  • 이상의 연구 결과로 미루어 볼 때, 산돌배 열매추출물은 대식세포에서 TLR2와 TLR4를 자극하여 MAPKs 신호전달을 활성화하여 NO, iNOS, IL-1𝛽, IL-6 및 TNF-α와 같은 면역증진 인자의 생성을 유도하고, 대식세포의 포식작용을 활성화시키는 것으로 판단된다. 따라서 산돌배 추출물은 대식세포의 활성화를 통해 인체의 면역시스템을 강화할 수 있으므로, 향후 면역 보조제나 면역증진을 위한 기능성 식의약품 개발을 위한 소재로 활용이 가능할 것으로 생각한다.

Immunostimulatory Activity of Hibiscus manihot Flower in Mouse Macrophages, RAW264.7 Cells

  • In this study, we investigated whether Hibiscus manihot flowers (HMF) exhibits immunostimulatory activity in RAW264.7 cells. HMF increased the production of immunostimulatory factors such as NO, iNOS, IL-1β and TNF-α in RAW264.7 cells. TLR2 and TLR4 blocked HMF-mediated production of immunostimulatory factors in RAW264.7 cells. In addition, the inhibition of MAPK signaling pathway reduced HMF-mediated production of immunostimulatory factors. From these results, HMF is thought to promote the production of immunostimulatory factors through activating TLR2/4/MAPK signaling in macrophages. It is believed that HMF can be developed as an agent related to immune enhancement in the future.

Immunostimulatory Activity of Syneilesis palmata Leaves through Macrophage Activation and Macrophage Autophagy in Mouse Macrophages, RAW264.7 Cells

  • Syneilesis palmata (SP) is a traditional medicinal plant. SP has been reported to have anti-inflammatory, anticancer, and anti-human immunodeficiency virus (HIV) activities. However, there is currently no research available on the immunostimulatory activity of SP. Therefore, in this study, we report that S. palmata leaves (SPL) activate macrophages. Increased secretion of both immunostimulatory mediators and phagocytic activity was observed in SPL-treated RAW264.7 cells. However, this effect was reversed by the inhibition of TLR2/4. In addition, inhibition of p38 decreased the secretion of immunostimulatory mediators induced by SPL, and inhibition of TLR2/4 decreased the phosphorylation of p38 induced by SPL. SPL augmented p62/SQSTM1 and LC3-II expression. The increase in protein levels of p62/SQSTM1 and LC3-II induced by SPL was decreased by the inhibition of TLR2/4. The results obtained from this study suggest that SPL activates macrophages via TLR2/4-dependent p38 activation and induces autophagy in macrophages via TLR2/4 stimulation.

Immune-Enhancing Effect and Anti-Obesity Activit of Kadsura japonica Fruits

  • Under the COVID-19 pandemic, interest in immune enhancement and anti-obesity is increasing. Thus, in this study, we investigated whether Kadsura japonica fruits (KJF) exhibits immunostimulatory activity and anti-obesity activity. KJF increased the production of immunostimulatory factors and phagocytosis in RAW264.7 cells. Inhibition of TLR2 and TLR4 blocked KJF-mediated production of immunostimulatory factors in RAW264.7 cells. In addition, the inhibition of MAPK and PI3K/AKT signaling pathway reduced KJF-mediated production of immunostimulatory factors, and the activation of MAPK and PI3K/AKT signaling pathway by KJF suppressed the inhibition of TLR2/4. KJF attenuated the lipid accumulation and the protein expression such as CEBPα, PPARγ, perilipin-1, adiponectin, and FABP4 related to the lipid accumulation in 3T3-L1 cells. In addition, KJF inhibited excessive proliferation of 3T3-L1 cells and protein expressions such as β-catenin and cyclin D1 related to cell growth. These findings indicate that KJF may have immunostimulatory activity and anti-obesity activity.

