDDM). The inhibitory effect of Smilacis Glabrae Rhizoma (SGR) were examined on production of nitric oxide (NO), prostaglandin $E_2$ $(PGE_2)$, synthase (iNOS), cyclooxygenase-2 (COX-2), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and NF-${\kappa}$B activation in Raw 264.7 cells. Methods: Raw 264.7 cells were pretreated with SGR(20, 50, 100 ${\mu}g$/ml), and then cultured with lipopolysaccharides (LPS). Cell viability was measured by MTT assay; inhibition of NO, $PGE_2$, and TNF-${\alpha}$ production were measured by Griess reagent and enzyme-linked immunosorbent assay(ELISA). Induction of COX-2 and iNOS were determined by western blotting analysis. Inhibition of NF-${\kappa}$B was measured by immunofluorescence assay (IFA). Results: SGR inactivated NF-${\kappa}$B, and inhibited the production of NO, iNOS, and $PGE_2$. Inhibition of COX-2 and TNF-${\alpha}$ could not be confirmed. Conclusions: From the above result. SGR was found to have an anti-inflammatory effect of inhibition of NO, iNOS, and $PGE_2$ production via inhibition of NF-${\kappa}$B.

동맥경화 예방과 치료를 위한 연구시도: Nitric Oxide의 역활 -광 유도 nitric oxide(PIANO)의 혈관이완에 따른 cyclic GMP의 증가 (Possible Role of Nitric Oxide in Prevention of Atherosclerosis: Photo-induced adequate nitric oxide (PIANO)-mediated relaxation involves cyclic GMP increment)

  • 본 연구의 목적은 광 유도에 의한 nitric oxide (PIANO)유리가 혈관이완에 대해 cyclic GMP (cGMP)가 관여하는 지의 여부와 아울러 ${\alpha}$-수용체를 통한 수축에 PIANO가 어떻게 작용하는 지를 파악하고자 하였다. In vitro 실험에서 흰쥐의 대동맥을 준 최고농도의 phenylephrine (PE)으로 수축시킨 후 nitric oxide 생성을 변화시키는 약물이나 광민감성 (photosensitizing) 약물에 대한 반응을 등장력 변화로 기록하였다. PIANO에 의한 혈관이완은 광노출 강도와 기간 및 광민감성 약물농도에 비례하여 증가하였고 cGMP의 증가를 수반하였다. PE에 의해 증대되는 phosphatidylinositide(PI) 전환은 PIANO에 의해 억제되었다. 이상의 결과는 cGMP의 증가로 인해 PIANO에 의한 혈관이완이 일어나며 ${\alpha}$-아드레날성 수용체 자극에 의한 PI 전환의 억제현상은 cGMP 증가의 결과로 생각할 수 있다. 결론적으로 PIANO에 의한 혈관이완은 cGMP의 증가로 인함을 확인할 수 있었다.

혈액순환과 관련하여 nitric oxide 생성과 angiotensin converting enzyme 활성에 미치는 Acai berry 에탄올 추출물의 영향 (Effects of Acai Berry Ethanolic Extracts on Production of Nitric Oxide and Activity of Angiotensin Converting Enzyme Related to Blood Circulation)

  • 본 연구의 목적은 혈관계 순환을 촉진시키는 약효제를 발굴하는 것이다. 아사이베리(Acai berry) 에탄올 추출물(ABEE)의 항산화 효과 뿐만 아니라 토끼의 폐에서 유래한 angiotensin converting enzyme (ACE)의 활성 및 대식세포와 혈관내피세포에서 nitric oxide 생성에 대한 효과가 본 연구에서 조사되었다. 먼저 환원력과 지질과산화에 대한 ABEE의 항산화효과가 관찰되었다. 뿐만 아니라, ABEE는 hydroxyl radical에 의하여 유발된 DNA 산화에 대한 보호효과를 보여주었다. 더욱이 ABEE는 0.01%의 농도에서 약 50% 정도의 angiotensin converting enzyme 활성에 대한 억제효과를 발휘하는 것으로 나타났다. 혈관내피세포에서 ABEE는 nitric oxide 의 생성을 증가시켰으나 대식세포에서는 lipopolysaccharide에 의하여 생성된 nitric oxide 생성을 감소시켰다. 게다가 superoxide dismutase (SOD)-2와 -3 발현수준은 ABEE 처리에 의하여 증가되었으나 SOD-1의 발현수준은 일정하였다. 더욱이 nitric oxide synthases-1 (NOS-1)의 발현수준은 ABEE 처리에 의하여 증가되었으나, 유발 효소인 NOS-2의 발현수준은 일정하였다. 또한 SOD의 전사인자인 Nrf-2의 발현수준은 ABEE에 의하여 증가되었다. 그러므로 이러한 결과들은 ABEE가 이상의 작용을 경유하여 혈액순환을 촉진시킬 수 있고, 혈관의 건강을 위한 큰 잠재성을 가지고 있다는 것을 암시하고 있다.

