Cyanobacteria Community and Growth Potential Test in Sediment of Lake Paldang (팔당호 퇴적층 남조류의 군집과 남조류 성장잠재성 분석)

  • We analyzed cyanobacterial community including akinete, resting cell, and benthic cyanobacteria in sediment of Bukhan, Namhan-River, Paldang-Lake, and Kyeongan-Stream and compared the physicochemical factors for water and pore water. We also performed cyanobacteria growth potential test through incubating sediment. As a result of physicochemical analysis, the concentrations of nutrients were similar for each depth of Bukhan-River. For Namhan-River and Lake Paldang sites, the concentrations of TP and DTN in bottom and deep water had higher levels. DTN in water body composed of NO3-N(73%) while DTN in pore water composed of NH3-N(77.8%). Benthic cyanobacteria in the sediment such as Oscillatoria tenuis, O. limosa, Phormidium tenue, Pseudanabaena limnetica, and Lyngbya sp. were dominant (between 0.0∼243.3×103 cells/g, w/w). Cell densities of cyanobacteria in sediment depth of 0∼2 cm in most sites were higher compared to those in other depths. The cell density of cyanobacteria in sediment correlated with pH, conductivity, BOD5, TP, DTP, and chl. a. Increased phytoplankton and organic matters were found to be able to inhibit the growth of benthic cyanobacteria. Results of cyanobacteria growth potential test after incubating sediment revealed that harmful cyanobacteria (Anabaena, Aphanizomenon, Microcystis, and Oscillatoria) appeared at 7 days post culturing. Base on these results, the methods used in this study are considered to be able to determine the appearance of harmful cyanobacteria.

Effect of Red Bean Protein and Microbial Transglutaminase on Gelling Properties of Myofibrillar Protein (적소두단백질(Red Bean Protein)과 Transglutaminase를 첨가한 돈육 근원섬유 단백질의 물성 증진 효과)

  • The effects of soy protein isolate (SPI) and red bean protein isolate (RBPI) on gelling properties of pork myofibrillar protein (MP) in the presence of microbial transglutaminase (MTG) were studied at 0.45 M NaCl. MP paste was incubated with MTG (0.1%) at various levels (0.1, 0.3, 0.5, and 1%) of SPI and RBPI before incubating at $4^{\circ}C$ for 4 h. The rheological property results showed that MP gel shear stress increased with increasing RBPI concentration. Cooking yield (CY) of the MP gel increased with increasing RBPI and SPI, whereas gel strength (GS) was not affected by adding RBPI or SPI. Thus, effects of incubation time (0, 4, 8, 10, and 12 h) were measured at 0.1% SPI and RBPI. GS values of the MP gel at 10 and 12 h were similar and were higher than those of the others. CY values were highest when RBPI (0.1%) was added, regardless of incubation time. The protein patterns indicated that incubating the MP with MTG for 10 h resulted in protein crosslinking between MP and RBPI or SPI. Based on these results, RBPI and SPI could be used as an ingredient to increase textural properties and cooking yield of meat protein gel.

Studies on Cultural Characteristics Pestalotiopsis theae causing Leaf Blight on Oriental Persimmon Tree (단감나무 둥근갈색무뉘병원균 Pestalotiopsis theae의 배양적 특성)

  • Culture conditions affecting mycelial growth and sporulation of P. theae, SP2, SP3 and P. longiseta which causing leaf blight on oriental persimmon leaf blight have been investigated. The optimum temperature for the mycelial growth and sporulation on potato dextrose agar was $25{\sim}30^{\circ}C$ in all the fungi, but was inhibited and finally arrested at 10 and $30^{\circ}C$. The optimum pH for mycelial growth and sporulation were ranged at pH 4.5~5.0 and 5.0~6.0 in all the fungi. Lenonian agar, potato sucrose agar and oatmeal medium were good culture media for the mycelial growth and sporulation of all the fungi. The effective sources of nitrogen and carbon for the mycelial growth were tryptone, glycine, starch, dextrose, galactose and lactose. Glutamic acid, peptone and tryptone were good nitrogen sources for sporulation of the fungi. Sucrose, starch and galactose were also good carbon sources for sporulation. Generally, vitamins had no effect on mycelial growth and sporulation. The pH of the potato dextrose broth inoculated with SP2, SP3 and P. theae and P. longiseta was changed from 7.0 to 4.5~4.7 and 5.0~5.4 after incubating for 10 days, respectively. But, the initial pH of the medium adjusted to 5.0 was lowered to 4.5~4.7 after incubating for 10 days.

