This study was conducted to investigate the effects on fermentation characteristics of rumen microorganism by different types and levels of lignosulfonate treated soybean meal (LSBM) in in vitro test and rumen simulation continuous culture (RSCC) system in dairy cows. The experiment I was control and 12 treatments (each with 3 replications) in vitro test to demonstrate composition of different types of treatments with lignosulfonate (Desulfonate, Na, Ca and solution) and levels (2, 4 and 8%) of soybean meal in the dairy cow diet. LSBM source treatments in the dairy cow diet showed pH value, $NH_3$-N concentration and total VFA concentration lower than control at all levels and incubation times (p<0.05). Dry matter digestibility of LSBM source treatments showed lower than control (p<0.05). Gas production and rumen microbial synthesis was decreased by rumen microbial fermentation for incubation times. Undegradable protein (UDP) concentration of all LSBM treatments was decreased for incubation times, and significantly higher than control (p<0.05). In the experiment II compared diets of the control, LSBM Na 2%, LSBM Sol 2%, which are high performance to undegradable protein (UDP) concentration experiment I in vitro test, and heated treatment lignosulfonate (LSBM Heat) 2% in the dairy cow diet from four station RSCC system ($4{\times}4$ Latin square). A rumen microbial fermentation characteristic was stability during 12~15 days of experimental period in all treatments. The pH value of LSBM treatments was higher than control treatment (p<0.05). The $NH_3$-N concentration, VFA concentration and rumen microbial synthesis of LSBM treatments were lower than control (p<0.05). The undegradable protein (UDP) showed LSBM Na 2% (45.28%), LSBM Sol 2% (43.52%) and LSBM Heat 2% (43.49%) higher than control (41.55%), respectively (p<0.05). Those experiments were designed to improve by-pass protein of diet and milk protein in the dairy cows. We will conduct those experiments the in vivo test by LSBM treatments in dairy cows diet.
In order to determine the effect of fermentation by the mycelia of fungal species, Formitella fraxinea and Sarcodon aspratus, on the in vitro dry matter digestibility and pH of mixtures with sawdust plus 20% wheat bran w/w, on dry matter basis to use as a feedstuff or an additive including fungal mycelium into a feedstuff. The mixtures were unfermented (UF) and fermented by Formitella fraxinea(FF) and Sarcodon aspratus(SA) for two weeks at $29^{\circ}C$ in a incubator. Fungal fermentation products were added to the basal diet to the level of 0, 1, 3 and 5%, w/w of diets each. The in vitro dry matter digestibilities, soluble sugar contents and pH of fermentation fluids were measured at 24, 48 and 72hr after fermentation begin. Neutral detergent fiber(NDF) contents in mixtures were lower for SA and UF(80.4 and 82.2%) than for FF(88.3%) (P<0.05). In vitro DM digestibility for 48h was higer for SA(21.2%) than for UF and FF(17.9 and 12.2%). The in vitro dry matter digestibility for 24hr was higher for diets added with FF 1% as 49.18%, and lower for diet added with FF 5%(43.07%) than basal diet(44.98%)(P<0.05), and tended to be higher for the diets added with fungal products. The pH of in vitro fermentation fluids for 24 and 48 hrs fermentation were lower for diets added with all FF and SA than for UF(P<0.05). However, those for 72 hrs fermentation were higher for SA 1%(6.74) than other diets(P<0.05). The soluble sugar concentration of in vitro fermentation fluid was not different between diets for 24 hr fermentation. However, those were higher for all additive diets than basal diet for 48 and 72 hrs fermentation(P<0.05). It could be concluded that dairy cow's diets added with fungal fermentation products have positive effects, and expected it will be more beneficial if more fungal mycelium was contained.

The objective of this study was to the effect of four medicinal plants (Rheum palmatum, Pharbitidis semen, Reynoutria japonica, Tribulus semen) supplementation on methane reduction and ruminal fermentation in in vitro batch culture method. Each medicinal plant was supplemented 5% on a substrate basis in the bottle, then filled with buffered rumen fluid. Incubation was conducted for 24 hours in a shaking incubator (39℃, 120 rpm). The ruminal pH values were not significantly different between the control and treatment groups. However, the digestibility of the feed was significantly higher in the group supplemented with medicinal plants than control group. Methane production (mL/g of digested dry matter) and total gas production (mL) was significantly lower in the treatment group compared to the control group in Tribulus semen group. Total volatile fatty acids concentration were significantly higher in all treatment groups than control group, and acetate concentration was significantly higher in all treatment groups than control group except for Rheum palmatum group. Propionate concentration was significantly higher in all treatment groups than control group, while butyrate concentration was significantly higher in Rheum palmatum group than control group. Ammonia nitrogen concentration was significantly higher in all treatment groups than control group. In conclusion, the addition of medicinal plants did not negatively impact rumen fermentation, and the results indicate that Tribulus semen has potential as a feed additive for reducing methane emissions.

