금앵근(金櫻根)이 Monosodium Urate로 유발된 백서(白鼠)의 통풍(痛風)에 미치는 영향 (Effects of Radix Rosae Laevigatae on the Monosodium Urate(MSU)-induced Gout Model in Rats)

  • The aim was to identify the inhibitory effects of Radix Rosae Laevigatae(RRL) on Monosodium Urate(MSU)-induced gout in rats. After pretreatment of RRL I (125mg/kg), RRL II (50mg/kg) for seven days followed by injection of MSU solution, the various indicators related to gout were measured, such as hematological and serum levels and including joint inflammation. Also, it was investigated whether RRL directly inhibits the activity of xanthine oxidase in vitro. As a result of this study, RRL didn't show the cytotoxicity on cell proliferation, but it showed significant inhibition of activity of xanthine oxidase in vitro. RRL slightly inhibited joint inflammation induced by MSU though not with statistical significance. RRL partially decreased MSU-induced BUN, creatinine, WBC, ESR elevation and significantly decreased MSU-induced AST, ALT, uric acid in serum. These results suggest that RRL has therapeutic effects that are applicable to prevention and treatment of gout, and should be further investigated.

패장약침(敗醬藥鍼)의 암전이 억제 및 면역 조절 효과에 관한 실험적 연구 (Experimental Studies on Antimetastatic and Immunomodulating Effects of Patriniae Radix Herbal-acupuncture)

  • Objectives : This study was designed to investigate the antimetastatic and immunomodulating effects of Patriniae Radix. Methods : Acute and subacute cytotoxicity experiment of Patriniae Radix was performed. Antimetastatic experiment was administered in vitro and in vivo. To observe the immunomodulating effects of Patriniae Radix, FACS analysis and ELISA assay were performed. Results : There was no acute and subacute toxicity responses in mouse treated with Patriniae Radix. Antimetastatic experiment in vitro and in vivo showed that Patriniae Radix has antimetastatic effects. This research revealed that Patriniae Radix mediate cellular immunity response. As compared with control, the population of total T cell, helper T cell, cytotoxic T cell and macrophage were increased. The production of Th 1 type cytokines from splenocyte and cytokines which is associated with anti-tumor activity form macrophage were increased significantly. Conclusion Patriniae Radix Herbal-acupuncture appears to have considerable activity on the treatment of liver metastasis from colon26-L5 carcinoma cell line, and deserves further evaluation in this setting.

Newly Designed Six-membered Azasugar-Containing Phosphorothioate Oligonucleotide as a Potent AIDS Therapeutic Drug

  • A series of modified oligonucleotides containing a phosphorothioate (P=S) backbone and a six-membered azasugar (6-AZS) as a sugar substitute in a nucleotide were synthesized and tested for their ability to inhibit the human immunodeficiency virus type I(HIV-l) in vitro without the aid of any transfecting agents. While P=S oligonucleotides with natural nucleotides had little anti-HIV-l activity, the six-membered azasugar nucleotide (6-AZN)-containing P=S oligonucleotides (AZPSONs) potently inhibited the HIV-l/SHIV replication and syncytium formation (ECso = 0.02-0.2 /lM) without cytotoxicity up to 100 /lM. DBM-2198, the most effective in anti-HIV-l activity among the AZPSONs, consists of random sequence and five 6¬AZNs evenly distributed in 18 nucleotides. DBM-2198 showed strong antiviral activity against, not only laboratory strains, but also primary isolates and even drug-resistant strains of HIV-I. DBM-2198 was much more effective than ddI or ddC in its anti-HIV-l activity in vitro. Particularly noteworthy is that the anti-HIV-l activity of DBM-2198 was better than that of AZT with respect to its long-lasting efficacy after a single treatment. Nevertheless, the antiviral activity of the AZPSONs was very specific to HIV-I. Poliovirus, or even simian immunodeficiency virus (SIV), was not inhibited by the AZPSONs. Taken together, our results strongly suggest that AZPSON can be used as a safe and effective AIDS-therapeutic drug against a broad spectrum of HIV -1 strains.

비브리오속 균주들에서외 세포의 효소의 분포 (Distribution of Extracellular Proteases from Various Vibrio Species)

  • The members of the genus Vibrio include harmless aquatic strain as well as strains capable of causing infections in human and fish. Pathogenic mechanisms are only understood for Vibrio cholerae O1 and O139 and not for the majority of Vibrio species. Twelve clinical and nonclinical strains were examined by in vitro and in vivo experiments for the importance of extracellular enzymes as a virulence determinant of Vibrio species. In vivo cytotoxicity assay was performed by injecting approximately $10^{8}$ cells/mL into mice (BALB/c). V. harvyi and V. vulnificus showed 100% lethality within 3hr after bacterial injection. V. fluvialis and four strains of V. parahaemolyticus showed 50% lethality within 4hr. V. mimicus, V. alginolyticus and V. furnissii revealed 30% lethality within 9hr. Nonclinical strains, V. campbellii and V. ordalii, did not show any lethality. In vitro protease and hemolytic activities were also good indicators for clinical and nonclinical strains of Vibrio species. The clinical strains showed much higher activities than nonclinical strains. The activity of some clinical strains of re-isolates was evidently increased. Most clinical strains had $\beta$ hemolytic activity. The results demonstrate that the prevalent distribution of extracellular proteases in pathogenic Vibrio sp. implies their importance as a virulence determinant.