  • Silymarin, a polyphenolic flavonoid antiodant, has been shown to have anti-inflammatory, hepatoprotective, and anticarcinogenic effects. In the present study, we report the inhibitory effect of silymarin on nitric oxide (NO) production and inducible nitric oxide synthase (iNOS) mRNA expression in macrophages. In vivo administration of silymarin attenuated NO production of peritoneal macrophages in lipopolysaccharide (LPS)-treated mice.(omitted)

  • Effects of nitric oxide and hypoxia on ethoxyresorufin deethylase in Hepa I cells and MCF-7 human breast cancer cells were examined. TCDD treatment have resulted in the stimulation of ethoxyresorufin deethylase activity based on fluorometry in Hepa I in dose and time dependent manner. 0.1 nM TCDD showed maximal stimulation of ethoxyresorufin deethylase activity and 24 hour treatment also showed maximal stimulation of ethoxyresorufin deethylase activity. In MCF-7 human breast cancer cells, untreated cells showed high basal level of ethoxyresorufin deethylase activity. TCDD treatment to MCF-7 cells resulted minor stimulation of ethoxyresorufin deethylase activity compared to that in Hepa I cells. Nitric oxide and hypoxia inhibit TCDD effects on ethoxyresorufin deethylase activity in both cell lines. And also flavonoids, such as quercetin showed an inhibition of ethoxyresorufin deethylase activity that is stimulated with TCDD or 3-Methylcholanthrene. Estrogen and estrogen metabolite such as 16 a-estriol and 2-hydroxyestradiol also affects the ethoxyresorufin deethylase activity in MCF-7 cells.

Inhibition of Nitric Oxide Production, iNOS and COX-2 Expression of Ergosterol Derivatives from Phellinus pini

  • Ergosta-4,6,8(14),22-tetraen-3-one (1), ergosta-7,24(28)-dien-3-ol (2), and 5,8-epidioxyergosta-6,22-dien-3-ol(3) were isolated from the fruit body of Phellinus pini. Their structures were based on spectroscopic methods including IR, MS, and NMR (1D and 2D). These compounds were screened for their ability to inhibit nitric oxide (NO) production in LPS-activated RAW 264.7 cells. Compounds 1, 2, and 3 reduced NO production in the assay with $IC_50$ values of 29.7 ${\mu}M$ (1), 15.1 ${\mu}M$ (2), and 18.4 ${\mu}M$ (3) respectively. They also suppressed the expression of protein and m-RNA of iNOS and COX-2 in a dose dependent manner by western blot analysis and RT-PCR experiment in LPS-activated microglial cells.

JNK/SAPK Is Required in Nitric Oxide-Induced Apoptosis in Osteoblasts

  • Nitric oxide(NO) induces apoptosis in human osteoblasts. Treatment with exogenous NO donors, SNAP (S-Nitroso-N-acelylpenicillamine) and SNP (sodium nitroprusside), to MG-63 osteoblasts resulted in apoptotic morphological changes, as shown by a bright blue-fluorescent condensed nuclei and chromatin fragmentation by fluorescence microscope of Hoechst 33258-staining. The activities of caspase-9 and the subsequent caspase-3-like cysteine proteases were increased during NO-induced cell death. Pretreatment with Z-VAD-FMK (a pancaspase inhibitor) or Ac-DEVD-CHO (a specific caspase-3 inhibitor) abrogated the NO-induced cell death. The NO donor markedly activated JNK, a stress-activated protein kinase in the human osteoblasts. This study showed that the inhibition of the JNK pathway markedly reduced NO-induced cell death. But neither PD98059 (MEK inhibitor) nor SB203580 (p38 MAPK inhibitor) had any effect on NO-induced death. Taken together, these results suggest that JNK/SAPK may be related to NO-induced apoptosis in MG-63 human osteoblasts.

Anti-inflammatory Activities of Chopi (Zanthoxylum piperitum A.P. DC) Essential Oil: Suppression of the Inducible Nitric Oxide Synthase and Cellular Adhesion

  • The aim of this study is to elucidate the anti-inflammatory activities of chopi (Zanthoxylum piperitum A.P. DC.) essential oil. Essential oil (EO) of chopi was extracted by steam distillation method, and its major constituents were limonene and geranyl acetate. Chopi-EO decreased approximately 38% of nitrite production, as compared to the lipopolysaccharde (LPS)-induced nitrite production. However, chopi-EO and its components did not quench nitric oxide (NO) chemically in cellfree system, and markedly inhibited approximately 40.4% of inducible nitric oxide synthase (iNOS) mRNA transcription. In addition, the inhibition of E-selectin gene transcription by chopi-EO caused the suppression of cellular adhesion. These results suggest that chopi-EO may exert potential anti-immunological inflammatory activity.

Antioxidant and Anti-inflammatory Activities of Equisetum hyemale

  • Equisetum hyemale L. has been prescribed widely as a traditional medicine for the treatment of inflammatory diseases such as rheumatoid arthritis, conjunctivitis, pyelonephritis. In order to identification the mechanism, we examined an antioxidant and anti-inflammatory activity of 85% methanol extract of E. hyemale. In this study E. hyemale exhibited strong scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical, superoxide radical, and nitric oxide. To elucidate the anti-inflammatory properties of E. hyemale, we investigated the inhibition effects of nitric oxide and IL-6 by E. hyemale in IFN-gamma and LPS-stimulated mouse peritoneal macrophages. E. hyemale suppressed nitric oxide, IL-6 production and iNOS expression dose-dependently without notable cytotoxic activity. These data suggest that E. hyemale might be useful in inflammatory diseases by inhibiting the free radicals and inflammatory mediators.
