비타민 E 보충이 여자 운동선수들의 항산화능과 면역능력에 미치는 영향 (Effects)

  • This study was designed to investigate the effects of vitamin E supplementation on radical scavenger activity and immune responses in female judo athletes(n=18). The age and sex matched sedentary students were used as controls(n=15). The initial plasma vitamin E concentration, lipid peroxide level and radical scavenger activity (RSA) were not different between two groups. The supplementation of $\alpha$-tocopheryl acetate(400IU/d) for 4 weeks significantly increased plasma vitamin E concentration of the subjects in both groups. In addition, the decrease in thiobarbituric acid reactive substance(TBARS)(p<0.05) and enhancement of RSA(p<0.05) were observed in both groups. Percentages of total T-cells in the athletic group was lower than that in the control group(p<0.05). However, the ratio of CD4 / CD8(helper T-cal $l^pressor T-cell) was higher in the sports group, due to more decrease in CD8 subset than in CD4 subset. The concentrations of IgG and IgM in the sports group were significantly lower than those in the control group(p<0.05). Therefore, severe training of the athletic group seems to be associated with the changes of supplementation. In conclusion, vitamin E supplementation (400IU/d, 4 weeks) of the female university students improved antioxidative activities of the blood, by decreasing lipid peroxide and enhancing radical scavenger activity. Percentages of T lymphocytes and IgG and IgM concentrations in the athletic group were lower than those in the control group. Meanwhile, vitamin E supplementation had no effects on immune status in both groups.ps.

Ginsan Enhances Humoral Antibody Response to Orally Delivered Antigen

  • Background: There have been several reports describing the capability of ginseng extracts as an adjuvant. In this study, we tested if ginsan, a polysaccharide extracted from Panax ginseng, was effective in enhancing antibody response to orally delivered Salmonella antigen. Methods: Ginsan was treated before oral salmonella antigen administration. Salmonella specific antibody was determined by ELISA. mRNA expression was determined by RT-PCR. Cell migration was determined by confocal microscopy and flow cytometry. COX expression was detected by western blot. Results: Ginsan treatment before oral Salmonella antigen delivery significantly increased both secretory and serum antibody production. Ginsan increased the expression of COX in the Peyer's patches. Various genes were screened and we found that CCL3 mRNA expression was increased in the Peyer's patch. Ginsan increased dendritic cells in the Peyer's patch and newly migrated dendritic cells were mostly found in the subepithelial dome region. When COX inhibitors were treated, the expression of CCL3 was reduced. COX inhibitor also antagonized both the migration of dendritic cells and the humoral immune response against oral Salmonella antigen. Conclusion: Ginsan effectively enhances the humoral immune response to orally delivered antigen, mediated by CCL3 via COX. Ginsan may serve as a potent vaccine suppliment for oral immunization.

The emerging role of lncRNAs in inflammatory bowel disease

  • Dysregulation of long noncoding RNA (lncRNA) expression is linked to the development of various diseases. Recently, an emerging body of evidence has indicated that lncRNAs play important roles in the pathogenesis of inflammatory bowel diseases (IBDs), including Crohn's disease (CD) and ulcerative Colitis (UC). In IBD, lncRNAs have been shown to be involved in diverse processes, including the regulation of intestinal epithelial cell apoptosis, association with lipid metabolism, and cell-cell interactions, thereby enhancing inflammation and the functional regulation of regulatory T cells. In this review, we aim to summarize the current knowledge regarding the role of lncRNAs in IBD and highlight potential avenues for future investigation. We also collate potentially immune-relevant, IBD-associated lncRNAs identified through a built-by association analysis with respect to their neighboring protein-coding genes within IBD-susceptible loci. We further underscore their importance by highlighting their enrichment for various aspects of immune system regulation, including antigen processing/presentation, immune cell proliferation and differentiation, and chronic inflammatory responses. Finally, we summarize the potential of lncRNAs as diagnostic biomarkers in IBD.

