Purpose: The objective of this study was to examine the affected period and the amount of bone formation during osteogenesis of intramembranous bone using low-intensity pulsed ultrasound (LPUS) $in$$vivo$. Methods: Xeno-bone (Bio-oss) and autogenous bone were grafted bilaterally into mini-pig mandibles. The left mandible served as the control and the other mandible was treated with 3 MHz, 160 mW (output, 0.8 mW) ultrasound stimulation for 7 days 15 minutes per day. The mini-pigs were sacrificed at 1, 2, 4, and 8 weeks, and micro computed tomography (${\mu}CT$), a microscopic examination, and a statistical analysis were performed on the specimens. Results: Based on a computerized image analysis of the ${\mu}CT$ scans, the experimental group had an average 150% more new bone formation than that in the control group. The effect of LPUS continued during the post operative 2 weeks. The histomorphological microscopic examination showed similar results. Conclusion: Our results suggest the LPUS had an effect on early intramembranous bone formation in vivo.
Recently, small-animal imaging technology has been rapidly developed for longitudinal screening of laboratory animals such as mice and rats. One of newly developed imaging modalities for small animals is an x-ray micro-CT (computed tomography). We have developed two types of x-ray micro-CT systems for small animal imaging. Both systems use flat-panel x-ray detectors and micro-focus x-ray sources to obtain high spatial resolution of $10{\mu}m$. In spite of the relatively large field-of-view (FOV) of flat-panel detectors, the spatial resolution in the whole-body imaging of rats should be sacrificed down to the order of $100{\mu}m$ due to the limited number of x-ray detector pixels. Though the spatial resolution of cone-beam CTs can be improved by moving an object toward an x-ray source, the FOV should be reduced and the object size is also limited. To overcome the limitation of the object size and resolution, we introduce zoom-in micro-tomography for high-resolution imaging of a local region-of-interest (ROI) inside a large object. For zoom-in imaging, we use two kinds of projection data in combination, one from a full FOV scan of the whole object and the other from a limited FOV scan of the ROI. Both of our micro-CT systems have zoom-in micro-tomography capability. One of both is a micro-CT system with a fixed gantry mounted with an x-ray source and a detector. An imaged object is laid on a rotating table between a source and a detector. The other micro-CT system has a rotating gantry with a fixed object table, which makes whole scans without rotating an object. In this paper, we report the results of in vivo small animal study using the developed micro-CTs.

This study evaluated the structure and quality of osteoporotic vertebral bone. To induce osteoporosis, eight rats were ovariectomized (OVX). All rats were divided into two groups (Normal group: 4, OVX group: 4). Total lumbar vertebrae for each rat were scanned by in-vivo ${\mu}CT$ at 0, 4 and 8 weeks. Morphological characteristics (BV/TV, Tb.Th, Tb.N, Tb.Sp and SMI) were calculated by in-vivo ${\mu}CT$ image analyzer. Three dimensional finite element models were analyzed to investigate bone strength of OVX and Normal groups. Moreover, the elastic modulus was quantitatively analyzed to evaluate the quality changes of osteoporotic bone. In the OVX group, BV/TV, Tb.Th and Tb.N were significantly decreased at all the lumbar over time (p<0.05). We also investigated a contrary tendency in Tb.Sp and SMI, compared to the above results in each group. A degree of alteration of mechanical characteristics in OVX group was decreased over measuring time (p<0.05). Bone quality presented by distribution of elastic modulus was improved in the Normal group more than OVX group. The findings of the present study indicated that both bone structure and quality of whole lumbar could be tracked and detected by analyzing the morphological and biomechanical characteristics of bones, based on a nondestructive method.

This study investigated the effect of red ginseng extract on metastasis of colon cancer cells in vitro and in vivo. Wound healing migration, cell motility, invasion, and activity, protein expression, and mRNA expression of matrix metalloproteinases (MMPs) were examined in SW480 human colon cancer cells. SW480 cells were cultured with or without $100{\mu}g/L$ PMA in the absence or presence of various concentrations (100, 200, or $300{\mu}g/mL$) of red ginseng extract. Red ginseng extract treatment caused signifi cant suppression of cell motility and invasion (p<0.05) in SW480 cells. Red ginseng extract inhibited MMP-2 and MMP-9 activity and their protein and mRNA expression in a dose-dependent manner (p<0.05) in SW480 cells. For experimental metastasis, BALB/c mice were injected intravenously with CT-26 mouse colon cancer cells in the tail vein, and were orally administered various concentrations (0, 75, 150, or 300 mg/kg body weight) of red ginseng extract for 3 weeks. Numbers of pulmonary nodules were signifi cantly decreased in mice that were fed red ginseng extract (p<0.05). Plasma MMP-2 and MMP-9 activity signifi cantly decreased in response to treatment with red ginseng extract in mice (p<0.05). These data suggest that red ginseng extract may be useful for prevention of cancer invasion and metastasis through inhibition of MMP-2 and MMP-9 pathways.

