In-situ Stabilization of Heavy Metal Contaminated Farmland Soils Near Abandoned Mine, using Various Stabilizing Agents: Column Test Study (폐광산 주변 중금속 오염 농경지 토양복원을 위한 다양한 첨가제의 안정화 효율 비교: 컬럼시험연구)

  • This study concerned remediation of heavy metal contaminated farmland soils near abandoned mine, using stabilization method, with particular emphasis on the remediating the soils contaminated with multi-elements. In this study, stabilizing heavy metals based on 'In-situ chemical fixation' has been applied to the soil collected from an abandoned mine in Korea, using column test, with various stabilizing agents, including $FeSO_4$, $KMnO_4$, sludge (collected from coal mine drainage treatment pond), zero-valent iron (ZVI), zeolite and $CaCO_3$. Sixty five-days operation of the flow-through columns yield $FeSO_4\;+\;KMnO_4$ and zeolite are efficient on reducing As leaching from the soil. ZVI and sludge are reducing the leaching of Cu. Although $FeSO_4\;+\;KMnO_4$ seem to be efficient for most heavy metals, high pH in the initial stage of test enabled high leaching of the heavy metals, whereas fixation of the heavy metals maintain throughout the rest of the test period, with increasing pH up to around 6. Addition of some alkaline agent may inhibit the low pH during the application. The column test was also run as two set: one set incubated with deionized water for 72 hours prior to starting the test, and the other without incubation. The incubated set demonstrated better stabilizing efficiency, indicating the potential optimized operation method.

Identification of Infectious Fungus in the Artificial Cultivation Rooms of Paecilomyces tenuipes (눈꽃동충하초 재배상 내에 발생된 오염균의 동정)

  • A infectious fungus (J131) recently isolated from artificial cultivation farms of Paecilomyces tenuipes is known to cause a great yield loss and deterioration of products. In this study, its occurrence and characterization were made. The isolate (J131) was identified as Paecilomyces farinosus (Holm ex S. F. Gray) Brown & Smith. Colonies of the isolate on PDA grew moderately fast, attaining a diameter of 19mm within 14 days at 24${\pm}$1$^{\circ}C$. Vegetative hyphae are smooth-walled and hyaline. Conidiophores mainly arose from the submerged mycelium, usually measuring 1.3-1.5$\mu\textrm{m}$ wide and consisted of verticillate branches with whorls of 2 to 4 phialides. Phialides of 3.8-5.8${\times}$1.5-1.8 $\mu\textrm{m}$, with a swollen basal portion tapered into a distinct neck of about 0.6-0.8 $\mu\textrm{m}$ wide. Conidia arisen from the phialides, were ellipsoidal to fusiform, smooth-walled, hyaline, 1.8-2.4${\times}$1.1-1.4$\mu\textrm{m}$. .When silkworm larvae were injected with concentration of 1${\times}$10$\^$8/ conidia/$m\ell$ of infectious fungus J131, infection rate was 41.3% and synnemata formation rate was 23.6%. Black spots were appeared on the surface of silkworm larvae in 5-9 days after injection. The surface of larvae was covered with hyphae which were gradually turned into yellow from white color during the incubation period of 3-4 days. 1 to several 4-6mm long synnemata of yellow or orange color were formed on the surface. The matured synnemata were ramified from its end and produced much conidia of white color.

  • The purpose of this study was to investigate the knowledge, attitude, infection management intention and educational needs among nurses with no experience of new respiratory infectious diseases (NRID: SARS and MERS). Data were collected from 162 nurses working at the general hospitals in B city using structured questionnaires from October 10 to 31, 2017. The infection management intention of NRID was high in those who were over 30 years old, married, highly educated, and had a total working experience of more than 5 years. Nurses' infection management intention for NRID showed a positive correlation between knowledge of NRID (r=.27, p<.001) and attitude toward NRID (r=.65, p<.001). In other words, the higher the knowledge score for NRID, the more positive the attitude and the higher the infection management intention. In addition, the knowledge score related to incubation period, treatment, isolation, and release criteria was low while the educational needs were high. Therefore, in preparation for the possibility of NRID relapse and other NRID in the future, a systematic program addressing these educational needs for nurses should be periodically implemented to enhance infection management.