The present study was conducted to investigate the effects of seaweeds on in vitro ruminal fermentation characteristics and methane gas production. Five seaweeds (Dictyota dichotoma, DD; Chrysymenia wrightii Yamada, CW; Codium fragile, CF; Sargassum fusiforme, SF; Gracilaria vermiculophylla, GV) were obtained from National Institute of Fisheries Science (NIFS) in South Korea. The ruminal fluids were collected from 3 rumen-cannulated Hanwoo steers (average 12-months-old). The buffered ruminal fluids (50 mL) were incubated with substrates (0.4 g of concentrate and 0.1 g of rice straw in dry matter basis) and seaweeds (5% of substrates) at 39℃ for 24 and 48 hours. The total gas and methane production of all treatments incubated for 24 hours were not affected by the seaweed. However, methane production (mL/g of digested dry matter) in the CW and CF treatments incubated for 48 hours was decreased compared to control (p<0.05). Additionally, the ruminal pH of all treatments incubated for 24 and 48 hours was lower than control (p<0.05). There was no signigicant difference in total VFA concentration at 24 hours of incubation, but it was higher in the CF treatment at 48 hours of incubation (p<0.05). The dry matter digestibility of all treatments incubated for 24 and 48 hours were not affected by the seaweed. In conclusion, Codium fragile reduced in vitro methane production without negative effects on rumen fermentation characteristics.
Ground rice, barley and corn were fed separately to the ruminally cannulated Hanwoo (Korean native cattle) for comparing their in situ and in vitro degradabilities, microbial growth, pH and gas production. It has been found that nearly all the dry matter (DM) and organic matter (OM) in barley and rice disappeared during 24 hr suspension in the rumen, but those in corn were only reduced by around 67%. Water soluble DM and OM fractions(‘a’), ranked from highest to lowest was corn, then rice and finally barley, but the order was reversed for content ‘b’, degradable fraction during time ‘t’. Judging by the degradation parameter of ‘b’ fraction, degradation rates per hour of DM and OM for barley were 38.3% and 37.2% respectively, significantly higher than those for rice (7.7% and 5.6%) and corn (4.1% and 1.3%). In general, results obtained from in vitro degradability of DM and OM were lower than those from in situ trials, but the ranking order of degradability was in agreement between both trials. In particular, ground rice has relatively lower in vitro microbial growth than corn or barley, but exhibited higher gas production. In addition, in vitro microbial growth of ground rice increased with up to 12 hr of incubation period, thereafter experienced a decrease with extended incubation time. pH of in vitro solution of rice decreased following 9 hr of incubation but gas production increased rapidly during the same period. From the results of DM and OM degradabilities and pH changes of in vitro solution with incubation time, it is concluded that rice represents a good source of energy for stability of rumen fermentation.
In order to investigate the effects of supplemental ionic surfactants in in vitro ruminal fermentation, N-Lauroylsarcosine sodium salt(N-LSS) and sodium dodecyl sulfate(SDS) for negative(-) ionic surfactant, and hexadecylpyridinium chloride monohydrate(HPCM) and hexadecyltrimethyl ammonium bromide(HTAB) for positive (+) ionic surfactant were supplemented by 0.05% and 0.1% into the Dehority’s artificial medium containing rice straw(1mm) as a substrate. In vitro DM digestibility, the growth of rumen mixed microbes, pH, cumulative gas production and SEM(Scanning Electron Microscopy) observation of microbial attachment on rice straw particle were investigated through the experiment composing 9 treatments (two supplemental levels of two positive ionic(+) surfactant, two supplemental levels of two negative(-) ionic surfactant) including the control. The sample collection was at 6, 12, 24, 48 and 72 h post fermentation with 3 replications per treatments. DM digestibility in treatments supplemented (+) or (-) surfactants almost stopped afterward 12 h fermentation, in vitro DM digestibility at 72 h post fermentation in the ionic surfactants was at half level of that of the control(P<0.05). Accumulative gas production in in vitro was less(P<0.05) with addition of ionic surfactants compared to the control. The amount of rumen mixed microbes recovered from in vitro incubation fluid pleateaued at 12 h post fermentation for the positive (+) ionic surfactants, but steadily increased as fermentation time elapsed for the control. Rumen microbial growth rate was significantly(P<0.05) low in the negative(-) ionic surfactant compared to the control. pH of the incubation fluid was ranged from 6.02 to 7.20, and was the highest in the negative(-) ionic surfactants, and was the lowest in the control(P<0.05). In SEM observation, rumen microbial population attached on rice straw particle was less with addition of ionic surfactants than the control. In conclusion we could not found any positive effects of negative- and positive- charged surfactants on rumunal fermentation characteristics and rumen microbial growth rates.
