본 연구는 꾸지뽕 열매로부터 분리한 조다당류(CTPS)가 초기 면역반응에 중추적인 역할을 수행하는 큰포식세포에서 활성화를 유도하는지에 관한 여부를 알아보기 위하여 선천 및 적응면역에서 중추적인 역할을 수행하는 큰포식세포에 CTPS를 처리하여 세포 증식률, 산화질소 분비능 및 사이토카인($TNF-{\alpha}$ 및 IL-6) 분비능이 증가되는 것을 확인하였다. 또한 활성화된 큰포식세포의 세포 표면에서 발현되는 CD80과 CD86의 발현과 탐식세포의 항원 제시에 밀접한 관련이 있는 주조직적합성 복합체(MHC class I 및 II)의 발현이 CTPS 처리구에서 유의적으로 증가되는 것으로 관찰되었다. 이러한 산화질소 분비능과 사이토카인의 증가 원인에 관한 면역기전 분석 결과, MAPKs의 인산화와 $NF-{\kappa}B$의 핵 내 이동성을 증가시켜 면역활성을 증가시키는 것으로 관찰되었다.

The alkali-soluble glucan of the yeast cell wall contains $\beta-(1,3)-$ and (1,6)-D-linkages and is known to systemically enhance the immune system. In the previous study [6], in order to isolate cell wall mutants, a wild-type strain was mutagenized by exposure to ultraviolet light, and the mutants were then selected via treatment with laminarinase $(endo-\beta-(1,3)-D-glucanase)$. The mass of alkali- and water-soluble glucans produced by the mutant was measured to be 33.8 mg/g of the dry mass of the yeast cell. Our results showed that the mutants generated the amount of alkali-soluble glucan 10-fold higher than that generated by the wild-type. Structural analysis showed that the alkali-soluble glucan from the mutants was associated with a higher degree of $\beta-(1,6)-D-linkage$ than was observed in conjunction with the wild-type. Yeast cell wall $\beta-glucan$ was shown to interact with macrophages via receptors, thereby inducing the release of tumor necrosis factor alpha $(TNF-\alpha)$ and nitric oxide. Alkali-soluble $\beta-glucans$, both from water-soluble and water-insoluble glucan, exhibited a higher degree of macrophage activity with regard to both the secretion of tumor necrosis factor alpha $(TNF-\alpha)$ and nitric oxide and direct phagocytosis, than did the positive control ($1{\mu}g$ of lipopolysaccharide).
Acute respiratory virus infectious diseases are a growing health problem, particularly among children and the elderly. Much effort has been made to develop probiotics that prevent influenza virus infections by enhancing innate immunity in the respiratory tract until vaccines are available. Lactobacillus plantarum GB-LP2, isolated from a traditional Korean fermented vegetable, has exhibited preventive effects on influenza virus infection in mice. To identify the molecular basis of this strain, we conducted a whole-genome assembly study. The single circular DNA chromosome of 3,284,304 bp was completely assembled and 3,250 protein-encoding genes were predicted. Evolutionarily accelerated genes related to the phenotypic trait of anti-infective activities for influenza virus were identified. These genes encode three integral membrane proteins, a teichoic acid export ATP-binding protein and a glucosamine - fructose-6-phosphate aminotransferase involved in host innate immunity, the nonspecific DNA-binding protein Dps, which protects bacteria from oxidative damage, and the response regulator of the three-component quorum-sensing regulatory system, which is related to the capacity of adhesion to the surface of the respiratory tract and competition with pathogens. This is the first study to identify the genetic backgrounds of the antiviral activity in L. plantarum strains. These findings provide insight into the anti-infective activities of L. plantarum and the development of preventive probiotics.
Physical activity is a primary cancer control strategy that has received little attention to date. However, an Increasing number of epidemiological studies have proposed that physical exercise may be beneficial by enhancing anticancer immune system responses. We investigated the effects of acute exercise on changes in nitric oxide (NO) production and inducible nitric oxide synthase (iNOS) expression. The amounts of NO generated by abdominal macrophages in mice were measured after exercise. Thirty-two mice, which were challenged with thioglycollate broth to activate peritoneal macrophages, were randomly assigned to control, exercise and recovery groups. The mice exercised on a motor-driven treadmill for 3 consecutive days, either moderately (18m/min, 30 min/day, 5% grade) or severely (18-35m/min, 60 min/day, 5% grade). The mice were killed immediately after exercise or after 6 hrs of recovery. Nitric oxide was quantified by the Griess assay. The exercised mice showed higher levels of NO generation than those of the control mice, but the intensity of exercise had no significant effect on NO generation. Mice allowed six hours of recovery after exercise showed higher levels of NO generation than that of animals sacrificed immediately after exercise, but there were no significant differences in NO generation with variations in the intensity of exercise. Increased levels of iNOS were found in the exercised groups, and this was greatest in the groups allowed six hours of recovery compared to those groups sacrificed immediately after exercise. The results of this study suggest that acute exercise may enhance an immune response by inducing macrophage-derived NO generation; these results support the epidemiological findings which support the benefits of exercise in the prevention and control of cancer. Further study is needed to determine the physiological significance of these findings, which could be applied to the use of therapeutic exercises to assist in the prevention and control of cancer.

