Establishment of Measurement of Human Cytomegalovirus with in situ ELISA (in situ ELISA를 이용한 사람세포거대바이러스의 측정법 개발)

  • Infection with human cytomegalovirus (HCMV) is of considerable clinical relevance after placental transmission and in immunosuppressed patients such as transplant recipients or patients with AIDS. The rapid detection method of HCMV has been required to overcome the time-consuming methods such as classical plaque assay or other immunological methods. This study was performed to establish the in situ ELISA, in which human lung fibroblasts infected with HCMV were fixed and used directly as antigen in 96 well culture plate. Expressed HCMV antigens were detected with HCMV-specific monoclonal antibodies. This method could detect HCMV dose-dependently upto $3{\times}10^2\;pfu/ml$. Antiviral activity of ganciclovir could be assayed within the known range of effective dose. This result showed that HCMV could be quantitated by in situ ELISA. The chemical, which was selected on the basis of component analysis in natural product, was tested to have the anti-HCMV activity by in situ ELISA, and three among five samples were found to have anti-HCMV activity with the dose-dependent manner. Conclusively in situ ELISA could be useful method for quantitation of HCMV and screening antiviral activity of samples to HCMV.

  • The importance of ozone monitoring has been growing due to the polar ozone depletion and increasing tropospheric ozone concentration over many Asian countries, including South Korea. In-situ measurement of the vertical ozone structure has advantages for ozone research, but observations are not sufficient. In this study, ozonesonde measurements were performed from October to November in Yongin during the GMAP (The GEMS Map of Air Pollution) 2021 campaign. The procedure for ozonesonde preparation and initial analysis of the observed ozone profile are documented. The observed ozone concentrations are in good agreement with previous studies in the troposphere, and they capture the stratospheric ozone distribution as well, including stratosphere-troposphere exchange event. These balloon-borne in situ measurements can contribute to the evaluation of remote sensing measurements such as Geostationary Environment Monitoring Spectrometer (GEMS). This document focuses on providing essential information of ozonesonde preparation and measurement for domestic researchers.

  • Polymer electrolyte membranes (PEMs) are key components to determine electrochemical fuel cell performances, in addition to electrode materials. The PEMs need to satisfy selective transport behaviors to small molecules including gases and protons; the PEMs have to transport protons as fast as possible, while they should act as hydrogen barriers, since the permeated gas induces the thermal degradation of cathode catalyst, resulting in rapid electrochemical reduction. To date, limited tools have been used to measure how fast hydrogen gas permeates through PEMs (e.g., Constant volume/variable Pressure (time-lag) method). However, most of the measurements are conducted under vacuum where PEMs are fully dried. Otherwise, the obtained hydrogen permeance is easily changeable, which causes the measurement errors to be large. In this study, hydrogen permeation properties through Nafion212 used as a standard PEM are evaluated using an in-situ measurement system in which both temperature and humidity are controlled at the same time.

  • Complex modal testing is employed for the in-situ parameter identification of a four-axis active magnetic bearing system while the system is in operation. In the test, magnetic bearings are used as exciters as well as actuators for feedback control. The experimental results show that the directional frequency response function, which is properly defined in the complex domain, is a powerful tool for identification of bearing as well as modal parameters. It is also shown that the position and current stiffnesses can be accurately estimated using the relations between the measured forces, displacements, and currents.

  • A series of column experiments was conducted to develop a monitoring system for in-situ and realtime measurement of ozone transport in unsaturated porous media using a fiber optic sensor. The calibration of the fiber optic transflection dip probe (FOTDP) system was successfully carried out at various ozone concentrations using a column with length of 30 cm and diameter of 5 cm packed with glass beads, which don't react with gaseous ozone. The breakthrough curves (BTCs) of ozone were obtained by converting the normalized intensity into ozone concentration. The FOTDP system worked well for in-situ monitoring of gas phase ozone at various water saturations and in presence of soil organic matter (SOM). However, the FOTDP system did not measure the ozone concentration at more than 70% water saturation.

  • An in-situ stress measurement interferometer is constructed and used to measure the intrinsic stress in Ti$O_2$ thin films during their growth by ion-assisted deposition. It is found that the stress increases with the momentum transferred by the ion beam to the growing film and is fairly well agreed with Windischmann's model. The variation of the stress with thickness is qualitatively explained in terms of the balance between the compressive stress produced by the ion beam and the surface diffusion determined by the surface temperature.

  • In-situ stress measurement using AE (Acoustic Emission) and DRA (Deformation Rate Analysis) is usually carried out under uniaxial loading in the laboratory and it consumes delay time from drilling to testing. Therefore, it should be considered how the lateral stress and delay time influence on the test results for the in-situ stress determination. As the delay time increased, the accuracy of estimating the pre-stress decreased. The pre-stress of the specimen loaded only axially was determined within an error of less than 9% (using AE) and 4% (using DRA). And the specimen on which axial pre-stress and the confining pressure were loaded had an error of less than 17% (using AE) and 14% (using DRA). The results of AE and DRA for field specimens were very similar with each other but smaller than those of hydraulic fracturing method.

  • This paper shows the reduction effect of tile earthquake-induced liquefaction potential of soils that improved by Vibro-Replacement Stone Columns. The Vibro-Replacement Stone Columns method transforms soft cohesive soils into a composite mass of compacted granular or crushed stone columns by using vibrating equipment and water jets. This study investigated and analyzed the behavior of the stone columns and composite ground using the results of in situ test and measurement at the job-site. This paper shows the evaluation of the earthquake-induced liquefaction potential of soils using in situ test. There are different types of in situ test used in the evaluation the liquefaction potential. In the particular study the Standard penetration test, and Cone penetration test were used. The N value of Standard Penetration test has been used all over for a very long time. The evaluation of the liquefaction of soil was performed using the worldwide renewed Cone penetration test

  • Non-contacting capacitive sensors, based on principle of the cross capacitor, for measuring small displacement less than 1.95$\pm$0.5 mm have been fabricated and characterized. To overcome disadvantages of the existed capacitive sensors, the new sensor is consisted of 4-electrodes which are formed 2 active electrodes and 2 guard electrodes on a sapphire plate with diameter 17mm and thickness 0.7 mm, and are symmetrically situated with constant gap of 0.2 mm among the electrodes. The sensor is evaluated to be correlation coefficient of 0.9987 for the range of 1.95$\pm$0.5 mm and that of 0.9995 for 1.95$\pm$0.25 mm range. This sensor can be used for in situ measurements in the mechanical mirror polishing with precision less than $\pm$1${\mu}{\textrm}{m}$.