TNF$\beta$ Induces Cytotoxicity of Antibody-Activated CD$4^+$T-lymphocytes Against Herpes Virus-Infected Target Cells

  • We have extended our previous work that cross-linking CD4 molecules using specific MAb induced antigen nonspecific, MHC unrestricted killing of virally infected target cells by CD$4^+$We have extended our previous work that cross-linking CD$4^+$ molecules using specific MAb induced antigen nonspecific, MHC unrestricted killing of virally infected target cells by CD$4^+$ T cells. The killing activity of antibody activated CD$4^+$T cells was completely blocked by herbimycin A, a protein tyrosine kinase (PTK) inhibitor, but not by bisindolylamaleimide, a protein kinase C (PKC) inhibitor. Herbimycin A treated human or bovine peripheral blood CD$4^+$T cells lacked PTK activity and failed to kill virally infected target cells even after cross-linking of CD4 molecules. The CD$4^+$cross-linking failed to induce effector cell proliferation or the transcription of TNF${\beta}$ Upregulation of TNF${\beta}$ was induced by incubating the antibody activated effector cells with BHV-1 infected D17 target cells for 10 h. Anti-TNF${\beta}$ antibody partially abolished (13-44%) the direct effector cell-mediated antiviral cytotoxicity. However, this antibody neutralized 70 to 100% of antiviral activity of effector and target cell culture supernatants against BHV-1 infected D17 cells. The inhibition level of the antiviral activity by the antibody was dependent on the effector and target cell ratio. These results support the hypothesis that increased p$56^ICK enzyme activity in effector cells transduces a signal critical for effector cell recognition of viral glycoproteins expressed on the target cells. Following target cell recognition, lytic cytokines known to participate in target cell killing were produced. A better understanding of the killing activity displayed by CD$4^+$T lymphocytes following surface receptor cross-linking will provide insight into the mechanisms of cytotoxic activity directed toward virally-infected cells.T cells. The Effect of Bacillus-Fermented Scutellariae Radix Acupuncture Solution on Interleukin Production in Mouse Macrophage Stimulated by Lipopolysaccharide (바실러스균 발효황금약침액이 Lipopolysaccharide로 활성화된 마우스 대식세포의 인터루킨 생성에 미치는 영향)