Isolation and Identification of Taxol, an Anticancer Drug from Phyllosticta melochiae Yates, an Endophytic Fungus of Melochia corchorifolia L.

  • Phyllosticta melochiae, an endophytic fungus isolated from the healthy leaves of Melochia corchorifolia, was screened for the production of an anticancer drug, taxol on modified liquid medium and potato dextrose broth medium in culture for the first time. The presence of taxol was confirmed by spectroscopic and chromatographic methods of analysis. The amount of taxol produced by this fungus was quantified by high performance liquid chromatography. The maximum amount of fungal taxol production was recorded as $274{\mu}g/L$. The production rate was increased to $5.5{\times}1,000$ fold than that found in the culture broth of earlier reported fungus, Taxomyces andreanae. The fungal taxol extracted also showed a strong cytotoxic activity in the in vitro culture of tested human cancer cells by apoptotic assay. The results designate that the fungal endophyte, P. melochiae is an excellent candidate for an alternate source of taxol supply and can serve as a potential species for genetic engineering to enhance the production of taxol to a higher level.

  • 인체 면역증강제로 활용 가능성을 조사하기 위해 R. aquatilis AY2000 균이 생산하는 당단백질인 AYS의 면역세포에 대한 독성과 면역증강 효과를 조사하였다. 그 결과 AYS는 적혈구에 대한 용혈작용과 hPBMC에 대한 증식 또는 사멸효과는 나타내지 않았으나, 배양 중인 hPBMC를 응집시키는 효과를 나타내었다. 또한 AYS는 in vitro에서 hPBMC에 작용하여 염증성 cytokine인 IL-6, IFN-${\gamma}$, TNF-${\alpha}$와 IL-5 생성을 유도하였고, 생쥐에서 AYS는 alum에 비해 항-BSA의 생산력을 더욱 증가시켰다.

  • 피부 자극과 부식 검사에 사용되는 동물실험을 대체하기 위한 방법으로 인공피부가 개발되어 왔다. 최근에 본 연구진은 녹각교를 함유하는 새로운 인공피부를 구축하였다. 현재 연구에서는 녹각교를 포함하는 인공피부를 사용하여 물질의 독성도 검사를 수행하였다. 그리하여 sodium dodecylsulfate (SDS) 또는 sodium carbonate를 인공피부에 도포하였고 표피의 손상 정도를 H&E와 면역조직화학 염색을 통하여 평가하였다. 인공피부의 표피는 SDS와 sodium carbonate에 의해 농도 의존적으로 영향을 받았다. 더 나아가 이들 물질에 의하여 p63의 발현이 감소하였다. 그러므로 녹각교를 함유하는 인공피부는 동물실험을 대체할 수 있는 모델로써 사용될 수 있고 in vitro에서 자극과 부식 검사 시험법의 발전에 도움을 줄 것으로 예상된다.

초미세먼지(PM2.5)로 유도된 in vitro 세포 독성에 대한 감태(Ecklonia cava) 추출물의 보호 효과 (Cellular protective effect of Ecklonia cava extract on ultra-fine dust (PM2.5)-induced cytoxicity)

  • 감태(Ecklonia cava) 추출물의 in vitro 항산화효과 및 초미세먼지로 유도된 세포 독성에 대한 비강(RPMI-2650), 폐(A549) 및 뇌신경(MC-IXC) 세포에서의 세포보호효과를 검증하였다. 감태는 총 페놀성화합물(물 추출물: 189.32, 80% 에탄올 추출물: 418.08 mg GAE/g of dried weight) 및 총 플라보노이드(물 추출물: 44.93, 80% 에탄올추출물 111.22 mg RE/g) 함유량을 각각 나타내었으며, ABTS/DPPH 라디칼 소거활성 및 지방질과산화 억제효과에서도 우수한 효과를 나타냈다. 또한, 감태 추출물은 초미세먼지로 유도될 수 있는 세포 내 산화적스트레스를 저해함으로써 세포 사멸을 효과적으로 억제시킬 수 있는 천연 소재로 판단되며, 80% 에탄올 추출물뿐만 아니라 물 추출물 또한 산업적 활용가능성이 기대된다.
