Free radical scavenging activity of some Mangroves available in Bangladesh

  • The crude alcoholic extracts of various parts of five different Bangladeshi mangrove plants (Amoora cucullata, Caesalpinia bonducella, Cerbera odollam, Derris uliginosa and Sonneratia caseolaris) were screened in-vitro for anti-oxidant activity using the 1,1-diphenyl-2-picryldydrazyl-hydrate (DPPH) free radical scavenging assay. Of all of them, the ethanolic extracts of the leaves of Derris uliginosa and Sonneratia caseolaris showed potential antioxidant activity ($IC_{50}$: 94.406 and 26.30 mg/ml respectively) whereas the ethanolic extracts of aerial parts of Derris uliginosa, barks of Cerbera odollam and leaves and stems of Caesalpinia bonducella showed moderate activity ($IC_{50}$: 125.89, 211.35 and 301.99 mg/ml respectively). Mild anti-oxidant activity was observed with the methanolic extract of leaves of Amoora cucullata and ethanolic extracts of fruits of Cerbera odollam.

In Vitro Genotoxicity Assessment of a Novel Resveratrol Analogue, HS-1793

  • Resveratrol has received considerable attention as a polyphenol with various biological effects such as anti-inflammatory, anti-oxidant, anti-mutagenic, anti-carcinogenic, and cardioprotective properties. As part of the overall safety assessment of HS-1793, a novel resveratrol analogue free from the restriction of metabolic instability and the high dose requirement of resveratrol, we assessed genotoxicity in three in vitro assays: a bacterial mutation assay, a comet assay, and a chromosomal aberration assay. In the bacterial reverse mutation assay, HS-1793 did not increase revertant colony numbers in S. typhimurium strains (TA98, TA100, TA1535 and TA1537) or an E. coli strain (WP2 uvrA) regardless of metabolic activation. HS-1793 showed no evidence of genotoxic activity such as DNA damage on L5178Y $Tk^{+/-}$ mouse lymphoma cells with or without the S9 mix in the in vitro comet assay. No statistically significant differences in the incidence of chromosomal aberrations following HS-1793 treatment was observed on Chinese hamster lung cells exposed with or without the S9 mix. These results provide additional evidence that HS-1793 is non-genotoxic at the dose tested in three standard tests and further supports the generally recognized as safe determination of HS-1793 during early drug development.

천초근 에탄올 추출물의 항산화 효능 및 대장암 세포 억제 효과 (Anti-colorectal Cancer and Anti-oxidant Activities of Rubiae radix Ethanol Extract in vitro)

  • 본 연구는 꼭두서니의 뿌리인 천초근의 물 추출물과 에탄올 추출물을 이용하여 대장암 세포에 대한 암세포 성장 억제 및 사멸효과가 있는지 알아보고자 수행하였다. ERA(천초근 에탄올 추출물)은 폴리페놀($45.77{\pm}2.03mg/g$)과 플라보노이드($22.82{\pm}1.33mg/g$)를 함유하고 있었으며, $H_2O_2$에 의해 증가된 ROS(reactive oxygen species)를 억제하는 효과를 나타냈지만 WRA(천초근 물 추출물)은 효과가 없었다. 또한 ERA는 $500{\mu}g/m{\ell}$의 농도로 대장암 세포주(HCT-116)에 처리했을 때 세포사멸을 유도할 뿐만 아니라 caspase-3 단백질 활성화, DNA fragmentation 및 apoptotic cell death를 일으키는 것으로 확인되었다. 이는 ERA가 HCT-116 세포주에 대해 apoptosis(세포자멸사)를 통해 항암효과를 나타내는 것으로 생각되지만 다른 연구결과들과 비교하였을 때 농도 대비 효능이 미미하다. 따라서 천초근 에탄올 추출물에 대장암 세포의 성장을 억제하는 유효성분을 분석하여 그 효능을 탐색하는 추가실험이 필요할 것으로 생각된다.

접골산(接骨散)이 골절치유에 미치는 영향 (Effects of Jeopgolsan (JGS) Extract on Fracture Healing)