  • Objectives : The purpose of this study is to investigate the effect of Bacillus-fermented Scutellariae Radix acupuncture solution (SB) on interleukin(IL) production in mouse macrophage stimulatedby lipopolysaccaride(LPS). Methods : Productions of interleukins were measured y High-throughput Multiplex Bead based Assay with Bio-plex Suspension Array System based on $xMAP^{(R)}$(multi-analyte profiling beads) technology. To begin with, cell culture supernatant was obtained after treatment with LPS(1 ${\mu}g/mL$) and SB for 24 hour. Then, it was incubated with the antibody-conj${\mu}g$ated beads for 30 minutes. And detection antibody was added and incubated for 30 minutes. After incubating for 30 minutes, Strepavidin-conjugated Phycoerythrin(SAPE) was then added. Incubating for another 30 minutes, the level of SAPE fluorescence was analyzed on Bio-plex Suspension Array System. Results : The results of the experiment are as follows. SB significantly inhibited the LPS-induced production of IL-3($9.15{\pm}0.35$ pg/mL) by $6.92{\pm}0.05,\;7.21{\pm}0.11,\;6.96{\pm}0.33,\;and\;7.45{\pm}0.74$ pg/mL at the concentration of 25, 50, 100, and 200 ${\mu}g/mL$ in mouse macrophage RAW 264.7 cells (p<0.05). SB significantly inhibited the LPS-induced production of IL-5($7.30{\pm}0.48$ pg/mL) by $6.50{\pm}0.29,\;6.30{\pm}0.25,\;6.30{\pm}0.25,\;and\;5.80{\pm}0.25$ pg/mL at the concentration of 25, 50 100, and 200 ${\mg}g/mL$ in RAW 264.7 cells (p<0.05). SB significantly inhibited the LPS-induced productiion of IL-9($17.26{\pm}0.19$ pg/mL) by $15.01{\pm}0.43$ pg/mL at the concentration of 25 ${\mu}g/mL$ in RAW 264.7 cells(p<0.05). SB significantly inhibited the LPS-induced productioh of IL-13($187.80{\pm}2.90$ pg/mL) by $152.80{\pm}4.25,\;172.80{\pm}3.97,\;162.10{\pm}6.67,\;and\;165.30{\pm}11.80$ pg/mL at the concentration fo 25, 50, 100, and 200 ${\mu}g/mL$ in RAW 264.7 cells(p<0.05). SB significantly inhibited the LPS-induced production of IL-17($18.30{\pm}0.95$ pg/mL) by $13.30{\pm}1.25,\;13.80{\pm}1.11,\;13.30{\pm}0.75,\;and\;14.00{\pm}1.08$ pg/mL at the concentration of 25, 50 100, and 200 ${\mu}g/mL$ in RAW 264.7 cells(p<0.05). SB significantly inhibited the LPS-induced production of IL-23($43.90{\pm}0.83$ pg/mL by $39.50{\pm}1.26,\;38.00{\pm}1.78,\;and\;39.60{\pm}2.49$ pg/mL at the concentration of 25, 100, and 200 ${\mu}g/mL$ in RAW 264.7 cells(p<0.05). Conclusions : These results suggest that SB has anti-inflammatory activity related with its inhibition of IL-3, IL-5, IL-13, IL-17, and IL-23 production in macrophages.

Institutional Milieu of High-Tech Venture Business Cluster in a Metropolitan Area: the Case of Daegu City in Korea (지방대도시 벤처생태계의 제도적 및 문화적 환경: 대구지역을 사례로)

  • In this paper we attempt to explore the institutional and clutural milieu of high-tech venture business cluster in Daegu City and provide policy implications for creating sustainable high-tech venture businesses cluster. Although since 1998 the Korean government has made explicitly a great deal of efforts to help attract and create high-tech new start-ups in non-capital cities such as the support of business incubating centers and the establishment of real service centres, it reveals that such policies are not so successful in creating a sustainable venture ecology. One of the most critical resions is that such policies are centred simply on building physical infrastructure such as incubating space and service centre with less concern with promoting venture business cluster reflecting cultural and institutional specificities in a given region. In this respect we argue that the focus of industrial policies to support promoting high-tech venture business clusters in non-capital cities urgently need to move towards to make it for firms to encourage entrepreneurship driving innovation and to create social capital incucing trust and networks amongst firms, universities, public &privite supporting agencies, together with the building up of physical infrastructure the existing policies have done.

  • With the tourism industry stagnating due to the recent COVID-19 virus issue, the importance of preparing a plan to resume and revitalize the tourism industry after the end of the COVID-19 is being emphasized. In particular, this researcher paid attention to the development of the horse industry as it can contribute to the differentiation of services as it is an experience-oriented tourism, and it can be said to be a field with a high potential for sustainable development in the future. However, in the domestic horse industry, legal problems related to installation, industrial structural problems organized around horse racing, and demand-based problems were scattered. Therefore, this researcher suggested a way to improve these problems, improve the quality of life of the people, and lead the horse industry to increase the income of rural residents. As a representative example, a legal improvement plan for the expansion and growth of equestrian facilities was presented, and a strategic incubating infrastructure construction plan was presented to support horse industry workers to nurture the horse industry as the 6th industry on their own. This movement will lead to qualitative growth through the conversion of the horse industry into local cultural contents, and not only to expand the base of public demand for the horse industry, but also to maximize the creation of added value through diversification of the horse industry, leading to qualitative growth of the horse industry.