인삼다당체 진산과 유사구조 다당체의 면역증강활성작용 (Comparison between Immunostimulatory Activity and Molecular Structure of Different Polysaccharides)

  • Background: We previously reported that ginsan, a polysaccharide extracted from Panax ginseng had an immunostimulatory activity such as mitogenic activity, activation of macrophages and killer cells, and production of a variety of cytokines which resulted in antitumor and antiseptic effects. We further purified $\alpha$-(1$\longrightarrow$6)-glucan and $\beta$-(2$\longrightarrow$6)-fructan from the ginsan with size exclusion and ion-exchange column chromatography successively. In this study, we performed the structure-based activity of ginsan by comparison with known polysacchrides such as $\beta$-glucan, curdlan, laminarin, levan, dextran, lentinan and OK-432. Methods: To investigate the immunostimulatory activity of several polysaccharide compounds, we investigated the stimulation of lymphocytes proliferation, the generation of activated killer cells and the secretion of nitrites from activated macrophages. Results: Of polysaccharides tested, curdlan and ginsan stimulated lymphocyte proliferation, suggesting that the molecular weight and composition of polysaccharide are dependent on the mitogenic activity. The production of nitric oxide was significantly increased in curdlan, levan, ginsan and its fraction, indicating that fructan has also capacity to activate macrophages and may devote to kill pathogens. In addition, the activation of macrophages was seemed to be independent of molecular weight of polysaccharide. The generation of AK cells was exhibited in order of curdlan, OK-432> F1, ginsan, F3> levan> etc. The AK activity may be dependent on molecular weight and composition of polysaccharides. Conclusion: Unfortunately, purified polysaccharide from ginsan were less active on immunostimulatory activity than mixed compounds of polysaccharides. From the viewpoint of structure and activity relationships, we found several characteristic features.

In vitro Immunostimulatory Activity of Bok Choy (Brassica campestris var. chinensis) Sprouts in RAW264.7 Macrophage Cells

  • Bok choy is one of Brassica vegetables widely consumed worldwide. Brassica vegetables have been reported to exert various pharmacological activities such as antioxidant, anti-cancer and cardioprotective activity. However, studies on immunostimulatory activity of bok choy sprout have not been conducted properly. Thus, in this study, we investigated in vitro immunostimulatory activity of bok choy sprout extract (BCS) using mouse macrophage RAW264.7 cells. Our results showed that BCS increased the production of immunomodulators such as NO, iNOS, IL-1β, IL-6, IL-12, TNF-α and MCP-1, and phagocytic activity in RAW264.7 cells. BCS activated MAPK, NF-κB and PI3K/AKT signaling pathways. However, BCS-mediated production of immunomodulators was dependent on JNK, NF-κB and PI3K/AKT signaling pathways. the mRNA expression of TLR2 were significantly increased by BCS, TLR2 inhibition by anti-TLR2 dramatically suppressed the production of immunomodulators by BCS. In addition, TLR2 inhibition by anti-TLR2 significantly reduced BCS-mediated phosphorylation level of AKT, JNK and NF-κB. From these results, BCS may have immunostimulatory activity via TLR2-MAPK, NF-κB and PI3K/AKT signaling pathways. Therefore, BCS expected to be used as a potential immune-enhancing agent.

사람 면역세포를 활용한 생식 원료 및 제품의 체질 맞춤형 면역 활성 탐색 (Screening of Personalized Immunostimulatory Activities of Saengsik Materials and Products Using Human Primary Immune Cell)

  • 사상의학은 한국 고유의 전통 한의학으로 4가지 체질 즉 태양인, 태음인, 소음인, 소양인으로 분류한다. 새로운 기능성 생식제품을 개발하기 위하여 태음인(Taeumin), 소양인(Soyangin) 그리고 소음인(Soeumin)의 혈액으로부터 분리한 면역세포(림프구와 대식세포)를 이용하여 생식 원료(쌀, 보리, 율무, 콩, 팥, 차조, 가지, 무, 당근, 표고버섯, 양송이버섯, 목이버섯)와 생식제품의 면역 활성을 연구하였다. 생식원료와 생식제품 추출물들을 처리한 면역세포에서 세포 증식율, NO와 TNF-${\alpha}$의 증가를 면역 활성의 지표로 사용하였다. 쌀(milled rice), 팥(adzuki bean), 무(radish) 그리고 표고버섯(Lentinus edodes)을 태음인을 위한 생식 원료로 선택하였고, 보리(barley), 콩(soybean), 가지(eggplants) 그리고 양송이버섯(Agaricus bisporus)을 소양인을 위한 생식 원료로 선택하였으며, 율무(Job's tears), 팥(adzuki bean), 당근(carrot) 그리고 목이버섯(Auricularia auricula-judae)을 소음인을 위한 생식 원료로 선택하였다. 이러한 결과를 근거하여 생식제품 개발을 위하여 생식 원료의 종류와 배합비율을 달리하여 배합비 1~6(Mix 1~6)을 개발하였다. 태음인의 면역 활성은 배합비 1, 배합비 2, 배합비 3, 배합비 4, 배합비 5 또는 배합비 6 추출물을 처리한 면역세포 중에서 배합비 2(Mix 2; 쌀 60%, 팥 20%, 무 10%, 표고 10%) 추출물을 처리한 면역세포에서 가장 높았다. 소양인의 면역 활성은 6개의 배합비(Mix 1~6) 추출물 중에서 배합비 4(Mix 4: 보리 60%, 콩 20%, 가지 10%, 양송이 10%) 처리에 의해 최고로 높았다. 배합비 6(Mix 6: 율무 60%, 팥 20%, 당근 10%, 목이 10%)를 처리한 면역세포의 면역 활성 증가는 다른 5개의 추출물들(Mix 1~5)보다 높았다. 따라서 배합비 2, 4 그리고 6은 체질 맞춤형 면역 활성생식제품을 위한 혼합물로 사용할 수 있을 것이다.