Development of suitable substrate of Sparassis latifolia for bottle cultivation (꽃송이버섯 병재배 적합 배지 개발)

  • This study sought to identify the optimum substrate composition for the stable bottle cultivation of Sparassis latifolia. The main substrate was fermented larch sawdust. Six nutrient sources were mixed at a maximum volume ratio of 20%. The fresh weight of fruit body was the highest at 128.5 g for GMSL69033 and 126.6 g for 'Neoul' in the treatments of beet pulp and corn flour in a volume ratio of 15:5. In addition, the total cultivation period was 94 days, which was shorter than that required for other treatments. The selected substrate characteristics were pH 4.7, C:N (carbon to nitrogen) ratio of 106:4, moisture content of 70%, and air filling porosity of 38%. We plan to develop new income items through research on mycelial incubation and fruit body growth conditions.

The Antimicrobial Activity of Fermented Extracts from Korean Dendropanax morbifera (국내 황칠나무 발효 추출물의 항균력 평가)

  • We investigated the fermentation conditions for extracts of leave/branches and sap from Korean Dendropanax morbifera (D. morbifera) using Lactobacillus plantarum (L. plantarum) ilchiwhangchil 1785 and L. plantarum ilchiwhangchil 2020. Log growth phase cultured L. plantarum ilchiwhangchil 1785 and L. plantarum ilchiwhangchil 2020 were used for fermentation. The pH and growth of the microorganisms in broth were monitored during the fermentation period. The results revealed that the optimum fermentation conditions for 20 wt% of leave/branches extracts and 1 wt% of sap extract was 2 days incubation at $37^{\circ}C$. The minimum inhibitory concentration (MIC) method and a disk diffusion assay were used to evaluate the antimicrobial activity of the fermented extracts of the leave/branches and sap against Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa. The antimicrobial activity increased in all three strains grown on the medium containing fermented extracts of the leave/branches and sap as compared with that of the strains grown on medium containing non fermented extracts. Furthermore, the antimicrobial activity increased in proportion to the contents of the fermented extracts. Our data suggest that fermented extracts of leave/branches and sap of D. morbifera have applications as natural bio functional materials, such as preservatives, cosmetic materials, and natural packaging materials.

Estrus synchronization and artificial insemination in Korean black goat (Capra hircus coreanae) using frozen-thawed semen

  • Presently, there is an increased demand for livestock products all over the world which has led to more devotion on improving livestock population. Although goats have been bred for a long time in Korea, but there is not much research conducted on traditional Korean black goat (Capra hircus coreanae) compared to other livestock populations. Mutton consumption has been dramatically changing from medicinal use to edible meat and this trend directs the black goat populations declining and also mutton import quantities are increasing consistently. The present study introduced a new estrus synchronizing technique with subsequent artificial insemination (AI) for Korean black goats to enable crossbreeding with non-native breeds for the small or subsistent farmers. Our data highlighted that, the percentage of motile sperm from the electro-ejaculated samples declined significantly after freezing and melting. In addition, the sperm motility significantly declined with regard to sperm incubation period (0, 5, 60, and 120 min at 37℃) and was negatively correlated (64.2 ± 7.9%, 63.3 ± 5.8%, 49.9 ± 6.3%, and 35.9 ± 7.6%, respectively) in frozen-thawed sperm samples. Moreover, the E2 levels were unchanged even 24 h after controlled internal drug releas (CIDR) withdrawal. But, 48 h and 72 h after CIDR removal, E2 levels increased significantly. These data helps us to consider the two time points for AI; CIDR removal after 24 h, at which E2 decreases, and after 48 h, as the time at which progesterone increases. Additionally, the AI after 48 h of CIDR removal group exhibited significantly higher pregnancy and parturition rates (42.9%) compared to AI after 24 h after CIDR removal 28.6% group. In conclusion, these studies will propose an optimal estrus synchronisation process with subsequent timing of AI and also will promote the Korean black goat breeding industry.

