Evaluation of Biological Activities of Fermented Rice Bran from Novel Black Colored Rice Cultivar SuperC3GHi (슈퍼자미(SuperC3GHi) 미강 발효물의 생리활성)

  • Rice bran is by-product from the dehulling process of rice, an important staple food in Korea. Various studies have been reported immune-enhancing effects of rice bran cultured with Lentinus edodes. In particular black rice bran that contains anthocyanin has the beneficial effects on immune system. SuperC3GHi, a recently released black colored rice cultivar, has higher contents of cyanidine-3-glucoside (C3G), a strong antioxidant than those of currently cultivated black rice cultivars. In this study, we investigate the beneficial effects of fermented SuperC3GHi rice bran (C3G-F) with Lentinus edodes. Using in vivo and in vitro model for inflammation and allgergy, anti-inflammatory and anti-allergic effects of C3G-F were determined. C3G-F induced tumor suppressive effect on Sarcoma-180 cell and B16 melanoma cell injected to mice. These results suggest that the capacity of C3G-F seems to act as a potent immunomodulator and with the absence of notable side-effects, C3G-F could be used as a novel functional food ingredient having possible therapeutic effects against immunological disorders.

In Vitro Screening for Compounds Derived from Traditional Chinese Medicines with Antiviral Activities Against Porcine Reproductive and Respiratory Syndrome Virus

  • Seventeen compounds derived from traditional Chinese medicines (TCMs) were tested for their antiviral activity against porcine reproductive and respiratory syndrome virus (PRRSV) in vitro. Visualization with the cytopathologic effect (CPE) assay and the 3-(4, 5-dimethyithiazol-2-yl)-2,5-diphenyltetrazolium bromide test were used to determine the 50% cytotoxic concentration ($CC_{50}$) and 50% effective concentration ($EC_{50}$) in cultured Marc-145 cells. Among the tested compounds, chlorogenic acid and scutellarin showed potential anti-PRRSV activity. The $EC_{50}$ values were $270.8{\pm}14.6{\mu}g/ml$ and $28.21{\pm}26.0{\mu}g/ml$ and the selectivity indexes were >5.54 and 35.5, respectively. The time-of-addition and virucidal assay indicated that the anti-PRRSV activity of the two compounds could be due to their inhibiting the early stage of virus replication and/or inactivating the virus directly. The inhibition of the virus attachment was not observed in the adsorption inhibition assay. The inhibition ratios of chlorogenic acid and scutellarin were, respectively, 90.8% and 61.1% at the maximum non-cytotoxic concentrations. The results have provided a basis for further exploration of their antiviral properties and mechanisms in vivo. We believe that the chlorogenic acid and scutellarin have a great potential to be developed as new anti-PRRSV drugs for clinical application.

A Comparative Study on the Physiological Activities of Auricularia spp. (목이버섯 품종간 생리활성 비교 연구)

  • This study investigated the physiological effects of three species of Auricularia known as Auricularia polytricha (JNM21001), Auricularia auricula-judae black (JNM21002), and Auricularia auricula-judae brown (JNM21012). In the ORAC assay, Auricularia spp. showed antioxidant activities in the order of JNM21001>JNM21012>JNM21002. All Auricularia spp. strongly inhibited the action of ${\alpha}$-amyloglucosidase up to 60%. In order to further test in vivo anti-obesity effects, high fat diet induced ICR obese mice fed a diet containing 20% fat were used. All Auricularia spp. supplementation during high fat diet feeding significantly reduced body weight gain, epididymal fat pads weight, and lowered the food efficiency ratio compared to the high fat control (HFC) group. In particular, the group fed with JNM21012 had a lower average daily body weight gain of 0.45 g/day, demonstrating similar levels to the normal diet fed group. The group fed with JNM21012 significantly reduced lowered serum triglycerides (42%), total cholesterol (81%), and LDL-cholesterol level (66%) compared with the HFC group.

Evaluation of DNA Damage and Repair Kinetics in the Earthworm (Eisenia fetida) Exposed to Radiation and Mercury (방사선과 수은에 의해 유도된 Eisenia fetida 체강세포의 DNA 손상 및 수복 평가)

