흑삼릉의 수치에 따른 Aldose Reductase 및 항산화활성에 미치는 영향 (Effects of Sparganii Rhizoma Processed on Rat Lens Aldose Reductase and Anti-oxidant Activities)

  • Sparganium stoloniferum Buch.-Ham. (Sparganiaceae) is a perennial herb which has been used as treatments for menstrual disorder, galactagogue and endometriosis in folk remedies. The Sparganii Rhizoma processed, heated with vinegar, used to different purpose compared with origin in medicine. In order to estimate the different effects between processed (PSR) and unprocessed (USR) Sparganii Rhizoma, extraction and systemic fractionation of the rhizomes were archived. Inhibitory effects of the extracts as well as the fractions of PSR and USR on rat lens aldose reductase and on rat platelet aggregation were investigated for the prevention or the treatment of chronic diabetic complications. Their antioxidant effects, measured using lipid peroxidation of liver tissue and radical scavenging activities on 1,1-diphenyl-picrylhydrazyl in vitro, were evaluated. The most of biological activities tested except rat platelet aggregation, fractions from PSR were shown to exhibit stronger activities than those from USR. It seems that caused by changing of the chemical components by heating process in conditioned with acetic acid. Compounds isolated were shown to significant inhibitory activity on rat lens aldose reductase. Inhibitory activity of compounds isolated on rat lens aldose reductase have been tested in vitro. $IC_{50}$/ (6.31 $\mu$M) of cerebroside I (V) was nearly equal to that (6.32 $\mu$M) of a reference compound, tetrametbylene glutaric acid (TMG).

제주도 자생식물들에 대한 미백 및 항산화 효능 탐색 (Preliminary Screening of Some Jeju Island Native Plants for Whitening and Antioxidant Activity)

  • 본 연구에서는 제주도에서 자생하는 37 종의 식물추출물에 대한 미백 및 항산화 효능을 조사하였다. 미백 효능은 in vitro tyrosinase 저해활성과 B16-F1 세포를 이용한 멜라닌 합성 저해효과를 조사하여 확인하였고, 항산화 효능은 DPPH (1,1-diphenyl-2-picrylhydrazyl)를 이용한 전자공여능 측정방법과 DCF-DA (dichlorofluorescin diacetate)를 이용한 V79-4 폐 섬유아세포에서 활성 산소종 소거작용 능력을 조사하여 측정하였다. 세포실험 조건에서 식물추출물들의 투여농도에 따른 세포독성 정도를 조사하기 위하여 MTT (3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide) assay를 수행하였다. 미백 효능조사 실험에서 우수한 결과를 나타내었던 8종의 식물 추출물들은 토끼를 이용한 국소 독성 시험 및 인체 피부 자극 임상 시험에 적용하여 안전성을 조사하였다. 연구결과에서 높은 미백과 항산화 효능을 동시에 나타내었던 비수리(Lespedeza cuneata), 제주광나무(Ligustrum lucidum (stem)), 산뽕나무(Morus bombycis (stem)), 꿀풀(Prunella vulgaris var. lilacina)의 식물추출물들은 화장품 원료화에 매우 유리한 조건을 갖고 있음을 확인하였다.

  • Peroxiredoxin(PRX)은 원핵세포에서 진핵세포에 이르기까지 세포 내부적으로 발생된 과산화물로부터 자신을 보호하는 중요한 항산화단백질이다. 포유류에서는 아직까지 여섯 개의 다른 동형체가 밝혀졌으며, 조류에서는 아직 발표된 바가 없다. 이 실험을 통해 최초로 조류의 PRX 단백질군들의 특성을 분석하였다. 생물정보분석기법을 통해 알아본 결과, 조류에서는 최소한 진화적으로 보존된 4개의 다른 PRX 단백질로 구성됨을 알 수 있다. 또한 닭의 PRXs로 in vitro 실험을 진행한 결과, 포유류의 것과 비슷한 항산화 활성을 나타냄을 알 수 있었다. 닭의 PRX는 조직 비특이적으로 발현하였으며, 이는 항산화 물질의 피해로부터 모든 조직을 보호하기 위한 필수적 요소이기 때문일 것으로 추정된다. 결론적으로, 생물정보분석기법을 통하여 추정할 수 있는 닭의 기능성 유전자군을 효과적으로 찾을 수 있고, in vitro 실험을 통하여 그 기능을 확인할 수 있었다.

하늘타리(Trichosanthes kirilowii Maxim.) 부위별 추출물의 항산화 및 항염증 활성 (Anti-Oxidant and Anti-Inflammatory Activities of Various Organ Extracts from Trichosanthes kirilowii Maxim.)