  • As a result of examining the germination rate between ginseng varieties, Jagyongjong varieties had the highest germination rate, and Yeonpung. had the lowest germination rate. In the ginseng seed germination rate experiment, the highest germination rate and growth condition were shown in artificial soil conditions of the ratio of Peatmoss 6.5: Pearlite 2: Masato 1.5. Good soil conditions require adequate soil moisture forces during the incubation period. The cultivation of ginseng medicinal crops requires optimal soil breathability, soil pH, and soil stabilization, which are important for root breathing. Microbial activity in the soil has a great influence on the growth of ginseng. The optimum pH of the soil for ginseng cultivation is 5.0-5.5 As a result of the experiment, the soil remained in an appropriate range after a month. In general, when the EC concentration value of the soil for ginseng cultivation is 0.2 mS/cm or more, growth deteriorates, and when the EC concentration value is 0.5 mS/cm or more, concentration obstacles such as root decay occur. As a result of the analysis, the higher the concentration value of EC, the more likely it is to interfere with ginseng growth.

Breeding of a new cultivar of White Flammulina velutipes, 'Seolhan' (흰색 팽이 신품종 '설한' 육성)

  • Flammulina velutipes is the representative edible mushroom in Korea. It makes up around 18% of the total mushroom production and accounts for about 36% of the total mushroom export. Despite being one of the most industrially important mushroom in Korea, the distribution rate of domestic cultivar was low (~31.6%) in 2020. Therefore, we need a new domestic cultivar with high quality as the main export mushroom. We developed a new white domestic cultivar, 'Seolhan', bred by mating two monokaryotic strains isolated from Baekseung and Woori-1ho. The optimum mycelial growth temperature was 25℃ and Seolhan had higher mycelial growth than the control at 20 and 25℃. In bottle cultivation, the optimum temperature of incubation was from 15℃ to 17℃ and the growth temperature of Seolhan was 7-8℃, which was slightly higher than that of the control. The yield of Seolan was 226.5±8.0 g, an increase of 18.4% compared to the control. The period of primodia formation also was a couple of days ahead. In morphological traits, the pileus of Seolhan was smaller than that of the control, the stipe was thicker, and the color brighter. As the results of replacement culture and genetic diversity test, it was confirmed that Seolhan was genetically different from other white varieties.

Selection of optimum conditions and distribution temperature of complete substrates of Pleurotus species for export to Southeast Asia (동남아 수출을 위한 느타리류 완성형배지의 최적 조건 및 운송 온도)

  • Pleurotus species are the most consumed and cultivated mushrooms in Korea. Although oyster mushrooms (P. ostreatus) can be cultivated automatically, their storability is slightly lower than that of king oyster mushrooms (P. eryngii) and winter mushrooms (Flammulina velutipes); therefore, the export proportion of oyster mushrooms is very low. Since Korean mushrooms are highly preferred across Southeast Asian, the export of oyster mushrooms in the form of complete substrates is expected to be more promising than that of fresh mushroom. Here, 1 and 2.5 kg complete substrates of P. ostreatus 'Soltari' and P. sajor-caju 'Sambok' were prepared and stored at different temperature from 0 to 15℃ for 10 days. Thereafter, the formation of fruiting bodies was induced. Since the 2.5 kg complete substrates required 70 days of incubation, their mycelia were at an advanced age and their fruiting bodies did not grown normally. When 70%-incubated complete substrates were stored at 5-10℃, the growth was faster and more uniform and stable fruiting bodies were formed. Export test of complete substrates to Vietnam using distribution containers set at 0℃ and 15℃ revealed that the growth period was shortened by 1-2 days when the distribution containers were set at 15℃ and the yield of 'Soltari' increased by approximately 10%. In addition, even though the yield of 'Sambok' was similar between treatments at 0℃ and 15℃, the quality of fruiting bodies from 15℃-distributed complete substrates was much better than that of those from 0℃-distributed substrates.

Evaluation of medium composition for developing an export-quality complete medium for Lentinula edodes (표고 수출용 완성형 배지의 적합 배지조성 선발)

  • This study aimed to replace the imported Chinese complete medium for Lentinula edodes with a newly-developed complete medium that is suitable for export as well. Three media compositions that supported fast mycelium growth and higher density, compared to that in the control, were chosen. Culture in the T13 medium reduced the incubation period for 'Nongjingo' by 12 days and for 'Cham-aram' by 10 days, compared to that required for the control; in addition, the number of days required for browning was greatly reduced for both varieties. The quantity of each mixed medium was increased according to the composition from the 1st to the 5th cycle by 5.9% for 'Nongjingo' and 12.6% for 'Cham-aram' in T13, compared to that in the control. A mixed medium comprising oak sawdust + Douglas fir sawdust + corn flour (40:40:20, v/v) was selected as the most suitable complete medium.