A Study on the Current Conditions of Local Independent Bookstores and Methods to Vitalize the Bookstores : Focused on Local Independent Bookstores in Gwangju (지역 독립서점 현황 및 활성화 방안 : 광주지역을 중심으로)

  • In Korea, the independent publication market has grown since it came into existence in the 1990s. In 2008, the country's first independent bookstores selling independent publications opened. As of late 2019, there were a total of 466 independent bookstores in Korea and the number of independent bookstores rapidly increased for the last decade. However, the closing rate of independent bookstores is rapidly increasing. This trend is also shown in independent bookstores in Gwangju. Recently, the closing rate of independent bookstores in Gwangju is more than twice as high as the national average. Against this backdrop, this study intended to identify the current conditions of independent bookstores in the city, analyze the problems facing them, and find solutions. Independent bookstores in Gwangju currently make various efforts such as book curation, management of various supplementary programs, and book markets. However, the bookstores have problems including the curation and programs focused on specific areas, focus on limited generations, dependence on publications from the Seoul metropolitan area. To address these problems, this study proposed solutions such as diversifying publications through planning and developing of local contents, strengthening the taste-based salon nature, organizing networks of independent bookstores, and systematically incubating independent bookstores. At the system level, this study also suggested a need for local bookstore certification system, a complete execution of a fixed book price system, and a fair provision rate.

Directions for Eco-friendly Utilization and Industrialization of Fishery By-products (수산부산물의 발생·처리 실태 및 산업화 방향)

  • The study aims to identify the generation and treatment of fishery by-products in Korea and suggests future directions and strategies for their eco-friendly utilization and industrialization. First, the study focuses on the identification of the generation and their treatment in Korea since merely few study were conducted and they did not provide enough information regarding the overall generation and treatment at the national level. According to the estimation, Korea generates 800 thousand to 1,200 thousand tones of fishery by-product every year. The fishery by-products generated at large seafood markets and processing facilities are used or processed as fish meal and feed, but those generated from households and small seafood restaurants are currently treated as food waste. In addition, inadequately treated fishery by-products cause various problems such as spoiling urban landscape, creating odor and incubating pest. After identifying the generation and treatment of fishery by-products, the study suggests directions for the formulation of infrastructure for transition into resource circulation society, minimization of dumped waste and their eco-friendly recycling as resources, diversification of recycled goods and development into a high-value added industry. Finally, the study suggests detailed strategies for the directions such as establishment of legal and institutional foundation, separation of fishery by-products from wastes, development of technology tailored for commercialization, introduction of pilot projects for industrialization and cultivation of social enterprises.

Antioxidative Activity of Water Extract of Different Parts of Acanthopanax divaricatus var, albeofructus (흰털오가피 부위별 물추출물의 항산화활성)

  • Acanthopanax species have traditionally been used as a tonic, a sedative as well as in the treatment of rheumatism, hypertension and diabetes. In the present study, oxidative stress was induced in Vero cells by incubating the cells with glucose and the cell viability was measured by MTT assay. The concentration of glucose which 50% of cell viability was 125 mM $(IC_{50})$ and the cell viability was increased to $87.6{\pm}8.8%$ by treatment of the extracts of Acanthopanax divaricatus var. albeofructus. The antioxidative activity of water extract of different parts of the Acanthopanax plant was investigated by DPPH (1,1-diphenyl-2-picrylhydrazyl) assay, xylenol orange assay, TBARS (thiobarbituric acid reactive substances) assay and enzyme (superoxide anion and catalase) assay. Each extract (leaves, root, stem and fruits) of the plant showed free radical and $H_2O_2$ scavenging activity. The extract also inhibited lipid peroxidation and recovered enzyme (superoxide anion dismutase and catalase) activity in Vero cells treated with glucose.