  • The single cell gel electrophoresis (SCGE) assay is a microelectrophoretic technique for assessments of DNA damage at the level of the individual eukaryotic cell. The SCGE assay, due to its simplicity, sensitivity and need of a few cells, has advantages compared to other genomic damage assays such as sister chromatid exchange, chromosomal aberration and micronucleus test. In this study, investigated were the levels of DNA damage and the repair kinetics in the coelomocytes of Eisenia fetida treated with HgCl2 and ionizing radiation by means of the SCGE assay. For detecting DNA damage and repair in coelomocytes, earthworms (E. fetida) were irradiated with six doses of ${\gamma}$-rays (0, 2.5, 5, 10, 20 and 50 Gy) and in vivo exposed to mercuric chloride at 0, 80 and 160 mg $kg^{-1}$ for 48 hours. Then the Olive tail moments were measured during 0~12 hours after irradiation and 0~72 hours after Hg treatment. The results showed that the more the oxidative stress was induced by mercury and radiation, the longer the repair time was required. Also, the results suggest that the SCGE assay may be used as an important tool for comparison of the sensitivity of different species to oxidative stresses.

Anti-wrinkle Effect of Morinda citrifolia (Noni) Extracts (노니 추출물의 주름개선 효과연구)

  • Wrinkle formation is mainly attributed to the environmental factors such as UV rays, air pollution, smoking and stress etc. Especially, UV rays induce premature skin aging which is characterized by deep wrinkle, leathery dryness etc. Recently, researches on the wrinkle formation and its prevention have been the main theme in cosmetics fields. We have studied the various plant extracts having anti-wrinkle effects and finally showed that Noni (Morinda citrifolia) extracts have the efficacy of promoting the type I collagen synthesis in normal human fibroblast, using PICP assay. We purified one active compound from Noni extracts and identified its structure. It was identified as 6,7-Dimethoxy-2H-1-Benzopyran-2-one; scopoletin by $^1H-NMR,\;^{13}C-NMR,$ IR, Mass analysis. Scopoletin increased collagen synthesis in a dose dependent manner (89.5% at $0.2{\mu}g/mL$). In order to verify the anti-aging effectiveness of the cream containing 3% noni extracsts, we performed the in vivo test with some female volunteers for 12 weeks. It reduced the signs of aging, especially face wrinkles. From these results, we conclude that the noni extracts could be used as an useful anti-wrinkle agent.

Acute Toxicity of Leuconostoc Citreum GR1 Isolated from Kimchi in Mice (김치로부터 분리한 Leuconostoc citreum GR1의 마우스에 대한 급성독성)

  • The present study was carried out to investigate the in vivo single-dose acute toxicity of Leuconostoc citreum GR1 (Leuc. citreum GR1), a lactic acid bacteria isolated from kimchi, in ICR male and female mice. The test article was orally administered once to both sexes of mice. The mortalities, clinical findings, autopsy findings, and body weight changes were monitored daily for two weeks. The male and female mice were gavaged with Leuc. kimchi GR1 of four doses (625, 1,250. 2,500 and 5,000 mg/kg). The oral $LD_{50}$ of Leuc. citreum GR1 was considered higher than 5,000 mg/kg. No significant changes in the general conditions, body weights, clinical signs and presence of gross lesions were observed in both sexes of mice to whom Leuc. citreum GR1 was administered orally. The results indicated that the 5,000 mg/kg dose of Leuc. citreum GR1 showed no adverse effect.

Anti-lipase and Lipolytic Activities of EtOH Extract from Juniperus rigida (노간주나무 에탄올추출물의 지방 흡수 억제 및 지방분해 효능)

  • Obesity is an important risk factor that significantly increases mortality and disease rates in the cardiovascular disease, diabetes, and various diseases. So far, the most powerful way to inhibit fat absorption is pancreatic lipase inhibitors. In this study, we investigated the anti-obesity effect of the extract of Juniperus rigida. Juniperus rigida extract (JRE) had a inhibitory effect on pancreatic lipase activity ($IC_{50}$=8.63 ${\mu}g$/ml). In in vivo oil-emulsion loading test, this extract also inhibited the intestinal fat absorption. In addition, we measured inhibitory effects of JRE on activity of phosphodiesterase (PDE) and hormone sensitive lipase (HSL) among the important enzymes associated with lipolysis. JRE strongly inhibited PDE activity ($IC_{50}$=4.56 ${\mu}g$/ml), whereas inhibitory effect on HSL activity was very weak compared with orlistat. As a result, JRE inhibited the absorption of fat by inhibiting the activity of pancreatic lipase and induced lipolysis through inhibition of PDE activity. Therefore, we suggest that Juniperus rigida may be a potential therapeutic agent improving obesity.