  • In this study, antioxidant and anti-inflammatory activities of the extract of different organs such as seed, fruit and root from Trichosanthes kirilowii Maxim. (TKM) were investigated in vitro. Among organs of TKM, the methanol extract of seed showed weak 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities with $10.4{\pm}0.1%$ and $25.5{\pm}1.1%$ at $50{\mu}g/mL$ and $100{\mu}g/mL$, respectively, and strong 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulphonicacid) (ABTS) radical scavenging activities with $72.4{\pm}6.3%$ and $96.4{\pm}2.3%$ at $50{\mu}g/mL$ and $100{\mu}g/mL$, respectively. Anti-inflammatory effects of the extracts were investigated by measuring nitric oxide (NO) production, mRNA expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in LPS-induced Raw 264.7 macrophage cells. The methanol extracts of seed and fruit at $50{\mu}g/mL$ potently suppressed LPS-stimulated production of NO and reduced the expression of iNOS mRNA. The methanol extracts of seed and fruit also reduced the expression of COX-2 mRNA remarkably. Therefore, seed and fruit of TKM may be utilized for a wide range of health benefits associated with antioxidant and anti-inflammatory activities.

감초 육성품종 뿌리 추출물의 In vitro 활성 평가 (Evaluation of the In vitro Activity of Glycyrrhiza Cultivar Roots)

  • Background: Glycyrrhiza radix (licorice root) have been used as an oriental medicine material for long time, and its protective effects on oxidative stress, inflammation and cognition deficit have been recently reported. However, the cultivation of Glycyrrhiza species as medicinal crops is associated with some problems such as low productivity and early leaf fall, etc. To resolve this problems, Glycyrrhiza cultivars have been developed by direct hybridization of each Glycyrrhiza species by Korean researchers. The present study was conducted to compare the Glycyrrhiza cultivar radix (Dagam, Sinwongam and Wongam) for their anti-oxidation, anti-inflammation, and cognition improvement effects and levels of liquiritin, isoliquiritigenin and licochalcone in order to select an excellent cultivar as a material resource. Methods and Results: For evaluating the inhibitory efficacies of the Glycyrrhiza cultivar extracts on oxidative stress and inflammation in BV2 cells, we measured their reactive oxygen species (ROS) production and nitric oxide (NO) release after treating them with lipopolysccharide. The scavenging activities on 2,2-diphenyl-1-picrylhydrazyl (DPPH) and peroxynitrite ($NOO^-$) radicals were evaluated. Cell proliferation and N-methyl-D-aspartate receptor (NMDAR) inhibition were analyzed. The total phenol, liquiritin, isoliquiritigenin and licochalcone A content in the extracts of the three culivars were quantified. Furthermore, the correlation coefficient between the activities and contents of total phenol, liquiritin, isoliquiritigenin and licochalcone A were also calculated. The results indicated that Sinwongam exhibited potent anti-oxidant, anti-inflammatory and NMDAR inhibititory activities. Sinwongam also showed higher total phenol and licochalcone A contents than the other cultivars. Among the three cultivars, Dagam exhibited a positive effects on NO release inhibition, cell proliferation and contents of liquiritin and isoliquiritigenin. Conclusions: Sinwongam is expected to be the most useful resource as a functional material for anti-oxidation/anti-inflammation and cognition improvement among the three studied licorice cultivars.

치마버섯을 이용한 진피 발효 배양물의 항산화 및 항염 효과 (Anti-Oxidant Effect and Anti-Inflammatory of Fermented Citrus unshiu Peel Extract by using Schizophyllum commune)

  • 감귤은 국내 및 동남아시아 지역에서 많이 즐기는 식품 중 하나이며, 이러한 감귤류의 껍질을 건조시킨 것을 진피라고 한다. 이러한 진피는 한방에서는 이뇨작용, 비장기능을 강화하는 것으로 알려져 있으나, 천연 플라보노이드를 다량 함유하고 있어 학계에서 여러 분야로 연구가 되고 있는 실정이다. 본 연구에서는 플라보노이드 배당체를 함유한 진피 추출물을 ${\beta}$-glucosidase 효소를 이용하여 아글리콘(aglycone)으로 변환 할 수 있도록 치마버섯 배양을 실시하였다. 진피 발효 배양액을 HPLC를 이용하여 분석하였으며, UVA에 의한 광손상 보호능을 섬유아세포를 이용하여 측정하였다. 진피 발효 배양액은 COX-2, LOX-5와 같은 항염증 관련 대사에도 관여하여 염증완화에도 도움을 줄 수 있음을 확인하였다. 또한 각질 형성세포를 이용한 interleukin-$1{\alpha}$를 측정한 결과 진피발효배양액 처리군의 저해활성이 더 높았다. 이러한 결과로 미루어 볼 때, 진피발효 배양액은 피부에 대한 항염 및 항산화 효과가 우수한 것으로 사료된다.