  • Objectives The purpose of this study was to evaluate the effect of Jeopgolsan (JGS) extract on anti-oxidant, anti-inflammatory activities in RAW 264.7 cells and on factors related with fracture healing in skull fractured rat. Methods Experimental animals were divided into four groups: normal group without any treatment (Normal), contral group were treated orally with distilled water (Control), Experimental group were treated orally with JGS at a concentration of 200 mg/kg/day (JGS 200) and Experimental group were treated orally with JGS at a concentration of 200 mg/kg/day (JGS 400). Rats in each group except the normal group were induced fractures in the skull. The 1,1-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid (ABTS) radical scavenging activity were measured to evaluate antioxidant activity. The production of nitric oxide (NO), $interleukin-1{\beta}$ ($IL-1{\beta}$), interleukin-6 (IL-6) and tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) in the RAW 264.7 cells were measured to evaluate anti-inflammatory activity. The production of osteocalcin calcitonin, carboxy-terminal telepeptides of type II collagen (CTX II), transforming growth $factor-{\beta}$ ($TGF-{\beta}$), bone morphogenetic protein-2 (BMP-2), Insulin and alkaline phosphatase (ALP) in serum of rats were measured to evaluate the effects of fracture healing at 0, 2, 4, and 6th week. X-rays were taken every 3 week from 0 to 6th week to evaluate fracture healing effect. Results 1. No cytotoxicity was observed. 2. DPPH and ABTS radical scavenging activity were increased in a concentration dependent manner, indicating anti-oxidant effect. 3. NO, $IL-1{\beta}$, IL-6, and $TNF-{\alpha}$ were not significantly changed, indicating no anti-inflammatory effect. 4. Osteocalcin, Calcitonin, $TGF-{\beta}$ and ALP were significantly increased in the experimental groups. 5. CTX II, insulin were significantly decreased in the expermental groups. 6. Radiologic examination showed that union of fracture was promoted. Conclusions From above results, JGS showed significant results in factors related with fracture healing and radiologic examination. Threfore, JGS is expected to be effective in the treatment of fracture.

Santalum album Linn wood and its oil: An aromatic Unani traditional medicine with versatile pharmacological activities

  • Santalum album Linn. [Family: Santalaceae] is commonly known as white sandalwood, sandal safaid and safed chandan. It is one of the most valuable trees and second costliest wood in the world. Sandalwood and its oil is extensively used in the Unani and other traditional systems of medicine as it has blood purifier, anti-inflammatory, analgesic, exhilarant, cardiotonic, antiseptic, nervine tonic and expectorant properties. It is used in skin, cardiac, liver, gastrointestinal, respiratory, integument and urogenital disorders. These uses are supported and proven by many in vitro or in vivo studies. The proven pharmacological activities of S. album are antimicrobial, anti-oxidant, anti-inflammatory, antimutagenic and anti-fatigue. The research has proven that sandal oil or its constituents have anti-microbial activity. Sandalwood oil showed skin cancer preventive effect in mice and its constituent alpha santalol showed the anticancer property. The methanolic extract of wood was confirmed for antioxidant, free radical scavenging, analgesic and anti-inflammatory activities. ${\alpha}$ and ${\beta}$ santalols present in sandal oil showed sedative effects. Sandalwood tea had a significant effect on heart muscles of frog and showed increased myocardial contractility. Its oil showed significant changes in hepatic xenobiotic metabolizing enzymes. Sandalwood oil and its major constituents showed less acute oral and dermal toxicity in laboratory animals. Hence, the aforementioned studies justify the uses of sandalwood and its oil mentioned in the classical Unani literature. However, further clinical trials are suggested to confirm its efficacy and safety in humans.

백단향추출물의 항산화, 항혈소판 응집 및 혈전 용해능에 관한 연구 (Preventive Effects of Santalum album L. Extracts on Oxidation, Platelet Aggregation and Thrombosis)

  • Objectives : To evaluate the in vitro scavenging activity, inhibitory effect of LDL oxidation of pro-oxidant reactive species, anti-platelet aggregative effects and anti-thrombosis effects in response to treatment with SA using various screening methods including biological and non-biological oxidants. Methods : The antioxidant activity concerning extract from SA was studied with in vitro methods by measuring the antioxidant activity by TEAC, measuring the scavenging effects on reactive oxygen species (ROS) [superoxide anion, hydroxyl radical] and on reactive nitrogen species (RNS) [nitric oxide and peroxynitrite] as well as measuring the inhibitory effect on $Cu^{2+}$-induced human LDL oxidation and the inhibitory effect on thrombin-induced platelet aggregation and thrombosis. Results : SA extracts were found to have a potent scavenging activity regarding oxidative stress as well as an inhibitory effect towards LDL oxidation, platelet aggregation, and thrombosis. Conclusions : The SA extracts have anti-oxidative and anti-atherosclerotic effects in vitro system, which can be used for developing pharmaceutical drugs against oxidative stress and atherosclerosis.