This study showed the change of mechanical characteristics of cortical bone as well as trabecular bone of an OVX rat. Detecting and tracking changes of mechanical characteristics could greatly contribute to an experiment test for the trabecular and cortical bone in the lumbar vertebrae of an OVX rat by using In-vivo Micro-CT.
Objectives: This study investigated the anti-osteoarthritic effects of Kyejiinsam-tang (hereinafter referred to KIT) on the monosodium iodoacetate (MIA)-induced osteoarthritis rats. Methods: Anti-oxidative effects of KIT were measured by scavenging activities of DPPH, reactive oxygen species (ROS) and nitric oxide (NO). Scavenging activities of anti-oxidation in lipopolysaccharide (LPS)-treated RAW 264.7 cells were also measured for inhibitory effects against the production of inflammatory mediators (tumor necrosis factor-${\alpha}$, interleukin-$1{\beta}$, interleukin-6). Osteoarthritis was induced in rats by injecting MIA in the knee joint. Rats were divided into a total of 4 groups (n=6). The normal group were not treated at all without inducing osteoarthritis whereas the control group were induced for osteoarthritis by MIA and oral medicated physiological saline per day. The positive comparison group was injected with MIA and after 7 days, 2 mg/kg of Indomethacin. The experimental group was injected with MIA and after 7 days was medicated with 34 mg/kg of KIT. Indomethacin and KIT were orally-medicated for each substance a total of 4 weeks, once per day. Weight-bearing on hind legs was measured every week after MIA injection. At the end of the experiment (5 weeks after MIA injection), micro CT (computed tomography)-arthrography and histopathological examinations on the articular structures of knee joint were performed. The effect on inflammatory cytokines and immunological cells in synovial fluid was measured. Volume of cartilage was measured by micro CT-arthrography. Injury to synovial tissue was measured by H & E (hematoxylin and eosin), Safranin-O immunofluorescence. Results: 1. Cytotoxicity against hFCs was insignificant. 2. KIT showed the potent full term for DPPH. 1. NO was significantly reduced by KIT (at 100, $200{\mu}g/m{\ell}$) and ROS was also reduced, but not significantly, by KIT (at $200{\mu}g/m{\ell}$). 2. IL-6 and IL-$1{\beta}$ were significantly reduced by KIT (at 100, $200{\mu}g/m{\ell}$) and TNF-${\alpha}$ was also reduced, but not significantly, by KIT (at $200{\mu}g/m{\ell}$). 1. In hind legs weight-bearing measurement, level of weight increased. 2. Functions of liver and kidney were not affected. 3. IL-$1{\beta}$ was significantly reduced and TNF-${\alpha}$, IL-6 were also reduced but not significantly. 4. PGE2 (prostaglandin E2), LTB4 (leukotriene B4) were significantly reduced in the KIT group. 5. MMP-9 (matrix metalloproteinase-9), TIMP-1 (tissue inhibitor of metalloproteinases-1) and Osteocalcin were significantly reduced in the KIT group. 6. Destruction of cartilage on micro CT arthrography was reduced but had no significant differences. 7. Histopathologically, injury to synovial membrane of the KIT group was decreased and proteoglycan content of KIT group was increased. Conclusions: According to this study, Kyejiinsam-tang has inhibiting effect on the progression of arthritis in MIA-induced osteoarthritis rat. Kyejiinsam-tang has anti-oxidants and anti-inflammation effects, and is related to inhibiting the activity of inflammatory cytokine and injury of volume in cartilage.
Although tectorigenin (TG), a major compound in the rhizome of Belamcanda chinensis, is conventionally used for the treatment of inflammatory diseases, its effects on osteogenesis and osteoclastogenesis have not been reported. The objective of this study was to investigate the effects and possible underlying mechanism of TG on in vitro osteoblastic differentiation and in vivo bone formation, as well as in vitro osteoclast differentiation and in vivo bone resorption. TG promoted the osteogenic differentiation of primary osteoblasts and periodontal ligament cells. Moreover, TG upregulated the expression of the BMP2, BMP4, and Smad-4 genes, and enhanced the expression of Runx2 and Osterix. In vivo studies involving mouse calvarial bone defects with ${\mu}CT$ and histologic analysis revealed that TG significantly increased new bone formation. Furthermore, TG treatment inhibited osteoclast differentiation and the mRNA levels of osteoclast markers. In vivo studies of mice demonstrated that TG caused the marked attenuation of bone resorption. These results collectively demonstrated that TG stimulated osteogenic differentiation in vitro, increased in vivo bone regeneration, inhibited osteoclast differentiation in vitro, and suppressed inflammatory bone loss in vivo. These novel findings suggest that TG may be useful for bone regeneration and treatment of bone diseases.

In photodynamic therapy (PDT), oxygen plays important role. Because of singlet oxygen which is produced by activated photosensitizer after laser irradiation of specific wavelength. The aim of this study is to find how oxygen concentration affects anticancer effect in PDT. Groups were divided into PDT with oxygen applied group and only PDT applied group. PDT with oxygen applied group supplied oxygen for 15 minute before laser irradiation. In vitro, CT-26 cell was incubated with various concentration of photofrin $(50.0{\sim}0.05{\mu}g/ml)$ and was irradiated with 632nm diode laser 6hr after application of photofrin. The cell viability of two groups was assessed by MTT assay. In vivo, CT-26 cell line was transplanted into the subcutaneous tissue of BALB/c mouse. The anticancer effect of two groups was measured by tumor volume change. In vitro study, the cell viability was significantly decreased at $1.56{\sim}3.13{\mu}g/ml$ in PDT with oxygen applied group. In vivo study, the PDT with oxygen applied group significantly higher reduction rate of tumor volume 7 days after PDT compared to PDT only group. The high oxygen concentration might enhance the anticancer effect of the photodynamic therapy.