  • Nypa fruticans wurmb (NF) has been used as traditional medicinal food in Asian countries. Especially, NF has been used for conventional medicine to treat inflammatory periodontal diseases. Previous studies have been shown that NF has large amount of useful constituents such as phenolic acids, polyphenols and flavonoids. Also, NF is known as having medicinal effects such as anti-oxidant, anti-inflammatory and cholesterol-lowering effects. NF has recently been attracted to use complementary medicinal food on inflammatory diseases in Korea. However, there are no obvious effects in inflammatory and metabolic diseases also mechanisms has been studied yet. The purpose of this study was to investigate the anti-inflammatory effects of NF and steamed-NF (SNF), which recently has been used as health food, using Human keratinocyte cell line (HaCaT) and Human mast cell line (HMC-1). The cytotoxicities of NF and SNF were measured by using MTT assays in HaCaT cells and HMC-1 cell. To evaluate anti-inflammatory effects of NF and SNF, HaCaT cells were stimulated with tumor necrosis factor $(TNF)-{\alpha}$ and Interferon $(IFN)-{\gamma}$. Also, HMC-1 cells were stimulated with phorbol-12-myristate-13-acetate (PMA) and A23187 calcium ionophore (A23187) to induce allergic inflammation. Inflammatory cytokine were measured by enzyme-linked immunosorbent assay (ELISA). In this result, the extract of NF and SNF (0.01 - 1mg/ml) did not show cytotoxicity in HaCaT cells and HMC-1 cells. In addition, the NF and SNF suppressed the production of interleukin (IL)-6 and IL-8 in HaCaT cells at highest concentration. Furthermore, the treatment of SNF significantly inhibited the secretion level of IL-8 in PMA plus A23187-stimulated HMC-1 cells compared with NF treatment group. These results suggest that the extract of NF and SNF may serve as a potential therapy for skin inflammatory diseases.

안지오텐신 전환효소활성 저해에 의해 항고혈압 효과를 갖는 신품종 감자 구이벨리 추출물 (Anti-hypertensive Activity of New Potato(Solanum tuberosum) Variety of Gui Valley Via Inhibition of Angiotensin Converting Enzyme)

  • This study was attempted to investigate the anti-hypertensive activity of Solanum tuberosum, cv. Gui valley. Ethanol extract of Gui vally (EGV) increased free radical scarvenge activity up to 91.6% of control at $200{\mu}g/m{\ell}$. It's anti-oxidant activity is similar to 10 uM of ascorbic acid, well known as antioxidant. EGV inhibited Angiotensin-I-converting enzyme (ACE) activity in vitro. ACE plays a important role in regulation of blood pressure and ACE inhibitors are important for the treatment of hypertension. Anti-hypertensive activity of EGV in vivo was assessed in lead acetate (LAT)-induced hypertensive rats for 8 weeks. Elevated blood pressure in control group was significantly decreased by EGV at 200 mg/kg. Also, ACE activity in blood was also suppressed by EGV treatment. Taken together, these results suggest that EGV has an anti-hypersive activity via inhibition of ACE and can be used for the treatment or prevention of hypertension.

HepG2 cell에서 quercetin의 HO-1 발현을 통한 mitochondria의 생합성 유도 효과에 관한 연구 (Quercetin Induces Mitochondrial Biogenesis via HO-1 Expression in HepG2 Cell)

  • Flavonoids show diverse bioactivities, such as anti-oxidant, anti-cancer, anti-allergic, anti-inflammatory, and anti-viral. Quercetin is one of the flavonoids present in a wide range of plants, especially onions and consumed all over the world. Recently, it is known that quercetin induces mitochondrial biogenesis in vivo and in vitro. However, detail mechanism of these actions remains unknown. We investigated quercetin's effects on mitochondrial biogenesis in HepG2 cells, and determined the mechanisms involved. We found that quercetin treatment induced the expression of mitochondrial biogenesis activators, $PGC-1{\alpha}$, NRF-1, TFAM, and mitochondrial proteins, cytochorome c and complex IV (COXIV). Moreover, amount of mitochondrial DNA was also increased by quercetin. Quercetin has been known to induce heme oxygenase (HO)-1 in several types of cells. Here, we found quercetin induces HO-1, and inhibition of HO-1 or CO, which is product of HO-1, decreased quercetin-induced mitochondrial biogenesis such as induction of $PGC-1{\alpha}$, NRF-1, TFAM, cytochorome c, COXIV, and mitochondrial DNA. These findings imply that quercetin can increase mitochondrial biogenesis via HO-1/CO system. High glucose results in dysfunction of mitochondria biogenesis. In the present study, 25 mM glucose decreased mitochondrial biogenesis and this damage was restored by quercetin. Conversely, inhibition of HO-1 or CO inhibited quercetin-induced mitochondrial biogenesis rescue. These results suggest that quercetin enhances mitochondrial biogenesis via HO-1/CO system and hence, can rescue mitochondria from damage by high glucose.