Heat-Killed Lactobacillus plantarum KCTC 13314BP Enhances Phagocytic Activity and Immunomodulatory Effects via Activation of MAPK and STAT3 Pathways

  • Identification of novel probiotic strains is of great interest in the field of functional foods. Specific strains of heat-killed bacteria have been reported to exert immunomodulatory effects. Herein, we investigated the immune-stimulatory function of heat-killed Lactobacillus plantarum KCTC 13314BP (LBP). Treatment with LBP significantly increased the production of $TNF-{\alpha}$ and IL-6 by macrophages. More importantly, LBP was able to enhance the phagocytic activity of macrophages against bacterial particles. Activation of p38, JNK, ERK, $NF-{\kappa}B$, and STAT3 was involved in the immunomodulatory function of LBP. LBP treatment significantly increased production of $TNF-{\alpha}$ by bone marrow-derived macrophages and splenocytes, further confirming the immunostimulatory effect of LBP in primary immune cells. Interestingly, the immunomodulatory effects of LBP were much stronger than those of Lactobacillus rhamnosus GG, a well-known probiotic strain. These results indicate that LBP can be a promising immune-enhancing functional food agent.

Efficacy of immune-strengthening functional drinks in top-level athletes: a questionnaire survey-based research

  • [Purpose] Functional beverages are intended to support those who want to maintain optimal physical condition and improve their quality of life through the enhancement of heart health, immunity, and digestion. The purpose of this study was to investigate the performance of top-level athletes consuming immune-strengthening conditioning nutritional drinks. [Methods] A total of 107 top-level athletes (baseball (56 players), pro volleyball (17), athletics (16), cycling (8), golf (6), and fencing (6)) participated in the experiment. They consumed an immune-enhancing functional beverage once a day for 8 weeks and responded to a survey before, during, and after drinking the beverage. [Results] Three total aspect-based subfactors were drawn from 24 questions in the factor analysis: physical, satisfaction with mental stability, and activity in performance. The physical, mental stability and performance changes of athletes significantly increased in period 2 (4 weeks after intake) and period 3 (after 8 weeks of intake). [Conclusion] We evaluated the efficacy of a new conditioned beverage containing Lactobacillus B240 and protein in improving the performance and physiological utility of top athletes. This functional drink may gain popularity among those seeking health benefits and improved exercise performance.

Hibiscus syriacus Leaves Upregulate p62/SQSTM1 through TLR4/p38, JNK, and NF-κB/Nrf2 Signaling Pathway in RAW264.7 Cells

  • Autophagy contributes to enhancing the immune system (innate and adaptive immune system) against foreign pathogens. Autophagy of macrophages is used as a major indicator for developing vaccine adjuvants to increase the adaptive immune response. In this study, HSL increased p62/SQSTM1 expression. Inhibition of TLR4, p38, JNK, and NF-κB blocked HSL-mediated increase of p62/SQSTM1. HSL activated p38, JNK, and NF-κB signaling, but HSL-mediated activation of p38, JNK, and NF-κB signaling was reversed by TLR4 inhibition. In addition, HSL increased Nrf2 expression, but HSL-mediated Nrf2 expression did not occur in the inhibition of TLR4, p38, JNK, and NF-κB. Taken together, it is believed that HSL-mediated autophagy may be dependent on activating Nrf2 expression via TLR4-dependent activation of p38, JNK, and NF-κB in macrophages.

  • 본 연구에서 우리는 작약 뿌리 추출물이 TLR4/PI3K/Nrf2 신호전달을 통해 p62/SQSTM1을 증가시켜 대식세포에서 자가포식을 유도한다는 것을 확인하였다. 대식세포의 자가포식 유도는 선천성과 적응성 면역 반응 간의 연결을 강화해 백신 보조제 개발에 있어서 중요한 표적으로 사용되기 때문에, 작약 뿌리 추출물은 백신개발에 필수적인 백신보조제로의 활용이 가능할 것으로 생각된다.