An in vitro study was conducted to examine the effects of the addition of activated charcoal (AC) on the ruminal fermentation characteristics, nutrient disappearance, and ruminal gas production. AC was added at the levels of 0.00, 0.25, and 0.50 % to each of the four types of diets (roughage/concentrate ratio : 8/2, 6/4, 4/6 and 2/8), respectively. Although not significant, ruminal pH tended to increase by adding AC, and as the concentrate level increased, ruminal pH decreased (P<0.05). Acetate concentration and acetate/propionate molar ratio tended to decrease in AC diets. but molar % of propionate tended to increase by the addition of AC. Ruminal degradation of dry matter, crude protein, NDF, ADF, and hemicellulose in AC diets tended to increase than in non-AC diet, however, no tendency in ruminal degradation of crude fat was observed. As the concentrate level increased, rumunal degradation of dry matter and nutrients in AC diets increased significantly(P<0.05). Ruminal gas production tended to decrease in the 0.50 % AC diets, however, it tended to increase in high roughage diets. Although there appeared some beneficial effects in adding AC to ruminant diets in this study, more works should be done with AC before we can make clear conclusion on the use of AC in the ruminant diets.

This study was conducted to determine the effects of supplementing whole oilseeds to the alcohol fermented feedstuff based on rye hay on in vitro rumen fermentation of Hanwoo steers. Experiments were arranged with four treatment groups, T1 (without any oilseeds), T2 (supplemented with cottonseed), T3 (supplemented with linseed) and T4 (supplemented with soybean). The pH decreased at 6 and 12h in vitro incubation in T2 and at 3 and 12h in vitro incubation in T4 (p<0.05). However, no decrease of the pH was observed at any in vitro incubation in T3 (p>0.05). Ammonia concentration increased at 6h in vitro incubation in T2 and at 6 and 9 h in vitro incubation in T4 (p<0.05). Concentrations of acetate, butyrate and propionate increased at 3, 9 and 12h in vitro incubation in T4 and at 3h in vitro incubation in T3 (p<0.05). This study indicates that the supplementation of whole oilseeds to alcohol fermented feedstuff based on rye kay can improve the in vitro rumen fermentation.
This study was conducted to evaluate effects of halogenated compounds on in vitro rumen fermentation characteristics and methane emissions. A fistulated Holstein cow of 650 kg body weight was used as a donor of rumen fluid. Five kinds of halogenated compounds (bromochloromethane (BCM), 2-bromoethane sulfonic acid (BES), 3-bromopropanesulfonic acid (BPS), chloroform (CLF), and pyromellitic diimide (PMDI) known to inhibit methyl-coenzyme M reductase activity were added to an in vitro fermentation incubated with rumen fluid. The microbial population including bacteria, protozoa, and fungi were enumerated, and gas production including methane and fermentation characteristics were observed in vitro. The pH values ranged from 6.25 to 6.72 in all the treatments, and these showed a similar level at 48 hr. The total gas production in the treatments showed a similar pattern with C at 48 hr, whereas methane production in the treatments was lower (p<0.05) than C. Concentrations of total volatile fatty acids (VFAs) and propionic acid were higher (p<0.05) in the treatments than in C at 12 hr. Therefore, halogenated compounds (BCM, BES, BPS, CLF, and PMDI) inhibited in vitro methane emissions by inhibiting methanogens in the rumen. Further studies on safety are needed.
In order to know the effects of Garlic, Scallion, Flavonoid, Urushiol, Anthocyanidin and Bio-MOS?? on pathogenic microbes and rumen anaerobic microbes, the growth rate of pathogens (including Escherichia coli O157, Salmonella paratyphi, Listeria monocytogenes and Staphylococcus aureus) and in vitro rumen microbial growth, gas production, ammonia concentration, carboxymethylcellulase(CMCase) activity, and microbial populations were investigated.The growth of pathogens was inhibited by supplementation of 0.1% Flavonoid, Scallion or Bio-MOS?? as biological active materials. And Scallion and Flavonoid had powerful antimicrobial properties on the pathogens applied in paper disc method.Although few effects by biological active materials disappeared in rumen fermentation in vitro, CMCase activity removed with supplementation of 1% of Flavonoid which had antimicrobial property in paper disc method. Scallion, having powerful antimicrobial property on pathogens and no inhibiting on rumen fermentation, might be a source in development of natural antimicrobial agent for ruminants.