Scab Resistance of Some Pear Genetic Resources and Inter Specific Hybrid Seedlings in Field Condition (포장조건에서 주요 배 유전자원과 종간교배 집단의 검은별무늬병 저항성)

  • This study was accomplished to select breeding materials among the pear germplasm 6 species 43 accessions and check out the expression of scab incidence of inter specific hybrids of 5 crosses 609 seedlings for scab resistance breeding. Fruitlets scab incidence of P. pyrifolia, P. bretschneideri and P. ussuriensis were 59.5, 73.4 and 63.0% respectively, and also fruitlets scab incidence of interspecific hybrid cultivars between P. pyrifolia and P. bretschneideri were 77.7%. However, fruitlets scab incidence of P. communis and interspecific hybrid accessions between P. pyrifolia and P. communis were significantly less than 15%. As a result of the Duncan test, scab resistance of pear accessions were classified into two groups: P. communis and their hybrid were resistance and P. bretschneideri, P. pyrifolia, P. ussuriensis, and among their hybrid were susceptible. Also, it showed the same trend in the seedlings. Scab incidence of 'Seolwon' ${\times}$ 'Chuwhangbae' and 'Supergold' ${\times}$ 'Mantienhong' in P. pyrifolia seedlings showed more than 80%, while that of interspecific hybrid between P. pyrifolia and P. communis showed about 2%. It showed scab resistance of P. communis transferred to hybrid seedlings and expressed to be dominant in their seedlings. Namely, P. communis was good breeding materials for scab resistance.

Anti-wrinkle Effect of Cosmetics Containing Duchesnea indica Extract (사매추출물을 함유하는 화장품의 주름 개선 효과)

  • In this study, we investigated anti-oxidative effects of Duchesnea indica extracts by using Oxygen Radical Absorbance Capacity (ORAC). The extracts were prepared with 0 %, 30 %, 50 %, 70 % and 100 % aqueous ethanol respectively. The 30 % EtOH D. indica extract showed higher ORAC activity than the other extracts. Therefore, we performed in vitro studies on cytotoxicity of NIH-3T3 cells and MMP-8 collagenase inhibition using by the 30 % EtOH extract. The 30 % EtOH extract showed no cytotoxicity and significant inhibition on MMP-8 collagenase. And we performed clinical studies for the anti-wrinkle effect of the Di-Wrinkle Free Cream. The cream formula was prepared with 2 % arbutin and 1 % D. indica extract. Twenty one healthy women volunteers, ages of 35 and 50, applied the cream on their faces twice a day for 8 weeks. The skin was evaluated with PRIMOS (phaseshift rapid in vivo measuring of human skin) system and analyzed by the student's paired t-test. The wrinkles on the eye region were reduced by 13 % based on the PRIMOS system after 8 weeks. In the safety study of the Di-Wrinkle Free Cream, no symptoms were observed such as erythema, edema, scaling, itching, stinging, burning, tightness and prickling by visual observation and medical examination of volunteers for 8 weeks. Moreover, there was no noticeable skin disorder during experience period. These results suggested that D. indica extracts could be applied as cosmeceuticals effective for anti-wrinkle.

The Antibacterial Activity Against Fish Pathogen of Paenibacillus sp. MK-11 Isolated from Jeju Coast (제주연안으로부터 분리한 Paenibacillus sp. MK-11의 어류 질병 세균에 대한 항균활성 탐색)

  • In this study, we isolate and identify bacteria from seawater collected from Jeju coast, to evaluate the antimicrobial activity against the fish pathogenic bacteria. 14 bacterial strains were isolated and identified using physiological, biochemical and molecular tools. Antibacterial activity of all the 14 isolates were screened against four major fish pathogens namely, two Gram-positive: Streptococcus iniae, Streptococcus parauberis and two Gram-negative: Vibrio anguillarum, Edwardsiella tarda. Results revealed that among the 14 isolates, MK-11 was found to have antibacterial activity against S. iniae, S. parauberis, V. anguillarum Particularly, S. iniae was susceptibility with the MIC value of $250{\mu}g/ml$. The biochemical and physio-chemical results reveal that MK-11 had the sugar-alcohol disassemble ability of the D-sorbitol and D-mannitol. Also the utilization of the yeast extract, sorbitol and di-potassium phosphate were noted to be high. The optimum culture condition such as pH and temperature was recorded as pH 6.0, $25^{\circ}C$ and along with 1% NaCl which differs from the previous reports particularly in nutrient resolutions. As results of the analysis of 16S rDNA sequences, MK-11 show the high similarity with Paenibacillus polymyxa, P. jamilae, P. brasilensis 99.78%, 99.43%, 99.39%, repectively. Hence, in the present study, the isolated Paemibacillus sp. MK-11 from Jeju seawater possesses the antibacterial activity against fish pathogens and it could be used as a new antibiotic agents against the gram positive fish pathogens.