4계통 재래종 닭고기의 화학적 특성 및 면역활성 (Comparison of Chemical Composition and Immune-enhancing Activity of the Four Lines of Korean Native Chickens)

  • 본 연구는 4종의 재래닭인 연산오계, 현인흑계, 횡성약닭, 황봉의 일반성분, 카르노신, 안세린 함량 및 면역활성을 비교하기 위하여 실시하였고, 대조구로 백색 레그혼을 사용하였다. 시험용 닭을 모두 동일한 조건에서 13주간 사육하여 도계한 후 화학적 성분조사를 실시하였다. 일반성분 분석에서는 4종의 재래닭이 백색레그혼에 비하여 조지방 함량이 높았으며, 콜레스테롤 함량은 현인흑계가 다른 품종에 비하여 높았고 횡성약닭이 유의적으로 낮았다(p<0.05). 면역증진 효능을 측정하기 위하여 대식세포를 이용하여 실험한 결과는 재래닭 4종의 가슴육에서만 대식세포 증식 및 NO 억제 효능이 있었다. 대식세포 탐식작용은 현인흑계와 횡성약닭가슴육에서 유의적으로 증가되었고, 염증 관련 사이토카인(IL-4, IL-10, $IFN-{\gamma}$) 유전자 발현은 백색레그혼에 비하여 4종의 재래닭에서 억제효과가 컸다. 결론적으로 4종류의 재래닭이 백색레그혼에 비하여 면역증진 효능이 높았으며, 특히 가슴육에서 효능이 있었다. 재래닭 품종 간에도 일반성분 및 면역조절능에 차이가 있는 것으로 나타났다.

Enhancing T Cell Immune Responses by B Cell-based Therapeutic Vaccine Against Chronic Virus Infection

  • Chronic virus infection leads to the functional impairment of dendritic cells (DCs) as well as T cells, limiting the clinical usefulness of DC-based therapeutic vaccine against chronic virus infection. Meanwhile, B cells have been known to maintain the ability to differentiate plasma cells producing antibodies even during chronic virus infection. Previously, ${\alpha}$-galactosylceramide (${\alpha}GC$) and cognate peptide-loaded B cells were comparable to DCs in priming peptide-specific $CD8^+$ T cells as antigen presenting cells (APCs). Here, we investigated whether B cells activated by ${\alpha}GC$ can improve virus-specific T cell immune responses instead of DCs during chronic virus infection. We found that comparable to B cells isolated from naïve mice, chronic B cells isolated from chronically infected mice with lymphocytic choriomeningitis virus (LCMV) clone 13 (CL13) after ${\alpha}GC$-loading could activate CD1d-restricted invariant natural killer T (iNKT) cells to produce effector cytokines and upregulate co-stimulatory molecules in both naïve and chronically infected mice. Similar to naïve B cells, chronic B cells efficiently primed LCMV glycoprotein (GP) 33-41-specific P14 $CD8^+$ T cells in vivo, thereby allowing the proliferation of functional $CD8^+$ T cells. Importantly, when ${\alpha}GC$ and cognate epitope-loaded chronic B cells were transferred into chronically infected mice, the mice showed a significant increase in the population of epitope-specific $CD8^+$ T cells and the accelerated control of viremia. Therefore, our studies demonstrate that reciprocal activation between ${\alpha}GC$-loaded chronic B cells and iNKT cells can strengthen virus-specific T cell immune responses, providing an effective regimen of autologous B cell-based therapeutic vaccine to treat chronic virus infection.

Immune Enhancement Effect of Asterias amurensis Fatty Acids through NF-κB and MAPK Pathways on RAW 264.7 Cells

  • Asterias amurensis is a marine organism that causes damage to the fishing industry worldwide; however, it has been considered a promising source of functional components. The present study aimed to investigate the immune-enhancing effects of fatty acids from three organs of A. amurensis on murine macrophages (RAW 264.7 cells). A. amurensis fatty acids boosted production of immune-associated factors such as nitric oxide (NO) and prostaglandin E2 in RAW 264.7 cells. A. amurensis fatty acids also enhanced the expression of critical immune-associated genes, including iNOS, $TNF-{\alpha}$, $IL-1{\beta}$, and IL-6, as well as COX-2. Western blotting showed that A. amurensis fatty acids stimulated the $NF-{\kappa}B$ and MAPK pathways by phosphorylation of $NF-{\kappa}B$ p-65, p38, ERK1/2, and JNK. A. amurensis fatty acids from different tissues resulted in different levels of $NF-{\kappa}B$ and MAPK phosphorylation in RAW 264.7 cells. The results increase our understanding of how A. amurensis fatty acids boost immunity in a physiological system, as a potential functional material.