In this work, we wanted to develop a probiotic from famous longevity villages in Korea. We visited eight longevity villages in Korea to collect fecal samples from healthy adults who were aged above 80 years and had regular bowel movements, and isolated lactic-acid-producing bacteria from the samples. Isolated colonies that appeared on MRS agar containing bromophenol blue were identified by means of 16S rRNA sequencing, and 102 of the isolates were identified as lactic-acid-producing bacteria (18 species). Lactobacillus fermentum was the most frequently found species. Eight isolates were selected on the basis of their ability to inhibit the growth of six intestinal pathogens (Escherichia coli O157:H7, Salmonella enterica subsp. enterica Typhimurium, Salmonella enterica subsp. enterica Enteritidis, Enterococcus faecalis, Staphylococcus aureus, and Listeria monocytogenes) and their susceptibility to 15 antimicrobial agents. Among these eight isolates, four Lactobacillus fermentum isolates were found not to produce any harmful enzymes or metabolites. Among them, Lactobacillus fermentum isolate no. 24 showed the strongest binding to intestinal epithelial cells, the highest immune-enhancing activity, anti-inflammation activity, and anti-oxidation activity as well as the highest survival rates in the presence of artificial gastric juice and bile solution. This isolate, designated Lactobacillus fermentum PL9988, has all the characteristics for a good probiotic.

The growth and health of the fetus and neonate are directly influenced by the nutritional and physiological status of sows. Sows are often under catabolic conditions due to restrict feeding program during pregnancy and low voluntary feed intake during lactation. The current restrict feeding program, which aims at controlling energy intake during gestation, results in an inadequate supply of dietary protein for fetal and mammary gland growth. Low voluntary feed intake during lactation also causes massive maternal tissue mobilization. Provision of amino acids and fatty acids with specific functions may enhance the performance of pregnant and lactating sows by modulating key metabolic pathways. These nutrients include arginine, branched-chain amino acids, glutamine, tryptophan, proline, conjugated linoleic acids, docosahexaenoic acid, and eicosapentaenoic acid, which can enhance conception rates, embryogenesis, blood flow, antioxidant activity, appetite, translation initiation for protein synthesis, immune cell proliferation, and intestinal development. The outcome is to improve sow reproductive performance as well as fetal and neonatal growth and health. Dietary supplementation with functional amino acids and fatty acids holds great promise in optimizing nutrition, health, and production performance of sows and piglets. Chronic arsenicosis is a major environmental health hazard throughout the world, including India. Animals and human beings are affected due to drinking of arsenic contaminated ground water, due to natural mineral deposits, arsenical pesticides or improperly disposed arsenical chemicals. Arsenic causes cancer with production of free radicals and reactive oxygen species (ROS) that are neutralized by an elaborate antioxidant defense system consisting of enzymes and numerous non-enzymatic antioxidants. Dietary antioxidant supplements are useful to counteract the carcinogenesis effects of arsenic. Oyster mushroom lectins can be regarded as ingredients of popular foods with biopharmaceutical properties. A variety of compounds have been isolated from mushrooms, which include polysaccharides and polysaccharopeptides with immune-enhancing effects. Lectins are beneficial in reducing arsenic toxicity due to anticarcinogenetic roles and may have therapeutic application in people suffering from chronic exposure to arsenic from natural sources, a global problem that is especially relevant to millions of people on the Indian subcontinent.
Objectives In human myeloid leukemia cells, there are no specific features of apoptosis compared with apoptosis in other cell types. Solanum nigrum L.(SNL) is a deciduous tree, which is widely distributed in Korea with reported anti-tumor, anti-inflammatory and non-specific immune-enhancing properties. Although the plant has been clinically used for treating a variety of diseases, its bioactive ingredients are unknown and its mode of action potential has never been investigated. Thus anti-tumor property of methanol extract was investigated. Methods In this study, anti-tumor property of methanol extract was investigated by determining its in vitro growth-inhibitory effects on human myeloid leukemia cells. XTT proliferation assay, DNA fragmentation, immunoblot analysis, densitometric analysis were used. Results 1. The methanol fraction of the extracts of SNL induced mitochondria-mediated apoptosis in human myeloid leukemia cells. 2. The methanol fraction exhibited relatively higher cytotoxic activity in a dose-dependent manner than chloroform, and hexane fraction. 3. Typical ladder profile of Oligonucleosomal fragments were appeared. 4. The secreted cytosolic cytochrome C level was increased by treatment of methanol fraction. Conclusions Methanol fraction of SNL is capable of inducing apoptosis in human myeloid leukemia cells.
The purpose of this study is to investigate the effect of a mixture of fermented Artemisiae Argi Folium and fermented Sophorae Radix (FAS) on hydrogen peroxide production within mouse macrophage Raw 264.7 with ethanol (EtOH) and nicotine. Artemisiae Argi Folium is known to have the antibacterial, immune-enhancing properties. And Sophorae Radix is also known to have the antibacterial, anti-inflammatory, anti-allergic properties. EtOH and nicotine are the ones of toxicants which could impair immunocytes like macrophage. EtOH and nicotine reduce the intracellular production of hydrogen peroxide ($H_2O_2$) of Raw 264.7. FAS increased the production of hydrogen peroxide reduced by EtOH and nicotine within Raw 264.7. These results indicate that FAS could restore the immuno-activity of macrophage impaired by EtOH and nicotine.

To develop a composition for enhancing immunity, based on alcohol extracts of the bumblebee as an active ingredient, bumblebee ethanol extracts were evaluated for their protective effect in chronic models of inflammation, adjuvant induced rat arthritis. B. terrestris worker extract (SDIEX) and, B. hypocrita sapporoensis lava and pupa extract (SPDYBEX), significantly decreased paw edema in arthritic rats, at a dose 100 mg/kg, respectively. The cytokine levels related inflammation of COX-2, $sPLA_2$, VEGF, and TNF-${\alpha}$, were decreased, compared to positive control, indomethacin (5 mg/kg). Histopathological data demonstrated decreases inflammatory activity, hind paw edema, and repaired hyaline articular cartilage in DRG over a 2 wk administration. HPLC and GC-MS analysis of SDIEX and SPDYBEX revealed the presence of cantharidin.