Acanthopanax sessiliflorus stem confers increased resistance to environmental stresses and lifespan extension in Caenorhabditis elegans

  • BACKGROUND/OBJECTIVES: Acanthopanax sessiliflorus is a native Korean plant and used as a traditional medicine or an ingredient in many Korean foods. The free radical theory of aging suggests that cellular oxidative stress caused by free radicals is the main cause of aging. Free radicals can be removed by cellular anti-oxidants. MATERIALS/METHODS: Here, we examined the anti-oxidant activity of Acanthopanax sessiliflorus extract both in vitro and in vivo. Survival of nematode C. elegans under stress conditions was also compared between control and Acanthopanax sessiliflorus extract-treated groups. Then, anti-aging effect of Acanthopanax sessiliflorus extract was monitored in C. elegans. RESULTS: Stem extract significantly reduced oxidative DNA damage in lymphocyte, which was not observed by leaves or root extract. Survival of C. elegans under oxidative-stress conditions was significantly enhanced by Acanthopanax sessiliflorus stem extract. In addition, Acanthopanax sessiliflorus stem increased resistance to other environmental stresses, including heat shock and ultraviolet irradiation. Treatment with Acanthopanax sessiliflorus stem extract significantly extended both mean and maximum lifespan in C. elegans. However, fertility was not affected by Acanthopanax sessiliflorus stem. CONCLUSION: Different parts of Acanthopanax sessiliflorus have different bioactivities and stem extract have strong anti-oxidant activity in both rat lymphocytes and C. elegans, and conferred a longevity phenotype without reduced reproduction in C. elegans, which provides conclusive evidence to support the free radical theory of aging.

흡연 시 인삼, 쑥, 솔잎 추출물이 폐 세포의 구조와 항산화 효소에 미치는 영향 (The Effects of Extracts from Ginseng, Wormwood and Pine needle in Pulmonary Structure and Anti-oxidant Enzyme in Smoking)

  • 본 실험에서 메탄올을 추출 용매로 한 인삼, 쑥, 그리고 솔잎 추출물을 수동분무기를 이용하여 호흡을 통해 직접 실험동물에게 투여하였을 경우 그 효과가 폐의 구조를 유지하는 데 도움을 줄뿐만 아니라 항산화 효소에도 길항적인 효과를 나타냄을 알 수 있었다. 즉, 추출물을 흡입시킴으로서 세포막의 지질과산화를 억제하여 세포의 구조와 기능을 유지함으로써 흡연으로 인한 세포의 파괴 및 불규칙적인 구조 이상을 줄일 뿐만 아니라 추출물에 포함되어 있는 인삼의 saponin, 쑥의 sesquiterpene과 cineol, 솔잎의 rutin 등의 항산화제의 작용에 의해 흡연으로 생긴 유해 라디칼의 분해를 촉진시키거나 항산화 효소의 활성을 증가시킴으로써 폐의 정상적인 기능에 도움을 주는 것으로 생각된다. 특히, 지금까지 연구되어진 in vitro나 in vivo에서의 경구투여나 주사를 통한 방법이 아니라 호흡을 통해 직접 폐에 투입함으로써 그 결과가 빠르게 나타날 수 있을 것으로 생각된다.

Anti-Asthmatic Effects of Portulaca Oleracea and its Constituents, a Review

  • Objectives: The medicinal plants are believed to enhance the natural resistance of the body to infections. Some of the main constituents of the plant and derived materials such as, proteins, lectins and polysaccharides have anti-inflammatory effects. Portulaca oleracea (P. oleracea) were used traditionally for dietary, food additive, spice and various medicinal purposes. This review article is focus on the anti-asthmatic effects of P. oleracea and its constituents. Methods: Various databases, such as the PubMed, Scopus, and Google Scholar, were searched the keywords including "Portulaca oleracea", "Quercetin", "Anti-inflammatory", "Antioxidant", "Cytokines", "Smooth muscle ", and " Relaxant effects " until the end of Jul 2018. Results: P. oleracea extracts and its constituents increased $IFN-{\gamma}$, IL-2, $IFN{\gamma}/IL-4$ and IL- 10/IL-4 ratio, but decreased secretion of $TNF-{\alpha}$, IL-4 and chemokines in both in vitro and in vivo studies. P. oleracea extracts and quercetin also signifcantly decreased production of NO, stimulated ${\beta}$-adrenoceptor and/or blocking muscarinic receptors in tracheal smooth muscles. Conclusion: P. oleracea extracts and quercetin showed relatively potent anti-asthmatic effects due to decreased production of NO, inflammatory cytokines and chemokines, reduced oxidant while enhanced antioxidant markers, and also showed potent relaxant effects on tracheal smooth muscles via stimulatory on ${\beta}$-adrenoceptor or/and blocking muscarinic receptors.

In vitro anti oxidant activity of methanol extract of Clerodendrum infortunatum Linn

  • The antioxidant potency of methanolic extract Clerodendrum infortunatum Linn. (MECI), which are widely used in the Indian indigenous system of medicine for different purposes, was studied. The antioxidant potential was evaluated using different established in vitro antioxidant tests viz. determination of total amount of polyphenolics compounds, DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging, nitric oxide scavenging, superoxide anion radical scavenging, hydroxyl radical scavenging and reductive power assay. It was found that MECI contain a high amount of polyphenolics and possesses significant free radical scavenging activity in all the assay. The higher activity was may be due to presence of richest amount of polyphenolics and flavonoids in it.