국가별 마늘의 체질에 따른 면역효능 및 항산화 활성 (Immunostimulatory and Antioxidant Activities of Steamed Garlic Grown in Different Countries)

  • 본 연구에서는 소양인, 태음인 그리고 소음인 지원자의 혈액으로부터 분리한 면역세포에서 한국, 일본 및 미국에서 재배한 생마늘(RG)과 증숙마늘(SG) 70% 에탄올추출물에 대한 면역활성과 항산화능을 탐색하였다. 70% 생마늘 추출물(RGE)은 세포 증식율, NO와 $TNF-{\alpha}$ 생성능이 소음인에 가장 높았고, 태음인과 소양인 순으로 나타났다. 반면에, 증숙마늘 추출물(SGE)은 각 개인별 체질에 영향을 미치지 않으면서 생마늘 추출물보다 면역활성이 더 높았다. 뿐만 아니라 국가별로 재배한 생마늘 및 증숙마늘의 면역효능은 나라별로 차이가 있었으며 한국산이 가장 높은 면역활성을 나타내었고 일본산, 미국산 순으로 나타났다. 이러한 결과는 항산화 효능에서도 유사한 경향을 나타내었다.

  • Syneilesis palmata (SP) has been used as a traditional medicinal plant and vegetable. SP was reported to exert pharmacological activities such as anti-inflammation, anti-cancer, and anti-HIV. However, there are no studies on the immunostimulatory activity of SP. Thus, in this study, we report that S. palmata leaves (SPL) induce the activation of macrophages. An increase in both secretions of immunostimulatory mediators and phagocytotic activity was observed in SPL-treated RAW264.7 cells. However, this was reversed by inhibition of TLR2/4. In addition, the p38 inhibition reduced the SPL-mediated secretion of immunostimulatory mediators, and the SPL-mediated p38 activation was blocked by the TLR2/4 inhibition. SPL augmented both p62/SQSTM1 and LC3-II. TLR2/4 inhibition blocked the SPL-mediated increase of p62/SQSTM1 and LC3-II. These findings indicate that SPL may activate macrophages through TLR2/4-dependent p38 activation and activate autophagy through TLR2/4 stimulation.

Immunostimulatory Activity of Hibiscus syriacus L. Leaves in Mouse Macrophages, RAW264.7 cells, and Immunosuppressed Mice

  • Under the COVID-19 pandemic, interest in immune enhancement is increasing. Although the immune-enhancing activity of plants of the genus Hibiscus has been reported, there is no study on the immune-enhancing activity of H. syriacus. Thus, in this study, we investigated the immune-enhancing activity of Hibiscus syriacus leaves (HSL) in mouse macrophages, RAW264.7 cells, and immunosuppressed mice. HSL increased the production of immunostimulatory factors such as nitric oxide (NO), inducible nitric oxide synthase (iNOS), interleukin-1β (IL-1β), and tumor necrosis factor-α (TNF-α) and activated the phagocytosis in RAW264.7 cells. The HSL-mediated production of immunostimulatory factors was dependent on toll-like receptor 4 (TLR4), p38, and c-Jun N-terminal kinase (JNK) in RAW264.7 cells. In the immunosuppressed mouse model, HSL increased the spleen index, the levels of the cytokines, and the numbers of lymphocytes, neutrophils, and monocytes. Taken together, HSL may be considered to have immune-enhancing activity and be expected to be used as a potential immune-enhancing agent.
